市販の7種類のマスクについて, 動物実験施設洗浄室における粉塵と空中細菌に対する捕集効率を検索した。マスクの捕集効率は, 10μm以下の粉塵重量に対しては平均19~50%, 粉塵の粒子径別にみた捕集効率は0.3μmレベルで22~71%, 1μmレベルで47~90%, 5μmレベルで90~99.6%であった。また, 空中細菌に対するマスクの捕集効率は35~81%であった。試験したマスク間の比較では, グラスウール製手術用, 3枚重ねの合成繊維製およびグラスウール挿入の28枚重ねのガーゼマスクは一般に高い効率を示した。合成繊維一枚のマスク, 18枚重ねのガーゼマスクおよびガスマスクは低い効率を示した。

During the 18th century, raising the "nezumi" rodent became so prevalent in Japan that two guidebooks were published on the topic. The first guidebook was entitled Yoso-tama-no-kakehashi (1775) and the second was entitled Chinganso-date-gusa (1787). It remains unclear in these texts whether the term nezumi was used to refer to the rat (Rattus norvegicus) or the mouse (Mus musculus). In this review, I explore Yoso-tama-no-kakehashi (English translation: A bridge to obtaining novel jewel-like nezumi). It was written by the owner of "Shunpo-do" and comprises two volumes; the first is 34 pages in length and the second is 14 pages. It introduces the nezumi and then provides details on novel varieties and the methods that were used to raise them. The nezumi dwells in peoples' homes. It is noteworthy that the "norako" species is classified in the same group as the nezumi. The norako is smaller than the nezumi. Its alias is "hatsuka-nezumi", a term which is still used in Japan today when referring to the mouse. This indicates that when the guidebook was written people distinguished the rat from the mouse by identifying the rat using the word nezumi and the mouse using the word norako. Moreover, I recently confirmed that the rat varieties which are introduced in Yoso-tama-no-kakehashi, such as "white", "spotted", "black bear-like", "deer-spotted", and "cracked-mark", can be found in modern laboratory rats. Taken together, it is very likely that the term nezumi is used to refer to the rat in Yoso-tama-no-kakehashi and that this was indeed a guidebook on the rat.

We conducted a survey on 3,096 members of the public in order to find out about public awareness concerning the use of animals in scientific research in Japan and statistically analyzed the results. Regarding the necessity of experiments, research, and educational activities using animals, 55–62% respondents answered that “development of medicine and medical technology for human beings”, “development of medicine and medical technology for animals”, “practical training at medical schools”, and “practical training at schools of veterinary medicine” were necessary, while 9–12% respondents answered that they were not necessary. These results showed that the Japanese public can dispassionately accept that animal experiments are necessary. Regarding the image of animal experiments, 50–70% respondents also supported animal experiments aimed at “advances in science and medicine”, “securing of human health and safety”, and “pursuit of economic interest”. On the other hand, when faced with questions that featured emotional language, a majority (51–57% of people) felt that “animal experiments are painful and cruel acts” and that “experimental animals are to be pitied”. This survey showed that the majority of the Japanese public can accept the necessity of animal experiments, but experts and researchers involved in animal experiments should consider seriously the large number of respondents that agreed with emotive descriptions of animal research.

マウスの寿命に及ぼす無菌状態と食餌制限の影響について検討した。実験動物は雄のICR系無菌およびSPFマウスを用いた。制限食の開始時期は生後5週とし, 自由摂取群と制限食群とに分けた。制限食群の食餌は自由摂取群の摂取量の80℃6 (4.5g/日) を毎日与えた。平均寿命は自由摂取群のSPFマウスでは75.9週, 無菌マウスでは88.9週, 制限食群のSPFマウスでは117.5週, 無菌マウスでは109.6週であった。また, 体重を計測した結果, 制限食群は自由摂取群と比べて, SPFおよび無菌マウス共に低値の成績が得られた。この結果, 離乳直後からの食餌制限は成熟を遅らせ, 成長期間が長くなり, 寿命が延びている可能性が考えられる。一方, 無菌マウスの平均寿命はSPFマウスの平均寿命と比べて, 自由摂取群では長く, 制限食群では短かった。この成績から, 無菌状態と食餌制限の組合せでは顕著な延命効果は認められなかったが, 各々単独では平均寿命の延長に影響を及ぼしている可能性が示唆された。

Sea urchin embryos have been one of model organisms to investigate cellular behaviors because of their simple cell composition and transparent body. They also give us an opportunity to investigate molecular functions of human proteins of interest that are conserved in sea urchin. Here we report that human disease-associated extracellular matrix orthologues ECM3 and QBRICK are necessary for mesenchymal cell migration during sea urchin embryogenesis. Immunofluorescence has visualized the colocalization of QBRICK and ECM3 on both apical and basal surface of ectoderm. On the basal surface, QBRICK and ECM3 constitute together a mesh-like fibrillar structure along the blastocoel wall. When the expression of ECM3 was knocked down by antisense-morpholino oligonucleotides, the ECM3-QBRICK fibrillar structure completely disappeared. When QBRICK was knocked down, the ECM3 was still present, but the basally localized fibers became fragmented. The ingression and migration of primary mesenchymal cells were not critically affected, but their migration at later stages was severely affected in both knock-down embryos. As a consequence of impaired primary mesenchymal cell migration, improper spicule formation was observed. These results indicate that ECM3 and QBRICK are components of extracellular matrix, which play important role in primary mesenchymal cell migration, and that sea urchin is a useful experimental animal model to investigate human disease-associated extracellular matrix proteins.

The Center for Animal Resources and Development (CARD), Kumamoto University was established in 1998. We provide advanced research support services for the mouse-based biomedical research community via an official and a premium mouse bank system. To efficiently manage these mouse banks, we have actively developed and modified basic mouse reproductive techniques. We shall introduce these techniques in this paper.

Cell labeling technologies, including the Cre/loxP system, are powerful tools in developmental biology. Although the conventional Cre/loxP system has been extensively used to label the expression of specific genes, it is less frequently used for labeling protein-protein interactions owing to technical difficulties. In the present study, we generated a new Gal4-dependent transgenic reporter mouse line that expressed Cre recombinase and a near-infrared fluorescent protein, miRFP670. To examine whether this newly generated transgenic mouse line is applicable in labeling of protein-protein interaction, we used a previously reported transgenic mouse lines that express Notch1 receptor with its intracellular domain replaced with a yeast transcription factor, Gal4. Upon the binding of this artificial Notch1 receptor and endogenous Notch1 ligands, Gal4 would be cleaved from the cell membrane to induce expression of Cre recombinase and miRFP670. Indeed, we observed miRFP670 signal in the mouse embryos (embryonic day 14.5). In addition, we examined whether our Cre recombinase was functional by using another transgenic mouse line that express dsRed after Cre-mediated recombination. We observed dsRed signal in small intestine epithelial cells where Notch1 signal was suggested to be involved in the crypt stem cell maintenance, suggesting that our Cre recombinase was functional. As our newly generated mouse line required only the functioning of Gal4, it could be useful for labeling several types of molecular activities in vivo.

Forward genetics is a powerful approach based on chromosomal mapping of phenotypes and has successfully led to the discovery of many mouse mutations in genes responsible for various phenotypes. Although crossing between genetically remote strains can produce F2 and backcross mice for chromosomal mapping, the phenotypes are often affected by background effects from the partner strains in genetic crosses. Genetic crosses between substrains might be useful in genetic mapping to avoid genetic background effects. In this study, we investigated single nucleotide polymorphisms (SNPs) available for genetic mapping using substrains of C57BL/6 and BALB/c mice. In C57BL/6 mice, 114 SNP markers were developed and assigned to locations on all chromosomes for full utilization for genetic mapping using genetic crosses between the C57BL/6J and C57BL/6N substrains. Moreover, genetic differences were identified in the 114 SNP markers among the seven C57BL/6 substrains from five production breeders. In addition, 106 SNPs were detected on all chromosomes of BALB/cAJcl and BALB/cByJJcl substrains. These SNPs could be used for genotyping in BALB/cJ, BALB/cAJcl, BALB/cAnNCrlCrlj, and BALB/cCrSlc mice, and they are particularly useful for genetic mapping using crosses between BALB/cByJJcl and other BALB/c substrains. The SNPs characterized in this study can be utilized for genetic mapping to identify the causative mutations of the phenotypes induced by N-ethyl-N-nitrosourea mutagenesis and the SNPs responsible for phenotypic differences between the substrains of C57BL/6 and BALB/c mice.

The cold storage of two-cell embryos is a useful technique for transporting genetically engineered mice without the shipment of live animals. However, the developmental ability of cold-stored embryos decreases with prolonged storage periods. Therefore, the transported embryos must be readily transferred to recipient mice upon arrival. The cryopreservation of cold-transported embryos may improve the flexibility of the schedule of embryo transfer. In this paper, we examined the viability and developmental ability of vitrified-warmed mouse embryos at the two-cell stage after cold storage in refrigerated temperatures for 0, 24, 48, 72, or 96 hours. The viability of vitrified-warmed embryos after cold storage was comparable to vitrified-warmed embryos without cold storage. Vitrified-warmed embryos after cold storage also developed normally to pups by embryo transfer. In addition, live pups were obtained from vitrified-warmed embryos after cold-transportation from Asahikawa Medical University. In summary, cold-stored embryos can be used for the transportation and archive of genetically engineered mice.

To collect rat mutations and increase the value of the rat model system, we introduced fancy-derived mutations to the laboratory and carried out genetic analyses. Six fancy rats were shipped from a fancy rat colony in the USA and used as founders. After initial crosses with a laboratory strain, TM/Kyo or PVG/Seac, inbreeding started and 6 partially inbred lines, including 2 sublines, were produced as Kyoto Fancy Rat Stock (KFRS) strains. During inbreeding, we isolated 9 mutations: 5 coat colors, American mink (am), Black eye (Be), grey (g), Pearl (Pel), siamese (sia); 1 coat pattern, head spot (hs); 2 coat textures, Rex (Re), satin (sat); and an ear pinnae malformation, dumbo (dmbo). Genetic analyses mapped 7 mutations to particular regions of the rat chromosomes (Chr): am to Chr 1, sia to Chr 1, sat to Chr 3, Re to Chr 7, g to Chr 8, dmbo to Chr 14, and hs to Chr 15. Candidate gene analysis revealed that a missense mutation in the tyrosinase gene, Ser79Pro, was responsible for sia. From mutant phenotypes and mapping positions, it is likely that all mutations isolated in this study were unique to the fancy rat. These findings suggest that fancy rat colonies are a good source for collecting rat mutations. The fancy-derived mutations, made available to biomedical research in the current study, will increase the scientific value of laboratory rats.

Thyroid cancer in children, the most common endocrine malignancy, shows aggressive behavior and has a high recurrence rate after surgical ablation. Radioactive iodine (RAI) treatment is the most effective primary modality for medical ablation of juvenile thyroid cancer, and leads to intentional hypothyroidism. Although several negative impacts of hypothyroidism have been reported in children in response to other antithyroid agents, the combined effects of RAI exposure and hypothyroidism, on growing bones specifically, are unknown. In this study, we investigated the effect of RAI-induced hypothyroidism on the long bones during the pubertal growth spurt using immature female rats. Female Sprague-Dawley rats were randomly divided into a control group, and an RAI-treated group fed with RAI (0.37 MBq/g body weight) twice via gavage. After 4 weeks, we observed a significantly-reduced serum free thyroxine level in the RAI group. The latter group also displayed decreased body weight gain compared to the control. In addition, the lengths of long bones, such as the leg bones and vertebral column, as well as bone mineral content, were reduced in the RAI-treated animals. Our results confirm the negative impacts of RAI-induced thyroid deficiency during puberty on longitudinal bone growth and bone mineralization.

Humanized mice are widely used to study the human immune system in vivo and investigate therapeutic targets for various human diseases. Immunodeficient NOD/Shi-scid-IL2rγnull (NOG) mice transferred with human hematopoietic stem cells are a useful model for studying human immune systems and analyzing engrafted human immune cells. The gut microbiota plays a significant role in the development and function of immune cells and the maintenance of immune homeostasis; however, there is currently no available animal model that has been reconstituted with human gut microbiota and immune systems in vivo. In this study, we established a new model of CD34+ cell-transferred humanized germ-free NOG mice using an aseptic method. Flow cytometric analysis revealed that the germ-free humanized mice exhibited a lower level of human CD3+ T cells than the SPF humanized mice. Additionally, we found that the human CD3+ T cells slightly increased after transplanting human gut microbiota into the germ-free humanized mice, suggesting that the human microbiota supports T cell proliferation or maintenance in humanized mice colonized by the gut microbiota. Consequently, the dual-humanized mice may be useful for investigating the physiological role of the gut microbiota in human immunity in vivo and for application as a new humanized mouse model in cancer immunology.
