A novel adsorbent for the selective removal of endotoxin from various protein solutions has been prepared. The adsorbent comprises aminated poly(γ-methyl L-glutamate) (PMLG) spheres. The larger is the number of incorporated amino groups, the larger is the endotoxin-adsorption capacity of the adsorbent, the smaller is the pore size (exclusion of molecular weight; Mlim) of the adsorbents, the smaller is the number of various proteins adsorbed. When Mlim was smaller than 300 (as the molecular weight of polysaccharide) and the amino-group content was 3.2meq/g, the adsorbent selectively removed endotoxin from various endotoxin-containing samples, such as protein solutions, without affecting the effective substances, even at such a high ionic strength (μ) of 0.2-0.4. Consequently, aminated PMLG spheres with a certain pore size are considered to be a useful adsorbent for the removal of endotoxin from drugs and fluids for injection.

In this study, a simultaneous assay for catecholamines and their metabolites in the brain was established using liquid chromatography–mass spectrometry (LC-MS). To achieve complete separation, a cation-exchange/reversed-phase mixed-mode copolymer resin column containing 0.81 wt% sulfo groups was used for the simultaneous LC-MS assay. The analyzed catecholamines were dopamine (DA), norepinephrine (NE), and epinephrine (E), while the metabolites lacking amino groups were 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), and 3-methoxy-4-hydroxyphenylglycol (MHPG). The metabolites were separated and detected using LC-MS, on columns with and without sulfo groups. However, we could not achieve adequate separation of catecholamines on both columns using a gradient elution of 0 – 50 (v/v)% methanol containing 0.1 (v/v)% formic acid (FA). When volatile ion-pairing reagents were added to the mobile phase, they improved the retention and detection of catecholamines on the sulfonated mixed-mode column. Under optimized elution conditions, which involved a linear gradient elution of water containing 0.1 (v/v)% FA to 50 (v/v)% acetonitrile in 50 mM ammonium formate at 40°C and a 0.20 mL/min rate, all six target molecules were simultaneously detected within 25 min, when using negative mode LC-MS on a sulfonated mixed-mode column. The limits of detection (LODs) for DA, NE, E, DOPCA, HVA, and MHPG were determined to be 20.7, 12.6, 74.6, 1110, 18.7, and 3196 nM, respectively. Moreover, the established LC-MS assay allowed the detection of endogenous DA, NE, and HVA, in normal mouse brain samples at concentrations higher than 20, 9, and 4 pmol/mg, respectively.

Many studies have demonstrated that the dietary supplementation of polyamines, especially spermidine (SPD), prevents age-related diseases. Rice bran is rich in polyamines and their amounts could be increased by fermentation with Aspergillus oryzae (A. oryzae). In this study, we developed a method for the determination of putrescine (PUT), SPD and spermine (SPM) in rice bran samples by liquid chromatography/electrospray ionization–tandem mass spectrometry (LC/ESI-MS/MS) after derivatization with 4-(N,N-dimethylaminosulfonyl)-7-fluoro-2,1,3-benzoxadiazole (DBD-F). The derivatization improved the LC retention and ESI-MS/MS detectability of the polyamines, and consequently enabled precise and accurate quantification. Using this method, we found that the SPD content increased to 158% due to fermentation with A. oryzae, while the content of PUT and SPM decreased. SPD is known as the polyamine playing a central role in cell proliferation and growth, and therefore has health benefits. The fermented rice bran might be a good material for functional foods aimed at SPD supplementation.

Simple and rapid tools for antibody detection are beneficial for therapeutic monoclonal antibody development. Using a synthetic cationic antibody-recognizing peptide, a label-free colorimetric assay for antibody detection was established, in which a solution containing silver nanoparticles (AgNPs) changes color depending on particle aggregation/dispersion. Among the peptide probes we previously screened as IgG binding, one (NKFRGKYK) has four cationic amino acids and a pI of 10.46. Hence, we hypothesized that the peptide would both bind IgG and induce anionic AgNP aggregation via electrostatic interactions. This dual functionality of the IgG binding peptide could be useful for colorimetric detection. The detection of IgG in a solution containing culture media was investigated, and IgG was successfully detected at 100 to 500 nM within 2 min. This method is promising for high-throughput selection of IgG-producing cells since it is fast, readily observable, and does not involve complicated nanoparticle functionalization.

Di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium [DPPH] is widely used as a standard for measuring the number of free radicals. Here, we evaluated the number of free radicals of “DPPH” reagents from three manufacturers by effective magnetic moment method. Interestingly, the reagents from different manufacturers had varying temperature dependencies for both magnetic moment and g-value at low temperatures. As a result, the maximum relative difference among the three reagents on the number of free radicals per unit mass was 20%. Carbon hydrogen nitrogen (CHN) analyses, high-resolution EPR measurements, FT-IR measurement, and NMR measurement confirmed that a major component of only one among the three reagents was “pure” DPPH. The evaluated purity based on free radical content was 0.998 kg kg−1 with expanded uncertainty of 0.036 kg kg−1. The other two reagents were found to be contaminated by several % of benzene in the DPPH crystal structure.

The inorganic arsenic (i-As) in grape products, in particular juice, wine and raisins, collected from the Japanese market was investigated. The concentrations of total As in nine grape concentrated juices ranged from 3 to 20 ng g−1, and more than 80% of the As was inorganic according to the results of speciation by HPLC-ICP-MS. Among them, four samples contained more than 10 ng g−1 of i-As, although 10 ng g−1 of i-As is the limit for apple juice recommended by the U.S. Food and Drug Administration. Moreover, more than 10 ng g−1 of i-As was found in some of the wine and raisin samples, with the total As concentrations ranging from 17 to 37 ng g−1. When fresh grapes were analyzed, As was mainly concentrated in the pericarp and a small amount was found in the fruit, although no As was observed in the branches and the juice. When grapes, including the pericarp, are processed, the As concentration in the products may increase during production processes such as drying, concentration, and ripening.

Double-viewing-position single-particle inductively coupled plasma–atomic emission spectrometry (DVP-SP-ICP-AES) measures emission intensity at two ICP vertical positions simultaneously using a single photomultiplier tube. A particle travelling up the ICP gives two consecutive temporal emission peaks. The Yb II 328.937-nm emission intensity of the two peaks for single Yb2O3 particles of diameter of 200 – 2000 nm are plotted against each other in a correlation plot. The correlation is poor when the gas temperature at the lower observation position is approximately the boiling point of the particles. Poor particle vaporization at the center of the central channel occurs because the gas temperature is 400 K lower than the temperature at the rim. The correlation is improved by shifting the observation positions up or using helium–argon mixed carrier gas to increase the gas temperature. Gas temperature is an important parameter for precise single particle-ICP measurements. DVP-SP-ICP-AES can be used to identify poor particle vaporization without the need of temperature measurement.

球状シリカビーズ表面にグルコースオキシダーゼ(GOD)とグルコアミラーゼ(GAM)を共有結合で固定化し,ミニカラムに充てんしてミクロなバイオリアクターをつくり,高速液体クロマトグラフの分離カラムの後に接続した.グルコースはGODの触媒作用により過酸化水素とグルコノラクトンを,マルトースはGAMによりグルコースを生じ,更にこのグルコースがGODにより過酸化水素を生じる.生成した過酸化水素を電気化学検出器でアンペロメトリックに測定し,グルコースとマルトースの間接定量を試みた.本方法の検出感度は,GOD固定化カラムを用いた化学発光-FIA法やGOD・GAM固定化カラムを用いた化学発光-FIA法とほぼ同等であり,示差屈折率検出器を用いるHPLC法よりも約500倍高かった.最小検出量は絶対量でグルコースが10ng,マルトースが15ng(いずれも<I>S</I>/<I>N</I>=3)であった.本法は酵素の触媒作用,電気化学検出器の選択的で高感度な検出を利用しているため,前処理が極めて簡略化でき,試料を希釈するだけでよかった.市販の麦芽飲料,麦芽入り豆乳,ノンアルコールビール中のグルコースとマルトースの定量に応用し,良好な結果が得られた.

The title complex, [(PPh<sub>3</sub>)<sub>2</sub>Ag(<i>μ</i>-N<sub>3</sub>)<sub>2</sub>Ag(PPh<sub>3</sub>)<sub>2</sub>], has been synthesized and structurally characterized by single-crystal X-ray diffraction. The silver atoms are joined through a double <i>μ</i><sub>1,3</sub>-azido (N<sub>3</sub>) bridge. To each silver atom are also coordinated two P atoms from two triphenylphosphine molecules. The coordinated polyhedron around silver is a distorted tetrahedron.

The increase in the use of therapeutic monoclonal antibodies (mAbs) and antibody-drug conjugates (ADCs) has made the detailed bioanalysis of these drugs essential not only for planning optimal therapeutic programs for clinical practice, but also for evaluating the biological equivalencies in the development of other biosimilars. The ligand binding assays that are widely in use now are being replaced rapidly by the highly accurate, sensitive, and selective analytical method using a mass spectrometer. This review will discuss the progress in and challenges observed during the development of a mass spectrometry-based bioanalytical method for therapeutic mAbs and ADCs.
