食品には,さまざまな機能性をもった化合物が多く含まれている.そのため,疾病の予防や症状の緩和に寄与する目的として,機能性を付与した食品が多く開発されている.化学物質や食品成分は,無毒性量(NOAEL)や許容一日摂取量(ADI)が設定され,さらに,医薬品は,開発段階において他の物質との相互作用についても詳しく調査されている.しかし,食品成分については,さまざまな成分が含まれているため,単体の安全性は確保されていても,相互作用については,すべてを検証し評価することは困難なものとなっている.そこで本稿では,食品に含まれる抗酸化物質に注目し,相互作用によって生じる活性酸素種及び活性窒素種について紹介する.

本研究では,インクジェットプリンティングを用いて一つのポリジメチルシロキサン(Poly(dimethylsiloxane), PDMS)製マイクロ流路内に2種類の反応試薬を固定化した1ステップ均一系競合型バイオアッセイマイクロデバイスを開発した.インクジェットプリンターは試薬をナノリットルサイズの液滴として正確な量を位置選択的に吐出するため,互いに反応する2種類の試薬を混ざることなく微少空間内に独立して固定化できる.そのため,先行研究でデバイスを作製する際に課題となった,相互に反応する試薬を固定化した二つのマイクロ流路を精密に組み合わせる操作が不要となる.本研究ではその一例として,ビオチン固定化酸化グラフェン(BG)と蛍光標識ストレプトアビジン(F-SA)を1本のマイクロ流路内に固定してビオチンアッセイマイクロデバイスを試作し,基礎検討として試薬の固定化位置や試薬及び添加剤種の評価を行った.その結果,液滴を流路壁面寄りに吐出すると再現性よく流路底面の両サイドに試薬固定できること,添加剤としてトレハロースが適すること及び,試料導入操作のみでの1ステップ検出で,検出下限は0.68 ng mL−1となり,先行研究と比較して約1/600倍改善することを明らかにした.

For materials development with high-level structural regulations, the emerging concept of nanoarchitectonics has been proposed. Analytical sciences, including sensing/detection, sensors, and related device construction, are active targets of the nanoarchitectonics approach. This review article focuses on the two features of interface and nanostructures are especially focused to discuss nanoarchitectonics for analytical science. Especially, two selected topics, (i) analyses on molecular sensing at interfaces and (ii) sensors using self-assembled supramolecular nanostructures, are exemplified in this review article. In addition to recent general examples, specific molecular recognition at the air–water interface and fabrication of sensing materials upon self-assembly of fullerene units are discussed. Descriptions of these examples indicate that nanoarchitectonics and analytical science share common benefits, and therefore, developments in both research fields should lead to synergies.

To establish a guideline for the design of cell culture substrates to control human mesenchymal stem cell (MSC) differentiation, we quantitatively characterized the heterogeneity in the responsiveness of MSCs to the elastic modulus of culture substrates. We analyzed the elastic modulus-dependent dynamics of a mechanotransducer, YAP, and an osteogenic differentiation factor, RUNX2, in three different MSC lots using a styrenated gelatin gel with controllable elastic modulus. The percentage of cells with YAP in the nucleus increased linearly with increases in the elastic modulus, reaching a plateau at 10 kPa for all the lots analyzed. The increase in the percentage with the substrate elastic modulus was described by the same linear function. The percentage of cells with RUNX2 nuclear localization also increased linearly with increases in the substrate elastic modulus, plateauing at 5 kPa, although the regression lines to the linearly increasing regions varied between lots. These similarities and differences in YAP and RUNX2 dynamics among cell populations are basis to design the substrate elastic modulus to manipulate YAP and RUNX2 localizations.

昭和33年10月,太平洋海域で漁獲した汚染度のいちじるしいマグロ類肝臓を入手したので,これらのうちの4検体についてTomskinsのイオン交換樹脂法および沈殿法によつて放射性核種を分離し,半減期,吸収およびエネルギーの測定によって<SUP>59</SUP>Fe,<SUP>65</SUP>Zn,<SUP>115m</SUP>Cdおよび<SUP>90</SUP>Sr+<SUP>90</SUP>Yを確認し,さらにそれぞれを定量した.<BR><SUP>65</SUP>Znならびに<SUP>59</SUP>Feは,<SUP>55</SUP>Feとともにビキニ海域で漁獲された魚類からすでに検出されている.また<SUP>113</SUP>Cd,<SUP>113m</SUP>Cdおよび<SUP>115m</SUP>Cd<SUP>9</SUP>がマグロ類から定性的に検出されているが,今回は<SUP>115m</SUP>Cdのみを確認し,定量をおこなった.<BR><SUP>65</SUP>Znおよび<SUP>59</SUP>Feは明らかに誘導放射性核種であり,また<SUP>115m</SUP>Cdはfission product中の生成率はきわめて少ないと考えられるにもかかわらず,これらが汚染の主要原因となっていることは興味深い.<BR>今回分析に供した以外の生肝臓数検体についてγ線スペクトルを測定した結果,<SUP>65</SUP>Znおよび<SUP>59</SUP>Feはいずれの検体についても共通に認められたが,さらに未確認ではあるが<SUP>54</SUP>Mnと思われるピークを認めた.

In HPLC analyses of soluble dietary fiber, desalting processes using open, mixed-bed ion-exchange columns are time-consuming and labor-intensive. We developed and validated a simple desalting method using tandem cation/anion exchange SPE cartridges. We found that combining Bond Elut Jr SCX (upstream) and Bond Elut PSA (downstream) cartridges provided adequate desalting of test solutions. The developed method was then validated in an inter-laboratory study. Five test samples were prepared by mixing food matrixes with purified soluble dietary fiber and treated to generate solutions to test the desalting process. These solutions were then analyzed by eight different laboratories. The results demonstrated that the developed method is simple and reliable for desalting samples containing 140 to 945 mg/100 mL of soluble dietary fiber in preparation for HPLC analysis of soluble dietary fiber.

Sample preparation is a highly important and integral part of bioanalysis for cleaning up the complex biological matrices and thereby minimizing matrix effect. Matrix effect can jeopardize the precise quantification and adversely affect the reliability of liquid chromatography–mass spectrometry-based analytical results by alteration of analyte ionization. Matrix components result in suppression or enhancement of the intensity of analyte response. In spite of the high specificity and selectivity of tandem mass spectrometry, a relatively higher concentration of coeluted matrix elements present in biofluids may alter the efficiency of quantification of a bioanalytical method. Numerous literature reports different types of sample preparation techniques employed in bioanalysis. In this review, the strategies for selection of the appropriate sample clean-up technique in bioanalysis are discussed extensively. A paradigm shift in the arena of sample preparation and bioanalytical approaches involving the liquid chromatography–mass spectroscopic technique has been scrutinized. Current trends and possible future advancements in the field of biological sample extraction methods, including instrumental techniques are analyzed in detail.

Measurement of single molecule interaction is performed by various micromanipulation techniques. However, a complex operation is required to perform the measurement, so the measurement throughput is low and a large amount of time is required for statistical analysis. In this paper, we propose a method for measuring intermolecular interactions that applies a tensile force to the intermolecular bond by using centrifugal force and can easily measure binding force. The binding force of an antigen–antibody reaction system was measured by the use of the proposed method as a proof of concept. The measured binding forces were distributed in the range of 19 – 133 pN. The values obtained by use of the proposed method were consistent with the conventional method. This result is promising for applications that require further development, such as the development of a biosensor as a novel measuring technique that is based on the intermolecular interaction measurement.

Cannabis is an important industrial plant, in addition to its illicit drug use. Compound Δ9-THC (Δ9-tetrahydrocannabinol) is mainly responsible for the hallucinogenic effect on humans. The aminoalkylindole group cannabimimetics targets at the same physiological receptors to mimic the analgesic effects of Δ9-THC. Since there is no reliable colorimetric test to detect these synthetic cannabimimetics on site, a simple colorimetric assay for (aminoalkyl)indole group-containing drugs was developed, based on the silica/sulfuric acid-catalyzed Ehrlich reaction of (aminoalkyl)indoles with p-dimethylaminobenzaldehyde. The electrophilic substitution reaction of indoles with carbonyl compounds resulting in the formation of bis(indolyl)alkanes in an acid-catalyzed reaction has been used for the first time for their spectrophotometric determination by color change from yellow to purple/blue. The method was statistically validated against liquid chromatography tandem mass spectrometry, and applied to certain (aminoalkyl)indole derivatives, with 0.5 – 2.5 μg mL−1 detection limits for AM-2201, JWH-081, MAM-2201, JWH-018, JWH-210, JWH-122, 5F-PB-22 and XLR-11.

Tandem mass tags (TMT) are usually introduced at the levels of isolated proteins or peptides. Here, for the first time, we report the labeling of whole cells and a critical evaluation of its performance in comparison to conventional labeling approaches. The obtained results indicated that TMT protein labeling using intact cells is generally possible, if it is coupled to a subsequent enrichment using anti-TMT antibody. The quantitative results were similar to those obtained after labeling of isolated proteins and both were found to be slightly complementary to peptide labeling. Furthermore, when using NHS-based TMT, no specificity towards cell surface proteins was observed in the case of cell labeling. In summary, the conducted study revealed first evidence for the general possibility of TMT cell labeling and highlighted limitations of NHS-based labeling reagents. Future studies should therefore focus on the synthesis and investigation of membrane impermeable TMTs to increase specificity towards cell surface proteins.

Therapeutic peptides and diagnostic agents with their molecular size below the renal clearance threshold suffer from short blood circulation time. Here, we report a novel design of peptide-based ligand with a strong binding affinity to human serum albumin (HSA), which can be used as a tag to extend the blood circulation of small-size molecules. We designed ligands with dual alkyl groups connected with a negatively charged spacer. The ligands showed both higher binding affinity to HSA and a higher retention in mice blood than that of a single alkylated peptide.

A new analytical methodology for a simple and efficient on-line preconcentration of trace inorganic anions in water and salt samples prior to ion chromatographic determination is proposed. The preconcentration method is based on partition/ion-exclusion chromatographic ion stacking (PIEC ion stacking) with a hydrophilic polymer gel column containing a small amount of fixed anionic charges. The developed on-line PIEC ion stacking–ion chromatography method was validated by recovery experiments for the determination of nitrate in tap water in terms of both accuracy and precision, and the results showed the reliability of the method. The method proposed was also successfully applied to the determination of trace impurity nitrite and nitrate in reagent-grade salts of sodium sulfate. A low background level can be achieved since pure water is used as the eluant for the PIEC ion stacking. It is possible to reach sensitive detection at sub-μg L−1 levels by on-line PIEC ion stacking–ion chromatography.
