The effects of various tannins and related compounds on the actions of adrenocorticotropic hormone (ACTH) and insulin in fat cells isolated from rats were investigated. Hydrolyzable tannins such as geraniin, mallotusinic acid, chebulinic acid and chebulagic acid enhanced the ACTH-induced lipolysis at the concentration of 20 μg/ml or 5 μg/ml. On the other hand, these hydrolyzable tannins had no effect on the insulin-stimulated lipogenesis from glucose. Condensed tannins such as Ss-tannin 1 and RSF-tannin H showed weak inhibitory effects on the ACTH-induced lipolysis, while they enhanced the insulin-stimulated lipogenesis from glucose. Based on these results, the relationship between the structures and physiological actions of these tannins is discussed.

Three new piscicidal triterpenens named irisgermanicals A, B and C along with seven known iridal-type triterpenes including iripallidal and iriflorental were isolated from the bark of Iris germanica rhizome, upon bioassay-guided fractionation using the Medaka (killie-fish; Oryzias latipes), and their bicyclic structures were elucidated based on the spectral analyses. Irisgermanicals B and C were characterized as geometrical iriflorental and iripallidal isomers, respectively, concerning the α, β-unsaturated aldehyde moiety. The piscicidal activity was observed for bicyclic iridals, among which iriflorental exhibited most potent activity.

Large-size secondary metabolites with dimeric-oligomeric and/or dendrine-type structure from higher plants are regarded as natural nanomolecules which are constructed in a bottom-up pathway from small molecules under biological condition. Among them are polyphenols classified as tannins which have large molecular weights ranging from 500 to 4000, and strong affinity to proteins, alkaloids and heavy metals to form complex molecules (precipitates). They constitute two major groups : one is condensed tannins (proantocyanidins), which are composed of flavan-3-ol units linked through C-C bond, and the other is hydrolyzable tannins, which are principally multiple esters of D-glucose with gallic acid and its oxidative metabolites. This paper reviews the diverse structural characteristics of both tannins, especially ellagitannin oligomers up to pentamers, and examples of their biological activities. Formation of insoluble and soluble complex between tannins and proteins are also described.

キシメジ科スギヒラタケの化学成分に関する研究の一環として,本キノコ中のUV検出成分についてHPLC分析を行った.UV検出による主要ピークの1つを単離したところ,各種分光法の結果に基づき,共役トリエン構造を有するα-エレオステアリン酸であると同定した.α-エレオステアリン酸は,試験した他の8種の食用キノコからは検出されなかったため,スギヒラタケ特有の脂肪酸であることが示唆された.またスギヒラタケおよび他の食用キノコ中の遊離長鎖脂肪酸について,2-ニトロフェニルヒドラジドへ変換後,HPLC分析した.スギヒラタケの主要脂肪酸としてオレイン酸が検出され,α-エレオステアリン酸のほか,リノール酸,パルミチン酸,ステアリン酸などの飽和長鎖脂肪酸が検出された.

The scavenging effects of twenty-five tannins including low-molecular polyphenols on the superoxide anion radical (O_2^-) generated in the hypoxanthine-xanthine oxidase system were estimated by electron spin resonance (ESR) measurements of the adducts formed by 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) and the radical. The scavenging effects of tannins and related polyphenols having ortho-trihydroxyl (pyrogallol) structure [galloyl, hexahydroxydiphenoyl (HHDP) groups in hydrolyzable tannins, galloyl group in acylated proanthocyanidins, and the B-ring of some flavan-3-ols] were stronger than the effects of unacylated proanthocyanidins. The effects of tannins and related polyphenols on the superoxide anion radical were also compared with those on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Each tannin in an ethanol solution of DPPH radical reduced the intensity of the signal of the DPPH radical, and gave a weak signal assignable to a radical derived from that tannin, in a similar way to the appearance of the signal of dl-α-tocopherol radical, accompanied with reduction of the signal of DPPH radical, in a mixture of dl-α-tocopherol and the DPPH radical. In contrast to the case of the superoxide anion radical, the effects of unacylated proanthocyanidins on DPPH radical were comparable with those of the other types of tannins. The scavenging effects of all of the tannins and related polyphenols tested in the experiments on DPPH radical were stronger than that of dl-α-tocopherol.
