There are many huge macromolecular complexes in living organisms. They are often hard to crystallize because of their size, complexity and heterogeneity. Cryo-electron microscopy (cryo-EM) is a suitable method to analyze the structures of such biological macromolecules, because it can be applied to various forms of samples, e.g. two-dimensional crystal, helical assembly, spherical virus, dispersed particle, cell organelle and cell, although attainable resolution depends on the system. In this review, I introduce these techniques and examples of the structure analysis, and briefly review the perspective of cryo-EM.

The crystal structures of polymorphisms of free amino acids have been reviewed. The polymorphisms were classified into three types: The first, including DL-methionine, DL-norleucine and DL-a-amino-butyric acid, has polymorphs of different packing arrangement of molecular double layers which is rigidly bound by hydrogen bonds. In this type, the polymorphs can interconvert in solid state by temperature. The second type, glycine, L-cystine and L-histidine, has polymorphs in which hydrogen bonding schemes are different although molecular conformations are similar. The third type, L-cystein, L-aspartic acid and L-glutamic acid, has polymorphs with different hydrogen bonding schemes and different molecular conformations. In this type, metastable form converts irreversibly to stable form by solvent-mediated transformation mechanism.

Chocolate consists of solid particles such as cocoa, sugar, and milk solids which are dispersed in a continuous fat phase. The major fat in chocolate is cocoa butter, and it is crystallized at room temperature, so chocolate is a food which we eat crystals. Therefore, fat crystals play a great role in chocolate quality and storage stability. In this paper, polymorphism of cocoa butter and the transformation of polymorphic forms are reviewed including recent results.

In-situ synchrotron diffraction experiments were conducted in order to clarify the formation process of α'-martensite from the γ-phase induced by external strain, combined with Lorentz transmission electron microscopy (TEM) and high-resolution TEM observations. Lorentz TEM observation revealed that α'-martensite exist near the defect structures such as dislocations and stacking faults in the parent γ-phase. In addition, it is clearly demonstrated that ε-martensite with hexagonal symmetry appears as an intermediate phase during the plastic deformation of SUS304 stainless steel. Our experimental results suggested that the interfaces between twin structures of the γ-phase presumably play a crucial role in the formation of ε-martensite.

Protein crystallography has been leading the structural biology field by solving molecular structures at atomic resolution. However, it has been limited in the integration of structural information of macromolecules with a dynamic nature, which can adopt various conformations in time scales of motion even in large complexes. Therefore, it is critical to use multiple methodologies to understand biological phenomena, and connect the interfaces of molecules at both molecular and atomic resolution in macromolecular assembly. “Correlated structural analysis” is a concept in structural life science to fill the gaps between structural biology techniques. In Japan, the Platform for Drug Discovery, Informatics, and Structural Life Science has been launched with diverse expertise in bioinformatics, sample production, and structure determination. The platform supports not only conventional structural studies via X-ray crystallography, but also correlated structural analysis by integrating protein crystallography, small angle X-ray scattering, NMR, and electron microscopy data.

With the sequencing of the genomes of various species, attention has turned to the structure, properties, and functional activities of proteins. However, rapid progress in the area of proteomics is premised on the availability of sufficient amounts of a large number of proteins. Here we described a novel cell-free system from wheat embryos for the high-throughput screening/ synthesis of gene products. Our system should open up many possibilities in the post-genome era.

10 years have past since a system for experiments under high pressure and temperature using synchrotron radiation, named MAX80, was planned to be installed in the Photon Factory. Many scientifically interesting activities in various fields have been done using MAX80. The development of this system has been performed intensively by high pressure community of Japan in cooperation with synchrotron radiation group. The process of the development, outline of the system and typical applications are described.

Thermostable direct hemolysin (TDH) is a major virulence factor of Vibrio parahaemolyticus that causes pandemic food-borne enterocolitis mediated by seafood. TDH exists as a tetramer in solution, and it possesses extreme hemolytic activity. Here, we present the crystal structure of the TDH tetramer at 1.5 Å resolution. The TDH tetramer forms a central pore with dimensions of 23 Å in diameter and ∼50 Å in depth. π-cation interactions between protomers comprising the tetramer were indispensable for hemolytic activity of TDH. The N-terminal region was intrinsically disordered outside the pore. Molecular dynamics (MD) simulations suggested that water molecules permeate freely through the central and side channel pores. These findings imply a novel membrane attachment mechanism by a soluble tetrameric pore-forming toxin.
