Mass spectrometry is a powerful methodology for chemical screening to directly quantify substrates and products of enzymes, but its low throughput has been an issue. Recently, an acoustic liquid-handling apparatus (Echo®) used for rapid nano-dispensing has been coupled to a high-sensitivity mass spectrometer to create the Echo® MS system, and we applied this system to screening of SARS-CoV-2 3CL protease inhibitors. Primary screening of 32,033 chemical samples was completed in 12 hours. Among the hits showing selective, dose-dependent 3CL-inhibitory activity, 8 compounds showed antiviral activity in cell-based assay.

To develop a method for predicting the skin color of grape berries of three cultivars of Vitis labrusca L. × Vitis vinifera L. grown in Japan, we investigated the relationship between skin color and air temperature in the grape production areas of 18 prefectures. When mean air temperature during the 40 days before harvest date was ≥24°C, the skin color ratings of ‘Kyoho’, ‘Pione’, and ‘Suzuka’ were significantly negatively correlated with temperature. Skin color ratings decreased by about 1 unit per 1°C increase; at a given mean air temperature during this period, the rating of ‘Suzuka’ was higher (by 0.7 units) than that of ‘Kyoho’, which was higher (by 1.0 unit) than that of ‘Pione’. Because an approach to predict harvest date has not been established, we developed a method to predict skin color at harvest based on air temperature after the full-flowering date. We found the times that had a strong negative correlation between the mean air temperature and the skin color rating at harvest was 43 days from 50 DAF (days after full flowering) for ‘Kyoho’, 46 days from 46 DAF for ‘Pione’, and 42 days from 52 DAF for ‘Suzuka’. We obtained a linear regression equation for the relationship between the skin color rating at harvest and the mean air temperature during the periods. If the full-flowering date is known, it is possible to predict skin color at harvest by using this equation and the predicted air temperature after full flowering. We also developed a method for predicting anthocyanin contents in berry skins at harvest using significant regressions among the skin color rating, the skin anthocyanin content, and mean air temperature.

Persimmon (Diospyros kaki) is a tree crop species that originated in East Asia, consists mainly of hexaploid individuals (2n = 6x = 90) with some nonaploid individuals. One of the unique characteristics of persimmon is the continuous accumulation of proanthocyanidins (PAs) in its fruit until the middle of fruit development, resulting in a strong astringent taste even at commercial fruit maturity. Among persimmon cultivars, pollination-constant and non-astringent (PCNA) types cease PA accumulation in early fruit development and become non-astringent at commercial maturity. PCNA is an allelic trait to non-PCNA and is controlled by a single locus called the ASTRINGENCY (AST) locus. Previous segregation analyses indicated that the AST locus shows hexasomic inheritance; a recessive allele, ast, at this locus confers PCNA. Here, we report a shuttle mapping approach to delimit the AST locus region in the hexaploid persimmon genome by using D. lotus, a diploid relative of D. kaki, as a reference. A D. lotus F1 population of 333 individuals and 296 D. kaki siblings segregating for the PCNA trait were used to map the AST region using haplotype-specific markers covering the AST region. This indicated that the AST locus is syntenic to an approximately 915-kb region of the D. lotus genome. In this 915-kb region, we found several candidates for AST that were revealed from the fruit transcriptome of a population segregating for the PCNA trait. These results could provide important clues for the isolation of AST in hexaploid persimmon.

The pellicle of the Japanese chestnut (Castanea crenata Sieb. et Zucc.) is difficult to peel, with the exception of the recently developed ‘Porotan’, whose pellicle peelability (PP) is high and controlled by a single major gene. The objective of the present study was to identify any genetic differences in PP in Japanese chestnuts with difficult-peeling pellicles. We detected a significant genetic difference in PP (evaluated as the nut surface area that peeled away without scraping with a hand knife after deep frying in cooking oil) among five Japanese chestnut cultivars grown in Tsukuba, Japan, using two trees in 2 years, and among three cultivars grown in five locations using a single tree in 1 year. We evaluated the PP of 32 Japanese chestnut cultivars/selections and one wild clone (Shibaguri-37) using a single tree in 3 years to quantify the variation. The broad-sense heritability of mean values over the 3 years was estimated as 0.67. Shibaguri-37 had the highest PP. The suggested new genes controlling the variation in PP have high potential in terms of breeding strategy for easy-peeling pellicles as an alternative to the major gene of ‘Porotan’, the use of which is likely to result in inbreeding.

‘Hasshu’, a dwarf budsport that originated from the leading persimmon cultivar ‘Hiratanenashi’ (Diospyros kaki Thunb.), was discovered in Japan in 2005. Although ‘Hiratanenashi’ is seedless because of anisoploidy (2n = 135 = 9x), ‘Hasshu’ produces some small normal seeds. In this study, we investigated differences in the morphological characteristics of the vegetative organs and fruits and in the ploidy level between ‘Hasshu’ and ‘Hiratanenashi’. The shoot length, internode length, and leaf size of ‘Hasshu’ were smaller than those of ‘Hiratanenashi’. ‘Hasshu’ bore smaller flowers than those of ‘Hiratanenashi’ in late May, and had consistently smaller fruit than ‘Hiratanenashi’ at all fruit development stages. ‘Hasshu’ ripened in late October, similar to ‘Hiratanenashi’. Both ‘Hiratanenashi’ and ‘Hasshu’ are pollination-variant astringent (PVA) cultivars. Except for the deletion of one allele at ssrdk10, no differences were detected between the two cultivars at four simple sequence repeat (SSR) marker loci. By flow cytometric analysis and chromosome observation, we confirmed that ‘Hasshu’ was octoploid (2n = 120 = 8x), indicating that it was both a dwarf and a ploidy-reduction mutation. These results suggest that recovery of the ability to produce some normal seeds by ‘Hasshu’ may have been caused by the change in ploidy from anisoploid to isoploid.
