L-Ascorbic acid 2-phosphate (Asc 2-P), a long-acting vitamin C derivative, stimulates collagen accumulation, cell proliferation, and formation of a three-dimensional tissue (dermis) by skin fibroblasts. Investigation of molecular mechanisms of Asc 2-P action indicated that it stimulates maturation of extracellular matrix (ECM) by activating transcription of genes for type I collagen which form a back-bone structure of the ECM. Asc 2-P also stimulates differentiation and maintains differentiated phenotypes of various kinds of cells of epithelial and mesenchymal origin. Asc 2-P may be useful to clarify new physiological functions of vitamin C and investigation of the molecular mechanisms.

Wernicke's encephalopathy was first described by Wernicke in 1881. However, the disease is often overlooked in our daily clinical activities, since Wernicke's encephalopathy can not be definitely diagnosed in an emergency room. If Wernicke's encephalopathy is not treated properly with vitamin B_1, patients will usually suffer from Korsakoff syndrome or sometimes fall into death. Vitamin B_1 deficiency has been thought to be overcome by an abundant food supply or effective vitamin B_1 treatment. Increased alcohol consumption or unbalanced diet has introduced a new aspect of vitamin B_1 deficiency, especially Wernicke's encephalopathy which often leads to dementia. To prevent dementia due to Wernicke's encephalopathy, it has been recommended that a large dose of vitamin B_1 is administered to patients with consciousness disturbance. This article summarizes the pathophysiology, diagnosis and symptoms of Wernicke's encephalopathy as well as the dose of vitamin B_1 administered to patients with Wernicke's encephalopathy and the side effect of the vitamin B_1 therapy.

In urine the vitamin C values obtained by using the α,α'-dipyridyl method were often elevated due to urinary reducing substances. Ascorbate oxidase is highly specific for oxidizing vitamin C. An improved method using ascotbate oxidase is described for the determination of urinary vitamin C. The method is specific, sensitive and reproducible, and it is also rapid and simple enough for many measurements. Using the method the urinary vitamin C was determined for 176 male students, 61 female students, 352 male staff members and 81 female staff members of our university. There was little difference in overall frequency distribution of vitamin C values in the four groups. However, somewhat lower vitamin C levels were found in students compared with staff members, and in male groups compared with female groups.

Folates are essential cofactors that are required for the provision of one-carbon moieties in key biosynthetic and epigenetic processes. Mammals cannot synthesize folates. Hence, dietary sources must meet metabolic needs, necessitating an efficient intestinal absorption mechanism. It has been suggested that absorption of folates involves carrier-mediated process with a low-pH optimum. Its molecule entity had long been unclear, but it was only recently that proton-coupled folate transporter (PCFT/SLC46A1), which was originally cloned as a heme transporter, was redefined as a transporter that mediates the translocation of folates across the cellular membrane by a proton-coupled mechanism. Since this discovery, studies on PCFT/SLC46A1 have been progressing rapidly, accumulating evidence for its role as the major folate transporter in the intestine. In this review, we summarize up-to-date information on this newly identified transporter, and describe its physiological significance in intestinal absorption and cellular distribution of folates and antifolate drugs such as methotrexate.
