Bifidobacterium sp. strain SEN was isolated and characterized by hydrolytic conversion of sennosides to sennidins (Akao et al., Appl. Environ. Microbiol., 60, 1041 (1994)). The sennoside-hydrolyzing capacity of the strain SEN was disappeared following the addition of glucose to the media in spite of good bacterial growth and potent activity hydrolyzing p-nitrophenyl β-D-glucopyranoside (pNPG). In a fructose-containing medium, no such suppressing effect was shown. Following a 10 h incubation in 50 mM potassium phosphate buffer (pH 7.4), the sennoside-hydrolyzing activity of the bacterium increased, dose-dependently, with the addition of sennoside B. Inhibition of the substrate-induced increase in sennoside-hydrolyzing activity was observed following the addition of some antibiotics (chloramphenicol, streptomycin, and rifampicin). In particular, chloramphenicol completely inhibited the increase of sennoside-hydrolyzing activity while 38% pNPG-hydrolyzing activity remained. It is suggested that the strain SEN produces two different β-glucosidases of which the sennoside-hydrolyzing enzyme is inducible. In addition, the glucosides pNPG, esculin, salicin, or amygdalin stimulated the induction of the sennoside β-glucosidase, but less markedly than sennoside. Sennidin A or sugars (glucose, fructose, cellobiose, or maltose) did not induce the enzyme.

Inhibitory effects of caffeoylquinic acids isolated from leaves of Artemisia species and other related compounds on the lipoxygenase dependent peroxidation of linoleic acid were studied by kinetic and electron spin resonance(ESR) measurements. The order of inhibition activity was as follows : 3,5-dicaffeoylquinic acid (ID_50=2.0×10^-5 M)=rosmarinic acid > geraniin (2.8×10^-5 M)>α-tocopherol (3.7×10^-5 M)>chlorogenic acid (7.5×10^-5 M)=caffeic acid>ferulic acid (2.5×10^-4 M). This order coincided well with that obtained from the measurement of radical scavenging activities of these compounds against 1,1-diphenyl-2-picryl hydrazyl. The inhibition profile of these compounds on lipid peroxidation in the lipoxygenase system was quite similar to those obtained previously in the biological systems of rat liver mitochondria and microsomes. In the separate ESR measurements in alkaline dimethyl sulfoxide solution, all caffeoyl-quinic acids exhibited relatively stable ESR signals assigned as a radical derived from the one-electron oxidation of dihydroxyphenyl group. From these results, it was concluded that the radical scavenging mechanism is commonly operative in both chemical and biological peroxidation systems.

The spectral karyotyping (SKY) method is a novel molecular cytogenetic technique which simultaneously discerns entire chromosomes. In order to elucidate the origins of micronuclei induced under hyperthermic conditions in human lymphocyte culture, peripheral blood cells were cultured at 40℃ or 42℃ for 3-24 h, using the cytokinesis-block method with cytochalasin B. The induced micronuclei were identified by the fluorescence in situ hybridization (FISH) and SKY methods. At 42℃ for more than 6 h, the frequency of occurrence of micronuclei in binucleated cells rose with increasing incubation time. By the FISH method, 83.3% of micronuclei induced in 24 h culture at 42℃ were shown to be positive for the human centromeric probes. By the SKY method, each micronucleus induced under the hyperthermic conditions was identified unequivocally and shown to contain a specific chromosome. These results suggest that the micronuclei induced under the hyperthermic conditions in human lymphocyte culture contain chromosomes which do not migrate to the poles at the anaphase of the cell cycle because of the breakdown of the spindle apparatus.

Synthetic studies on salamander alkaloids isolated from animal venom in the skin gland of Salamandra maculosa LAURENTI were performed. Syntheses of three groups of alkaloids, one with a bicyclo-oxazolidine, one with a carbinolamine, and another with a pure 3-aza-A-homo steroid, are discussed. Regio- and stereospecific syntheses of the alkaloids are discussed in connection with the construction of the bicyclo-oxazolidine system of the major alkaloids and in connection with Beckmann rearrangement of steroid 3-ketoximes for the syntheses of the other alkaloids. A theoretical approach to the resolution of the geometrical isomers of oximes by adsorption chromatography is also discussed.

From the aerial part of Stachys sieboldi MIQ. (Labiatae), three glycosides were isolated and characterized as isoscutellarein. 4'-methyl ether 7-O-β-(6'''-O-acetyl-2"-allosyl) glucoside (1), isoscutellarein 7-O-β-(6'''-O-acetyl-2"-allosyl) glucoside (2), and acteoside (8). The effects of these glycosides on the activity of hyarulonidase were also examined.

During screening for the development of drugs from natural products, methanolic extract of tuber of Stachys sieboldii MIQ. (Labiatae) significantly inhibited the lethal induced by KCN in mice. The methanolic extract was fractionated by column chromatography to identify the active constituents. Acteoside and stachysoside C, phenylethanoid glycoside, have a significant effect on the KCN-induced anoxia model.

Cardenolides in the seeds, bark, and leaves of Cerbera manghas L. were investigated. From the seeds, tanghinigenin glycosides were isolated along with known digitoxigenin glycosides, cerberin, neriifolin, thevetin B, and 2'-O-acetyl thevetin B. From the barks of root and stem, gentiobiosyl thevetoside, glucosyl thevetoside, and thevetoside of tanghinigenin and 17βH-tanghinigenin were obtained. The cardenolides in the air-dried leaves were found to vary with seasons. 17βH-neriifolin is major and preferable to neriifolin in July, while 17βH-deacetyltanghinin and deacetyltanghinin present in the leaves of Feb.

Several amino acids and ammonia were allowed to react with L-ascorbic acid or D-glucose in 0.1 M phosphate buffer (pH 7.0) at 37°C for 2 months. The reaction mixtures were submitted to mutagenic assay and structural analysis. The browning products from L-tryptophan (0.25M) and L-ascorbic acid (1M) showed a DNA-damaging potency to B. subtilis. They also gave a mutagenic activity to S. typhimurium TA 100 but not TA 98. By extracting the reaction products with benzene and ethyl acetate, followed by purifying them through silica-gel column and thin layer chromatographies (Kiesel-gel 60), 6 β-carbolyl and 4 furyl derivatives were identified. Among them, 1-(2-furyl)-9H-pyrido-[3, 4-b] indole exhibited a mutagenic activity to S. typhimurium TA 100 with or without S-9 mixture and 1-(2-furyl)-9H-pyrido [3, 4-b] indole-3-carboxylic acid showed the activity to S. typhimurium TA 100 only in the absence of S-9 mixture.
