Leucomycin A<SUB>3</SUB> (I) was oxidized with active manganese dioxide to dehydroleucomycin A<SUB>3</SUB> (II) which was identical with magnamycin B. Acid treatment of I yielded demycarosylleucomycin A<SUB>3</SUB> (VII), which was converted to a triacetate showing mass spectrum M<SUP>+</SUP> (m/e 725), and this observation confirmed the structure of I. The diene system of I was assumed to have a trans-trans configuration on the basis of its NMR spectrum, and a full structure of I was proposed with stereochemistry.

A series of new antibiotics, Kinamycin A, B, C, and D which are mainly effective against gram-positive bacteria, were extracted with chloroform from the broth filtrate of Streptomyces murayamaensis sp. nov. HATA et OHTANI. Kinamycin C (I), C<SUB>24</SUB>H<SUB>20</SUB>O<SUB>10</SUB>N<SUB>2</SUB>, m/e 496 (M<SUP>+</SUP>), was determined to have an 8-hydroxynaphthoquinone skeleton, nitrile, acetoxyl, and tertiary methyl groups from its ultraviolet (UV), infrared (IR), and nuclear magnetic resonance (NMR) spectra, and was further found to have a unique structure of N-C-N from some chemical reactions and X-ray diffraction. Structure relationship among I and other components A, B, and D was assumed to be due to the difference in the number and position of the acetoxyl group from analyses of IR, NMR, and mass spectra of their acetylated compounds. The antimicrobial activity of the four kinamycins increases with the decreasing number of acetoxy group, in the order of kinamycin C, A, D, and B. In addition, some derivatives obtained during structural studies on I were found to have nearly equal or increased antimicrobia1 activity compared with I and kinamycin D (XI).

The effects of geniposide isolated from Gardeniae Fructus ("San-shi-shi"in Japanese) on metabolic alterations in high sugar diet-fed rats were investigated. It was found that the oral administration of the high sugar diet caused hyperlipemia, liver injury with elevation of glutamic pyruvic transaminase (GPT) in the serum, and the accumulation of lipid peroxide in the liver. Geniposide reduced the serum triglyceride, lipid peroxide, phospholipid, glucose, insulin and GPT in the high sugar diet-fed rats. It was found that the administration of geniposide reduced the sucrase activity in the small intestine and the deposition of lipid peroxide in the liver.

The effects of various tannins and related compounds on the actions of adrenocorticotropic hormone (ACTH) and insulin in fat cells isolated from rats were investigated. Hydrolyzable tannins such as geraniin, mallotusinic acid, chebulinic acid and chebulagic acid enhanced the ACTH-induced lipolysis at the concentration of 20 μg/ml or 5 μg/ml. On the other hand, these hydrolyzable tannins had no effect on the insulin-stimulated lipogenesis from glucose. Condensed tannins such as Ss-tannin 1 and RSF-tannin H showed weak inhibitory effects on the ACTH-induced lipolysis, while they enhanced the insulin-stimulated lipogenesis from glucose. Based on these results, the relationship between the structures and physiological actions of these tannins is discussed.

The effects of oral administration of flavonoid components of Scutellariae Radix on serum and liver lipid levels of rats treated with ethanol were investigated. It was found that wogonin reduced serum triglyceride level, and that baicalein and baicalin, the major components of the drug, decreased total cholesterol, free cholesterol and triglyceride contents in the liver. Baicalein increased high density lipoprotein-cholesterol (HDL-ch) in the serum of the ethanol-treated rats. In addition to these in vivo experiments, the actions of wogonin, baicalein and baicalin on catecholamine-induced lipolysis in isolated fat cells were investigated. It was found that the three flavones inhibited noradrenaline-induced lipolysis in isolated fat cells. The relationship between these in vivo and in vitro experiments is discussed.

Trichothecene mycotoxins are chemically related sesquiterpenoids with 12,13-epoxy-trichothecene nuclei. T-2 toxin, nivalenol, deoxynivalenol, and satratoxins are isolated as causative toxicants of food-borne diseases such as moldy-corn toxicosis in U.S., Akakabi (red-mold) toxicosis in Japan, vomiting and feed-refusal diseases in U.S., and satratoxicosis in the Central Europe, respectively. Currently, the TLC, HPLC, GC and GC-MS are introduced for chemical analyses of the trichothecene mycotoxins in food and feed, and the accumulated data strongly revealed that, among more than 60 kinds of trichothecenes, deoxynivalenol (=vomitoxin) and nivalenol, both being produced by Fusarium graminearum (Gibberella zeae in perfect stage), are major contaminants in wheat, barley, corn and other cereals and foodstuffs. In 1983,the Canadian goverment set a guideline for deoxynivalenol in wheat, and FDA (US) recommended "level of concern" for this mycotoxin. As for nivalenol, which is also a contaminant in domestic cereals along with deoxynivalenol and possesses more than ten times higher toxicity than deoxynivalenol, neither guideline nor "level of concern" is recommended by the Japanese goverment at present.

From the methanolic extract of leaves of Apocynum venetum L. var. basikurumon HARA, eight constituents were isolated and identified with respective authentic samples as succinic acid, chlorogenic acid, isoquercitrin, hyperoside, D-(-)-bornesitol, sucrose, β-sitosterol, and β-amyrin.

Protective effects of several hydroxyl radical (HO&middot;) scavengers and superoxide dismutase on alloxan-induced diabetes were studied in vivo. Thiourea, methylthiourea, dimethylthiourea and ethanol, HO&middot; scavengers (type 1), which easily cross cellular membranes, protected initial- and permanent-hyperglycemia. In contrast, mannitol and sorbitol (type 2), which can not cross cellular membranes, protected initial-hyperglycemia, but did not protect permanent-hyperglycemia. Furthermore, superoxide dismutase which is not expected to be incorporated into pancreatic &beta;-cell, strongly protected initial-hyperglycemia but partially protected permanent-hyperglycemia. These results suggest that HO&middot;primarily damages the &beta;-cell membranes by acting at or neat a site involved in insulin release, and subsequently damages the intracellular components. From these results, the possible protective mechanism of these scavengers was discussed.

目的:現在,国家戦略特区の過疎地等で認められている遠隔薬剤指導(オンライン服薬指導)は,オンライン診療と合わせて医療へのアクセシビリティ改善への寄与が期待されている。本研究では,実際にへき地で実証運用されているオンライン服薬指導を利用する患者が感じるその利点や課題点を,インタビュー調査により探索的に検討した。方法:2019年10月に,オンライン服薬指導を受けた経験のある患者を対象として,インタビューガイドに基づく半構造化インタビューを実施した。インタビュー内容は逐語録化し,コーディング,カテゴリ化(<>で示す)を複数の研究者で行い,質的に分析した。結果・考察:対象者の年代は60代が3名,90代が1名,性別は男女2名ずつであった。患者はオンライン診療・服薬指導に関して,<病院・薬局に行かずに薬の入手が可能>,<医療機関が遠い地域での診療の簡便化>といった地理的課題の解消による利点を挙げるだけでなく,<自分の都合で診療時間の設定が可能>,<働き世代の通院に掛ける時間の省略>といった利便性の向上についても実感していた。一方で,<高齢者のタブレット端末の操作の難しさ>が課題点として指摘された。へき地在住者は高齢で独居の場合が多いことから,高齢者を対象としてタブレット端末への抵抗感をなくしたり,操作を支援したりする取り組みが必要であると考えられた。本調査では,オンライン上での医師への相談から受診を勧められ,疾患に気づいた経験をもつ患者がいた一方で,これまで薬剤師との接点がなかったためか,オンライン上で薬剤師に相談した経験をもつ患者は少なかった。今後,へき地でのオンライン服薬指導の有用性を向上させるためには,へき地における医療や健康サポートにおける薬剤師の関わり方についても同時に検討していく必要があると考えられた。

To investigate the role of constitutive hsp70 in protein folding and to probe the supplementation by other hsps in this folding, yeast cells expressing reduced constitutive hsp70 proteins, ssa1ssa2,were transformed with a plasmid expressing a bacterial luciferase protein. With several independent clone cells of transformants, the levels of luciferase activity and some hsps, such as hsp104,hsp90,hsp70 and hsp26,were examined. The luciferase activity was significantly lower in ssa1ssa2 transformants than in the wild type (wt) cell transformed with the same plasmid. Among several clone cells of ssa1ssa2,the cells with higher luciferase activities exhibited higher amounts of Ssa4 which is known to be expressed instead of lacking Ssa1 and Ssa2. The luciferase activity closely correlated with the amount of Ssa proteins, more than with the amount of other hsps. It is suggested that consitutional Ssa "chaperones" are needed for the folding of proteins and, in cells lacking Ssa1 and Ssa2,the increased Ssa4 is thought to partly compensate for their role in the folding of luciferase in vivo.

Mammalian small heat shock protein (s-hsp) has been suggested to participate not only in stress tolerance but also in the growth regulation and differentiation of cells. To confirm the role of s-hsp in cell growth, we investigated the relationship between the expression of hsp26 and yeast cell growth. Cells lacking constitutive hsp70,ssa1ssa2,have been known to have a poor growth rate and to over-express hsp26 and some other hsps. We obtained several cell clones of ssa1ssa2 whose doubling times were different from one another. The amount of hsp26 was closely linked to the doubling time of ssa1ssa2 cells. This result suggests that the expression of hsp26 modulates the growth rate of yeast cells lacking constitutive hsp70,similarly to mammalian cells.

Two new melampolide-type sesquiterpene lactones, 8β-epoxyangeloyloxy-9α-ethoxy-14-oxo-acanthospermolide (1) and 8β-angeloyloxy-9α-ethoxy-14-oxo-acanthospermolide (2), were isolated from the leaves of yacon [Smallanthus sonchifolia (POEPP. et ENDL.) H. Robinson] along with eleven known melampolides, allo-schkuhriolide (3), enhydrin (4), polymatin A (5), fluctuanin (6), 8β-angeloyloxy-9α-acetoxy-14-oxo-acanthospermolide (7), 8β-angeloyloxy-14-oxo-acanthospermolide (8), 8β-methacryloyloxymelampolid-14-oic acid methyl ester (9), uvedalin (10), polymatin B (11), 8β-tigloyloxymelampolid-14-oic acid methyl ester (12), and sonchifolin (13). Their structures were established on the basis of spectroscopic evidence including 1D- and 2D-NMR experiments. All isolates were evaluated for inhibition of LPS-induced nitric oxide production in murine macrophage RAW 264.7 cells.
