This paper is aimed at evaluating the effect of prolonged mental stress on human physiological state. The human secretory immune and endocrine substances were introduced as “biomarkers” of such a prolonged stress. This relatively new field of study investigating the relationship and/or interaction between human mental and somatic states has been developed according to the development of the molecular analysis techniques, such as Enzyme-linked immunosorbent assay (ELISA), and showed that the several secretory hormones and immune substances could possibly be useful stress markers. However, what types of stressors and its duration could be evaluated by such biomarkers is not well understood. In this article, the effect of prolonged mental stress during long-term graduation examination of undergraduate students was investigated comparing with the effect of short-term mental stress by simple arithmetic task. As a result, one immune substance within saliva, immunoglobulin A, was showed as a possible stress marker for both long- and short-term stressors.

色が人間の内分泌系に及ぼす生理的効果について,唾液中に含まれるコルチゾールにより検証した.青・赤・透明の眼鏡を着用させた状態でストレス負荷試験を実施した(被験者内計画:n=23).その結果,青色による内分泌系 (HPA系)のストレス反応抑制効果が認められた.

爪に含まれるコルチゾールは過去の比較的長期的なホルモンの動態を反映すると考えられているが,その標準的な定量方法についてはまだ確立されたものがない。本研究では爪試料の粉砕粒度や抽出時間が爪試料から抽出されるコルチゾール量に与える影響を検討した。健常な男性14名から手の爪を採取した。爪試料を1, 4, 16分間粉砕し,粉砕粒度の条件(粗い,中程度,細かい)を設定した。また,粉砕した検体はメタノールにより抽出処理を行うが,本研究では抽出時間を1, 6, 24, 48時間の4条件に設定した。上澄み液にろ過処理を施し,蒸発乾固させた後に,最終的にコルチゾールの抽出量を酵素免疫測定により評価した。粉砕粒度と抽出時間を要因とした分散分析を行った結果,粉砕粒度が細かいほど,また抽出時間が長いほど,コルチゾールの抽出量が多いことが示された。また,粉砕粒度と抽出時間の交互作用が有意であり,爪試料が中程度以上に粉砕されていて,かつ抽出時間が48時間の条件では,粉砕粒度の影響は小さいことも示された。本研究では粉砕粒度や抽出時間が爪からのコルチゾールの抽出量に影響を与えることを明確に示した。爪からコルチゾールを測定する際はこれらの要因にも留意する必要性が示された。

The effect of an alerting odorant on the secretion of salivary cortisol during sleep and after awakening was investigated. Salivary cortisol was assessed because this glucocorticoid represents activation of the hypothalamus-pituitary-adrenal (HPA) system, which is a dominant stress reaction pathway in the body. Subjects were exposed to an alerting odorant, Thesaron®, during a 6h sleep period but not after awakening. Surprisingly, cortisol secretion after awakening was found to be suppressed, as well as during sleep. Exposure of this alerting aroma during sleep may have an impact on cortisol secretion after awakening by suppressing HPA activity after awakening.

(OBJECTIVE) The change in the secretion of stress-related bio-chemical substances, which are cortisol, immunoglobulin A (sIgA), and alpha-amylase, while asleep was regarded. As for the sample collection, we developed and proposed a saliva collection technique as a non-invasive and non-disturbing manner of sample collection while asleep, whereas the blood collection should impose greater psycho-physiological stresses to subjects. (METHOD) Subjects, 10 male university students, were instructed to take a 6-hour of sleep from 0:00 of the midnight to 6:00 in the morning at the environmental controlled room, where electrocardiogram (ECG) and blood pressure (BP) were measured, and saliva collection was made continuously in the time series. (RESULT) Salivary sIgA and alpha-amylase concentration obtained by saliva samples depicted a similar profile in the time series, i. e. gradually increase while subjects were asleep, and dropped to the initial level right after awakening. Meanwhile cortisol remained the same level while asleep and got started to increase right after awakening and reached at a peak around 40 minutes later. (CONCLUSION) We developed and proposed our own saliva collection technique for the purpose of non-invasive sample collection while asleep. As a developing result, the distinctive difference in the secretion of stress-related substances was illustrated;which might be derived from the difference in the stress-reaction pathways in our body.

精神的なストレスの評価指標としてコルチゾールというホルモンが注目されている.従来,同物質は血液,唾液,尿などの検体から定量されてきた.これに対し本研究では,皮膚に微細孔を形成し,経皮的にコルチゾールを定量する新たな手法を提案する.
