Decreased Mg2+ concentration has been implicated in altered vascular reactivity, endothelial dysfunction and structural remodeling, processes important in vascular changes and target organ damage associated with hypertension. Unlike our knowledge of other major cations, mechanisms regulating cellular Mg2+ handling are poorly understood. Until recently little was known about protein transporters controlling transmembrane Mg2+ influx. However, new research has uncovered a number of genes and proteins identified as transmembrane Mg2+ transporters, particularly transient receptor potential melastatin (TRPM) cation channels, TRPM6 and TRPM7. Whereas TRPM6 is found primarily in epithelial cells, TRPM7 is ubiquitously expressed. Vascular TRPM7 has been implicated as a signaling kinase involved in vascular smooth muscle cell growth, apoptosis, adhesion, contraction, cytoskeletal organization and migration, and is modulated by vasoactive agents, pressure, stretch and osmotic changes. Emerging evidence suggests that vascular TRPM7 function might be altered in hypertension. The present review discusses the importance of Mg2+ in vascular biology in hypertension and focuses on transport systems, mainly TRPM7, that might play a role in the control of vascular Mg2+ homeostasis. Elucidation of the relationship between the complex systems responsible for regulation of Mg2+ homeostasis, the role of TRPM7 in vascular signaling, and the cardiovascular impact will be important for understanding the clinical implications of hypomagnesemia in cardiovascular disease. The analysis of children’s activity-travel patterns has gained increasing attention in a variety of research fields. Most of the previous studies suggested that more compact development promotes less car-dependent travel among children. Unfortunately distinguishing what was a cultural artefact and what was a ‘universal’ truth of children’s travel is difficult as existing literature on children’s travel vary not just by country, but also by measures and analysis. The aim of this paper is to help bridge this gap of knowledge. Using the UK National Travel Survey and the Osaka Regional Area person trip datasets, this paper compares children’s travel behaviour in two countries with significantly different cultures. This paper discusses both consistencies and discrepancies of travel behaviours between the countries to highlight what factors are consistent despite cultural differences, and what may vary within cultures or different built environments.

Risperidone was compared with antipsychotics hitherto used for in vitro receptor binding using animal brain or cloned (human) receptors and in vivo receptor occupancy in rat and guinea pig brain following acute treatment. Both in vitro and in vivo, risperidone, 9-OH-risperidone, SM-9018, clozapine and clocapramine showed higher affinity for 5-HT2A- than for D2-receptors, whereas mosapramine, haloperidol, bromperidol and nemonapride had a slight to strong preference for D2- compared to 5-HT2A-receptors. In vivo, risperidone showed the highest potency for 5-HT2A-receptor occupancy; To obtain the same extent of D2-receptor occupancy, a 19-times higher dosage was required. 9-OH-Risperidone, the principal active metabolite of risperidone, showed a receptor occupancy profile comparable to that of risperidone. No regional selectivity for D2-receptor occupancy in mesolimbic vs nigrostriatal areas was detected for any of the compounds. Risperidone differed from the other compounds by the remarkably shallow slope of its D2-receptor dose-occupancy curve. A greater predominance of 5-HT2A-receptor vs D2-receptor occupancy and a more gradual occupancy of D2A-receptors differentiate risperidone from the other compounds. Both properties probably assist in preventing an extensive blockade of D2-receptors, the cause for extrapyramidal symptoms (EPS). The predominant 5-HT2A-receptor occupancy most likely underlies risperidone''s beneficial effects on the negative symptoms of schizophrenia and an adequately low D2-receptor occupancy adds to the treatment of positive symptoms with a low liability of EPS.

スギ(Cryptomeria japonica)花粉症は、日本国民の約3割に広がっているとの報告もあり、深刻な社会問題となっている。花粉症対策の一つとして、花粉の形成機構を解明し阻害することができれば、花粉の飛散量を減らすことが可能となる。本研究では、ゲノム編集技術の一つであるCRISPR/Cas9法を用いて、スギの花粉形成に関わる遺伝子に変異を導入し、花粉形成への影響を調べた。スギの花粉形成関連遺伝子を標的とするCRISPR/Cas9ベクターを構築し、アグロバクテリウム法により遺伝子組換えスギを作出した。遺伝子組換えスギのゲノムDNA中の標的遺伝子には欠失変異が見つかり、スギでもゲノム編集による遺伝子変異が生じることが分かった。夏季にジベレリンを散布し、遺伝子組換えスギの花芽形成を誘導した。標的遺伝子の両対立遺伝子に欠失変異が生じた遺伝子組換えスギでは雄花中に花粉が検出されなかったが、非組換えスギでは花粉が作られていた。従って、スギの花粉形成関連遺伝子に変異が起こることにより、無花粉になることが示された。本研究は、内閣府SIP次世代農林水産業創造技術により実施されました。

The Early Cretaceous Okurodani Formation, Tetori Group, near Shokawa village, Gifu Prefecture, Japan, has yielded a mixed assemblage of terrestrial and aquatic small vertebrates, including the fragmentary remains of a frog (ilium, vertebra) and a salamander (vertebra). These are the first Mesozoic lissamphibians recorded from Japan, and some of the oldest from Asia. The ilium is of rather primitive form and suggests a frog of basal grade, that is outside the Discoglossanura.

Twenty-four simulations were carried using the Meteorological Research Institute-Atmospheric General Circulation Model (MRI-AGCM) to predict the late 21st century climate under scenario A1B of the Special Report on Emissions Scenarios. Future climate analogues were identified for Central American capital cities using a recently developed nonparametric method. We used MRI-AGCM3.2H with a horizontal resolution of approximately 60 km, three convection schemes, four sea surface temperature distributions, and two initial conditions. Thus, the total ensemble size was 24, with a simulation period of 25 years. Most of the future analogues are at lower latitudes than their target cities, or near biological diversity and endemism hotspots like coral reefs and mangrove forests. Projected seasonal variations in surface air temperature and rainfall in Panama City were similar to the present-day climate of Soc Trang, located at the mouth of the Mekong River in Vietnam. The nonparametric method introduced in this study for identifying climate analogues can be utilized for impact assessments under a changing climate.

[Purpose] To characterize the case report evidence of Chiropractic BioPhysics® (CBP®) technique methods applied to increase cervical lordosis and improve forward head posture. [Methods] The CBP Non-profit website as well as PubMed and Index to Chiropractic literature were searched for case reports/series documenting the increase of cervical lordosis and improvement of forward head posture in the treatment of various craniocervical spinal disorders by CBP technique methods. [Results] Sixty patients were reported in 41 unique manuscripts detailing the improvement in cervical spine alignment by CBP technique methods. On average, there was a 14° improvement in cervical lordosis and a 12 mm reduction in forward head position after 40 treatments over 16 weeks with a 5-point reduction in pain rating scores. Thirty-eight percent of cases included follow-up showing only slight loss of lordosis, but maintenance of pain and disability improvements after an average of 1.5 treatments per month for 1.8 years. [Conclusion] An abundance of reports document improvement in craniocervical and other ailments by CBP methods that increase cervical lordosis. Routine radiographic imaging of the spine is recommended as it is safe and the only current practical method of screening for critical biomechanical biomarkers of sagittal spine alignment.

The generality of strength suggests that a “strong” individual will typically exhibit higher values of strength across a wide range of strength tasks for a given muscle relative to their weaker counterpart. This concept is often extended to adaptation, suggesting that increasing strength on a given movement or strength task with a given muscle should reflect an increase in other movements or tasks using that same muscle. The concept of a generality of strength adaptation appears less supported in the literature. Objective: To elaborate on recommendations for strength assessment, providing a focus on the “generality of strength” and the “generality of strength adaptation.”Design & Methods: We reviewed the literature on a generality of strength. In addition, we examined the resistance training lit erature to provide evidence and discussion on a generality of strength adaptation. Results/Conclusions: The generality of strength adaptation, even across strength skills using the same muscle on related movements seems quite low. Although some studies show a weak generality of strength adaptation and others show no generality of strength adaptation, it appears that increases in strength diminish as the strength assessment becomes farther removed from the actual training stimulus.

Background: This longitudinal study aimed to determine chronological changes in the seroprevalence of prior SARS-CoV-2 infection, including asymptomatic infections in Hiroshima Prefecture, Japan.Methods: A stratified random sample of 7,500 residents from five cities of Hiroshima Prefecture was selected to participate in a three-round survey from late 2020 to early 2021, before the introduction of the COVID-19 vaccine. The seroprevalence of anti–SARS-CoV-2 antibodies was calculated if at least two of four commercially available immunoassays were positive. Then, the ratio between seroprevalence and the prevalence of confirmed COVID-19 cases in Hiroshima was calculated and compared to the results from other prefectures where the Ministry of Health, Labour and Welfare conducted a survey by using the same reagents at almost the same period.Results: The numbers of participants in the first, second, and third rounds of the survey were 3025, 2396, and 2351, respectively and their anti-SARS-CoV-2 antibodies seroprevalences were 0.03% (95% confidence interval: 0.00–0.10%), 0.08% (0.00–0.20%), and 0.30% (0.08–0.52%), respectively. The ratio between the seroprevalence and the prevalence of confirmed COVID-19 cases in Hiroshima was 1.2, which was smaller than that in similar studies in other prefectures.Conclusions: The seroprevalence of anti–SARS-CoV-2 antibodies in Hiroshima increased tenfold in a half year. The difference between seroprevalence and the prevalence of confirmed COVID-19 cases in Hiroshima was smaller than that in other prefectures, suggesting that asymptomatic patients were more actively detected in Hiroshima.
