SPF発育鶏胚の高度病原性伝染性ファブリキウス嚢病ウイルス(hv IBDV)F539株及びDV86株に対する感受性を, 3つの接種ルートで, 従来型IBDV G691株と比較しながら調べた. その結果, 鶏胚のhv IBDV 2株に対する感受性はG691株より高く, 感染胚のほぼ100%が死亡し, 算出された50%胚致死量は50%胚感染量とほぼ同値を示した. 鶏胚に対する高い致死性はhv IBDVに特徴的な性状と思われた.

白色レグホーン種の原種をトリ白血病ウイルスの感染状態を基準に, 血清から抗体もウイルスも検出されない非感染鶏, 血清から抗体は検出されるが卵白からウイルスは検出されない感染・非排泄鶏, 卵白からウイルスが検出される感染・排泄鶏に分類し, 各群の生産性を比較した. その結果, 産卵率の低下, 卵白のハウ単位の低下, 卵殻の肥厚, 体重の軽量化等の生産阻害は感染・ 排泄鶏でのみ認められた.

2胚移植によって受胎した正常双胎妊娠牛(N牛), フリーマーチン妊娠牛(F牛)および反転性裂体妊娠牛(S牛)の妊娠全期間にわたる末梢血中プロジェステロン(P<SUB>4</SUB>), エストロン(E<SUB>1</SUB>)およびエストラジオール(E<SUB>2</SUB>)の消長を調べた. 血中P4濃度は, F牛が妊娠254日に双子を死産した際に急減した他は, 三者の間には相違がみられなかった. 血中E1濃度は, N牛においては好期の経過に伴い上昇し, 分娩日に最高値を示した後に急減し, F牛においてはN牛よりも低い値で推移し, 妊娠254日に突然上昇した後に急減した. また, S牛においてもN牛より低く, 変動を伴って推移した. 血中E<SUB>2</SUB>濃度は, 三者ともにE<SUB>1</SUB>より低く, かつE<SUB>1</SUB>濃度に平行して推移した. 血中E<SUB>1</SUB>, E<SUB>2</SUB>濃度は, 分娩後1日にN牛では両者とも20pg/ml以下の値に低下したが, F牛とS牛はE<SUB>1</SUB>が150kg/ml以上, E<SUB>2</SUB>が20pg/ml以上の値を示した. この成績は, 妊娠中の胎子数の判定同様子宮内の胞子の予後の判定においても, P<SUB>4</SUB>よりE<SUB>1</SUB>やE<SUB>2</SUB>の方が優れた指標となることを示唆している.

11才の雌ドーベルマンの喉頭に発生した腫瘍を摘出し, 検索した. 腫瘍は種々の大きさで多形の細胞のシート状配列で構成されていた. 腫瘍細胞はデスミン, ミオグロビン, アクチンに強陽性, ビメンチンに弱陽性, 高分子ならびに低分子のサイトケラチン, 上皮細胞膜抗原ならびにミオシンに陰性であった. 電顕的には, 一部の腫瘍細胞の細胞質に, クリステに富むミトコンドリが多数存在し, 不整形で電子密度が高くZ帯に類似した領域が認められた. 以上の所見から, 本腫瘍は横紋筋肉腫と診断された.

エジプトで飼育されている水牛, 牛および緬羊について, それらの第一胃内繊毛虫構成を調査した. その結果, 水牛では12属29種7型, 牛では10属28種11型および緬羊では7属18種6型がそれぞれ同定された. 牛と水牛では22種が共通してみられたが, 緬羊では12種が牛および水牛との共通種であった. 各家畜ともエントジニウム属, 特にE.simplex, E.nanellumおよびE.exigumが最も多くみられた. 以上から, エジプトの家畜反芻動物の第一胃内繊毛虫構成は, 水牛が熱帯地域の繊毛虫の一部を保有しているものの, 全体として, 温帯地域の反芻獣のそれと類似していることが明らかとなった.

2、4、6-trinitrobenzene sulfonic acid(TNBS)をラットの近位大腸内に投与すると、遠位大腸の伸展刺激に対する痛み感受性の有意な増大が認められた。TNBSに直接暴露された近位大腸には粘膜壊死と炎症性細胞浸潤が認められ、組織中ミエロパーオキシダーゼの有意な増加が認められた。一方、遠位大腸には粘膜壊死は発生せず、ミエロパーオキシダーゼ含量の増加も認められなかったが、トルイジンブルー陽性粘膜型肥満細胞が有意に増加していた。さらに遠位大腸組織を器官培養し、粘膜型肥満細胞脱顆粒の特異的なマーカーであるrat mast cell protease-2(RMCP-2)の遊離量を測定したところ、TNBS処置ラットでは正常対象値と比較して有意なRMCP-2遊離量の増加が認められた。また、肥満細胞安定化作用を持つデドキサントラゾールの皮下投与によりTNBS誘発大腸過敏は有意かつ用量依存的に抑制された。以上の成績から、脱顆粒亢進を伴った大腸粘膜肥満細胞浸潤によるメディエーター遊離亢進がTNBSによる大腸過敏症の発現に関与していることが示唆された。

ホルスタイン種子ウシ, 雌雄各4例の頚部心臓逸所症例を剖検し, 胸骨を軟X線学的に観察した。異常子ウシの生存期間は出生後3分から312日であった。心臓は頚部腹側で筋肉と皮膚に被われ, 心膜腔内に位置し, その二重心尖は前背方を, 心底は後腹方を向いていた。全例で, 大動脈弓からの主要動脈の分岐はイヌ型を示し, 多くの例で重複前大静脈と重複奇静脈も認められた。胸骨柄の幅は極めて広く, 胸郭前口も広かった。胸骨は前後に短く, 幅は広く, 13〜26個の胸骨片で構成され, 胸骨柄と胸骨体の骨片は対の様相を呈していた。胸腺の胸部は欠如し, 頚部は心臓の前背方に集合していた。線維性心膜から伸びる靭帯が, 前方では下顎骨と耳下腺筋膜に, 側方では頚筋膜に, また後方では第一肋骨あるいは胸骨柄に付着し, 心臓を保定していた。

A locally bred, 12-year-old, intact female Satsuma dog presented with generalized alopecia. Erythema, crusts and desquamation were observed primarily on the truck. Papules and erosions were present in the pinnae, and there were multiple areas of skin necrosis on the right forelimb. The cutaneous lesions had not responded to treatment with systemic antibiotics and prednisolone. The dog also had progressive anemia. Babesia gibsoni was detected in the blood, and the dog was treated with antiprotozoal agents. The skin lesions and anemia improved, but relapsed after the treatment was discontinued. Histopathological examination of skin biopsies revealed findings suggestive of early leukocytoclastic vasculitis or ischemic vasculopathy.

A 2-year-old male mixed dog was referred to us for further evaluation and treatment of a 4-week-history of oliguria and abdominal distension after a surgical repair of urethral injury. To relieve the urethral stricture, we placed a self-expanding aortic stent graft extension with a partial coverage with an expanded polytetrafluoroethylene (ePTFE). After the placement of the stent, the dog presented with a normal urinary voiding, despite the presence of urinary incontinence. The current case indicates that the ePTFE-covered, self-expanding ASGE is an effective intervention for the treatment of severe urethral stricture in the dog.

Two miniature dachshunds, a 7-year-old neutered male and an 8-year-old male, presented with chronic hematochezia and tenesmus. A solitary pedunculated or multiple diffuse colorectal polyps were identified by colonoscopy and resected by polypectomy. Inflammatory colorectal polyps (ICRPs) were diagnosed according to histopathological findings. Both cases were treated with immunosuppressive therapy, and the clinical signs were resolved, although the colorectal polyps remained to some extent. Several months after the initial diagnosis, both cases presented with recurrence of hematochezia and enlargement of the polyps. A second colonoscopic polypectomy was performed, and adenoma was diagnosed histopathologically in both cases. ICRPs are a nonneoplastic disease, but their long-term prognosis is unknown. Careful follow-up seems to be important, and repetitive biopsy is recommended when growth of polyps is identified in miniature dachshunds with ICRPs.

Fluoroquinolone resistance is mainly caused by mutations in quinolone resistance-determining regions of DNA gyrase and topoisomerase IV in Escherichia coli. The AcrAB-TolC efflux pump contributes to resistance against fluoroquinolone and other antimicrobials. In this study, we investigated a high-level mechanism of fluoroquinolone resistance in E. coli that was isolated from human clinical samples and canine fecal samples. E. coli strains with high levels of fluoroquinolone resistance have been found to be frequently resistant to cephalosporins. Strains with high-level fluoroquinolone resistance exhibited lower intracellular enrofloxacin (ENR) concentrations, higher expression of AcrA, and a greater reduction in the fluoroquinolone minimum inhibitory concentration for treatment with an efflux pump inhibitor. The frequency of strains with enhanced ENR resistance selection and the survival rate of E. coli in the presence of ENR in vitro were correlated well with AcrA protein expression levels in the parental strains. These results suggest that AcrAB-TolC efflux pump over-expression is related to high-level fluoroquinolone resistance and the selection of strains with enhanced fluoroquinolone resistance.

We established a homologous sandwich enzyme-linked immunosorbent assay (ELISA) to measure serum levels of canine ferritin. Our assay uses a rabbit anti-canine heart ferritin polyclonal antibody, and canine heart ferritin as a standard. Serum ferritin concentration in healthy dogs (n=163) was 789 ± 284 ng/ml (mean ± standard deviation), a value higher than reported previously. Confidence levels relating to repeatability, dilution and recovery for this method were high. Therefore, we believe our developed sandwich ELISA will be effective in evaluating serum levels of canine ferritin.

L-type amino acid transporter 1 (LAT1), an isoform of amino acid transport system L, transports branched or aromatic amino acids essential for fundamental cellular activities such as cellular growth, proliferation and maintenance. This amino acid transporter recently has received attention because of its preferential and up-regulated expression in a variety of human tumors, in contrast to its limited distribution and low-level expression in normal tissues. In this study, to explore the feasibility of using LAT1 expression as a molecular marker in mammary gland tumors (MGT), we performed a comparative study of LAT1 expression at the mRNA and protein levels in normal mammary gland cells and tumor cells. Conventional RT-PCR and Western blotting were performed on MGT tissues from 16 dogs and normal organs from nine healthy dogs. LAT1 expression was detected in ten of the 16 MGT patients. As is the case in human tissues, LAT1 showed limited expressional distribution in normal canine organs. For quantitative expressional comparison, extensive real-time RT-PCR was performed on mRNA samples from 53 MGT patients. The comparison demonstrated that LAT1 mRNA levels from MGT tissues were 20 times higher than those in normal mammary gland tissues. Additionally, histologically invasive MGT showed a higher expression of LAT1 than non-invasive tumors. These findings suggest that LAT1 could be a clinical marker and therapeutic target for invasive malignant MGT.

Canine serum amyloid A (SAA) is a useful diagnostic marker of systemic inflammation. A latex agglutination turbidimetric immunoassay (LAT) was validated for automated measurements. The aim of the study was to evaluate the clinical applicability of SAA measured by the LAT. SAA was measured in 7 groups of dogs with and without systemic inflammation (n=247). Overlap performance was investigated. Diagnostic performance was compared to body temperature and leukocyte markers. Clinical decision limits for SAA were estimated. In dogs with neurological, neoplastic or gastrointestinal disorders (n=143), it was investigated whether a higher proportion of SAA positive dogs could be detected in cases of complications with risk of systemic inflammation. Significantly higher concentrations of SAA were measured in dogs with (range [48.75; 5,032 mg/l]), compared to dogs without systemic inflammation [0; 56.4 mg/l]. SAA was a more sensitive and specific marker of systemic inflammation (area under the receiver-operating characteristic curve (AUC) 1.00), compared to body temperature (0.6) and segmented neutrophils (best performing leukocyte marker, 0.84). A clinical decision limit of 56.4 mg/l was established giving close to perfect discrimination between dogs with and without systemic inflammation. Higher proportions of SAA-positive dogs were observed in dogs with neurological, neoplastic and gastrointestinal disorders with complications known to increase risk of systemic inflammation, compared to uncomplicated cases. The automated LAT makes SAA applicable as a relevant diagnostic marker of systemic inflammation in dogs for routine random-access real-time use in a general clinical setting.

The purpose of the present study was to determine the prevalence of antibiotic-resistant enterococci in dogs and cats subjected to differing antibiotic pressures, and the prevalence of vancomycin resistance genes in isolates from these animals. Enterococci were isolated from fecal samples of 65 healthy dogs and 29 healthy cats brought to animal hospitals, from rectal swabs of 73 puppies and 15 kittens from five breeders and two pet shops, and from fecal samples of 20 dogs and 9 cats that were treated with antibiotics in Nippon Veterinary and Life Science University Animal Medical Center. The rates of resistance to ampicillin among isolates from the kitten-puppy group and healthy dog-cat group were 6.8 and 4.3%, respectively. In contrast, the rates of resistance to ampicillin in enterococci from the treatment group under antibiotic pressure were 37.5%. There was a significant difference between the antibiotic-treated group and the untreated group (P<0.01). Similarly, in the treatment group, the rate of resistance to enrofloxacin was extremely high (75.0%). In comparison, in the healthy group and kitten-puppy group, the rates of resistance to enrofloxacin were 23.4 and 12.1%, respectively. Among these groups, a significant difference was also observed in the apparent resistance rates (P<0.01). Vancomycin-resistant enterococci (VRE) harboring vanA or vanB were not detected in any groups. Therefore, contamination of VRE in dogs and cats is still considered to be minimal in Japan.

To verify availability of skin conductance (SC) as an indicator for the sympathetic nervous system (SNS) activity in dogs, the changes of SC and blood levels of stress-related hormones induced by drugs were compared. SC and cortisol, adrenaline and noradrenaline levels were measured in 5 dogs on 4 occasions with or without drug-induced sedation at 7 day-intervals (no treatment, intramuscular medetomidine 0.01 mg/kg, intramuscular acepromazine 0.1 mg/kg, and intravenous fentanyl 0.02 mg/kg). The fentanyl treatment produced significant higher levels of SC and plasma cortisol and adrenaline, compared with the other 3 treatments. The plasma noradrenaline level also tended to be higher following the fentanyl treatment. These results indicate that SC may reflect changes in the SNS activities in dogs.

To provide anatomical data on the nutrient foramen (NF), the authors observed the number, site and its index (SI), position, diameter and penetrating direction of the nutrient canal (NC) in the femur and tibia of German shepherd dogs. The femur often had more than two NF on the caudal surface with 30.0 to 86.7% of SI. The proximal NF of the femur was regarded to be the major NF, because it was present in most dogs and had the largest diameter. The tibia always had only one NF with 23.2 to 38.3% of SI on the lateral border. Their direction of the NC followed Bérard’s rule with the exception of the proximal NF of the femur. These results describe the anatomical condition of the NF and NC in the dog.
