研究の結果として今Review of International Studiesという英字雑誌に一つの論文(Enemies Within? : Alliances, 'Internal Threats' and Comprehensive Security in East Asia)の掲載が決まって修正作業中です。この論文は人間安保次元で同盟にともなう問題を扱っています。同盟の結果から生じる色々な軍隊に駐屯と基地を囲んだ問題が同盟にいかなる影響を及ぼすかを把握するのに目標を置いています。今までの研究が同盟は外部的威嚇に備えるために形成されると見ているが本論文は内部的威嚇という概念を導入してこのような新しい威嚇が全般的な同盟関係にどんな影響を及ぼすかを調査しました。その結果、冷静以後の同盟関係は外部的そして内部的威嚇の相関関係で同盟の水準が決定されるという結果を導き出しました。もう一つの論文(Humanitarian Power : An Identity in the Making in post-Cold War Japan and South Korea)を完成してJournal of Asian Studiesという英字雑誌に送りました。この論文は冷戦後日本と韓国の海外派兵の形態に対して研究しています。その結果日本と韓国は同盟の義務や国際的災難にともなう人道的支援のために非戦闘兵の派兵を推進してきました。このような一貫した行動は今後日本と韓国が人道的国家というアイデンティティを確立するという結果を導き出しました。本研究の関連研究でJapan Forumという英字雑誌に一つの論文(Constructing Fear : How the Japanese State Mediates Risks from North Korea)の掲載が決まって今印刷中です。この論文は日本国家が北朝鮮の色々な威嚇に対してどんな危機管理形態を見せるのかを研究しました。その結果日本政府は北朝鮮のミサイル発射、拉致、そして不審船の問題を処理しながら市民社会と特にマスコミの影響を受けて日本を普通国家で作る側に政策の方向を定めたという結果を導き出しました。この過程で威嚇を恐怖で作る現代社会の色々な行為者などに研究の焦点を合わせました。

Growth deficit (gd) is a recessive mutation that occurs spontaneously in the inbred NC/Sgn mouse strain. Because homozygotes (gd/gd) of both sexes are sterile, they must be produced by mating putative heterozygous carriers (+/gd) whose phenotypes are essentially the same as those of wild-type +/+ mice. The objectives of this study were to develop an efficient method that distinguished a gd allele from a wild-type allele and, if possible, to identify nucleotide substitutions responsible for the gd mutation. The location of the gd locus was estimated to be at 58.3 Mbp on chromosome 4, over which Musk is located. An A-to-G base substitution, which resulted in an M826V amino acid exchange, was identified within a tyrosine kinase domain of Musk. This base substitution disrupted a recognition site for NlaIII; this allowed for discriminating the gd allele from the wild-type allele using PCR-RFLP analysis. When 130 (C57BL/6J × NC/Sgn-gd) F2 mice were genotyped by PCR-RFLP analysis, all 32 growth-retarded F2 mice were judged to have the gd/gd genotype. Musk mutations are known to cause congenital myasthenia, which is accompanied by growth retardation, postnatal lethality, and development of a hunchback. These were the typical phenotypes of gd/gd mutants. Although we cannot rule out the possibility that the neighboring genes around the Musk locus are related to the gd phenotype, gd could possibly be classified as a mutant allele of Musk.

The taxonomy of the Trachysalambria curvirostris species group is clarified based on abundant material from East Asia and other localities in the Indo-West Pacific and the Mediterranean. The following six species, including one new species, are recognized : T. curvirostris (Stimpson, 1860), T. longipes (Paulson, 1875), T. aspera (Alcock, 1905), T. palaestinensis (Steinitz, 1932), T. albicoma (Hayashi and Toriyama, 1980), and T. nansei new species. For nomenclatural stability, a neotype of Trachysalambria curvirostris is designated. Species differences are discussed in detail and a key to these species is provided.

The purpose of this study is to facilitate the discovery of categories for packaging-design benefits and brand equity in order to further examine the impact relationship among categories in the two dimensions, as seen from the perspectives of packaging design experts. Finally, this article will develop research propositions that were used as a basis for formulating hypotheses in empirical studies. Research results showed that the dimension of packaging-design benefits included 13 categories that are aesthetic, cultural, symbolic, convenient, environmental, informative, visibility, protective, innovative, identifiable, systematic, memorable and displayable qualities. In the brand equity dimension, the literature review and coding analysis obtained 22 categories. In examining the categories relationship between packaging-design benefits and brand equity, this study found 68 category relationships between the two dimensions. Furthermore, analysis from the perspective of packaging design experts showed that the relationship between packaging-design benefits and brand recognition was the strongest and also the most important. Packaging design experts considered the influence of packaging-design benefits on brand awareness and brand association equally important. However, in terms of brand association, the relationship strength and importance of packaging-design benefits and non-product-based association was low compared to product-based association.

A 6-year-old ShihTzu presented with tonic-clonic cluster seizure. T2-weighted magnetic resonance (MR) images showed bilateral diffuse hyperintense lesions at the cerebral cortex with enlarged sulci. Computed tomography revealed a portosystemic shunt (PSS) and azygos continuation. Based on the clinical signs, blood examinations and diagnostic images, the dog was diagnosed with hepatic encephalopathy secondary to PSS. The neurologic signs were gradually improved after medical therapy for hyperammonemia. This is the first report of hyperintensity of the cerebral cortex on T2-weighted MR images associated with acute hepatic encephalopathy in a dog.

Vascular tumors and disorders, like angiomatosis, are rarely described in cetacean species. A retrospective histological study was carried out on lung samples from 35 common dolphins (Delphinus delphis) stranded in the Canary Islands coasts looking for morphological vascular changes and likely related causes. Twenty-five out of thirty-five (71%) common dolphins showed focal or multifocal angiomatosis-like lesions. A high association between this type of vascular proliferation and parasitic infestation was observed. In addition, a single pulmonary cavernous hemangioma not previously reported in common dolphins is presented.

Absorption spectroscopy has been applied to the measurement of the arc-heater plumes. Number density distributions of atomic metastable oxygen were obtained from the measured absorption line profile at 777.19 nm in argon-oxygen plumes generated by arcjet type arc-heaters. As a result, it was found that the oxygen is localized off axis at the nozzle exit, and diffuses from outside toward the centerline in the plume. Moreover, numerical simulation was conducted to investigate the diffusion process of the oxygen in the arc-heater. The result indicates that the maximum degree of oxygen dissociation would be at the level of 0.01%.

Haemophius parasuis is known to produce a group of virulence-associated autotransporter (AT) proteins, VtaAs; however, no other ATs have been characterized yet. On the basis of reported sequence of a putative espP2 gene for extracellular serine protease (ESP)-like protein of H. parasuis, this putative AT gene was successfully amplified from H. parasuis serotype 5 field strain HPS0819, cloned and sequenced. The confirmed ORF sequence showed 100% identity with the reported putative espP2 gene. The recombinant ESP-like protein purified from Escherichia coli with pET expression system was used for immunological characterization. An approximately 85 kDa antigen was detected in cultured H. parasuis by using antiserum to the purified ESP-like protein and antibodies against the recombinant ESP-like protein was detected in a selected serum from pigs with experimental H. parasuis infection. The results indicated that H. parasuis could produce ESP-like protein in vitro and in vivo. In an immune protection study using guinea pigs, 6 out of 10 animals immunized with the recombinant ESP-like protein survived after challenge with 5×109 bacteria of strain HPS0819 whereas 7 out of 10 animals immunized with formalin-inactivated H0819 bacterin survived after challenge. The results suggest that ESP-like protein could be one of vaccine antigen candidates for H. parasuis infection.

By a series of experiments, we identified a new member of locoweed family, Oxytropis serioopetala, which produces swainsonine, a phytotoxin harmful to livestock. In order to evaluate the toxicity of Oxytropis serioopetala, its extract was administered to ten rabbits by gavage at a dose of 1.5 mg/kg body weight as swainsonine once daily. After the 20th day, the rabbits appeared depressive and anorexia. Besides, intention tremors were apparent upon move. Their eyes were dull. The rear limbs were severely weak and even progressed to partial paresis. Activities of serum aspartate aminotransferase (AST), alanine transaminase (ALT), alkaline phosphatase (AKP) and urea nitrogen (BUN) levels in the poisoned rabbits increased significantly. Serum α-mannosidase (AMA) activity decreased markedly. Pathomorphological lesions in the locoweed-poisoned rabbits developed severe microvacuolation of visceral and neurological tissue. The extensive vacuolation was observed in liver, kidney and brain. These clinical and pathological features are similar to the symptoms of locoism.

Disease biomarker using blood is clinically important, since blood is easy to obtain from patients, thus it requires relatively less stress. However, blood generally reflects not only targeted diseases but also whole body status of patients. Thus, it is important which contents of blood are considered. Recently, miRNAs in blood, blood-borne miRNome, turns out to be promising candidates for blood based biomarker for diseases. In this paper, we propose a new method based upon principal component analysis to identify better candidates for miRNAs as blood based biomarker using miRNA expression profiles of patients. Our method based upon principal components analysis provides us better blood-borne miRNome to discriminate diseases from healthy controls. They are hsa-miR-425,hsa-miR-15b,hsa-miR-185, hsa-miR-92a, hsa-miR-140-3p, hsamiR-320a, hsa-miR-486-5p, hsa-miR-16, hsa-miR-191, hsa-miR-106b, hsa-miR-19b, and hsa-miR-30d and are previously extensively reported to be cancer/disease related miRNAs. We have found that these common miRNAs are expressive or suppressive significantly in most of diseases/cancers, but in diseases/cancers specific combinatory manner. It enables us to discriminate cancers/diseases from healthy control well.

MiRNAs are recently known to be critical players causing cell senescence, by regulating target genes. Thus inference of miRNAs critically regulating target genes is important. However, miRNAs critically regulating target genes are believed to have significant fold changes, typically upregulations, during cell senescence. In this study, we consider the target gene regulation by miRNAs together with miRNAs expression change during fibroblast IMR90 cell senescence. Then we found that the simultaneous consideration of two criterion lists more feasible miRNAs: i.e., miRNAs being more often reported to be down/upregulated and/or having biological backgrounds inducing cell senescence. Thus, the amount of target gene regulation, which can be inferred by the recently developed MiRaGE Server, is recommended to be considered together for the estimation of miRNAs critically contributing to cell senescence.

Proteomic analysis is very useful procedure to understand the bacterial behavior changes with reaction to the external environment. This is because most of genomic information of bacteria is devoted to code enzyme to control metabolic networks inside the individual cell. In this paper, we have performed proteomic analysis of Streptococcus pyogenes, which is known to be a flesh-eating bacteria and can cause several human life-threatening diseases. Its proteome during growth phase is measured for four time points under two different incubation conditions; with and without shaking. The purpose of it is to understand adaptivity to oxidative stress. Principal component analysis is applied and turns out to be useful to depict biologically important proteins for both supernatant and cell components.

Soil disease suppression is a worldwide important issue in order to realize stable food supply to people. In spite of that, no established indicators of soil disease suppression have been found out yet. This prevents us from controlling well soil state such that no diseases take place. In this paper, we have proposed a new biological indicator of soil disease suppression; the ability of bacteria to consume carbon resources, which can be automatically observed by Omunilog ID system during a duration of one or two days. This indicator turned out to distinguish disease suppressive soils from others. We have modeled these characteristic time developments of consumption of carbon resources by the simple ecological model where bacteria compete with each other for carbon resources. Measured ecological structure of soil bacteria can fit with the theoretical prediction well. In order to find characteristic features for each of soils, observed time developments are embedded into two dimensional space by non-metric multidimensional method. It results in the almost one dimensional arrangements of embedded points. By analyzing spacial distributions of each carbon resources in the embedded space, healthy soil turns out to have mostly uniform distribution along this one dimensional arrangement. Since sick soil and non-soil example have rather localized distributions, the ecological systems in more disease suppressive soil are both more diverse and more uniform. Since this indicator can be extremely easily and quickly obtained automatically, it is expected to use in order to validate many efforts to try to improve soil before any harvests very much.

本年度は、研究のコンセプトとデータ収集のための更なる知識、データ準備・分析に必要な理解を得るために、研究に関する更なる文献調査を行った。ガーナにおける複雑な土地利用変化の要因を理解するために、データ収集・分折の統合的なアプローチが必要であり、コミュニティの重要なステークホルダーとの協議、コミュニティとの交流、土地被覆変化の分析と炭素濃度測定のための地理情報システム(GIS)とリモートセンシングの使用を行った。また、研究成果報告として国連大学サステイナビリティ平和研究所の学術会議で最終発表を行った。ガーナへの現地調査は、主導・協働の共同研究の構築、生態域からのデータ収集、土地利用調査の遂行、土地被覆変化の原因調査、異なる土地被覆の炭第濃度のマッピングを目的とした。変化する景観と異なる土地被覆タイプにおける陸地の炭素濃度を測定するため、1990年と2000年にガーナの衛星画像(LANDSAT TM)から得られた土地被覆マップが使用された。研究結果としては、1990年から2000年のガーナにおいて、異なる土地被覆タイプに関する顕著な増加と減少、つまり農耕地、乾燥疎開林、森林農業・市街地に著しい増加が見られた。ガーナの林地はかなりの勢いで減少を続け、さらに林地の多くの地域が農業活動や、その他の土地被覆タイプに変化しており、多くの混地帯と水塊は上述した調査期間の間に干上がっていた。土地被覆タイブの変化はガーナを隔てる各地域における穀物生産、生息分布、土壌炭素濃度、生活形態に多大な影響を与えている。土地利用強度は、居住環境の豊かさ、不均一性、分断化、および形状複雑性の増加に起因している。部分的な土地利用変化とその他の要因による地域の食品保障格差はガーナの持続可能な開発にとって依然電要な問題である。基本的には6つの土地被覆タイプがガーナの炭棄貯藏の99パーセントを担っており、炭素の増減が総針貯蔵量に多大な影響を与えているのである。

Platelet-rich plasma (PRP) was administered to cattle with hoof defects (10-mm drill hole) to promote sole defect healing. Two types of gels were prepared. One was a mixed gel [compound of gelatin microspheres (GM) incorporated with PRP and alginate] and was used for the test group, while the other was an alginate gel used for the control group. Each hole was filled with one of these gels. The drill hole depth, pressure pain, and hardness of the regenerated tissue were measured for 3 weeks after treatment. These measurements improved significantly in the test group than in the control group. The results suggest that a compound of GM incorporated with PRP and alginate had a hoof-regenerating effect in cattle.

Porphyromonas gulae, a gram-negative black-pigmented anaerobe, is a pathogen for periodontitis in dogs. An approximately 41-kDa fimbrial subunit protein (FimA) encoded by fimA is regarded as associated with periodontitis. In the present study, the fimA genes of 17 P. gulae strains were sequenced, and classified into two major types. The generation of phylogenetic trees based on the deduced amino acid sequence of FimA of P. gulae strains along with sequences from several strains of Porphyromonas gingivalis, a major cause of human periodontitis, revealed that the two types of FimA (types A and B) of P. gulae were similar to type I FimA and types II and III FimA of P. gingivalis, respectively. A PCR system for classification was established based on differences in the nucleotide sequences of the fimA genes. Analysis of 115 P. gulae-positive oral swab specimens from dogs revealed that 42.6, 22.6, and 26.1% of them contained type A, type B, and both type A and B fimA genes, respectively. Experiments with a mouse abscess model demonstrated that the strains with type B fimA caused significantly greater systemic inflammation than those with type A. These results suggest that the FimA proteins of P. gulae are diverse with two major types and that strains with type B fimA could be more virulent.

To evaluate the relationship among immune status and increased morbidity and mortality, peripheral blood lymphocytes (CD3+, CD4+, CD8+, CD21+ cells) from 32 healthy dogs aged over 8 years were analyzed. Twenty-five dogs out of 32 were followed-up for 3 years after the analysis, and 14 dogs were found to be diseased and nine dogs died. There was no notable difference between the age of dogs that died compared with the ones that survived. The relative percentage of CD4+ and CD4+:CD8+ ratio decreased notably in dogs falling ill compared with healthy dogs. The relative percentage of CD3+ lymphocytes showed a notable decrease in dogs that died within 3 years in comparison with dogs that survived. In a discriminant analysis of morbidity and mortality, most patients were correctly classified as diseased or not and surving or dead, respectively. These results indicate that the immunophenotypes of peripheral blood lymphocytes in older dogs offer promise as parameters for evaluating mortality and morbidity.

研究実績(1)申請者は本研究の期間中に行った約300人のアンケート調査と約80人に対するインテンシヴな面接調査を完了し、現在テキスト分析を中心とした研究報告を執筆中であり、既に4本の論文を研究成果として発表し、さらに4本を執筆中である。この執筆中のものには岩波新書が含まれる。以上の調査研究から明らかとなったことは、次の通りである。日本における韓流ブームは、中年女性たちによるある種のサイバー文化反乱と位置づけることができる。彼女たちは、家庭や職場という既存の生活枠を飛び出し、広範な社会的ネットワークを形成しつつ互に交流することで、起伏に乏しい平凡な日常を精神的・物的にも超えようとしている。この越境的文化行動は、高度大衆消費社会の重圧のもと、グローバライゼーションとローカライゼーションの同時多発的な進行とによって変化していく政治的・社会的・文化的環境条件に適応するための、自己救済的な努力といえよう。彼女たちの日常から喚起されるある種の喪失感、あるいは願望に伴う"ヒステリー"を癒す遊びとしての韓流は、自己実現・自己回復に向けた意志的な取り組みであり、アイデンティティの表現なのである。彼女たちは、「見えない市民」としてプライベートな空間に留まることを拒み、自分の存在と欲求を表出するために文化的市民権を求めていると解釈できよう。彼女たちにとって韓流文化の消費は、現実が満たせずにいる自己の欲求を充足させ、充実した生のあり方を示してくれるものである。それはまた独自の政治的インプリケーションをもつものであった。(2)学会活動としては、2005、2006年度の「アジア学」学会(於米)で、研究の中間的成果を発表するとともに、2006年の7月には「インターアジア・カルチュラル・スターディズ」学会(於東京)および立命館大学、東京大学、同志社大学で開催されたシンポジュームで各々報告した。また立教大学では「誰のための『韓流』か」と題した講演を行った。
