Fish production is increasingly important to global food security. A major factor in maintaining health, productivity and welfare of farmed fish is the establishment and promotion of a stable and beneficial intestinal microbiota. Understanding the effects of factors such as host and environment on gut microbial community structure is essential for developing strategies for stimulating the establishment of a health-promoting gut-microbiota. We compared intestinal microbiota of common carp and rainbow trout, two fish with different dietary habits, sourced from various farm locations. There were distinct differences in the gut microbiota of carp and trout intestine. The microbiota of carp was dominated by Fusobacteriia and Gammaproteobacteria, while the trout microbiota consisted predominantly of Mollicutes and Betaproteobacteria. The majority of bacterial sequences clustered into a relatively low number of operational taxonomic units (OTUs) revealing a comparatively simple microbiota, with Cetobacterium, Aeromonas and Mycoplasma being highly abundant. Within each species, fish from different facilities were found to have markedly similar predominant bacterial populations despite distinctly different rearing environments, demonstrating intra-species uniformity and significant influence of host selectivity. This study demonstrates that in fish the host species imparts substantial impact in shaping the community structure of the intestinal microbiota.

Given their applicability in genetic engineering, undomesticated Bacillus strains are extensively used as non-natural hosts for chemical production due to their high tolerance of toxic substrates or products. However, they are difficult to genomically modify due to their low transformation efficiencies. In this study, the Bacillus-E. coli shuttle vector pHY300PLK, which is widely used in gram-positive bacteria, was adopted for genome integration in organic solvent-tolerant Bacillus isolates. The Bacillus-replicative vector was used to deliver homologous recombinant DNA and propagate itself inside the host cell, increasing the likelihood of genome integration of the recombinant DNA. Then, the unintegrated vectors were cured by cell cultivation in antibiotic-free medium with facilitation of nickel ions. The developed protocol was successfully demonstrated and validated by the disruption of amyE gene in B. subtilis 168. With an improved clonal selection protocol, the probability of clonal selection of the amyE::cat genome-integrated mutants was increased up to 42.0 ± 10.2%. Genome integration in undomesticated, organic solvent tolerant Bacillus strains was also successfully demonstrated with amyE as well as proB gene creating the gene-disrupted mutants with the corresponding phenotype and genotype. Not only was this technique effectively applied to several strains of undomesticated B. subtilis, but it was also successfully applied to B. cereus. This study validates the possibility of the application of Bacillus-replicative vector as well as the developed protocol in a variety of genome modification of undomesticated Bacillus species.

ガラガラヘビ<i>Crotalus durissus terrificus</i>の子宮にみられる回旋と筋肉収縮を,冬を越えて精子を貯蔵する(長期精子貯蔵)ための戦略として報告した.交尾期(秋季),濾胞の大きさ(5-30mm),排卵の時期(春季),妊娠期間(4-5ヵ月)など,雌の繁殖周期について述べた.雌の子宮には,交尾後すぐに形態学的,生理学的変化が観察された.この状態は1季節(冬季)続くだけで,排卵後に子宮に弛緩し,精子が卵管をさかのぼる.生殖管の解剖形態を記述し,温帯産の近縁種プレーリーガラガラヘビとの関連で,語句の用法について論議した.交尾期にはまだ受精の準備ができていない卵黄形成後の濾胞を,無駄にしないような生理学的過程の重要性を仮定すると,この種では繁殖に関する出来事が時間的に同調しないため,長期精子貯蔵をせざるを得ないと考えられる.

The purpose of this research is to investigate the meaning and effects of imitation in preschoolers' body expression. In our previous researches, we considered how preschool children create stories in a body expression play. Through these studies, it had been shown that imitation helps the development of the ability to create original expressions. We analyzed the cases and classified the patterns of imitation. Imitation of preschoolers during body expression could be divided in four patterns. (1) A child is seeking the chance and timing to begin moving. (2) A child enjoys mimicking some movement and, through it, communicating with other children. (3) A child is self-conscious of his movements and image. (4) A child is taking in an image or an idea of a movement new to him. It seems appropriate to not judge one of the four patterns as superior, but to understand that all four serve as a starting point for creative expression.

We report on the characteristics of 9 bb^^- pair events produced by a 600 GeV/c π^- beam and detected in the hybrid emulsion spectrometer of Fermilab experiment E653. The measured lifetimes for samples of 12 neutral and 6 charged beauty hadrons are τ_<b^0>=0.81^<+0.34+0.08'>_<-0.22-0.02> ps, and τ_<b^±>=3.84^<+2.73+0.80>_<-1.36-0.16> ps.

台湾の養殖ウシエビ(P.monodon)の稚エビにおけるMBV保有率は1984~86年の15~17%から1988年には80~85%に増加したが, 親エビは約50%であった。一方, P.penicillatusでは20~25%であった。MBVの実験感染によってウシエビの稚エビに死亡がみられた。感染したウシエビとP.penicillatusは発育ステージにかかわらず, ストレス下では生存率が有意に低下した。MBVに感染したウシエビの親は細菌の二次的感染によって斃死するように思われた

The rapid development of high-throughput techniques and expansion of bacterial databases have accelerated efforts to bring plant microbiomes into cultivation. We introduced plant-only-based culture media as a successful candidate to mimic the nutritional matrices of plant roots. We herein employed a G3 PhyloChip microarray to meticulously characterize the culture-dependent and -independent bacterial communities of the maize root compartments, the endo- and ecto-rhizospheres. An emphasis was placed on the preference of the growth of unculturable candidate divisions/phyla on plant-only-based culture media over standard culture media (nutrient agar). A total of 1,818 different operational taxonomic units (OTUs) were resolved representing 67 bacterial phyla. Plant-only-based culture media displayed particular affinity towards recovering endophytic over ectophytic rhizobacteria. This was shown by the slightly higher recovery of CFUs for endophytes on plant-only-based culture media (26%) than on standard culture media (10%) as well as the higher taxa richness and numbers of exclusive families of unculturable divisions/phyla. Out of 30 bacterial phyla (comprising >95% of the whole population), 13 were of a significantly higher incidence on plant-only-based culture media, 6 phyla of which were not-yet-cultured (Atribacteria, OP9; Dependentiae, TM6; Latescibacteria, WS3; Marinimicrobia, SAR406; Omnitrophica, OP3; BRC1). Furthermore, plant-only-based culture media significantly enriched less abundant and/or hard-to-culture bacterial phyla (Acidobacteria, Gemmatimonadetes, and Tenericutes). These results present conclusive evidence of the ability of plant-only-based culture media to bring the plant-fed in situ microbiome into the status of plant-fed in vitro cultures, and to widen the scope of cultivation of heretofore-unculturable bacterial divisions/phyla.

Malanjkhand copper project, Madhya Pradesh, India, generates a lot of lean sulphidic ores containing chalcopyrite (∼0.3% of copper) with traces of oxidized, semi-oxidized and secondary sulfides. EPMA analysis shows that the granitic rock samples have plagioclase feldspar, orthoclase feldspar and quartz as major constituents within which the ore minerals are sparsely distributed. Due to the complex structure and low solubility of the sulphide-bearing granitic rock, the dissolution of copper from the hard matrix is difficult. The present study deals with change in crystallographic and chemical nature of the mineral system present in the host rock. In an attempt to increase copper recovery, the ore was subjected to heating at 600°C under normal atmospheric conditions. This heat treatment resulted in enhanced copper recovery, up to 98% within 60 days of progressive leaching.
