Fcα/μR (CD351) is an Fc receptor for both IgA and IgM and forms an atypical dimer that is resistant to reduction by 2-mercaptoethanol or boiling. We previously demonstrated that the cytoplasmic portion of Fcα/μR is required for dimer formation and for its efficient cell-surface expression. However, the biochemical nature of these phenomena has not been determined. By using a BW5147 mouse cell line expressing deletion mutants of the cytoplasmic region of Fcα/μR, we found that the region spanning amino acids 504–523 was required for efficient cell-surface expression, whereas the region spanning amino acids 481–490 was required for dimmer formation. Immunoblotting analyses of transfectants simultaneously expressing Flag-tagged Fcα/μR and hemagglutinin-tagged Fcα/μR suggested that Fcα/μR does not form homodimers. Instead, our data suggest that Fcα/μR forms heterodimers with an as-yet-unknown molecule with a molecular weight of 60–70 kDa.

An l-glucose-utilizing bacterium, Paracoccus sp. 43P, was isolated from soil by enrichment cultivation in a minimal medium containing l-glucose as the sole carbon source. In cell-free extracts from this bacterium, NAD+-dependent l-glucose dehydrogenase was detected as having sole activity toward l-glucose. This enzyme, LgdA, was purified, and the lgdA gene was found to be located in a cluster of putative inositol catabolic genes. LgdA showed similar dehydrogenase activity toward scyllo- and myo-inositols. l-Gluconate dehydrogenase activity was also detected in cell-free extracts, which represents the reaction product of LgdA activity toward l-glucose. Enzyme purification and gene cloning revealed that the corresponding gene resides in a nine-gene cluster, the lgn cluster, which may participate in aldonate incorporation and assimilation. Kinetic and reaction product analysis of each gene product in the cluster indicated that they sequentially metabolize l-gluconate to glycolytic intermediates, d-glyceraldehyde-3-phosphate, and pyruvate through reactions of C-5 epimerization by dehydrogenase/reductase, dehydration, phosphorylation, and aldolase reaction, using a pathway similar to l-galactonate catabolism in Escherichia coli. Gene disruption studies indicated that the identified genes are responsible for l-glucose catabolism.

Mutations and polymorphisms of factor H gene (FH1) are known to be closely involved in the developmentof atypical hemolytic uremic syndrome (aHUS). Several groups have identified disease risk mutations andpolymorphisms of FH1 for the development of aHUS, and have investigated frequencies of aHUS in a numberof ethnic groups. However, such studies on Japanese populations are limited. In the present study,we analyzed FH1 in Japanese aHUS patients and healthy volunteers, and examined whether those variantsimpacted on a tendency for the development of aHUS in Japanese populations. Similar to previousstudies, we found that a high frequency of FH1 mutations, located in exon 23 of FH1, encodes short consensusrepeat 20 in C-terminal end of factor H molecule in patients with aHUS (40%), but not in healthyvolunteers. Interestingly, no significant differences in frequency of well-known disease risk polymorphismsfor aHUS were observed between healthy volunteers and aHUS patients. Our results suggestedthat although FH1 mutations relates to the development of Japanese aHUS in accordance with other ethnicstudies, other factor may be required for factor H polymorphism to be a risk factor of Japanese aHUS.

Neutrophils play a central role in host defenses against infectious microbial pathogens. However, molecular basis for their triggering signals has been incompletely understood. Here, we show molecular and functional characteristics of myeloid-associated immunoglobulin-like receptor (MAIR)-IV. MAIR-IV was preferentially expressed on neutrophils in the peripheral blood, bone marrow, peritoneal cavity and spleen. MAIR-IV physically associates with immunoreceptor tyrosine based activating motif (ITAM)-bearing adaptor FcRγ chain. Cross-linking MAIR-IV with anti-MAIR-IV monoclonal antibody significantly induced secretion of proinflammatory cytokines TNF-α and IL-6 from neutrophils. Thus, MAIR-IV may regulate activation of neutrophils and play an important role for innate immunity.

Fcα/μR is expressed not only in lymphoid, but also in non-lymphoid organs, including kidney. However, molecular and functional characteristics of Fcα/μR, particularly expressed in non-lymphoid organs, have remained unclear. Here we identified an isoform of Fcα/μR in the murine kidney on the C57BL/6J background. The kidney expressed only the isoform, which was not expressed in other lymphoid and non-lymphoid organs, and this isoform binds both IgA and IgM. Immunohistochemical analyses suggested that the Fcα/μR isoform was expressed in the tubular epithelial cells, but not in the glomeruli. This was confirmed by flow cytometry analysis of isolated tubular epithelial cells and by RT-PCR analyses using the separately excised glomerular and tubular regions by the laser microdissection system. These results suggest that Fcα/μR may not be involved in IgA deposition in glomerular mesangium cells in IgA nephropathy. Rather, it may play an important role in immunity in the renal tubular regions.

Datasets of structural damage and regional damage due to past earthquakes are not only useful to know the actual state of each event but also effective for strong ground motion predictions and damage mitigations of future earthquakes. In this paper, several considerations are illustrated concerning strong ground motion research and data processing analysis. Especially, the activity to make research results contribute to society is discussed based on three examples of earthquake damage data analysis. The examples shown here describe that the well known notion in general, what is called common knowledge, does not necessarily have high reliability, and that the lack of confirmation of the data accuracy misleads the conclusion of research results beyond common sense. From these examples, the importance of an empirical research policy as well as a reasonable research policy is discussed. And it is pointed out that the information exchange among the researchers in various fields will be very effective for the development of the strong ground motion research.

This paper examines the current educational effort, adopting the Balanced Scorecard (BSC). The BSC is a goal-setting and accountability tool that all schools (and eventually district departments) in the San Francisco Unified School District build to describe the efforts of each individual school site in meeting the goals and objectives of their respective district's Strategic Plan. It ensures that schools are focusing on key strategic actions to improve student achievements for all students and to disrupt the historic power of demographics. Mainly, student test scores now serve as the prevailing achievement. The BSC offers schools and the community at large the opportunity to design alternative measures. The importance of engaging the school community and all stakeholders in discussions to gain a shared understanding has been stressed. However, many difficulties confront the adoption of this new approach, which includes ensuring consistency, selection of evaluation methods, increased burdens on educators, and personnel allocation. It is contemplated that with more measures to assess performance, greater levels of accountability and school performance can also be achieved. The implementation of BSC in the San Francisco Unified School District began in 2009, and it deserves continued attention.

This paper proposes a semi-autonomous collision avoidance system for the prevention of collisions between vehicles and pedestrians and objects on a road. The system is designed to be compatible with the human-centered automation principle, i.e., the decision to perform a maneuver to avoid a collision is made by the driver. However, the system is partly autonomous in that it turns the steering wheel independently when the driver only applies the brake, indicating his or her intent to avoid the obstacle. With a medium-fidelity driving simulator, we conducted an experiment to investigate the effectiveness of this system for improving safety in emergency situations, as well as its acceptance by drivers. The results indicate that the system effectively improves safety in emergency situations, and the semi-autonomous characteristic of the system was found to be acceptable to drivers.
