Recent studies have revealed that decline in cellular nicotinamide adenine dinucleotide (NAD+) levels causes aging-related disorders and therapeutic approaches increasing cellular NAD+ prevent these disorders in animal models. The administration of nicotinamide mononucleotide (NMN) has been shown to mitigate aging-related dysfunctions. However, the safety of NMN in humans have remained unclear. We, therefore, conducted a clinical trial to investigate the safety of single NMN administration in 10 healthy men. A single-arm non-randomized intervention was conducted by single oral administration of 100, 250, and 500 mg NMN. Clinical findings and parameters, and the pharmacokinetics of NMN metabolites were investigated for 5 h after each intervention. Ophthalmic examination and sleep quality assessment were also conducted before and after the intervention. The single oral administrations of NMN did not cause any significant clinical symptoms or changes in heart rate, blood pressure, oxygen saturation, and body temperature. Laboratory analysis results did not show significant changes, except for increases in serum bilirubin levels and decreases in serum creatinine, chloride, and blood glucose levels within the normal ranges, independent of the dose of NMN. Results of ophthalmic examination and sleep quality score showed no differences before and after the intervention. Plasma concentrations of N-methyl-2-pyridone-5-carboxamide and N-methyl-4-pyridone-5-carboxamide were significantly increased dose-dependently by NMN administration. The single oral administration of NMN was safe and effectively metabolized in healthy men without causing any significant deleterious effects. Thus, the oral administration of NMN was found to be feasible, implicating a potential therapeutic strategy to mitigate aging-related disorders in humans.

The Paracalanus parvus (Claus, 1863) species complex is a common marine calanoid copepod found in the world’s oceans. Recent genetic analyses of its specimens from the world oceans revealed that the complex in the western North Pacific consists of three species, i.e., P. indicus Wolfenden, 1905, P. tropicus Andronov, 1977 and an undescribed species. We assign the last one to be Paracalanus orientalis n. sp. by comparing it with previous morphological descriptions of related species. Both sexes of the new species are fully described based on genetically identified specimens from the surface layer of the south of Japan. The new species can be reliably distinguished from P. indicus and P. tropicus by the hunchback shape of the female if it is clearly present. Among the three species, the high length:width ratio of the third exopod segment of leg 4 is also characteristic of the new species, which would be identifiable, in most cases, by having a ratio of 5.4 or more.

A new species of sapphirinid copepod, Sapphirina doliolettae n. sp. was described from the Kuroshio Extension region in the western North Pacific Ocean. The new species is similar to S. nigromaculata and S. scarlata with respect to the following characteristics: 5-segmented antennule, one inner marginal process on caudal rami, and slender endopods with two apical spines on leg 4. However, it can be distinguished from these congeners by a combination of the following characteristics: the relative length of each segment of the antennule and antenna in both sexes, the width of the fourth pedigerous somite in females, the shape of the anterolateral corner of the genital somite in males and terminal process length of 3rd endopodal segment of leg 2 in males. The mitochondrial cytochrome oxidase subunit I sequences of S. doliolettae showed high interspecific variabilities from other Sapphirina species, including S. nigromaculata (20.2%) and S. scarlata (21.6–21.8%).

High-intensity interval training (HIIT) has recently received much attention as a new option for aerobic training. Despite its smaller time requirement, HIIT has been reported to have a greater effect than continuous moderate-intensity training on fat loss, especially a decrease in truncal adiposity. We therefore examined whether long-term HIIT preferentially modulates truncal adiposity rather than peripheral adiposity, especially thigh adiposity, where local muscle energy consumption increased profoundly during HIIT. We also examined the association between changes in adipose tissue distribution and serum adiponectin level. Twelve healthy male participants (28-48 years old) were assigned to a group that performed HIIT using only a leg ergometer (L-HIIT, n = 7) or to a group that performed HIIT using both leg and arm ergometers (LA-HIIT, n = 5) twice weekly for 16 weeks. The training programs consisted of 8 to 12 sets of >90% VO2 peak for 1 min, with 1 min of very light active recovery. Body composition analyses as well as aerobic fitness and measurements of serum adiponectin were performed at baseline and after intervention. A linear improvement in aerobic fitness was observed along with a decrease in leg fat (5.4 ± 1.7 vs. 5.1 ± 1.7 kg, p < 0.05) near the main working muscles during HIIT in the combined (L+LA-HIIT) group. Moreover, there was an association of decrease in leg fat or thigh adiposity with improvement in aerobic fitness in the combined group (ρ = -0.59, p < 0.05; and ρ = -0.71, p < 0.05, respectively). Visceral adiposity was decreased in L-HIIT (115 ± 45 vs. 100 ± 47 cm2, p < 0.05), however no decrease was observed in total fat or truncal fat in either group. No change was observed in serum adiponectin concentration in either group. Changes in serum adiponectin were associated with changes in visceral adiposity in the combined group (ρ = -0.72, p < 0.01). Regional rather than whole-body fat loss was observed after a 16-week HIIT program.

A commutability confirmation test for the blood aldosterone measurement was performed on liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) as a designated comparison method (DCM) and four chemiluminescent enzyme immunoassay (CLEIA) measurement procedures based on metrological traceability. A conventional radioimmunoassay (RIA) and two measurement procedures of CLEIA which obtains RIA equivalent values were also compared. The relationship between the DCM value and the CLEIA value with respect to 120 pg/mL of the RIA value, which is the screening criterion of primary aldosteronism (PA) was clarified. For the correlation test, 75 samples of patient serum and plasma were used. Regression analysis revealed that the standardized LC-MS/MS and four CLEIA measurement procedures were in good agreement. This is the effect of measurement specificity and calibration using by certified reference material (CRM). The median of the LC-MS/MS corresponding to 120 pg/mL of RIA was 48.5 pg/mL. In the mean of standardized four CLEIA values corresponding to the 48.5 pg/mL of LC-MS/MS value was 47.51 pg/mL and the standard deviation (SD) was 2.93 pg/mL. However, the correlation between the RIA value and the RIA equivalent of the two measurement procedures by CLEIA differed depending on the measurement procedure. This is due to the influence of RIA measurement performance. Standardized CLEIA measurements are suitable for routine measurement procedure. When converting the LC-MS/MS equivalent value by the standardized CLEIA to the conventional RIA value, it is necessary to use the conversion formula.

A 25-month field survey was conducted to investigate the life cycle and seasonal population fluctuations in the poecilostomatoid copepod Hemicyclops spinulosus in the burrows of the ocypodid crab Macrophthalmus japonicus in the mud-flats of the Tama-River estuary, central Japan. On the basis of sample collections in the water column and from the crab burrows, it was confirmed that H. spinulosus is planktonic during the naupliar stages and settles on the bottom during the first copepodid stage to inhabit the burrows. Furthermore, the copepods' reproduction took place mainly during early summer to autumn with a successive decrease from autumn to winter. A supplementary observation on the burrows of the polychaete Tylorrhynchus heterochaetus suggested that these burrows are another important habitat of H. spinulosus. There were additional discoveries of male polymorphism and precopulatory mate guarding behavior by males, suggesting an adaptation in the reproductive strategy of this copepod to their narrow habitat spaces and low population densities, in contrast to the congeneric species H. gomsoensis, which co-occurs in the estuary but attains much larger population sizes and is associated with hosts having much larger burrow spaces.

Type 2 diabetes is a progressive disease and most patients with type 2 diabetes eventually need insulin therapy. The objective of this study was to clarify C-peptide immunoreactivity (CPR), a marker of beta cell function, as a predictor of requirement for insulin therapy. We conducted a retrospective study of 579 consecutive subjects with type 2 diabetes who were admitted to our hospital from 2000 to 2007 and were able to be followed up for at least 6 months after discharge. Fasting and postprandial serum CPR and urinary CPR levels had been measured during admission. Information about insulin therapy at the last visit was obtained from medical records. At the last visit, 364 subjects (62.9%) were treated with insulin. Mean interval between discharge and the last visit was 4.5 ± 2.3 years. A 9-year-old, spayed female Chihuahua was presented with a 1-week history of lethargy and anorexia. Abdominal ultrasonography and computed tomography found bilateral adrenal masses without metastasis. Serum cortisol levels that were sampled before and after an adrenocorticotropic hormone stimulation test were within reference ranges. Lethargy and anorexia completely resolved after short-term fluid therapy; the clinical signs did not occur for approximately 8 months until her sudden death. A postmortem examination revealed bilateral adrenocortical carcinomas and liver metastasis. Primary adrenocortical carcinomas developed in the dog met the definition of bilateral incidental adrenal gland masses (IAGMs). This is the first case report to demonstrate based on histological identification that adrenocortical carcinomas cause bilateral IAGMs in dogs.

雄ホロホロ鳥における繁殖能力の,年間を通じての推移を明らかにする目的で,交尾器の大きさ,精液量,精子濃度および血漿テストステロン濃度を1年間測定した。1. 交尾器の大きさは9月初旬まで徐々に増大し,換羽期において一度縮小したが,その後再び増大して12月下旬以降は成熟した大きさを維持した。また,交尾器の大きさと日長,気温および血漿テストステロン濃度とのいずれとも,有意な相関関係は認められなかった。2. 精液量および精子濃度は4月より上昇し,5月下旬から10月上旬にかけて高い値を維持した(精液量:9~38μl,精子濃度:6.51~29.9億/ml)。その後,換羽期と1月から2月にかけての期間において低い値を示し118 日本家禽学会誌22巻3号(1985)た。3. 血漿テストステロン濃度の推移は,日長の変化とよく対応したものであり,最高値(2.95±0.81ng/ml)および最低値(0.12±0.02ng/ml)は,それぞれ7月上旬と9月下旬の換羽初期に認められた。また,繁殖期,非繁殖期および換羽期における血漿テストステロン濃度の平均値は,それぞれ1.25±0.12ng/ml, 0.64±0.05ng/mlおよび0.20±0.05ng/mlであり,各値間には有意(P<0.01)差が認められた。4.日長,気温および血漿テストステロン濃度と各測定項目間の相関係数は表1に示した。

Chronic kidney disease (CKD) impairs physical performance in humans, which leads to a risk of all-cause mortality. In our previous study, we demonstrated that a reduction in muscle mitochondria rather than muscle mass was a major cause of physical decline in 5/6 nephrectomized CKD model mice. Because ghrelin administration has been reported to enhance oxygen utilization in skeletal muscle, we examined the usefulness of ghrelin for a recovery of physical decline in 5/6 nephrectomized C57Bl/6 mice, focusing on the epigenetic modification of peroxisome proliferator activated receptor gamma coactivator-1α (PGC-1α), a master regulator of mitochondrial biogenesis. The mice were intraperitoneally administered acylated ghrelin (0.1 nmol/gBW; three times per week) for a month. Muscle strength and exercise endurance were measured by using a dynamometer and treadmill, respectively. Mitochondrial DNA copy number was determined by quantitative PCR. The methylation levels of the cytosine residue at 260 base pairs upstream of the translation initiation point (C-260) of PGC-1α, which has been demonstrated to decrease the expression, was evaluated by methylation-specific PCR and bisulfite genomic sequencing methods after the ghrelin administration. Ghrelin administration improved both muscle strength and exercise endurance in the mice and was associated with an increase in muscle mass and muscle mitochondrial content. Ghrelin administration decreased the methylation ratio of C-260 of PGC-1α in the skeletal muscle and increased the expression. Therefore, ghrelin administration effectively reduced the physical decline in 5/6 nephrectomized mice and was accompanied with an increased mitochondrial content through de-methylation of the promoter region of PGC-1α in the muscle.

It remains unclear whether epithelial cell-derived cytokines, including interleukin (IL)-25, IL-33 and thymic stromal lymphopoietin (TSLP), contribute to development of canine chronic enteropathy (CE), which includes antibiotic-responsive enteropathy (ARE), food-responsive enteropathy (FRE) and inflammatory bowel disease (IBD). In the present study, we examined mRNA expression of il-25, il-33 and tslp in the duodenal and colonic mucosae of dogs with ARE, FRE and IBD. Real-time PCR analysis revealed that mRNA expression of il-33 was significantly lower in the duodenum in dogs with FRE than in healthy dogs. The results suggest that epithelial cell-derived cytokines may not be an inducer of Th2-type immunity in the gut of dogs with CE, and decreased expression of IL-33 may be involved in induction of FRE. Further studies are required to clarify roles of epithelial cell-derived cytokines, especially IL-33, in the pathogenesis of canine CE.

GPR56 is a member of the adhesion G protein-coupled receptor (GPCR) and is highly expressed in parts of tumor cells. The involvement of GPR56 in tumorigenesis has been reported. We generated agonistic monoclonal antibodies against human GPR56 and analyzed the action and signaling pathway of GPR56. The antibodies inhibited cell migration through the Gq and Rho pathway in human glioma U87-MG cells. Co-immunoprecipitation analysis indicated that the interaction between the GPR56 extracellular domain and transmembrane domain was potentiated by agonistic antibodies. These results demonstrated that functional antibodies are invaluable tools for GPCR research and should open a new avenue for therapeutic treatment of tumors.

The aim of this study was to clarify the relationship between baseline beta cell function and future glycated albumin (GA) to glycated hemoglobin ratio (GA/HbA1c) in patients with type 2 diabetes. In our retrospective cohort, 210 type 2 diabetic patients who had been admitted to our hospital and in whom HbA1c and GA had been measured at baseline and 2 years after admission were included in this study. Baseline beta cell function was assessed by postprandial C-peptide immunoreactivity index (PCPRI) during admission. With intensification of treatment during admission, HbA1c and GA were significantly decreased 1 year and 2 years after admission. While baseline HbA1c was not significantly correlated with HbA1c after 2 years, baseline GA/HbA1c was strongly correlated with GA/HbA1c after 2 years (r = 0.575, P <0.001). When the patients were divided into two groups according to median PCPRI, patients with low PCPRI showed higher GA/HbA1c both at baseline and after 2 years compared to those with high PCPRI. There was a significant negative correlation between PCPRI and GA/HbA1c after 2 years (r = -0.379, P <0.001). Multiple regression analysis revealed that PCPRI was an independent predictor of GA/HbA1c after 2 years. In conclusion, our findings suggest that lower beta cell function is associated with sustained higher GA/HbA1c ratio in patients with type 2 diabetes.
