In the present work a retrospective study on vegetal foreign bodies (VFBs) localization inside the tracheobronchial tree of forty-one dogs is reported. Totally 47 grass awns were found, and only three dogs presented more than one FB. Most of VFBs were found inside right bronchial ramifications (76.6%, no. 36), while 23.4% (no. 11) were found inside the ventro-caudal branch of the left cranial lobar bronchus. The impossibility of predicting the location, and the concomitant presence of more than one FBs in a single patient are conditions that impose to make always a careful evaluation of the whole bronchial tree.

The characteristics of surface antigens in canine hepatocellular carcinoma (cHCC) have not been clarified. The objective of this study was to investigate surface antigens, which are considered as stem/progenitor or cancer cell markers, in cHCC cell lines. Expression of various antigens including CD29, CD34, CD44, CD90, CD133 and Dlk-1 was assessed in four cHCC cell lines by flow cytometry. CD44, CD133 and Dlk-1 expression was detectable in all cell lines, and three cell lines expressed CD29. These results indicate that CD29, CD44, CD133 and Dlk-1 have potential as suitable markers in cHCC identification, suggesting that these findings will contribute to the establishment of an early diagnostic tool for the identification of hepatocellular maturation processes.

リスザルは神経研究や認知科学において最もよく利用されてきた実験用霊長類の1種である.従って,リスザルの習性や行動を詳細に知ることは,中枢神経系の退行や,機能障害を判定するのに有効である.本研究では,リスザルがウズラのゆで卵を取ってから食べるまでの一連の摂食行動(Eating series: ES)について観察し,連鎖の解析をした.実験1では72匹のリスザルの,のべ378回のESについて解析した結果,3通りの統計学的に有意な推移パターンが見られた.実験2では,8匹のリスザルを無作為に選びだし,各個体につき31〜36回のESについて解析した.その結果,リスザルはウズラの卵を押すまたはこするという動作を示すPr/Ruグループとそうでないno-Pr/Ruグループとに分けられた.また,Pr/Ruグループの中でも,常にこの動作を示す個体とそうでない個体とがいた.このことからリスザルは実験室内で各個体に特徴的なESを獲得し,発達させてきたことが推察された.

Mast cell tumors (MCTs) are the most common tumors in dogs, accounting for 16–21% of cutaneous tumors. Although several small molecule inhibitors, including imatinib mesylate, have been used for the treatment of MCTs, the response rate remains less than 50%. In this study, the effects of different selective signal inhibitors on MCT cell growth were evaluated using 4 different cell lines derived from dogs. We found that the phosphoinositide 3-kinase (PI3K) signaling pathway is crucial for the proliferation of MCT cells in the presence or absence of c-kit gene mutations. Here, we propose a novel therapeutic strategy to target the PI3K pathway for the treatment of canine MCTs.

The yeasts of the Malassezia genus are opportunistic microorganisms in the skin and auricular canal of human and animals, mainly cats, and can cause otitis externa and dermatitis disorders. The aim of this study was to evaluate the occurrence of different species of Malassezia in the external ear canal of cats with and without otitis externa. Thirty-one normal cats and 82 animals with otitis externa were clinically examined. Sterile cotton swabs were used to collect specimens from the external ear canal and streaked onto the surface of Sabouraud dextrose agar (SDA) and modified Dixon agar. Malassezia yeasts were isolated from 95.1% and 48.4% of the cats with and without otitis externa, respectively. The rate of isolation in affected animals versus normals was highly significant (P<0.05). Out of the 137 isolates obtained from cats with otitis, 57.7% were identified as M. pachydermatis (with significant frequency; P<0.05), 15.4% as M. obtusa, 11.4% as M. globosa, 7.3% as M. slooffiae, 4.1% as M. sympodialis, 2.4% as M. furfur and 1.6% as M. restricta. Malassezia species were frequently isolated from subjects with age range from 1to 4 years old (42.7%). Our finding of Malassezia isolates indicated that feline otitis externa can be associated with lipid-dependent Malassezia species in addition to the non lipid- dependent species M. pachydermatis.

The aim of this study was to verify the accuracy of echocardiography by dual-source computed tomography (DSCT). Seven normal beagles underwent DSCT and echocardiography. Echocardiographic measurements were obtained according to the American Society of Echocardiography guidelines. The DSCT images were reconstructed onto the same echocardiographic image plane by using a reconstruction program, and then anatomical measurements were obtained. Nonparametric analysis was used for verifying the significance of each of the measured parameters. The anatomical measurements obtained using echocardiography and DSCT were not significant (P>0.05), and the difference between the measurements obtained using both the methods were within 95% confidence intervals except those for interventricular septal thickness and left ventricular posterior wall thickness in end diastole. The reasons for these differences were considered to be the adjacent structures such as papillary muscles or chordae tendineae that may have influenced the echocardiographic findings, lower far-field image quality of echocardiography, low test-retest reproducibility of echocardiography, high-quality images of DSCT minimizing the motion artifact and the retrospective ECG gating technique of DSCT that offered an adequate timing decision for the systolic and diastolic phase during cardiac movement. Although there were differences in the measurements of interventricular septal thickness and left ventricular posterior wall thickness in end diastole obtained using echocardiography and DSCT, we could conclude that echocardiographic measurement is as accurate and reliable as DSCT for cardiac anatomical assessment.

The epidural distribution of iohexol (0.2 ml/kg) administered at thoracic vertebrae (Thoracic group) and lumbar vertebrae (Lumbar group) was compared by computed tomographic (CT) epidurography in dogs. The total spread of iohexol was similar between the 2 groups upon reaching a similar cranial level. The maximal CT values were higher at the C7/T1 and T4/T5 levels in Thoracic group, but they were higher at the T13/L1 and L4/L5 levels in Lumbar group (P<0.05). This result suggests that the distribution pattern of the drug administered epidurally at thoracic vertebrae and lumbar vertebrae is different in dogs.

Two Miniature Dachshunds, a 7-year-old neutered male and an 8-year-old male, presented with chronic hematochezia and tenesmus. A solitary pedunculated or multiple diffuse colorectal polyps were identified with colonoscopy and resected by polypectomy. Inflammatory colorectal polyps (ICRPs) were diagnosed according to histopathological findings. Both cases were treated with immunosuppressive therapy, and the clinical signs were resolved, although the colorectal polyps remained to some extent. Several months after the initial diagnosis, both cases presented with recurrence of hematochezia and enlargement of the polyps. A second colonoscopic polypectomy was performed, and adenoma was diagnosed histopathologically in both cases. ICRPs are a nonneoplastic disease, but their long-term prognosis is unknown. Careful follow-up seems to be important, and repetitive biopsy is recommended when the growth of the polyps is identified in Miniature Dachshunds with ICRPs.

We established a homologous sandwich enzyme-linked immunosorbent assay (ELISA) to measure serum levels of canine ferritin. Our assay uses a rabbit anti-canine heart ferritin polyclonal antibody, and canine heart ferritin as a standard. Serum ferritin concentration in healthy dogs (n=163) was 789 ± 284 ng/ml (mean ± standard deviation), a value higher than reported previously. Confidence levels relating to repeatability, dilution, and recovery for this method were high. Therefore, we believe our developed sandwich ELISA will be effective in evaluating serum levels of canine ferritin.

Use of firocoxib in dogs for postoperative pain control has not been published in any of the journals in Japan. A field study was conducted to evaluate the efficacy and safety of firocoxib in dogs in controlling pain associated with soft tissue surgery in Japan. The study followed a negative control, double-blind, multicenter clinical efficacy study using a randomized block design. A total of 131 client-owned dogs presented to the clinical practices for soft tissue surgery were enrolled. Sixty-nine dogs were allocated to the firocoxib-treated group and received 5 mg/kg of firocoxib orally on Day 0 before the surgery and once daily through Day 2, while 62 dogs were allocated to the non-treated group handled in a similar manner only without the firocoxib administration. Pain assessment took place on Day 0 before the surgery through Day 2. The primary efficacy variable was a success/failure variable based on whether the dog needed rescue medication (based on pain assessment after the surgery or Investigator’s judgment) and a significant difference between firocoxib-treated group (16.4%) and non-treated group (50.0%) (P=0.0031) was observed. There was no adverse event during the study that was considered to be related to the administration of firocoxib. This study indicated the clinical efficacy and safety profile of firocoxib administered to control pain associated with soft tissue surgery under field condition.

Canine polyneuropathy is a neurological disorder characterized by a dysfunction of multiple peripheral nerves. The etiology of the disease is diverse; it may occur in cases of infectious, immune-mediated, or hereditary conditions or in association with endocrinopathy, neoplasm, or chemical intoxication. It is often difficult to determine the etiology through clinical symptoms. The aim of this study is to investigate pathological differences among three canine polyneuropathy cases with each presumably having a different etiology. Cases included a 13-month-old female border collie (Dog No.1), a 21-month-old male chihuahua (Dog No.2), and an 11-year-old male beagle (Dog No.3). Clinical examinations revealed hindlimb ataxia and sensory loss in Dog No.1, forelimb paralysis and vertebral pain in Dog No.2, and paddling-gait and hypothyroidism in Dog No.3. Histopathologically, axonal swelling and pale myelin were observed in Dog No.1. Giant axons mimicking giant axonal neuropathy were obvious in Dog No.2. Dog No.3 showed atrophic axons and severe interstitial edema. Distributions of peripheral nerve lesions coincided with respective clinical symptoms. According to their clinical and pathological features, Dogs No.1 and No.2 were suspected of hereditary polyneuropathy, while Dog No.3 seemed to have hypothyroidism-associated polyneuropathy. As each case demonstrated unique pathological features, different pathogeneses of peripheral nerve dysfunction were suggested.

We evaluated transcervical insemination (TCI) using frozen semen by flexible endoscopy in dogs. Eight female and eight male beagles were used in this study. A flexible endoscope and a washing tube were used for TCI. A tracheal tube was used as an alternative to the penis and was an auxiliary for inserting the flexible endoscope. The mean time required to insert the washing tube into the external os of the uterus after inserting the endoscope into the tracheal tube was 7.5 min. Slight or mild pain was observed in all bitches during TCI. However, TCI could be easily performed with retention in all bitches and without sedation anesthesia. The tracheal tube was useful to ensure the visual field using air sufflation. Clinical signs suspicious of infection were not observed in any bitches from the TCI to the pregnancy diagnosis. The conception rate was 87.5%, and the mean number of fetuses was 6.3. TCI using a flexible endoscope in bitches was performed quickly with minimal invasiveness. We present a new method of TCI in dogs. This method should be studied in small and large breeds to obtain more detailed results.

Previous animal studies have shown that transplantation of mesenchymal stem cells (MSCs) into spinal cord lesions enhances axonal regeneration and promotes functional recovery. We isolated the MSCs derived from fat, bone marrow, Wharton’s jelly and umbilical cord blood (UCB) positive for MSC markers and negative for hematopoietic cell markers. Their effects on the regeneration of injured canine spinal cords were compared. Spinal cord injury was induced by balloon catheter compression. Dogs with injured spinal cords were treated with only matrigel or matrigel mixed with each type of MSCs. Olby and modified Tarlov scores, immunohistochemistry, ELISA and Western blot analysis were used to evaluate the therapeutic effects. The different MSC groups showed significant improvements in locomotion at 8 weeks after transplantation (P<0.05). This recovery was accompanied by increased numbers of surviving neuron and neurofilament-positive fibers in the lesion site. Compared to the control, the lesion sizes were smaller, and fewer microglia and reactive astrocytes were found in the spinal cord epicenter of all MSC groups. Although there were no significant differences in functional recovery among the MSCs groups, UCB-derived MSCs (UCSCs) induced more nerve regeneration and anti-inflammation activity (P<0.05). Transplanted MSCs survived for 8 weeks and reduced IL-6 and COX-2 levels, which may have promoted neuronal regeneration in the spinal cord. Our data suggest that transplantation of MSCs promotes functional recovery after SCI. Furthermore, application of UCSCs led to more nerve regeneration, neuroprotection and less inflammation compared to other MSCs.

A full-length cDNA sequence of canine L-type amino acid transporter 1 (Lat1) was determined from a canine brain. The sequence was 1828 bp long and was predicted to encode 485 amino acid polypeptides. The deduced amino acid sequence of canine Lat1 showed 93.2% and 91.1% similarities to those of humans and rats, respectively. Northern blot analysis detected Lat1 expression in the cerebellum at 4 kb, and Western blot analysis showed a single band at 40 kDa. RT-PCR analysis revealed a distinct expression of Lat1 in the pancreas and testis in addition to the cerebrum and cerebellum. Notably, Lat1 expression was observed in the tissues of thyroid cancer, melanoma and hemangiopericytoma. Although the cancer samples examined were not enough, Lat1 may serve as a useful biomarker of cancer cells in veterinary clinic.

腹部膨満を示し高窒素血症を呈した3か月齢の雌の三毛猫を剖検し, 右側腎臓腹側に尿臭を持つ黄色透明液体を容れる巨大な腎周囲嚢胞(8.5×6.0×4.5cm)を認めた.嚢胞内腔は不規則に拡張した腎盂と腎実質を貫く小孔で連絡していた.嚢胞は上皮に内張りされ, 壁は膠原線維と平滑筋で構成されており, 腎盂や尿管壁の構造に類似していた.嚢胞に接する腎実質間質には, リンパ系細胞の浸潤を認めた.嚢胞は発生異常による腎盂憩室と考えられた.

Trehalose has several novel anti-inflammatory and cell-protective functions. We hypothesized that lyophilized aspirin/trehalose could decrease the severity of aspirin-induced gastropathy. Thirteen dogs were assigned into aspirin, lyophilized aspirin/trehalose, and control groups, and the gastric lesions were assessed on gastroscopy with the modified Lanza scale. Another 6 dogs were used to measure the plasma aspirin concentration by high-performance liquid chromatography after aspirin or lyophilized aspirin/trehalose administration. The results indicated that lyophilized aspirin/trehalose induced less gastric ulceration than aspirin despite maintaining therapeutic concentrations of plasma aspirin in both the groups. Lyophilized aspirin/trehalose might be a solution to decrease aspirin-induced gastropathy.

A full-length cDNA sequence of canine Na-dependent neutral amino acid transporter (ASCT2) and its distribution were determined. The sequence was 2090 bp long and was predicted to encode 544 amino acid polypeptides. The amino acid sequence deduced from canine ASCT2 showed a 90% similarity to that of humans and mice. Northern blot analysis revealed ASCT2 expression in the kidney, heart, lung and muscles, and Western blot analysis using anti-human ASCT2 antiserum detected the bands at 60 and 65 kDa in membrane protein of the lung. RT-PCR analysis revealed ASCT2 expression in all the tissues examined.

It is well known that heart rate or arterial blood pressure may increase in response to surgical stimulation despite the absence of a purposeful movement. However, there is limited information regarding anesthetic requirement for blunting adrenergic response in dogs. This study was designed to compare the minimum alveolar concentrations of sevoflurane required to prevent autonomic response (MAC-BAR) and purposeful movement (MAC) in dogs. Sevoflurane MAC-BAR and MAC were determined in 5 beagle dogs by judging dogs' response to a noxious electrical stimulus applied to the gingiva. The sevoflurane MAC-BAR was significantly higher than MAC (3.33 ± 0.48 vs 2.10 ± 0.28%, P=0.005). These results suggested that autonomic responses occurred at sevoflurane anesthetic concentrations at which purposeful movements were absent.

Although the susceptibility of dermatophytes to antifungal drugs is well documented in humans, the effectiveness in animals has not been previously investigated. The in vitro susceptibility of 54 clinical isolates from animal dermatophytoses to ketoconazole (KTZ), itaconazole (ITZ) and terbinafine (TFN) was measured using microdilution assay (CLSI M38-A2 test) and by the E-test (KTZ and ITZ). All 3 drugs showed antifungal activity, while KTZ displayed the broadest minimum inhibition concentration (MIC) range (0.125–16 μg/ml) against M. canis and M. gypseum. The MIC of KTZ and ITZ was almost the same for human and animal isolates of T. mentagrophytes and T. rubrum. The MIC of TFN was almost the same for dermatophytes isolated from humans and animals.
