腸内細菌は消化管でビタミン,短鎖脂肪酸の生成を担うだけでなく胆汁酸の代謝変換をおこなう。腸内細菌による胆汁酸の代謝変換は消化管の胆汁酸組成変化を誘導し,核内受容体farnesoid X receptor(FXR)シグナルやタンパク分解シグナルを介して宿主の胆汁酸代謝動態の調節に関与する。また近年腸内細菌による胆汁酸代謝変換を介するシグナルが肥満の予防の標的になることが報告された。本稿では,腸内細菌依存的な胆汁酸シグナルの脂質恒常性への寄与について紹介する。

リパーゼは油脂工業において最も重要な酵素であり, 微生物リパーゼは商業化され, 一部油脂製造用として工業的に用いられている。近年, 基質特異性の高いリパーゼを用いた構造油脂の生産の検討などが多く行われ, たとえば, 生理活性が高いとされるEPA, DHAや中鎖脂肪酸, 新規油糧原料など多くの油脂原料が基質として利用されている。しかし, リパーゼは高価であり, また長期間使用による活性の低下が起こりやすく, 製造コストが比較的高い。コスト低減法としては, リパーゼを安価に製造したり, リパーゼの安定性を高めたり, リパーゼを活性化するなどの方法がある。リパーゼの安定性を高める方法としては, 安定化剤添加, 化学修飾, 固定化, 遺伝子操作などがあげられる。また, リパーゼの安定性に大きな影響を与える因子としては, 反応系の水分量, 反応温度や基質油の精製度, 過酸化物価などがあげられ, これらを調節することで安定性が向上する。リパーゼ反応の工業化例としては, 脂肪酸製造, カカオ代用脂, 中鎖・長鎖脂肪酸のトリアシルグリセロールなどがあげられる。実際の反応では, 化学反応と比較して基質特異性が高いことやマイルドな条件による反応で風味が良いなどの利点があるが, 精製度が低いリパーゼを用いると, 酵素中の不純物が反応油へ影響を及ぼすこともある。

Aloe vela leaf gel extract (AGE) are widely used as cosmetic and pharmaceutical ingredients because of its versatile skin care properties. In order to enhance the bioavailability of AGE, liposomes encapsulating AGE were prepared and examined for their interfacial and biochemical properties. The liposomes prepared from a soybean lecithin (SLP-WHITE, 1.0 wt%) by the Bangham method gave relatively a good trapping efficiency up to the AGE concentration of 0.5 wt%. The stable liposomes were then prepared from 1.0 wt% of SLP-WHITE and different concentrations of AGE by the mechanochemical method using a homogenizer and microfluidizer. The liposomes obtained from 0.25 wt% of AGE were confirmed to be small unilamellar vesicles with a diameter of less than 200 nm, and remained well dispersed for at least two weeks. The obtained liposomes encapsulating AGE were further examined for the effects on proliferation and type I collagen synthesis in normal human neonatal skin fibroblasts, NB1RGB cells. Liposomal AGE clearly showed higher proliferation rate than that of AGE alone. In addition, compared to the control, liposomal AGE significantly increased the collagen synthesis by 23%, while AGE alone showed a small effect. Liposomal AGE was also assayed for the effect on proliferation in normal human epidermal keratinocytes, NHEK(F) cells. Interestingly, liposomal AGE fractions containing 4 and 20μg/mL of the extract considerably increased the proliferation rate by 77% and 101%, respectively. In contrast, AGE alone fractions containing 4 and 20μg/mL of the extract increased the rate by 41% and 60%, respectively. Accordingly, the bioavailability and skin care properties of AGE will be significantly enhanced by liposome encapsulation, and the present liposomal AGE should have a great potential as an effective skin care formulation.

In the present study, mechanical phenomena on fractal agar gel were analyzed to understand the interfacial properties of hydrophilic biosurfaces. The evaluation of adhesion strength between the fractal agar gel surfaces showed that the fractal structure inhibits the adhesion between the agar gel surfaces. In addition, when the disintegration behavior of an agar gel block was observed between fractal agar gel substrates, the rough structure prevented the sliding of an agar gel block. These findings are useful for understanding the biological significance of rough structure on the biological surfaces.

Effects of rice bran oil on the oxidative and flavor stability of fish oil were investigated by the gas liquid chromatography-head space method. When fish oil blending with different ratio of rice bran oils was oxidized at room temperature in the dark, volatile compounds produced during autoxidation was measured by gas liquid chromatography. The amounts of volatile compounds were decreased with increased the ratio of blended rice bran oil as well as peroxide value. The level of propanal and acrolein which gave unpleasant flavor was also decreased with increased the ratio of blended rice bran oil. Especially, the level of propanal and acrolein and peroxide value were remarkably decreased when blending more than 75% of rice bran oil. Blending of rice bran oil improved the oxidative and flavor stabilities of fish oil.

Squalene (SQ), a main component of human sebum, is readily photooxidized by exposure to sunlight, producing six squalene monohydroperoxide (SQ-OOH) isomers. Despite its known connection to various skin conditions, few studies have sought to analyze SQ-OOH at the isomeric level. In this study, we aimed to develop a method to discriminate each SQ-OOH isomer with the use of tandem mass spectrometry (MS/MS). The six standard SQ-OOH isomers were prepared by photooxidizing SQ in the presence of rose bengal, a photosensitizer, and isolated by semipreparative high-performance liquid chromatography (HPLC). To purify each isomer, 2-methoxypropene, which reversibly reacts with the hydroperoxide group of SQ-OOH, was utilized. Product ion scanning was then performed on the standard SQ-OOH isomers in the absence and presence of the sodium ion. In the absence of the sodium ion, the fragmentation patterns produced by atmospheric pressure chemical ionization were similar between the isomers, whereas in the presence of the sodium ion by electrospray ionization, unique fragmentation patterns were achieved. Based on these fragment ions, HPLC-MS/MS multiple reaction monitoring analysis was conducted on a mixture of the standard SQ-OOH isomers. We achieved discrimination of SQ-OOH isomers with high selectivity and detected SQ-OOH isomers at nanogram levels. These results may improve our understanding of the effect of SQ-OOH on skin conditions as well as the mechanism behind SQ peroxidation.

ドコサヘキサエン酸 (DHA, 22 : 6n-3) は神経細胞膜リン脂質の構築成分であるn-3系必須不飽和脂肪酸であり, 正常な脳の発達や視力を維持するのに極めて重要である。脳内のDHAが欠乏すると, 神経伝達系, 膜結合型酵素やイオンチャネル等の活性, 遺伝子発現, およびシナプスの可塑性, 等に著明に影響を及ぼし, そのためにDHA欠乏は, 加齢に伴う脳機能異常, アルツハイマー病, うつ病, ならびにペルオキシソーム病などを誘発する。また, DHAの摂取不足は認知・学習機能を低下させるが, DHA摂取によりこの機能は回復するようである。疫学的研究によると, 魚の消費が少ないとアルッハイマー病の罹患率が高いことから, 魚油, とくにその主成分であるDHAによる神経保護作用が推察される。脳機能障害に作用するDHAの分子メカニズムは不明であるが, DHAを摂取すると, 動物の学習機能障害が改善し, またアルツハイマー病モデルラットやマウスでの学習機能障害が予防, さらには改善される。本論文では, 食餌性DHAによる記憶・学習機能向上効果, さらには臨床応用として脳機能障害の代表的疾患であるアルツハイマー病やうつ病との関連性について紹介する。

Palmitoleic acid is a promising bactericidal agent for cleansing products with alternative bactericidal abilities. In this study, we focus on the physical and biological activity of palmitoleic acid calcium salt (C16:1 fatty acid Ca salt) because it forms via an ion-exchange reaction between palmitoleic acid and Ca ions in tap water, and remains on the skin surface during the cleansing process. Here, we prepared C16:1 fatty acid Ca salt to investigate its crystal structure and physical and bactericidal properties. The Ca salt was a plate-shaped lamellar crystalline powder with a particle diameter of several micrometers to several tens of micrometers; it exhibited significant lubricity and alternative bactericidal activity against Staphylococcus aureus (S. aureus) and Propionibacterium acnes (P. acnes). We also examined other fatty acid Ca salts prepared from lauric acid (C12:0 fatty acid), palmitic acid (C16:0 fatty acid), and oleic acid (C18:1 fatty acid). The bactericidal activities and lubricity of the fatty acid Ca salts changed with the alkyl chain length and the degree of unsaturation. The C16:1 fatty acid Ca salt exhibited the strongest selective bactericidal ability among the four investigated fatty acid Ca salts. These findings suggest that C16:1 fatty acid and its Ca salt have potential applications in cleansing and cosmetic products.

エリスリトールは,ショ糖の約75%の甘味をもつ四炭糖の糖アルコールである。ブドウ糖を原料として酵母の発酵により生産される "ブドウ糖発酵甘味料" であり,糖質では唯一のカロリーゼロの甘味料である。消費者の健康志向を背景として,エリスリトールは,あらゆる分野での低カロリー製品の検討やシュガーレス菓子の検討などに利用されている。本稿では,エリスリトールの生理的特性や物理化学的特性を中心に紹介し,その特性を利用した使用例についても紹介する。

抗がん剤新薬の開発が困難になっているなか, 既存の抗がん剤にドラッグデリバリー技術を適用して, 有効性を高め, 副作用を低減する開発研究が積極的になされている。すでにLHRHアナログやドキソルビシン塩酸塩リボソーム製剤はがん治療の標準的ガイドラインに採用されている。また, 既存の抗がん剤の利便性を改善した製剤として, アルブミン結合型パクリタキセルナノ粒子製剤も上市されている。本稿では, これらのDDS製剤の物理化学的および臨床面での特徴を紹介する。

In this study, the chemical composition and anti-inflammatory activities of hydrodistilled essential oil from Farfugium japonicum were investigated for the first time. The chemical constituents of the essential oil were further analyzed by GC-MS and included 1-undecene (22.43%), 1-nonene (19.83%), β-caryophyllene (12.26%), α-copaene (3.70%), γ-curcumene (2.86%), germacrene D (2.69%), and 1-decene (2.08%). The effects of the essential oil on nitric oxide (NO) and prostaglandin E2 (PGE2) production in lipopolysaccharide (LPS)-activated RAW 264.7 macrophages were also examined. The results indicate that the F. japonicum essential oil is an effective inhibitor of LPS-induced NO and PGE2 production in RAW 264.7 cells. These inhibitory effects of the F. japonicum essential oil were accompanied by dose-dependent decreases in the iNOS and COX-2 mRNA expression. In order to determine whether F. japonicum essential oil can safely be applied to human skin, the cytotoxic effects of F. japonicum essential oil were determined by colorimetric MTT assays in human dermal fibroblast and keratinocyte HaCaT cells. F. japonicum essential oil exhibited low cytotoxicity at 100 μg/mL. Based on these results, we suggest that F. japonicum essential oil may be considered a potential anti-inflammatory candidate for topical application.

Imidazoline surfactants belong to the category of cationic surfactants. Cationic surfactants are often quaternary nitrogen salts and are widely used both in non aqueous systems and in applications such as textile softeners, dispersants and emulsifiers. In the class of imidazoline compounds there is a presence of a pendant group, an imidazoline head group and a hydrocarbon tail. This paper reviews the imidazoline and its salts as an emerging class of cationic surfactants. It deals with the synthesis, various properties that prove its use in laundry detergent applications like good detergency, resiliency, foaming property, softening property, rewettability, good storage stability and less irritating property. In addition the film forming and corrosion inhibition properties tend them to varied and numerous industrial applications.

The physicochemical properties, fatty acid profiles, content of tocopherol and sterol of the oils extracted from the nuts of Elaeagnus mollis Diels grown in different regions of China were studied in this work. The results indicated that the Elaeagnus mollis Diels nut oils contained about 0.2% sterols and the tocopherol contents were in the range of 119.6-128.6mg/100g. The nut oils were all rich in unsaturated fatty acids, especially oleic acid and linoleic acid. Furthermore, the main triacylglycerols species of the nut oils were all dilinoleoyl-monoolein (LOL), dioleoyl-monolinoleoyl (OLO) and trilinoleate (LLL). This work might be useful for developing applications for Elaeagnus mollis Diels nut oil.

The accumulation (incorporation) of paprika carotenoid in human plasma and erythrocytes was investigated. A paprika carotenoid supplement (14 mg/day) was ingested for 4 weeks by 5 young healthy volunteers (3 men and 2 women). After 2 weeks of carotenoid ingestion, the carotenoid levels in plasma and erythrocytes increased by 1.2-fold and 2.2-fold, respectively. Characteristic carotenoids found in paprika (capsanthin, cucurbitaxanthin A, and cryptocapsin) were detected in both plasma and erythrocytes. An oxidative metabolite of capsanthin (capsanthone) was also found in both plasma and erythrocytes.

Enterococcus faecalis is one of the major lactic acid bacterium (LAB) species colonizing the intestines of animals and humans. The characteristic odor of the volatile oils obtained from both the liquid medium after incubation (MAI) and liquid medium before incubation (MBI) in the cultivation process of E. faecalis was investigated to determine the utility of the liquid medium. In total, fifty-six and thirty-two compounds were detected in the volatile oils from the MAI (MAI oil) and MBI (MBI oil), respectively. The principle components of MAI oil were 2,5-dimethylpyrazine (19.3%), phenylacetaldehyde (19.3%), and phenylethyl alcohol (9.3%). The aroma extract dilution analysis (AEDA) method was performed using gas chromatography-olfactometry (GC-O). The total number of aroma-active compounds identified in the volatile oil from MBI and MAI was thirteen compounds; in particular, 5-methyl-2-furanmethanol, phenylacetaldehyde, and phenylethyl alcohol were the most primary aroma-active compounds in MAI oil. These results imply that the industrial cultivation medium after incubation of E. faecalis may be utilized as a source of volatile oils.

The combination method of high performance liquid chromatography / UV-Visible (Vis) absorption spectrometry by photodiode-array detector and atmospheric pressure chemical ionization mass spectrometry (LC/PAD/APCI-MS) were applied for the rapid identification of carotenoids obtained from several natural sources. APCI-MS spectra provided not only information of the molecule weight but also the presence of functional groups such as hydroxy, acetyl and glycosyl groups in carotenoids. On the other hand, UV-Vis spectra gave the information of chromophores which consist of the conjugated double bonds and end groups in carotenoids. This combination method allows the carotenoids to be characterized by retention time in HPLC, APCI-MS and UV-Vis spectral data so that it offers rapid and accurate method for the identification of carotenoids extracted from various natual sources.

Clove bud essential oil (CEO) and its major individual phenolic constituent eugenol were formulated as nanoparticles in water-based microemulsion systems. The oil titration method was used to incorporate different amounts of the oil and eugenol in the micellar solution of Tween-20. The Antioxidant and antimicrobial activities were evaluated using the DPPH* free radical scavenging assay and the agar disc dilution method, respectively. Results showed that microemulsion improved the evaluated activities of CEO and eugenol compared with the crude counterparts. Individual eugenol microemulsion was more effective than CEO microemulsion which contained only 61.7% eugenol among its constituents. The results of this study could have potential applications in water-based disinfectants, preservation and flavoring of food and in personal hygiene products. It may also have promising applications in the nutraceutical and functional beverage field.
