The aim of this study was to investigate the effects of percutaneous transplanted autologous neurogenically-induced bone marrow-derived mesenchymal stem cells (NIBM-MSCs) in paraplegic dogs without deep pain perception (DPP) secondary to external spinal trauma. Thirteen client owned dogs that had failed in improvement neurologically at least 42 days after conservative management, decompression and decompression-stabilization were included in the study. Each dog received two doses of autologous 5.0 × 106 NIBM-MSCs suspension, which were positive to 2',3′-Cyclic-nucleotide-3′-phosphodiesterase (CNPase) and Microtubule-associated protein 2 (MAP-2), as well as to Glial fibrillary acidic protein (GFAP) and beta III tubulin. The cells were injected into the spinal cord through the hemilaminectomy or laminectomy defects percutaneously with 21 days interval for 2 times. The results were evaluated using Texas Spinal Cord Injury Scale (TSCIS), somatosensory evoked potentials (SEP) and motor evoked potentials (MEP) at the admission time, cell transplantation procedures and during 2, 5, 7 and 12th months after the second cell transplantation. Improvement after cell transplantation in gait, nociception, proprioception, SEP and MEP results was observed in just 2 cases, and only gait score improvement was seen in 6 cases, and no improvement was recorded in 5 cases. All progresses were observed until 2nd month after the second cell transplantation, however, there was no improvement after this period. In conclusion, percutaneous transplantation of autologous NIBM-MSCs is a promising candidate modality for cases with spinal cord injury after spinal trauma and poor prognosis.

The great invasion of antibiotics in our food has become a crude problem due to the noncodified use of them in our farms for treatment of infections or as a food supplement to improve growth and animal output that inducing sever harmful health problems on human. This study aimed to evaluate the residues of three of the most commonly used antibiotics in poultry farms and effect of heat treatment on these residues. A total of 300 chicken meats, giblets and chicken meat products were examined using a microbiological method. The positive samples were subjected to high performance liquid chromatography (HPLC) for determination of oxytetracycline, gentamicin and tilmicosin residues. The antibiotics were validated according to the guidelines laid down by the joint FAO/WHO Expert Committee on Food Additives. The incidence of antibiotic residues by microbiological method were 22%, 32%, 54%, 24% and 6% in breast, thigh, liver, gizzard, and luncheon, respectively. The presence of oxytetracycline, gentamicin and tilmicosin residues above the maximum residue limits (MRLs) in 24, 5 and 14 samples, respectively. Although calculated estimated daily intakes (EDIs) for antibiotics presented showed lower exposure levels than the fixed values of acceptable daily intakes (ADIs), the presence of these residues even in lower ADIs on long run predisposes consumers to drug resistance and allergic reactions. Cooking methods reduce antibiotic residues in experimentally administered chicken meat with varying percentages from 35.17 to 74.27%.

The mechanical properties, volumetric shrinkage and depth of cure of a short fiber-reinforced resin composite (SFRC) were investigated in this study and compared to both a bulk fill resin composite (BFRC) and conventional glass/ceramic-filled resin composite (CGRC). Fracture toughness, flexural properties, volumetric shrinkage and depth of cure of the SFRC, BFRC and CGRC were measured. SFRC had significantly higher fracture toughness than BFRCs and CGRCs. The flexural properties of SFRC were comparable with BFRCs and CGRCs. SFRC showed significantly lower volumetric shrinkage than the other tested resin composites. The depth of cure of the SFRC was similar to BFRCs and higher than CGRCs. The data from this laboratory investigation suggests that SFRC exhibits improvements in fracture toughness, volumetric shrinkage and depth of cure when compared with CGRC, but depth of cure of SFRC was similar to BFRC.

It was previously shown that the potent new DNA-binding antibiotic, CC-1065, prolonged life span, but was not curative, when administered to mice bearing a variety of transplantable tumors. In this paper we show results of preliminary studies indicating that CC-1065 caused lethal delayed hepatotoxicity at therapeutic antineoplastic doses. In non-tumor-bearing mice toxic deaths were delayed ca 50 days after a single iv dose of 12.5μg/kg and as much as 70 days after 10μg/kg was given ip. Intravenous mouse LD50's were 9μg/kg, single dose, and 0.3μg/kg/day, five daily doses. Intraperitoneal LD50's were 0.53-6.90μg/kg, single dose, and 0.14μg/kg/day, five daily doses. Mice treated with high doses iv died within 12 days with frank hepatic necrosis, whereas delayed deaths at lower doses were associated with changes in hepatic mitochondrial morphology. This suggested that separate mechanisms of hepatotoxicity were operative at high and low dose ranges. Attempts to prevent the delayed toxicity of CC-1065 in the mouse by treatment with WR-2721, N-acetylcysteine, phenobarbital, Aroclor 1254, and 3-methylcholanthrene were unsuccessful; no effect on the LD50 or the times of death was observed. Lethal doses in the rabbit were similar on a body surface area basis to those in the mouse; evidence of hepatotoxicity was also observed in the rabbit.

Antioxidants are expected to improve cardiovascular disease (CVD) by eliminating oxidative stress, but clinical trials have not shown promising results in chronic CVD. Animal studies have revealed that reactive oxygen species (ROS) exacerbate acute CVDs in which high levels of ROS are observed. However, ROS are also necessary for angiogenesis after ischemia, because ROS not only damage cells but also stimulate the cell signaling required for angiogenesis. ROS affect signaling by protein modifications, especially of cysteine amino acid thiols. Although there are several cysteine modifications, S-glutathionylation (GSH adducts; -SSG), a reversible cysteine modification by glutathione (GSH), plays an important role in angiogenic signal transduction by ROS. Glutaredoxin-1 (Glrx) is an enzyme that specifically removes GSH adducts in vivo. Overexpression of Glrx inhibits, whereas deletion of Glrx improves revascularization after mouse hindlimb ischemia. These studies indicate that increased levels of GSH adducts in ischemic muscle are beneficial in promoting angiogenesis. The underlying mechanism can be explained by multiple targets of S-gluathionylation, which mediate the angiogenic effects in ischemia. Increments in the master angiogenic transcriptional factor, HIF-1α, reduction of the anti-angiogenic factor sFlt1, activation of the endoplasmic reticulum Ca2+pump, SERCA, and inhibition of phosphatases may occur as a consequence of enhanced S-glutathionylation in ischemic tissue. In summary, inducing S-glutathionylation by inhibiting Glrx may be a therapeutic strategy to improve ischemic angiogenesis in CVD.

本研究は、日本語の単語間の連想関係をマッピングして語彙知識の特徴を検討するものである。そのため、本研究の中心は、大規模日本語連想語データベース(JWAD)の構築を継続している(Joyce,2005a,2005b,2005c,2005d,2005e,2006,2007;Joyce&Miyake,2008)。連想語に関する質問紙調査を行って連想反応を収集し、連想反応の適切さによってコード化して、ターゲット単語に対して集合での延べ数・異なり数を集計するなどのような整理して漣想反応データをJapanese word association databaseの第一版(JWAD-V1)として公開した(Joyce,2005;2006;2007)。JWAD-V1は、反応者の50名、2,099のターゲット単語に対して、合計104,800の連想反応からなっている。複雑な連想ネットワークを明確にすること及び意味空間での階層構造を視覚化することの方法として、本研究は、大規模日本語連想語データベース(JWAD)に基づいた語彙連想マップの作成、または、JWADの連想ネットワーク表象にグラフクラスタリング方法を発展させた(Joyce&Miyake,2007,2008;Miyake&Joyce,2007a,2007b,in press;Miyake,Joyce,Jung,&Akama,2007)。グラフ理論やネットワーク分析結果、JWADの連想ネットワーク表象は、スケールフリーとスパース性の構造的な特徴を持っていることが明らかになった。語彙連想マップの開発にとって、連想反応の集合とグラフクラスタリングから得られる単語のクラスタリングは、有益な比較材料となりうる。語彙マップの発展・応用を検討する研究として、専門語の学習方法におけるバイリンガル語彙マップの有効性を示唆する結果が得られた日本語習得の研究(Joyce,Takano,&Nishina,2006;Takano,Joyce,&Nishina,2006,2007)、日本語辞書編集の研究(Joyce,2005b,2005d,2006;Joyce&Srdanovic,accepted)、日本語文字体系の研究(Joyce,2007)など行った。
