Contrast-enhanced ultrasonography (CEUS) of the hepatic vein can assess intrahepatic hemodynamic changes, and has been studied as a noninvasive method to assess the severity of portal hypertension and hepatic fibrosis in humans. However, few reports have described its usefulness in veterinary medicine. The purpose of this study was to characterize CEUS findings of the hepatic vein in normal dogs and assess the repeatability of this method both in a conscious group (n=6) and a sedated group (n=6). Sonazoid® (0.01 ml/kg) was used as a contrast agent and scanning of the hepatic vein was performed for 2 min. Time-intensity curves were generated from regions of interest in the hepatic vein. Four perfusion parameters were measured for quantitative analysis: hepatic vein arrival time (HVAT), time to peak (TTP), time to peak phase (TTPP) and wash-out ratio (WR). CEUS examinations were performed three times in each dog. The median (range) values of HVAT, TTP, TTPP and WR in the conscious group were 13.5 sec (9–22 sec), 12.5 sec (6–24 sec), 8 sec (6–13 sec) and 78.0% (60.7–91.7%), respectively. Median (range) values of HVAT, TTP, TTPP and WR in the sedated group were 12 sec (8–17 sec), 12.5 sec (9–17 sec), 9 sec (7–13 sec) and 84.1% (63.0–94.4%), respectively. The coefficients of variation of these parameters in the conscious and sedated groups were 7.6–29.7% and 11.8%–14.8%, respectively.

This study describes the imaging features and characteristics of caval foramen hernias in 7 dogs diagnosed by computed tomography (CT). On lateral radiographs, 6 of 7 dogs showed dome-shaped, broad-based, caudal mediastinal lesions. CT findings included caudal vena cava (CVC) compression (n=7), right lateral (n=6) or medial (n=1) liver lobe involvement, hepatic vein dilation (n=5) and biliary tract involvement (n=1) with partial (n=6) or entire (n=1) liver lobe hernias. A caval foramen hernia should be part of the differential diagnosis when the aforementioned imaging features are detected. CT is considered as a useful tool for diagnosis and evaluation in dogs with a caval foramen hernia.

A miniature dachshund aged 9 years and 7 months with a history of polyuria/polydipsia and depression was referred. General physical and neurological examinations revealed no obvious abnormalities. MRI of the brain revealed a large space-occupying lesion in the left frontal lobe. This was surgically removed and pathologically diagnosed as a primitive neuroectodermal tumor (PNET). Although the clinical signs had been improved, follow-up MRI revealed recurrence of the tumor. Lomustine was administered, but 1 year after surgery, the dog exhibited cluster seizures and died. This is the first reported case of a dog with PNET confined to the forebrain region treated by surgical resection in combination with chemotherapy, as observed by repeated follow-up MRI.

重篤な羽毛形成不全および脱羽を示すタイハクオウム(Cacatua alba)をオウム類嘴一羽毛病(PBFD)と診断した. 光顕で観察された核内および細胞質内封入体は, 電顕的に結晶状,渦巻き状,半円形状または同心円状配列を示すウイルス粒子で構成されていた. 超微形態学的解析により, 皮膚および羽包部から回収されたウイルスは,正二十面体で直径15〜20nmのウイルス粒子と確認された.

乳腺腫瘍は雌イヌにおいて高頻度に発生する腫瘍のひとつである.本研究では, 腫瘍マーカーとしての有用性を検討するためにイヌ乳腺腫瘍材料におけるc-kit遺伝子の発現を調べた.4種の哺乳動物のc-kit cDNAの塩基配列を参考にRT-PCR法のためのプライマーセットを合成し, イヌc-kit遺伝子を増幅した.正常イヌ脾臓の全RNAからのRT-PCR法により増幅したイヌc-kit遺伝子の大きさは756bpであり, 哺乳動物のcDNAの該当部分と高い相同性を示した.11例のイヌ乳腺腫瘍(腺癌, 良性混合腫および悪性混合腫)すべてにおいてc-kit mRNAの発現が検出された.イヌ乳腺腫瘍のうち, 腺癌におけるc-kit遺伝子の発現は, 悪性混合腫瘍に比べて有意に高かった.さらに, 15例の乳腺腫瘍以外の各種イヌ腫瘍組織標本におけるc-kitの発現も同様に検討した.肥満細胞腫の全例において, 高いmRNAの発現が認められたが, 残りの12例の腫瘍では, 5例で低レベルのRT-PCR産物が検出されたのをのぞいて, c-kit遺伝子の明らかな発現は見られなかった.以上の結果より, イヌ乳腺腫瘍では, c-kit遺伝子mRNAの発現が有意に高く, その診断上有用であると思われる.

白色レグホーン種の原種をトリ白血病ウイルスの感染状態を基準に, 血清から抗体もウイルスも検出されない非感染鶏, 血清から抗体は検出されるが卵白からウイルスは検出されない感染・非排泄鶏, 卵白からウイルスが検出される感染・排泄鶏に分類し, 各群の生産性を比較した. その結果, 産卵率の低下, 卵白のハウ単位の低下, 卵殻の肥厚, 体重の軽量化等の生産阻害は感染・ 排泄鶏でのみ認められた.

The aim of this study was to investigate the effects of percutaneous transplanted autologous neurogenically-induced bone marrow-derived mesenchymal stem cells (NIBM-MSCs) in paraplegic dogs without deep pain perception (DPP) secondary to external spinal trauma. Thirteen client owned dogs that had failed in improvement neurologically at least 42 days after conservative management, decompression and decompression-stabilization were included in the study. Each dog received two doses of autologous 5.0 × 106 NIBM-MSCs suspension, which were positive to 2',3′-Cyclic-nucleotide-3′-phosphodiesterase (CNPase) and Microtubule-associated protein 2 (MAP-2), as well as to Glial fibrillary acidic protein (GFAP) and beta III tubulin. The cells were injected into the spinal cord through the hemilaminectomy or laminectomy defects percutaneously with 21 days interval for 2 times. The results were evaluated using Texas Spinal Cord Injury Scale (TSCIS), somatosensory evoked potentials (SEP) and motor evoked potentials (MEP) at the admission time, cell transplantation procedures and during 2, 5, 7 and 12th months after the second cell transplantation. Improvement after cell transplantation in gait, nociception, proprioception, SEP and MEP results was observed in just 2 cases, and only gait score improvement was seen in 6 cases, and no improvement was recorded in 5 cases. All progresses were observed until 2nd month after the second cell transplantation, however, there was no improvement after this period. In conclusion, percutaneous transplantation of autologous NIBM-MSCs is a promising candidate modality for cases with spinal cord injury after spinal trauma and poor prognosis.

2胚移植によって受胎した正常双胎妊娠牛(N牛), フリーマーチン妊娠牛(F牛)および反転性裂体妊娠牛(S牛)の妊娠全期間にわたる末梢血中プロジェステロン(P<SUB>4</SUB>), エストロン(E<SUB>1</SUB>)およびエストラジオール(E<SUB>2</SUB>)の消長を調べた. 血中P4濃度は, F牛が妊娠254日に双子を死産した際に急減した他は, 三者の間には相違がみられなかった. 血中E1濃度は, N牛においては好期の経過に伴い上昇し, 分娩日に最高値を示した後に急減し, F牛においてはN牛よりも低い値で推移し, 妊娠254日に突然上昇した後に急減した. また, S牛においてもN牛より低く, 変動を伴って推移した. 血中E<SUB>2</SUB>濃度は, 三者ともにE<SUB>1</SUB>より低く, かつE<SUB>1</SUB>濃度に平行して推移した. 血中E<SUB>1</SUB>, E<SUB>2</SUB>濃度は, 分娩後1日にN牛では両者とも20pg/ml以下の値に低下したが, F牛とS牛はE<SUB>1</SUB>が150kg/ml以上, E<SUB>2</SUB>が20pg/ml以上の値を示した. この成績は, 妊娠中の胎子数の判定同様子宮内の胞子の予後の判定においても, P<SUB>4</SUB>よりE<SUB>1</SUB>やE<SUB>2</SUB>の方が優れた指標となることを示唆している.

抗ウマCRPウサギ血清を用いた単純放射免疫拡散(SRID)法により, 臨床的に正常なウマの加齢および周産期におけるCRPの血中動態および実験的に炎症を作出したウマにおけるCRPの血中動態を検討した. 初乳未摂取の新生子の血清CRP濃度は雌雄ともに測定限界以下であったが, 幼齢期に上昇し, 青壮年期に低値を示し, 以降やや増加して安定するという加齢性の変化を示した. 周産期には, 分娩時および分娩後2か月目から4か月目の血清CRP濃度上昇および分娩2か月前の低下を認めた. 炎症刺激後24時間目より血清CRP濃度は上昇し, 3〜5日目で処置前の3〜6倍に達し最高値を示した. その後低下し14日目には処置前値に回復した. 以上の成績から, ウマにおいでもCRPは急性相蛋白であることが判明した.

日本の子牛におけるウシトロウイルス(BoTV)の糞便への排出状況と下痢との関連性を調べた.同時にウシロタウイルス,ウシコロナウイルスとクリプトスポリジウムの排出状況も調べた.BoTVのスパイク遺伝子を標的としたRT-PCRにより,北海道の子牛下痢便99例中15例(15.2%)からBoTV遺伝子が検出された.正常便では114例中8例(7.0%)がBoTV陽性であった.北海道外の38例の子牛下痢便では9例(23.7%)が陽性を示した.北海道のBoTV陽性15例のうち11例では今回検査対象とした病原体のうちBoTVのみが検出された.BoTV陽性15例中11例の子牛は2週給以下であった.今回の調査で,ウシロタウイルスと下痢との関連性は認められたが,ウシコロナウイルスとクリプトスポリジウムと下痢との関連性は認められなかった.17例のBoTV陽性のRT-PCR産物をランダムに選択し,それらの塩基配列を決定して進化系統樹を作成したところ,日本のBoTVは少なくとも2グループに分類されることが示された.今回の結果から,ウシトロウイルスは日本に広く浸潤し,子牛,特に2週齢以下の子牛下痢の原因ウイルスの一つであることが示唆された.

Epigallocatechin gallate (EGCG) is the major polyphenolic compound of green tea. Polyphenolic compounds were extracted from the leaf of Camellia sinensis (Japanese green tea), and the minimum inhibitory concentration against canine oral bacteria was measured. Subsequently, we investigated the inhibitory effects of polyphenolic compounds and EGCG on the growth of canine oral bacteria. EGCG showed antimicrobial activity against a model bacterium, Streptococcus mutans. Our results indicate that EGCG can inhibit the growth and biofilm formation of S. mutans, and that EGCG does not interact with streptococcal lipoteichoic acid (LTA). Furthermore, our findings suggest that EGCG interacts with other component (s) of the bacterial membrane aside from streptococcal LTA to inhibit biofilm formation and damage biofilms.

イオタラム酸(IOT)の猫における体内消失を検討するために健常猫及び実験的腎不全猫にイオタラム酸(IOT: 118.3 mg/kg)を静脈内投与し, その血漿消失速度を測定した. 一区画モデルによる解析で実験的腎不全猫のIOT血中消失速度は2.16±0.240×10-3μg/ml/minと健常群の値(4.10±1.00×10-3μg/ml/min)に比較して有意な低値を示し, 血漿IOT消失速度は猫の簡便なGFR測定法となる可能性が示唆された.

Density gradient ultracentrifugation (DGUC) and gel electrophoresis are conventionally used to obtain lipoprotein profiles of animals. We recently applied high-performance liquid chromatography with a gel permeation column (GP-HPLC) and an on-line dual enzymatic system to dogs for lipoprotein profile analysis. We compared the GP-HPLC with DGUC as a method to obtain a feline lipoprotein profile. The lipoprotein profiles showed large and small peaks, which corresponded to high-density lipoprotein (HDL) and low-density lipoprotein (LDL), respectively, whereas very low-density lipoprotein (VLDL) and chylomicron (CM) were only marginally detected. This profile was very similar to that of dogs reported previously. Healthy cats also had a small amount of cholesterol-rich particles distinct from the normal LDL or HDL profile. There was no difference in lipoprotein profiles between the sexes, but males had a significantly larger LDL particle size (P=0.015). This study shows the feasibility of GP-HPLC for obtaining accurate lipoprotein profiles with small sample volumes and provides valuable reference data for healthy cats that should facilitate diagnoses.

Lymphoma is one of the most common malignant tumors in canine. Protein phosphatase 2A (PP2A), a well-conserved serine/threonine phosphatase, plays a critical role as a tumor suppressor. Perphenazine (PPZ) is one of the phenothiazines and widely used as an antipsychotic drug. Recently, it is reported that PPZ directly binds with scaffolding subunit of PP2A complex and exerts anti-tumor effects on human T cell acute lymphoblastic leukemia. However, the effect of PPZ on canine lymphoma has not been studied. Here, we investigated the potential therapeutic role of PPZ and its molecular mechanism in canine T-cell lymphoma. In canine T-cell lymphoma cell lines, UL-1 and Ema, PPZ decreased cell survival in a dose-dependent manner. Increased caspase 3 activity and Annexin V positive cells suggested that PPZ induced apoptosis. PPZ dephosphorylated Akt, MEK1/2 and ERK1/2. Akt inhibitor, but not MEK1/2 inhibitor and ERK1/2 inhibitor, induced cell death, indicating the importance of Akt dephosphorylation for the anti-tumor effect of PPZ. Finally, we observed enhanced PP2A activity by PPZ treatment. The present results for the first time revealed that PPZ induced canine lymphoma cells apoptosis through Akt dephosphorylation via PP2A activation. Our study suggests the possible therapeutic application of phenothiazines for canine T-cell lymphoma.

We compared the effects of acupuncture and electroacupuncture on cell proliferation and neuroblast differentiation using specific markers, Ki67 and doublecortin (DCX), in the subgranular zone of the dentate gyrus (SZDG) in 13-week old Wistar rats. Acupuncture and electroacupuncture were applied simultaneously in the acu-points, ST36 (Zusanli) and GV20 (Baihui), once a day for 3 weeks. Acupuncture and electroacupuncture at these acu-points significantly increased the number of Ki67-positive cells and DCX-immunoreactive neuroblasts compared to the control or sham acupuncture group. Electroacupuncture treatment significantly increased the number of well-developed (tertiary) dendrites in the SZDG compared to acupuncture treatment. These results suggest that both acupuncture and electroacupuncture increase neurogenesis in the normal, but that electroacupuncture has greater effects on neuroblast plasticity than acupuncture in the dentate gyrus.

The Babesia gibsoni heat shock protein 90 (BgHSP90) gene was cloned and sequenced. The length of the gene was 2,610 bp with two introns. This gene was amplified from cDNA corresponding to full length cording sequence (CDS) with an open reading frame of 2,148 bp. A phylogenetic analysis of the CDS of HSP90 gene showed that B. gibsoni was most closely related to B. bovis and Babesia sp. BQ1/Lintan and lies within a phylogenetic cluster of protozoa. Moreover, mRNA transcription profile for BgHSP90 exposed to high temperature were examined by quantitative real-time reverse transcription-polymerase chain reaction. BgHSP90 levels was elevated when the parasites were incubated at 43°C for 1 hr.

Canine degenerative myelopathy (DM) is characterized by progressive degeneration of the spinal cord. Although atrophic changes in the intercostal muscles were previously reported in the late stage of DM in Pembroke Welsh Corgis (PWCs), changes in respiratory function have not yet been examined. In the present study, we performed an arterial blood gas analysis and measured respiratory movements over progressive disease stages to document changes in respiratory function in DM-affected PWCs. We found that respiratory dysfunction progressed during the later stages of DM and correlated with a change in respiratory movement to the abdominal breathing pattern. These results suggested that hypoventilation occurred due to dysfunctional changes in the intercostal muscles and resulted in hypoxemia in the later stages of DM.

Clinical and histopathological characteristics of 16 dogs with nodal paracortical (T-zone) lymphoma (TZL) were evaluated. At initial examination, generalized lymphadenopathy was found in all dogs, and peripheral lymphocytosis was found in 10 of the 16 dogs. At initial diagnosis or during the disease course, 8 dogs (50%) were affected with demodicosis. Immunohistochemical analysis for CD3, CD20 and CD25 was performed for 6 dogs with TZL; the tumor cells were positive for CD3 and CD25 and negative for CD20. Median overall survival time was 938 days. A watchful waiting approach was adopted for 6 cases (38%), and 5 of the 6 dogs were still alive at the end of follow-up. The clinical course of TZL in dogs is generally indolent; however, many cases develop a variety of infectious and other neoplastic diseases after the diagnosis of TZL.

To investigate inhibitory effect for oral bacterial proliferation, we divided 12 dogs into 3 groups; scaling alone (C; control group), brushing (B) and application of toothpaste (P). Before scaling (Pre) and at 0 to 8 weeks after scaling (0–8 w), we collected oral bacteria from the dental surface every week and counted them using a bacterial counter. The results demonstrated a significant reduction in the number of oral bacteria for group B relative to Pre and group C, as well as for group P relative to group C at 5–7 w. Consequently, brushing may inhibit an increase in the number of oral bacteria,and toothpaste may be effective at a certain level, although not more than that of brushing.

The definitive hosts of Sarcocystis sinensis in water buffaloes have hitherto been unknown, but the close similarity of this species to the cat-transmitted Sarcocystis bovifelis in cattle suggested they were felids. In a previous study, two domestic cats were fed macroscopic sarcocysts of Sarcocystis fusiformis contained within or dissected from the esophageal muscles of water buffaloes, while no microscopic sarcocysts of S. sinensis were noticed. Both cats started shedding small numbers of sporocysts 8–10 days post infection (dpi) and were euthanized 15 dpi. Using a PCR-based molecular assay targeting the mitochondrial cox1 gene of S. fusiformis, both cats were shown to act as definitive hosts for this species. In the present study, DNA samples derived from oocysts/sporocysts in the intestinal mucosa of both cats were further examined by PCR for the presence of S. sinensis using 2 newly designed primers selectively targeting the cox1 gene of this species. All 6 DNA samples examined from each cat tested positive for S. sinensis. A 1,038-bp-long portion of cox1 was amplified and sequenced as 2 overlapping fragments from 5 of these DNA samples. The 5 sequences shared 99.3–100% identity with 7 previous cox1 sequences of S. sinensis obtained from sarcocysts in water buffaloes. Additionally, amplification of the ITS1 region with primers targeting various Sarcocystis spp., yielded amplicons of 2 different lengths, corresponding to those obtained from sarcocyst isolates of S. sinensis and S. fusiformis, respectively. This is the first study to show that cats act as definitive hosts for S. sinensis.
