In this study, we present dual-beam Doppler optical coherence angiography with variable beam separation. Altering beam distance, independently of the scanning protocol, provides a flexible way to select the velocity range of detectable blood flow. This system utilized a one-micrometer wavelength light source to visualize deep into the posterior eye, i.e., the choroid. Two-dimensional choroidal vasculature maps of a human subject acquired with different beam separations, and hence with several velocity ranges, are presented. Combining these maps yields a semi-quantitative visualization of axial velocity of the choroidal circulation. The proposed technique may be useful for identifying choroidal abnormalities that occur in pathological conditions of the eye.

A high-penetration swept-source optical coherence tomography (HP-SS-OCT) system based on a 1-μm short cavity laser is developed. Doppler OCT processing is applied, along with a custom-made numerical phase stabilization algorithm; this process does not require additional calibration hardware. Thus, our phase stabilization method is simple and can be employed in a variety of SS-OCT systems. The bidirectional blood flow and vasculature in the deep choroid was successfully imaged via two Doppler modes that use different time intervals for Doppler processing. En face projection image of squared power of Doppler shift is compared to ICGA, and the utility of our method is verified.

Dual-beam-scan Doppler optical coherence angiography (DB-OCA) enables high-speed, high-sensitivity blood flow imaging. However, birefringence of biological tissues is an obstacle to vasculature imaging. Here, the influence of polarization and birefringence on DB-OCA imaging was analyzed. A DB-OCA system without birefringence artifact has been developed by introducing a Faraday rotator. The performance was confirmed in vitro using chicken muscle and in vivo using the human eye. Birefringence artifacts due to birefringent tissues were suppressed. Micro-vasculatures in the lamina cribrosa and nerve fiber layer of human eyes were visualized in vivo. High-speed and high-sensitivity micro-vasculature imaging involving birefringent tissues is available with polarization multiplexing DB-OCA.

In this article, we consider a variety of inference problems for high-dimensional data. The purpose of this article is to suggest directions for future research and possible solutions about p n problems by using new types of two-stage estimation methodologies. This is the first attempt to apply sequential analysis to high-dimensional statistical inference ensuring prespecified accuracy. We offer the sample size determination for inference problems by creating new types of multivariate two-stage procedures. To develop theory and methodologies, the most important and basic idea is the asymptotic normality when p → ∞. By developing asymptotic normality when p → ∞, we first give (a) a given-bandwidth confidence region for the square loss. In addition, we give (b) a two-sample test to assure prespecified size and power simultaneously together with (c) an equality-test procedure for two covariance matrices. We also give (d) a two-stage discriminant procedure that controls misclassification rates being no more than a prespecified value. Moreover, we propose (e) a two-stage variable selection procedure that provides screening of variables in the first stage and selects a significant set of associated variables from among a set of candidate variables in the second stage. Following the variable selection procedure, we consider (f) variable selection for high-dimensional regression to compare favorably with the lasso in terms of the assurance of accuracy and the computational cost. Further, we consider variable selection for classification and propose (g) a two-stage discriminant procedure after screening some variables. Finally, we consider (h) pathway analysis for high-dimensional data by constructing a multiple test of correlation coefficients.

Mycalolides are tris-oxazole macrolides isolated from the marine sponge Mycale sp., which shows cytotoxic, antifungal, and actin-depolymerizing activities. To develop an efficient synthetic route of mycalolides and to evaluate its functional mechanism of biological activities, tris-oxazole macrolactone analogs of mycalolides were synthesized through the use of ring-closing metathesis (RCM). The presence/absence of protecting groups at C3, solvent polarity, and reaction temperature significantly affected the stereoselectivity of RCM (E/Z=2.5/1.0–1.0/2.5). The 19E- and 19Z-stereoisomers both exhibited moderate cytotoxicity against tumor cells, but neither showed significant actin-depolymerizing properties or antimycotic activity against pathogenic fungi. Thus, both the side-chain (actin-binding) moiety and the macrolactone moiety were suggested to be essential for the potent biological activities of the parent molecules.

We propose a methodology for estimating the residence time of groundwater based on bomb-produced 36Cl. Water samples were collected from 28 springs and 2 flowing wells located around Mt. Fuji, Central Japan. 36Cl/Cl ratios in the water samples, determined by accelerator mass spectrometry (AMS), were between 43 × 10−15 and 412 × 10−15. A reference time series of the above-background (i.e., bomb-derived) 36Cl concentration was constructed by linearly scaling the background-corrected Dye-3 data according to the estimated total bomb-produced 36Cl fallout in the Mt. Fuji area. Assuming piston flow transport, estimates of residence time were obtained by comparing the measured bomb-derived 36Cl concentrations in spring water with the reference curve. The distribution of 36Cl-based residence times is basically consistent with that of tritium-based estimates calculated from data presented in previous studies, although the estimated residence times differ between the two tracers. This discrepancy may reflect chlorine recycling via vegetation or the relatively small change in fallout rate, approximately since 1975, which would give rise to large uncertainties in 36Cl-based estimates of recharge for the period, approximately since 1975. Given the estimated ages for groundwater from flowing wells, dating based on a 36Cl bomb pulse may be more reliable and sensitive for groundwater recharged before 1975, back as far as the mid-1950s.

In this paper, the writer has attempted to analyze the educational thought of Thomas Tryon (1634-1703) in order to clarify an aspect of the historical stream of European thought of embryo education. Jean-Jacques Rousseau, citing Varro's discourse, said such as the following : Educit obstetrix, dit Varron; educat nutrix, instituit poedagogus, docet magister. Ainsi l'education, l'institution, l'instruction, sont trois choses aussi differentes dans leur objet que la gouvernante, le precepteur et le maitre. This shows explicitly that the term "educacion" traditionally meant the action of midwives and nurses. "Education" did not mean school-instruction but "san-iku (bringing forth and breeding up)" (Kunio Yanagida). Accordingly, in the first place, the history of education must be a history of "saniku" which clarifies the historical transformation of "education". Furthermore, the crucial and critical field of such history, though it might sound strange, is the history of embryo education, because it comprises the overlapping field both of "san" and "iku". In fact, historical materials of embryo education consist of books for midwifery and child-rearing. However, at least to Japanese, European history of embryo education discourses has not been familiar. On the other hand, we have already had many informations on Japanese history of embryo education ("taikyo"). Therefore, in Japan, exsists a tendency to think that embryo education discourses exisist in Japan rather than in Europe. But, in this paper, the writer presents one of embryo education discourses in the 17th-century England as an example for embryo education in Europe. Thomas Tryon, a Pythagorean and vegetarian, published his book A New Method of Educating Children in 1695,which was read by John Locke too. This book on education began with his discourse on embryo education, which was founded on two premises : (1) The "Impressions" that women make, whilst they are pregnant and go with the child, have a wonderful "Influence" upon their issue. (2) There is a strong and unconceivable "Sympathy" between the mother and the child. Because of these premises, he had to submit the discourse on embryo education at first, proposing six dietetic advices, physical and mental, toward mothers. Of course, his dietetic advices which were based upon his The Way to Health functioned as a manual for mothers' self-fashioning, naturally stressing on the importance of female education. In Tryon's educational thought, embryo education was an archtype and a foundation of his whole thought of education, which represented European tradition of education.
