We investigated the cell viability, proliferative capacity and neuronal differentiation potential of canine bone marrow stromal cells (BMSCs) after cryopreservation. BMSCs were cryopreserved using cryoprotectant solutions with 10% DMSO and 10% FBS (DF group) or without DMSO and FBS (DF-free group); fresh BMSCs were used as a control. The cell viability and proliferative capacity of BMSCs were similar in the DF-free and control groups, while those in the DF group were lower. In all groups, BMSCs differentiated into neuron-like cells that stained positive against neuron markers, and the mRNA expression levels of neuron markers increased after neuronal induction. In conclusion, cryopreservation with DF-free cryoprotectant solution did not diminish the cell viability, proliferative capacity or neuronal differentiation potential of canine BMSCs.

Conductive polymers have been studied extensively because of their attractive physical properties, such as conductivity, luminescent performance, and dielectric property. Poly(3,4-ethylenedioxythiophene) (PEDOT) is one of the most employed conductive polymers for applications, such as a buffer layer of organic electroluminescent devices, due to its high conductivity and electrical stability. In this study, we fabricated a conductive paper coated with PEDOT by direct polymerization onto a paper sheet. The conductive paper exhibited the electrical conductivity of 1.8 S/cm. Scanning electron microscopy images of the conductive paper showed two structures: thin polymer membranes attached to cellulose fibers at the surfaces, and thick polymer sheets extended through the void spaces between the fibers in the inner layers. Consequently, strong interactions between the PEDOT and the cellulose fibers enhanced mechanical properties of the conductive paper. Neutrophils play a central role in host defenses against infectious microbial pathogens. However, molecular basis for their triggering signals has been incompletely understood. Here, we show molecular and functional characteristics of myeloid-associated immunoglobulin-like receptor (MAIR)-IV. MAIR-IV was preferentially expressed on neutrophils in the peripheral blood, bone marrow, peritoneal cavity and spleen. MAIR-IV physically associates with immunoreceptor tyrosine based activating motif (ITAM)-bearing adaptor FcRγ chain. Cross-linking MAIR-IV with anti-MAIR-IV monoclonal antibody significantly induced secretion of proinflammatory cytokines TNF-α and IL-6 from neutrophils. Thus, MAIR-IV may regulate activation of neutrophils and play an important role for innate immunity.

Experimentalstudy on proton exchange membrane (PEM) electrolyzer was carried out focusing on the effect of pore structural properties of currentcollectors, such as porosity and pore diameter. Various titanium (Ti)-felt substrates with different porosities and pore diameters (measured by capillary flow porometry) were used as the anode currentcollector. Results show that when the mean pore diameter of the currentcollector was larger than 10 μm, the electrolysis performance improved with decreasing pore diameter. In contrast, changes in porosity had no significant effect on the cell performance when the porosity exceeded 0.50. The flow pattern of two-phase flow in the flow channel was discussed in terms of its relationship to bubble size and to pore diameter of the currentcollector. Finally, correlation between the calculated membrane resistance and the measured pore diameter of the currentcollectors suggest that larger bubbles generated from larger pores tend to become long bubbles in the channel, thus hindering the water supply to the membrane.

We analyzed the distribution of 11 periodontitis-related bacterial species in dental plaque collected from 176 Japanese dogs divided into young (less than 2 years of age), middle-aged (2–7 years of age) and elderly (more than 8 years of age) groups using a polymerase chain reaction method. Clinical examination revealed that no dogs in the young group were affected by periodontitis, whereas the rates for gingivitis and periodontitis were high in the middle-aged and elderly groups. In addition, the total numbers of bacterial species in the middle-aged and elderly groups were significantly greater than in the young group. Our findings suggest that age is an important factor associated with the distribution of periodontitis-related bacteria and periodontal conditions in dogs.

Inference of subcellular location only using sequence information is important. In this paper, we proposed the application of principal component analysis-based linear discriminant analysis for subcellular location information. It achieved the performance of Area Under the Curve under Receiver Operating Characteristic curve mostly more than 0.95 if less than 80% sequence identity non-redundancy was applied.

Type III secretion system (T3SS) effector protein is a part of bacterial secretion systems. T3SS exists in the pathogenic and symbiotic bacteria. How the T3SS effector proteins in these two classes differ from each other should be interesting. In this paper, we proposed the usage of principal component analysis based linear discriminant analysis that discriminates T3SS effector proteins between plant pathogenic, animal pathogenic and plant symbiotic bacteria by the accuracy of 0.77. We also hypothesized that the feature vector proposed by Yahara et al represents protein structure, possibly protein folds defined in Structural Classification of Proteins (SCOP) database.

Xenoturbella bocki, a marine animal with a simple body plan, has recently been suggested to be sister group to the Acoelomorpha, together forming the new phylum Xenacoelomorpha. The phylogenetic position of the phylum is still under debate, either as an early branching bilaterian or as a sister group to the Ambulacraria (hemichordates and echinoderms) within the deuterostomes. Although development has been described for several species of Acoelomorpha, little is known about the life cycle of Xenoturbella. Here we report the embryonic stages of Xenoturbella, and show that it is a direct developer without a feeding larval stage. This mode of development is similar to that of the acoelomorphs, supporting the newly proposed phylum Xenacoelomorpha and suggesting that the last common ancestor of the phylum might have been a direct developer.

There is strong demand to create wearable PC systems that can support the user outdoors. When we are outdoors, our movement makes it impossible to use traditional input devices such as keyboards and mice. We propose a hand gesture interface based on image processing to operate wearable PCs. The semi-transparent PC screen is displayed on the head mount display(HMD), and the user makes hand gestures to select icons on the screen. The user's hand is extracted from the images captured by a color camera mounted above the HMD. Since skin color can vary widely due to outdoor lighting effects, a key problem is accurately discrimination the hand from the background. The proposed method does not assume any fixed skin color space. First, the image is divided into blocks and blocks with similar average color are linked. Contiguous regions are then subjected to hand recognition. Blocks on the edges of the hand region are subdivided for more accurate finger discrimination. A change in hand shape is recognized as hand movement. Our current input interface associates a hand grasp with a mouse click. Tests on a prototype system confirm that the proposed method recognizes hand gestures accurately at high speed. We intend to develop a wider range of recognizable gestures.

Canonical correlation analysis (CCA) is a powerful tool for analyzing multi-dimensional paired data. However, CCA tends to perform poorly when the number of paired samples is limited, which is often the case in practice. To cope with this problem, we propose a semi-supervised variant of CCA named SemiCCA that allows us to incorporate additional unpaired samples for mitigating overfitting. Advantages of the proposed method over previously proposed methods are its computational efficiency and intuitive operationality: it smoothly bridges the generalized eigenvalue problems of CCA and principal component analysis (PCA), and thus its solution can be computed efficiently just by solving a single eigenvalue problem as the original CCA.

The TGF-β and Wnt pathways are involved in cell fate and tumorigenicity. A recent report indicated that a TGF-β target gene, TMEPAI (transmembrane prostate androgen-induced RNA), is possibly also a downstream target of Wnt signaling. Although TMEPAI was believed to be involved in tumorigenicity because of its blockage of TGF-β signaling, how TGF-β and Wnt signals affect the activation of the TMEPAI gene is not well understood. Herein, we show that the TMEPAI promoter is regulated synergistically by TGF-β/Smad and Wnt/β-catenin/T cell factor (TCF) 7L2. The critical cis-element for dual signals, termed TGF-β-responsive TCF7L2-binding element (TTE), is located in intron 1 of the TMEPAI gene. TCF7L2, but not Smad proteins, bound to TTE, whereas the disruption of TTE by mutagenesis remarkably counteracted both TGF-β and TCF7L2 responses. The introduction of mutations in critical Smad-binding elements blocked the activation of the TMEPAI promoter by TCF7L2. Furthermore, our DNA-protein interaction experiments revealed the indirect binding of TCF7L2 to Smad-binding elements via Smad3 upon TGF-β stimulation as well as its TGF-β-dependent association with TTE. We demonstrate that the Wnt/β-catenin/TCF7L2 pathway is preferentially able to alter the transcriptional regulation of the TGF-β-target gene, TMEPAI.

By adopting a network architecture in which not only a calling but also a called terminal can select a wavelength, a novel WDM network becomes possible. This we call Wavelength Assignment Photonic Network (WAPN). In this network wavelengths are a kind of network resources and according to requests from terminals, wavelengths are allocated or assigned to calls. In the system a wavelength used for a call is to be used for another call after the call is terminated. By supplying wavelengths to the home, a bitrate-free, protocol free or even transmission method free network can be realized. In this paper, from a viewpoint of S/N or Q factor, WAPN is evaluated with special focus on the node architecture -i.e., from the viewpoint of node size, number of switching stages, crosstalk level, and losses, because the allowable node size is the crucial issue to decide the whole network capacity. After brief explanations of this proposed system, the model for system evaluations will be established and a node system is to be evaluated for some practical parameter values considering especially traffic characteristics of a node. As a result of this study a node system with capacity more than 100 thousands erl (about 20 Tbps throughput) can be constructed using present available technologies, which will enable us to construct large WAPN network with radius of 2,000 km and subscribers of about 50 millions.

Background: Colestimide has been reported to lower blood glucose levels in patients with type 2 diabetes complicated by hypercholesterolemia. Aim: To examine the mechanism by which colestimide decreases plasma glucose levels in the above patients. Methods: A total of 16 inpatients with type 2 diabetes complicated by hypercholesterolemia received colestimide for 1 week after their plasma glucose levels stabilized. We measured plasma glucose, serum immunoreactive insulin (IRI), serum lipid, plasma glucagon, and plasma glucagon-like peptide-1 (GLP-1) levels. These variables at baseline and 1 week of colestimide administration were compared. Results: Preprandial plasma glucose levels (baseline: 132 ± 33 mg/dL vs. completion: 118 ± 43 mg/dL, P=0.073) tended to decrease after colestimide administration, while 1-hr postprandial plasma glucose levels (baseline: 208 ± 49 mg/dL vs. completion: 166 ± 30 mg/dL, P<0.001) and 2-hr postprandial plasma glucose levels (baseline: 209 ± 56 mg/dL vs. completion: 178 ± 39 mg/dL, P=0.015) decreased significantly at 1 week of colestimide administration. The 2-hr postprandial plasma GLP-1 level was significantly (P=0.015) higher at 1 week of colestimide administration as compared with the baseline level, while there were no significant changes in preprandial and 1-hr postprandial plasma GLP-1 levels. Conclusions: The GLP-1-increasing activity of colestimide may explain, at least in part, the mechanism of its blood glucose-lowering activity in patients with type 2 diabetes complicated by hypercholesterolemia.

he basic helix–loop–helix protein E2-2 is known to play a role in quiescence of endothelial cells (ECs). However, it is unclear how the activity of E2-2 is controlled in the cells. In this study, we identified FAM96B as an interaction partner of E2-2. FAM96B interfered with E2-2-mediated effects on luciferase reporter activities. Furthermore, the suppression of vascular endothelial growth factor receptor 2 promoter activity by E2-2 was rescued by the expression of FAM96B in a dose-dependent manner. Interestingly, FAM96B decreased the expression of ectopic and endogenous E2-2 proteins. Mutational analysis revealed that the middle region of FAM96B is required for the limited expression of E2-2 protein. When FAM96B was expressed in ECs, the EC migration, proliferation, and tube formation were potentiated. Taken together, these findings suggest that FAM96B acts as a regulator of E2-2 through the control of its protein expression.

An optically active, π-stacked poly[2,7-bis(4-t-butylphenyl)dibenzofulvene] having a preferred-handed helical conformation was synthesized by anionic polymerization. A thin film sample of the polymer exhibited broad-band white circularly polarized light (CPL) emission on photo excitation.

Porphyromonas gulae, a gram-negative black-pigmented anaerobe, is a pathogen for periodontitis in dogs. An approximately 41-kDa fimbrial subunit protein (FimA) encoded by fimA is regarded as associated with periodontitis. In the present study, the fimA genes of 17 P. gulae strains were sequenced, and classified into two major types. The generation of phylogenetic trees based on the deduced amino acid sequence of FimA of P. gulae strains along with sequences from several strains of Porphyromonas gingivalis, a major cause of human periodontitis, revealed that the two types of FimA (types A and B) of P. gulae were similar to type I FimA and types II and III FimA of P. gingivalis, respectively. A PCR system for classification was established based on differences in the nucleotide sequences of the fimA genes. Analysis of 115 P. gulae-positive oral swab specimens from dogs revealed that 42.6%, 22.6%, and 26.1% of them contained type A, type B, and both type A and B fimA genes, respectively. Experiments with a mouse abscess model demonstrated that the strains with type B fimA caused significantly greater systemic inflammation than those with type A. These results suggest that the FimA proteins of P. gulae are diverse with two major types and that strains with type B fimA could be more virulent.

We investigated the influence of autologous serum (AS)-supplemented medium on the proliferation and differentiation into neurons of canine bone marrow stromal cells (BMSCs). Canine BMSCs were cultured using α-MEM only, α-MEM with 10% fetal bovine serum (FBS), and 5, 10 and 20% AS-supplemented α-MEM. Growth of canine BMSCs was observed in all AS groups. The proliferation capacity of canine BMSCs in the AS groups was similar to that in the FBS group. No significant differences between the FBS and AS groups were observed in the percentage of the cells that changed to the neuron-like morphology and neuron-specific enolase-positive ratio after neuronal differentiation. Canine BMSCs cultured using AS-supplemented medium were able to proliferate and showed neuronal differentiation potency.

Adult urodele amphibians such as newts are capable of regenerating lost structures including their limbs. In these species, dedifferentiation of myofiber is essential for the regenerative process. Upon terminal differentiation, nuclei of myofiber (myonuclei) are withdrawn from cell cycle, but prior to dedifferentiation, myonuclei reenter the cell cycle. In contrast with urodele amphibians, it is generally accepted that mammalian myofibers are not able to dedifferentiate in response to muscle injury. A recent study has suggested that electroporation can induce dedifferentiation response of skeletal muscle in newt limbs. In the present study, we examined whether myonuclei of skeletal muscle of mammals are capable of reentering the cell cycle by means of electroporation. Electroporation was applied to tibialis anterior muscle of the rat with or without plasmid DNA. Histological analyses revealed that, while electroporation induces degenerative/regenerative responses in skeletal muscle irrespective of the presence of plasmid DNA, the expression of proliferating cell nuclear antigen (PCNA) in myonuclei was observed only in the presence of plasmid DNA. The present results indicate that myonuclei of skeletal muscle are capable of reentering the cell cycle and suggest that in vivo electroporation can induce dedifferentiation of mammalian skeletal muscle.
