Understanding the molecular mechanisms of DNA double-strand break (DSB) repair machinery, specifically non-homologous DNA-end joining (NHEJ), is crucial for developing next-generation radiotherapies and common chemotherapeutics for human and animal cancers. The localization, protein-protein interactions and post-translational modifications of core NHEJ factors, might play vital roles for regulation of NHEJ activity. The human Ku heterodimer (Ku70/Ku80) is a core NHEJ factor in the NHEJ pathway and is involved in sensing of DSBs. Companion animals, such as canines, have been proposed to be an excellent model for cancer research, including development of chemotherapeutics. However, the post-translational modifications, localization and complex formation of canine Ku70 have not been clarified. Here, we show that canine Ku70 localizes in the nuclei of interphase cells and that it is recruited quickly at laser-microirradiated DSB sites. Structurally, two DNA-PK phosphorylation sites (S6 and S51), an ubiquitination site (K114), two canonical sumoylation consensus motifs, a CDK phosphorylation motif, and a nuclear localization signal (NLS) in the human Ku70 are evolutionarily conserved in canine and mouse species, while the acetylation sites in human Ku70 are partially conserved. Intriguingly, the primary candidate nucleophile (K31) required for 5’dRP/AP lyase activity of human and mouse Ku70 is not conserved in canines, suggesting that canine Ku does not possess this activity. Our findings provide insights into the molecular mechanisms of Ku-dependent NHEJ in a canine model and form a platform for the development of next-generation common chemotherapeutics for human and animal cancers.

看護において,「安楽」という用語は「安全」と共に看護の目的とされ,実践場面においても看護目標としても多用されている言葉である。だが,「安楽」という用語は抽象的であり,研究者は看護者によってその意味するところが異なるという体験がしばしばあった。そこで,本研究においては,看護領域において用いられている「安楽」という用語を,実践場面で使用されている側面から検討し,その意味するところを明らかにすることとした。このことは,看護識者が「安楽」なケアの提供を意識化し,概念化する手がかりとなる。本研究の対象は,内科系外科系病棟看護師29名であり,質問紙を用いた半構成的面接法を行った。分析の視点は,Rodgersによる概念分析の方法に準拠し,『定義(Definition),先行するもの(Antecedents),帰結(Consequences),代替となる用語(Alternative Terms),関連する概念(Related Concept)』の5つの枠組を採用した。半構成的面接法によって得られた記述データを,Rodgersの5つの枠組毎に分析して,内容を抽出(原データ)し,その内容を要約した題目をつけ(コーディング1),更にそれらを類型化(コーディング2)した。その結果,29名の看護師から,29通りの「安楽」の定義が示された。「安楽」に"先行するもの"は,患者に関するものとして「苦痛」「不快」「不安]「環境」等が,看護師に関するものとして「安全」「看護技術」等が抽出された。「安楽」の"帰結"としては、「患者の満足」「自然治権力を高める」などが抽出された。"「安楽」の意味するもの"を, "「安楽」に先行するもの"を用いて,構造化した。看護師が患者により安楽なケアを提供するには,この構造図の要素を充実・強化する必要があることが示唆され,看護実践場面における「安楽」という用語の意味は,この構造図の範囲内において説明でき,連想されることが示された。

Permanent magnet synchronous (PM) motors are widely used for industrial applications. In the PM motor drives, it is difficult to achieve the wide speed and constant power operations, which are desired in traction and spindle drives, because the field-flux of PM motor is uncontrollable and as a result the field-weakening control is not available. The air-gap flux, however, can be weakened by the demagnetizing effects due to the d-axis armature reaction (flux-weakening control). In this paper, the wide speed control method of the interior PM (IPM) motor, which includes the maximum torque-per-ampere control and the flux-weakening control, is proposed. In the high-speed flux-weakening region, there are some cases where the control performances become worse and the control system becomes unstable because of the saturation of current regulator and the effects of magnetic saturation. The high performance current regulator compensating these effects is also proposed. The control performances of the proposed control system are confirmed by the several drive tests with respect to the prototype IPM motor.
