東北地方南部太平洋側特有の春季多照条件を効果的に利用する栽培法として, 4月下旬の田植えについて検討して来た。その結果, 従来の田植時期 (5月10日) に比べて, 生育, 収量が向上することがわかったため, 1977, 1978の両年現地実証試験を宮城県内で行なった。<br>両年次の早植時期 (4月下旬) における県内温度分布では, 最高気温は比較的高い14~17℃の範囲であるが, 平均気温は8~12℃, 最低気温は2~6℃の範囲に分布し, この時期には, 低温障害発生危険温度を示す。しかし, 水田水温の推定値からみれば, 最高水温は24~28℃, 平均水温11~15℃, 最低水温3~9℃となって, 最低水温のやや低い地帯があるが, 水田水温全体からみれば, 低温障害の恐れのある地帯は極あて少ないことがわかった。<br>なお, 主な試験地における水田水温 (地表温度) の平均温度はほとんど12℃以上で経過していた。<br>田植日から最高分げつ期までの積算値でみると, 日照時数では両年次で, 早植 (4月20日) は標準植 (5月10日) より約70~100時間多い。気温, 水田水温では両田植日間にあまり大きな差がない。早植はやや少ない気温を示しているが, 水田水温では, 早植が多くなっている。<br>最高分げつ期から出穂期まで間の積算値では, 日照, 気温, 水田水温とも田植日による差は小さい。この間に含まれる梅雨期の影響が大きい。<br>出穂後40日間の積算値では, 日照, 気温, 水田水温とも早植の方が標準値より多く, 登熟期間中の気象は早植の場合に好条件となった。<br>各試験田の初期生育量 (乾物重量) は, 早植ほど増加傾向を示し, さらに4月末までに田植えした場合の収量は両年次とも600kg/10a以上となり, 現地においても早植栽培の効果が確認された。

The Loop-mediated isothermal amplification (LAMP) method amplifies DNA with high simply, specificity, sensitivity and rapidity. In this study, A LAMP assay with 6 primers targeting a highly conserved region of the GRA1 gene was developed to diagnose <i>Toxoplasma gondii</i>. The reaction time of the LAMP assay was shortened to 30 min after optimizing the reaction system. The LAMP assay was found to be highly specific and stable. The detection limit of the LAMP assay was 10 copies, the same as that of the conventional PCR. We used the LAMP assay to develop a real-time fluorogenic protocol to quantitate <i>T. gondii</i> DNA and generated a log-linear regression plot by plotting the time-to-threshold values against genomic equivalent copies. Furthermore, the LAMP assay was applied to detect <i>T. gondii</i> DNA in 423 blood samples and 380 lymph node samples from 10 pig farms, and positive results were obtained for 7.8% and 8.2% of samples, respectively. The results showed that the LAMP method is slightly more sensitive than conventional PCR (6.1% and 7.6%). Positive samples obtained from 6 pig farms. The LAMP assay established in this study resulted in simple, specific, sensitive and rapid detection of <i>T. gondii</i> DNA and is expected to play an important role in clinical detection of <i>T. gondii</i>.

Given their applicability in genetic engineering, undomesticated Bacillus strains are extensively used as non-natural hosts for chemical production due to their high tolerance of toxic substrates or products. However, they are difficult to genomically modify due to their low transformation efficiencies. In this study, the Bacillus-E. coli shuttle vector pHY300PLK, which is widely used in gram-positive bacteria, was adopted for genome integration in organic solvent-tolerant Bacillus isolates. The Bacillus-replicative vector was used to deliver homologous recombinant DNA and propagate itself inside the host cell, increasing the likelihood of genome integration of the recombinant DNA. Then, the unintegrated vectors were cured by cell cultivation in antibiotic-free medium with facilitation of nickel ions. The developed protocol was successfully demonstrated and validated by the disruption of amyE gene in B. subtilis 168. With an improved clonal selection protocol, the probability of clonal selection of the amyE::cat genome-integrated mutants was increased up to 42.0 ± 10.2%. Genome integration in undomesticated, organic solvent tolerant Bacillus strains was also successfully demonstrated with amyE as well as proB gene creating the gene-disrupted mutants with the corresponding phenotype and genotype. Not only was this technique effectively applied to several strains of undomesticated B. subtilis, but it was also successfully applied to B. cereus. This study validates the possibility of the application of Bacillus-replicative vector as well as the developed protocol in a variety of genome modification of undomesticated Bacillus species.

Two carbohydrate binding surface sites (SBSs) on barley &alpha;-amylase 1 (AMY1) of glycoside hydrolase family 13 (GH13) displayed synergy in interactions with starch granules, thus being pivotal for hydrolysis of supramolecular substrates. Mutational analysis showed that SBS1 is more critical for the conversion of starch granules, while SBS2 has higher affinity than SBS1 for &beta;-cyclodextrin (&beta;-CD). Noticeably, the binding preference of &beta;-CD to SBS2 differed distinctly from that of maltooligosaccharides to the catalytic nucleophile mutant D180A AMY1. Binding energy maps at subsites -8 through +4 of the active site indicated remarkably elevated affinity due to the Y380A mutation at SBS2. The high-yield AMY2 expression variant A42P, made it possible to show that Tyr378&mdash;corresponding to Tyr380 in AMY1&mdash;has a role in interactions with starch granules, but not in &beta;-CD binding. Besides SBSs, dedicated starch binding domains (SBDs) mediate binding to starch granules. SBDs are currently categorised into 9 carbohydrate binding module (CBM) families. A novel CBM20 subfamily encountered in regulatory enzymes possesses characteristically low affinity for &beta;-CD. Although &alpha;-amylase is essential for starch mobilisation in germinating barley seeds, efficient degradation requires the concerted action of &alpha;-amylase, &beta;-amylase, limit dextrinase (LD) and possibly &alpha;-glucosidase. Limit dextrinase (LD) is encoded by a single gene and represents the sole debranching activity during germination. Recent expression of functional LD in <i>Pichia pastoris</i> makes biochemical and biophysical characterisation of this GH13 enzyme possible. An endogenous limit dextrinase inhibitor was cloned and produced recombinantly and demonstrated to have sub-nanomolar affinity for LD.

<B>目的</B> グローバル経済が進展する中,生活を規定する企業活動は持続的成長を図るために様々な方策を実施している.その一つが,企業の社会的責任(Corporate Social Responsibility: 以下CSRと呼ぶ)である.CSRが及ぶ範囲は,社会的公正性,倫理性,環境,人権と幅広いが,日本のCSRは環境問題に特化され,ステイクホルダーである労働者・消費者・家庭人・地域住民への社会的公正性や倫理性,人権に関する取組みが遅れている.本報告は,現在の生活経営学的課題をCSR視点で捉え直し,日本企業のCSRと生活経営学の今後のあり方を提起する.<BR><B>方法</B> 生活経営学的課題を収入(賃金)と働き方に絞り現状を把握する.CSRガイドラインとして多用されているGRI(Global Reporting Initiative)に含まれる「労働とディーセント・ワーク」指標に照らし,法令遵守としての最低賃金制度とその実態を検討する.<BR><B>結果</B> 雇用形態別労働者数においては非正規労働者の増加,特に男性の非正規労働者の増加が明らかとなり,賃金については男女格差と同時に雇用形態別格差が示された.日本では,多くの企業がCSR=法令遵守として捉えているので,賃金の基本的な法令である最低賃金法から現状を把握し,最低賃金未満の労働者率と法令違反率を示す.さらにGRIの「労働慣行とディーセント・ワーク」指標に照らしながら,日本企業の不足点を明らかにした.それらの改善のためには,多様なステイクホルダーが生活主体として企業と向き合い、CSRに関わる姿勢が重要であり,能動的な生活者視点すなわち生活経営学が果たす役割は大きい.

In the present study, biomolecule-stabilized Au nanoclusters were demonstrated as a novel fluorescence probe for sensitive and selective detection of pazufloxacin mesilate (PZFX) for the first time. The linear decrease in the fluorescence intensity of Au nanoclusters induced by PZFX allowed for the quantitative detection of PZFX in the range of 0.15 &mu;g/mL to 1 mg/mL, and the detection limit for PZFX was 0.2 &mu;g/mL. Circular dichroism spectroscopy and fluorescence decay studies were then performed to discuss the quenching mechanism. In addition, practical application of the present approach was also demonstrated for real samples, which suggested its great potential for accurate analysis of similar drugs.

<p>This paper describes a novel method for label-free mercury(II) ion detection based on exonuclease III-induced target signal recycling amplification using double-stranded DNA templated copper nanoclusters. The synthesized DNA-Cu nanoclusters were used with exonuclease III loop amplification technology for ultra-high sensitivity detection of mercury(II) ions, which were detected by significantly decreased fluorescence intensity. Under the optimal experimental conditions, there was a clear linear relationship between Hg<sup>2+</sup> concentration in the range of 0.04 to 8 nM and fluorescence intensity. The detection limit for Hg<sup>2+</sup> was 4 pM. In addition, the interference of other metal ions on the mercury(II) ion detection was also studied. To confirm the application of the fluorescent sensor, it was applied to determine the concentrations of mercury(II) ions in tap water, and the results showed that the method can be used to detect mercury(II) ions in water samples successfully.</p>

Nanogold particles of size 15 nm were used to label goat anti-human ceruloplasmin (GCP) to obtain an immunonanogold probe (AuGCP) for ceruloplasmin (CP). In a pH 7.8 citric acid-Na<sub>2</sub>HPO<sub>4</sub> buffer solution and in the presence of polyethylene glycol (PEG), an immunoreaction between AuGCP and CP took place, and the released nanogold particles aggregated to bigger clusters, which caused the resonance scattering (RS) intensity at 552 nm (<i>I</i><sub>552nm</sub>) to be enhanced greatly. The enhanced intensity &Delta;<i>I</i> was proportional to the CP concentration (<i>C</i><sub>CP</sub>) in the range from 0.0030 to 1.26 &micro;g/mL, with a regress equation of &Delta;<i>I</i> = 101.0<i>C</i><sub>CP</sub> + 1.8, a correlation coefficient of 0.9970, and a detection limit of 1.1 ng/mL CP. This simple and sensitive RS immunoassay was applied to the determination of CP in human plasma, with satisfactory results.
