It is necessary to develop a more efficient gene expression system for gene therapy. A plasmid DNA, using eukaryoic or mammalian promoters, requires to localize into nuclear for gene expression. However, it is difficult to entry into nuclear, because nuclear pore size is not sufficient against the size of plasmid DNA. In this study, to develop a novel cytoplasmic gene expression system that dose not require nuclear localization of plasmid DNA to transcription, we examined the characterization of T7 cytoplasmic gene expression system. When co-transfected with pT7-IRES-L(luciferase expression plasmid containing T7 promoter) and T7 RNA polymerase into LLCMK2 cells, the gene expression of pT7-IRES-L was observed rapidly within 6hr after transfection and significant level of luciferase activity was detected. In contrast, pRSVL, a common plasmid DNA consist of luciferase expression plasmid and Rous sarcoma virus promoter, required 24-48hr for induction of gene expression. The gene expression level of the T7 system was enhanced with an increase in the amount of T7 RNA polymerase. To increase and prolong the gene expression, a plasmid DNA(pT7 AUTO-2) which contained the T7 RNA polymerase gene driven by the T7 promoter was co-transfected with pT7-IRES-L and T7 RNA polymerase. The plasmid DNA(pT7 AUTO-2) dose-dependently enhanced the luciferase gene expression by pT7-IRES-L and T7 RNA polymerase. In addition, we attempted to optimize the cytoplasmic gene expression system. The optimal ratio for co-transfection of pT7-IRES-L and pT7 AUTO-2 was 1 to 3 (mole ratio). These results suggest that T7 gene expression system may be useful in many gene therapies where transient but rapid efficient gene expression is required.

Reflexivity is a concept which reflects oneself to others and determines oneself by the reflection of others. Giddens critiques empirical and functional methods of sociology, and suggests new rules of sociological methods by means of using the concept of reflexivity. Giddens also uses reflexivity to explain modern society. In modern society reflexivity works faster in the social system, and reliable certainty cannot be found anywhere. Giddens insists that post industrial society is not 'post-modernity' but 'high-modernity', a boundless working of reflexivity. In high-modernity, politics acts as an important role as rationality did in modernity. Giddens makes a close connection between sociological method and modern society by using 'reflexivity', which is the key word of both method of social thought and social theory of modernity. If reflexivity works more and more, we must doubt not only the foundation of social science but also that of logics and natural science. To pursue reflexivity more radically, we will see a more uncertain world than Giddens anticipates. In addition, we should pursue not simple modernization, which reflects Western rational modernization, but reflexive modernization, which reflects multiple cultures. When we understand that everything is reflected in our own culture, including our concepts, even the concept of 'reflexivity', and sympathize with other different cultures, we can subjectively and critically begin a new method of social thought. At the same time, we can reflexively construct a society for the future.

ウサギの臼歯不正咬合の発生要因を検討するため,発症群137例,対照群345例について疫学的に調査するとともに,食餌内容に関するアンケート調査を行い,発症群34例,対照群58例より回答を得た。発症群の平均体重は対照群よりも有意に低く,体格の小さなウサギほど臼歯不正咬合を発症しやすい可能性があるものと思われた。臼歯不正咬合の罹患率は雑種で高く,ロップイヤー系品種で低い傾向にあった。雄は雌に比べて罹患率が有意に高かった。発症群の平均乾草摂取量は対照群と比較して有意に少なく,過去の記述と一致して,乾草摂取量が少ないことは臼歯不正咬合発症の危険因子となりうることが推察された。食餌指導により乾草の摂取量を十分確保させること,さらに雑種や雄,そして体格の小さな個体には,より厳密な食餌指導を行うことが,臼歯不正咬合の発症予防となり,または発症後の進行防止に役立つものと思われた。

【目的】最近は胃瘻に対して慎重であるべきとの意見が散見されるが、過去の報告で胃瘻造設をした家族の満足度は60~ 70%と決して低くない。今回胃瘻造設に対する家族と医療従事者側の考えの違いをアンケートで検討した。【対象及び方法】1) H22.4.1~ H25.3.31に当院で PEG造設術を施行した患者家族、2) 一般病棟・療養病棟に勤務している看護師・介護士、3) 老健保健施設・特別養護老人ホームに勤務している看護師・介護士・介護員を対象にアンケートを実施した。【結果】家族51人、医療従事者側213人 (一般病棟85人、療養病棟27人、老健保健施設43人、特別養護老人ホーム58人) だった。胃瘻に満足しているのは家族では74.5%で、医療従事者側の一般病棟39.0%・療養病棟30.8%・老健保健施設27.5%・特別養護老人ホーム14.3%より有意に多かった。自分自身で胃瘻を希望する家族は31.3%で、医療従事者側の3.4~ 11.1%より有意に多かった。【結論】家族は医療従事者側より、胃瘻に対して肯定的な意見が多かった。医療従事者側では、介護施設で胃瘻に対して否定的な意見が多かった。

Chk1 is required for arrest of the mammalian cell cycle before mitosis in response to DNA damage or replication block. It is also implicated in regulation of cell cycle progression because it is essential for embryonic cell viability. With the use of mouse embryonic stem cells conditionally deficient in Chk1, we now show that this kinase is indispensable for the timing of mitotic initiation. Chk1 deficiency resulted in premature onset of mitosis through reduction of Cdc2 phosphorylation on Tyr15 as a result of increased Cdc25 activity and decreased Wee1 activity. Our results suggest that Chk1 regulates Cdc2 to establish proper timing of mitotic initiation during the mammalian embryonic cell cycle.

電磁波吸収材料は,不要な電磁波を効率良く吸収し熱に変換する材料である。電磁波の電界や磁界は物質中の電子が持つ電荷や磁気モーメントと相互作用し,電磁波のエネルギーが電子の移動や磁気モーメントの運動のエネルギーに変換されることで吸収が生じる。特に磁性損失材料の吸収機構である磁気共鳴について解説するとともに,最近の材料の開発動向についても触れる。

目的 : 大腿静脈カテーテル留置時に上行腰静脈へ迷入すると, 後腹膜血腫等の合併症を引き起こす可能性がある. しかし, カテーテルの迷入についてはあまり認識されていない.方法 : 2011年4月から2013年3月の間に当院で大腿静脈カテーテルを留置した患者107名を対象とし後ろ向きに検討した.結果 : 上行腰静脈への迷入は11/110回 (10.0%) で認め, 左側で5/34回 (14.7%) , 右側で6/76回 (7.9%) だった. 位置確認の腹部レントゲン検査で, カテーテルが側方へ変位している場合, 頭側に急峻に立ち上がる場合に迷入の可能性があった.結論 : カテーテル迷入は稀では無く, 迷入が疑われれば積極的に腹部CT, 側面レントゲン等の追加の検査が必要である.

We have successfully prepared Carbon/Fe-Co alloy composites starting from metal organic precursors synthesized by polymerized complex method. The carbon/Fe-Co alloy composites were obtained after the carbonization of the precursors heat-treated at ambient pressure and lower temperatures ranging from 500 to 700°C in N₂ gas flow. Crystalline sizes of both carbon matrix and Fe-Co alloy particles were determined by X-ray diffraction measurements as a function of the Co concentration and the carbonized temperature. The alloy particles were widely dispersed in the carbon matrix and the particle size carbonized at 600°C was about 20∼50 nm observed by scanning electron microscopy. The composite carbonized at 600°C had very small coercivity of 14 Oe and exhibited relatively large permeability spectra at frequency of GHz range.

This study sought to examine the reliability and validity of the Japanese version of the Behavior Rating Inventory of Executive Function (J-BRIEF). In this study, BRIEF was administered to evaluate executive function in everyday life in 91 subjects with autism spectrum disorder (ASD; ages 12–15; 72 boys) and 2,230 community samples (CS; ages 12–15; 1,083 boys). For this purpose, we applied categorical confirmatory factor analysis, which revealed that the scale was composed of two factors and eight subscales of the high test-retest stability. Reliability was confirmed using an external criterion (ADHD-Rating scale: ADHD-RS). Receiver operating characteristic analysis revealed an optimal cut-off of 118.5 (sensitivity = 0.811, specificity = 0.828). This study confirmed the reliability and the validity of J-BRIEF.
