A global property (i.e., pitch set) of a melody appears to serve as a primary cue for key identification. Previous studies have proposed specific local properties in a melody (e.g., the augmented fourth, the perfect fifth, etc.) that may function as further cues. However, the role of the latter in key identification is controversial. The present study was designed to investigate what kinds of local properties, if any, function as reliable cues for key identification. Listeners were asked to identify keys for 450 melodies that consisted of the same pitch set but differed in sequential constraints. Using multiple discriminant analyses, we evaluated relative contributions of as many kinds of local properties as possible (e.g., single intervals, single pitch classes in each sequential position, etc.). The results showed that, except for the pitch class of the final tone, for which interpretation should be taken cautiously, none of specific local properties examined contributed significantly to key identification. This finding suggests that, contrary to prior findings, key identification is derived from unidentified properties other than the specific local properties.

During the late 1970s, the El Niño-Southern Oscillation (ENSO) experienced a notable regime change, manifested by a change in amplitude, dominant ENSO period, and sea surface temperature anomaly (SSTA) propagation characteristics. The present study shows that these features of the ENSO regime change are associated with property changes of the canonical ENSO, i.e., cold-tongue (CT) type ENSO. Another signature of the ENSO regime change is manifested in the frequent occurrence of a warm-pool (WP) type ENSO that accompanies SSTAs centered over the central Pacific near the WP edge and exhibits characteristics differing from those of the CT ENSO. The distinct manifestations of the two types of ENSO detected in this ENSO regime change are clearly identifiable with the removal of the strong background decadal signal. Since the late 1970s, the WP ENSO has featured a weak eastward (westward) propagation of the SSTA center in the developing (decaying) phase, which makes no net contribution to the observed eastward propagation, and a 2-3 yr period compared to the 4-5 yr period of the CT ENSO. Observations strongly suggest that the WP and CT ENSO are independent quasi-biennial and quasi-quadrennial modes, respectively, of the tropical Pacific climate variability. Our observations also suggest that these two ENSO modes have coexisted actively since the late 1970s when either El Niño or La Niña can be separated into the two types.

Sex ratio distortion, which is commonly abbreviated as sex-ratio, has been studied in many Drosophila species, but the mechanism remains largely unknown. Here, we report on the sex-ratio mutant of D. simulans named excess of females (exf). The third chromosomal recessive mutation results in a sex ratio of approximately 0.2 or less (males/total). Cytological observation demonstrated that meiosis appeared to be completed normally, but that most Y chromosome-bearing nuclei failed to elongate during spermiogenesis, as revealed by fluorescence in situ hybridization using sex chromosome-specific probes. These aberrant nuclei contained membranous inclusions as revealed by electron microscopic analysis. Most of the aberrant exf spermatids failed to individualize and mature, suggesting that a later stage of spermiogenesis is involved in prevention of production of sperm with abnormal morphology. On the one hand, in exf seminal vesicles, sperm nuclei with a length of 5–8.5 μm were occasionally observed, in addition to those with wild-type sperm dimensions, that is, a length of approximately 10 μm. Thus, spermatids with less severe nuclear defects can escape elimination and be released into the seminal vesicles as mature sperm. Furthermore, we constructed His2AvD-GFP and ProtamineB-eGFP transgenic lines in D. simulans, and examined the processes involved in replacement of chromatin proteins over a time course, according to nuclear morphology. We found that both normal and abnormal sperm heads demonstrated equal chromatin replacement during late spermiogenesis. Our results suggest that exf belongs to a unique class of meiotic drive systems in that (1) intranuclear membranous inclusions cause failure of nuclear shaping of Y-bearing spermatids without affecting the histone-protamine transition, and (2) a portion of the aberrant spermatids differentiate into mature sperm; these are transferred to and stored by females.

QT prolongation, a risk factor for arrhythmias, can result from genetic variants in one (or more) of the genes governing cardiac repolarization as well as intake of drugs known to affect a cardiac K+ channel encoded by human ether-a-go-go–related gene (HERG). In this paper, we will report a case of drug-induced long QT syndrome associated with an H1-receptor antagonist, hydroxyzine, in which a mutation was identified in the HERG gene. After taking 75 mg of hydroxyzine for several days, a 34-year-old female began to experience repetitive syncope. The deleterious effect of hydroxyzine was suspected because QTc interval shortened from 630 to 464 ms after cessation of the drug. Later on, the patient was found to harbor an A614V-HERG mutation. By using the patch-clamp technique in the heterologous expression system, we examined the functional outcome of the A614V mutation and confirmed a dominant-negative effect on HERG expression. Hydroxyzine concentration-dependently inhibited both wild-type (WT) and WT/A614V-HERG K+ currents. Half-maximum block concentrations of WT and WT/A614V-HERG K+ currents were 0.62 and 0.52 μM, respectively. Thus, accidental combination of genetic mutation and intake of hydroxyzine appeared to have led to a severe phenotype, probably, syncope due to torsade de pointes.

To improve the yield of a Japanese tomato (Solanum lycopersicum) cultivar and determine how fruit yield changes as a result of grafting, we investigated the effects of a Dutch rootstock [‘Maxifort’ (S. lycopersicum × S. habrochaites): Mx] on the dry matter (DM) production and fruit yield of Dutch and Japanese cultivars. The Japanese cultivar (‘Momotaro York’: My) grafted onto Mx (My/Mx: scion/rootstock) had significantly higher fresh and dry weights of fruits per unit area than My/My. Fruit fresh weight yield per unit area was highly correlated with fruit dry weight (DW) yield (r = 0.96–0.97, P < 0.001), and DW yield was significantly correlated with total aboveground DM (r = 0.71–0.96, P < 0.001) and with DM allocation to the fruits (r = 0.52–0.75, P < 0.01). Total aboveground DM (TDM) was significantly and highly correlated with light-use efficiency (r = 0.98, P < 0.001). However, there was no significant correlation between light-use efficiency and the maximum photosynthetic rate, stomatal conductance, or the light-extinction coefficient. Although stomatal conductance significantly (P < 0.05) differed between the rootstocks at 57 and 119 days after transplanting (DAT), there was no significant difference in the maximum photosynthetic rate between the scion/rootstock combinations at 57 and 119 DAT. These results indicated that the fruit yield of My could be improved by grafting onto Mx, and that the increases in yield and TDM were mainly determined by the increase in light-use efficiency.

A 9-month-old intact female Yorkshire terrier dog was presented with episodic partial seizure-like cramping of the limbs. The patient’s episodes began six months previously; the interval between episodes became shorter, and the duration of the episodes increased. Various tests including neurologic examination, blood examination, abdominal radiography, ultrasonographic examination, angiographic computed tomography (CT) and brain magnetic resonance imaging (MRI) detected no remarkable changes. After these tests were conducted, the patient’s condition was suspected to be canine epileptoid cramping syndrome (CECS), which could be a form of paroxysmal dyskinesia (PD), and as a trial therapy, Science Diet k/d (Hill’s Pet Nutrition, Topeka, KS, U.S.A.) was prescribed. The clinical signs were dramatically reduced after diet therapy, and we diagnosed the patient with CECS. This is the first case report of CECS in a Yorkshire terrier dog.

To produce tomato fruits with 6% average Brix without decreasing yield, we investigated the effect of moderate salinity stress on Brix and yield in a single-truss, high-density tomato production system. Because tomato fruit development can be predicted from cumulative temperature, we also assessed cumulative temperature after anthesis as a potential indicator for determining the starting points of salinity stress treatments. When transverse diameters of the first fruit reached 4 cm (i.e., early increase treatment) or the first fruits were at the mature green stage (i.e., late increase treatment), nutrient solution electrical conductivity was slowly increased until the breaker stage from 1.8 dS·m−1 to 6.0 dS·m−1. Plants subjected to the late increase treatment produced tomato fruits with Brix values of 6% without reductions in marketable yield. We also increased nutrient solution electrical conductivity based on cumulative temperature after anthesis and found that early-treated plants produced tomatoes with higher Brix levels and yields than late-treated plants. In summary, moderate salinity stress to avoid excessive stress on plants increased sugar concentrations without decreasing fruit yield and resulted in tomato fruits with average Brix of about 6% when nutrient solution electrical conductivity was increased at a rate of 0.1 dS·m−1·day−1. Because seasonal differences in cumulative temperature influence the appropriate timing of salinity stress applications, further study is needed to optimize year-round growth under moderate salinity stress in single truss, high-density tomato production systems.

Workplace bullying has been a topic of increasing interest since the 90s. Several studies have contributed to a better understanding of the antecedents and consequences of being exposed to negative acts at work during a prolonged period of time. However, there is a lack of validated instruments in Italian to map workplace bullying. Consequently, the goal of the present study is twofold. First, the authors aim to validate the Italian version of the Negative Acts Questionnaire-Revised (NAQ-R: Einarsen and Hoel, 2001; Einarsen, et al., 2009). Second, the authors aim to establish the prevalence of workplace bullying. Results from 3,112 employees nested within 25 Italian organizations revealed that a reduced version of 17 items with a two-factor structure provided the best fit. Moreover, the prevalence of bullying was 15.2 per cent according to the criteria proposed by Mikkelsen and Einarsen (2001). Furthermore, significant correlations between bullying dimensions and organizational climate were found, which provided additional support for using the NAQ-Italian reduced version. Implications to prevent bullying and improve employees’ well-being using this method are discussed.

本年度は量子気体顕微鏡の技術を活用して、原子が2次元光格子中に欠陥なく並んでいる状態を生成する段階に踏み込んだ。まず、Bose-Hubbard模型で実現される超流動状態からMott絶縁状態への相転移を、飛行時間測定法を用いて観測した。超流動状態では原子の位相が揃っているため、波動関数が拡散するとともに干渉縞が現れる。ポテンシャルの深さを徐々に上げていくとMott絶縁状態が誘起され、原子の位置が確定されると同時に位相が不確定になり、干渉縞が消えていくことが確認された。実験において相転移が生じたポテンシャル深さは、理論的な予想と一致していた。Mott絶縁状態が誘起されても、系の温度が十分に低くなっていない場合、一つのサイトに2個以上の原子が入ったり、あるいは欠陥が生じたりしてしまう。私が構築した量子気体顕微鏡は、サイト内原子数が0か1かを区別することはできるが、原子数が1か2以上かを判断することはできない。そこで、光会合技術を導入することで、同一サイト中の2原子を分子に変換・排除し、原子数の偶奇を判定することにした。実際に光会合光を照射した後、量子気体顕微鏡を使って光格子中の原子分布を直接観測したところ、各サイトの原子数が予想に反して揺らいでいることが確認された。原子数のゆらぎを抑圧するためには系の温度を1nK程度まで下げる必要性がある。別途、系の加熱レートを評価したところ、30nK/sec程度であることが判明した。超流動相からMott絶縁相への相転移を誘起し、顕微鏡で観測するためには、1sec程度の時間が必要となるため、上記した超低温度下での観測は難しいと言える。光格子を生成しているレーザーを吸収・自然放出することによる不可避な加熱レートを見積もったところ、僅か50pK/secであり、測定された加熱レートが、主に、レーザーの周波数・強度ノイズや、実験系の音響振動といった技術的なノイズによるものであることが明らかとなった。

Ornamental cabbage (Brassica oleracea var. acephala f. tricolor) cultivars of Japanese seed companies are grown in Europe. Unpleasant odors from cut flowers have become a problem in this region. We investigated volatiles emitted from cut flowers of ‘Hatsubeni’ and ‘Haresugata’, two major Japanese ornamental cabbage cultivars in Europe, by gas chromatography-mass spectrometry (GC-MS). Dimethyl disulfide was identified as the major odor-active component emerging from plants as well as from vase water. Vase water was a major source of unpleasant scents, and odor development was prevented by changing the water frequently. We tested the effects of potential suppressors of dimethyl disulfide emission from cut flowers applied to vase water. Cyprodinil, an inhibitor of the synthesis of dimethyl disulfide from methionine, and aminooxyacetic acid, an inhibitor of enzymatic reactions involved in dimethyl disulfide biosynthesis, did not show any effects. In contrast, isothiazolinonic germicide, a cut-flower preservative, inhibited the rot of cut ends of flower stems and reduced dimethyl disulfide emission from cut flowers by 30–40%. In addition, this germicide significantly inhibited emissions from vase water. Isothiazolinonic germicide is a promising candidate inhibitor of unpleasant scents from cut flowers of ornamental cabbage.

The aim of the present article is to review the cardioprotective properties of cannabinoids, with an emphasis on the signaling pathways involved. Cannabinoids have been reported to protect against ischemia in rat isolated hearts, as well as in rats and mice in vivo. Although these effects have been observed mostly with a pre-treatment of a cannabinoid, we report that the selective CB2-receptor agonist JWH133 is able to reduce infarct size when administered either before ischemia, during the entire ischemic period, or just upon reperfusion. Little is known about the signaling pathways involved in these cardioprotective effects. Likely candidates include protein kinase C (PKC) and mitogen-activated protein kinases (MAPK) since they are activated during ischemia-reperfusion and contribute to the protective effect ischemic preconditioning. The use of pharmacological inhibitors suggests that PKC, p38 MAPK, and p42/p44 MAPK (ERK1/2) contribute to the protective effect of cannabinoids. In addition, perfusion with JWH133 in healthy hearts caused an increase in both p38 MAPK phosphorylation level and activity, whereas the CB1-receptor agonist ACEA was associated with an increase in the phosphorylation status of both ERK1 and ERK2 without any change in activity. During ischemia, both agonists doubled p38 MAPK activity, whereas ERK1/2 phosphorylation level and activity during reperfusion were enhanced only by the CB1-receptor agonist. Finally, although nitric oxide (NO) was shown to exert both pro and anti-apoptotic effects on cardiomyocytes, with an apparently controversial effect on myocardial survival, our data suggest that NO may contribute to the cardioprotective effect of some cannabinoids.

A twelve-year-old, spayed female Yorkshire Terrier with intermittent vomiting was diagnosed with regional granulomatous enteritis through histopathological examination. On ultrasonography and computed tomography, a focal thickened duodenal wall showed a mass-like appearance with indistinct wall layers. Marked uptake of 18F-fluorodeoxyglucose was observed from the mass on positron emission tomography-computed tomography. Regional granulomatous enteritis is a rare form of inflammatory bowel disease and may have similar imaging features with intestinal tumors. This is the first study describing the diagnostic imaging features of ultrasonography, computed tomography and positron emission tomography-computed tomography of regional granulomatous enteritis in a dog.

網膜色素変性(RP)は網膜視細胞の機能が障害される遺伝性の疾患群であり、遺伝性失明、また、60歳以下視覚障害者の最大の原因である。原因となる遺伝子変異は多数解明されているものの遺伝子診断を一般的な臨床検査とするまでにはまだまだ多くの課題が残っている。まず、全身疾患を伴わないRPについてこれまでに原因遺伝子が判明しているのは少なくとも40個と多数ある。RP患者の半分以上は弧発や遺伝形式不明であり、遺伝子診断や遺伝カウンセリングはほぼできないのが現状であり、多数の患者が遺伝子診断及び遺伝カウンセリングを受けられないのである。そこで、我々は効率とコストの面を考えながらdHPLCを用いた方法で遺伝形式を問わず全てのRP患者における網羅的な変異解析を試みた。約15%の弧発例や遺伝形式不明の患者で原因と思われる遺伝子変異を検出。結果はその内、18個が新規変異であった(Journal of Medical Genetics,2008)。効果的な治療法が現れない現状では、世界の研究者はES細胞やiPS細胞からin vitro分化してきた視細胞あるいは前駆細胞などを網膜細胞移植の重要なソースと期待しており、我々は世界初マウス、サル及びヒトES細胞から網膜視細胞への分化を効率よくできた(小坂田、高橋らNat Biotech 2008)。この成果により、視細胞分化誘導法及び移植細胞源に関する問題を解決できた。しかし、胚性幹細胞の生命倫理学的な配慮と移植における免疫拒否などの問題は残っている。そこで最近、皮膚などの組織体細胞から再生医療の大きな武器である「多能性幹細胞」を人工的に作ることに成功した(山中)。我々はこのips細胞を用いて視細胞の前駆細胞及び視細胞への分化に取り組み、ヒトips細胞から視細胞の分化に成功した(Hirami et al.,unpublished)。更に小分子を用いて新たな分化方法を効率よくできた(OsaRada F,Jin ZB et al.,unpublished)。これらの研究結果から、網膜色素変性症患者に対して遺伝子診断の臨床応用をでき、また、網膜再生の移植ソースとして視細胞分化を効率よくできた。

A total of 658 cattle in 6 provinces in the Philippines were screened for Anaplasma marginale infection by using a diagnostic heat-shock operon (groEL) gene-PCR assay. The screening-positive samples were further tested using the major surface antigen protein 1a (Msp1a) gene-PCR assay. Screening PCR results showed 130 cattle (19.8%) were positive for the A. marginale infection. Subsequent amplification using the Msp1a gene only showed 93 samples (14.1%) to be positive. In addition, 37 tandem-repeat structures, including 20 novel structures, and 41 distinct genotypes were identified. Interestingly, multiple infections of 4 different genotypes were also observed in A. marginale-infected cattle. The present study demonstrated the prevalence and characterization of diverse genotypes of A. marginale in the Philippine cattle.

Minimal reports are available on the relationship between blood lipids such as cholesterol, triglycerides, high-density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) and acne. Most of available literature was about the effect of drugs used in acne treatment on these parameters. In this work we determined plasma total cholesterol, triglycerides, HDL-C and LDL-C levels in 166 (83 males and 83 females) newly diagnosed untreated Jordanian acne patients and compared with 105 (52 males and 53 females) of age and sex matched healthy controls. Results indicated that acne patients, males and females, had significantly low plasma HDL-C levels (p=0.000). Plasma total cholesterol, triglycerides and LDL-C levels were shown to be within the normal range except for triglycerides and LDL-C levels in severe acne cases for both sexes, were shown to be significantly elevated compared with those in healthy controls (p=0.004 and 0.000 consequently). It has been noticed that there was a trend for plasma HDL-C of acne patients to decrease as the severity of acne condition increases. Our results indicated that acne patients have significant changes in the plasma lipids profile that should be considered in the pathogenesis as well as in the treatment of acne.

脊椎動物の胚発生において神経板は、新しく転写因子SOX2を発現した神経系原基細胞を後部神経板に付加させていくことによって伸長する。このSox2発現細胞は、原条前方部の両側に位置し神経系・中胚葉・表皮系の共通の前駆体であるStem zoneから生み出される。Sox2の発現を制御するエンハンサーのうちN-1エンハンサーは、Stem zoneと一致する領域とそこから生み出された直後の神経系原基細胞で活性を持つ。このことから、N-1エンハンサーの活性化とSox2発現の直後に起こる遺伝子調節は、Stem zoneから神経系原基細胞を生み出す過程に直接的な役割を果たしていることが考えられた。そこで、Stem zoneから神経系原基細胞を生み出す分子機構を明らかにするために、N-1エンハンサー欠失マウス胚を作成し、解析を行った。N-1エンハンサー欠失マウス胚でSox2の発現は、N-1エンハンサーが活性を持つ領域で欠失した。この結果から後部神経板尾部におけるSox2の発現はN-1エンハンサーに依存的であることが示された。しかし、このSox2発現の欠失は他のエンハンサーによって補完され、結果的には正常に神経管が形成された。次に、後部神経板尾部におけるSox2依存的な遺伝子調節を明らかにするために、8.5日胚、8 somite stageのN-1エンハンサー欠失マウス胚を使ってMicroarray解析を行った。その結果、幾つかのSox2依存的に制御される遺伝子の候補を同定することができた。これらの候補遺伝子について、さらに解析を進めることが、神経系原基細胞が生み出される分子メカニズムを明らかにし、Stem zoneについても新たな知見をもたらすことが予想される。

News articles usually represent a biased viewpoint on contentious issues, potentially causing social problems. To mitigate this media bias, we propose a novel framework for predicting orientation of a news article by analyzing social user behaviors in Twitter. Highly active users tend to have consistent behavior patterns in social network by retweeting behavior among users with the same viewpoints for contentious issues. The bias ratio of highly active users is measured to predict orientation of users. Then political orientation of a news article is predicted based on the bias ratio of users, mutual retweeting and opinion analysis of tweet documents. The analysis of user behavior shows that users with the value of 1 in bias ratio are 88.82%. It indicates that most of users have distinctive orientation. Our prediction method based on orientation of users achieved 88.6% performance in accuracy. Experimental results show significant improvements over the SVM classification. These results show that proposed detection method is effective in social network.
