The effect of buthionine sulfoximine (BSO), a glutathione biosynthesis inhibitor, on the acute lethal toxicity and urotoxicity induced by cyclophosphamide (CPA) was examined in mice. Pretreatment of mice with BSO (500 mg/ kg, i.p.) 5 hr prior to CPA resulted in enhanced lethality and urotoxicity of CPA. In contrast, administration of cysteamine decreased the lethality and urotoxicity of CPA.

The effects of three stressors of different categories, namely cold exposure, immobilization, and lipopolysaccharide (LPS) treatment, on sympathetic nerve activity were examined by assessing its biochemical index norepinephrine (NE) turnover in peripheral organs of C57BL/6 mice. NE turnover was assessed by measuring the decrease in the organ NE concentration 3 h after inhibition of catecholamine biosynthesis with α-methyl-p-tyrosine. NE turnover in brown adipose tissue (BAT) in the room temperature (23°C) control group was as high as that in the cold exposure (4°C) group. Similarly, the mRNA level of the thermogenic marker uncoupling protein 1 (UCP1) in the room temperature control group was as high as that in the cold exposure group. As sympathetic stimulation upregulates the UCP1 mRNA level, we thought that sympathetic nerve tonus in BAT was already accelerated at room temperature. To exclude factors affecting basal sympathetic nerve activity, mice housed at thermoneutral temperature (30°C) were used as controls for the subsequent experiments. In this condition, cold exposure accelerated NE turnover in the BAT, as well as heart and pancreas. The corticosterone level showed a higher trend in the cold exposure group in comparison to the control group. Immobilization accelerated NE turnover in the spleen, pancreas, and white adipose tissue and elevated the corticosterone level. LPS (3 mg/kg, i.p.) did not affect NE turnover in all peripheral organs but elevated the corticosterone level. In summary, the sympathetic nervous and adrenocortical responses to three stressors differed greatly. In particular, sympathetic responses showed clear organ-specific acceleration patterns. This important feature may improve our understanding of the multiplicity of biological responses.

A 75-year-old woman with rheumatoid arthritis (RA) presented with long-term painful erythema on the right upper arm and left elbow. The patient was diagnosed with intralymphatic histiocytosis (ILH) based on the biopsy findings. Because the patient was unresponsive to single-agent treatment with methotrexate, infliximab and etanercept, we switched to tocilizumab (TCZ) treatment, which induced remission of the ILH. Our case suggests that TCZ may be a treatment option for ILH in patients with RA.

A 7-month-old castrated male French Bull dog was presented with vomiting, lethargy, anorexia and weight loss of 2 weeks duration. The patient’s history and clinical manifestations of suspected hepatopathy were subjected to ultrasonography, radiography, biochemical investigations and cytology of hepatic lesion. The cytologic impression was hepatic lymphoma, which was later confirmed by histopathology. The neoplastic cells were strongly diffusely immunoreactive for PAX5, but not immunoreactive for CD3, and B lymphocyte specific clonal proliferation was detected using by assay of antigen receptor rearrangement. Large numbers of immunoreactive mature non-neoplastic lymphocytes were admixed with the neoplastic cell population. Therefore, the immunohistochemical results were definitively consistent with a T-cell rich B-cell lymphoma (TCRBCL). This is the first description of a hepatic TCRBCL in a juvenile dog showing a poor response to aggressive chemotherapy.

最近のコンピュータネットワークではWWWなど様々な情報資源を提供するサーバ(情報ベース)が自然発生的に点在しており、ユーザか移動計算機からこうした情報資源を自由に検索・加工したい、という要求が大きい。しかし、これら情報ベースはユーザが望む利用手段や情報構造をあらかじめ用意していないことが多く、結果としてほぼ全ての情報を移動計算機側からブラウズしがちである。この問題を解決するためには一般に仲介者と呼ばれる補助機構が必要である。我々はスクリプト言語による遠隔プログラムを使ってこの機構を提案してきた。図1が提案した仲介者機構による検索手順である。すなわち、ユーザは未知な情報ベースに対し、(1)始めに、ユーザは情報ベースへそのクラス定義を問い合わせ、その仕様(i.e. 情報構造と利用手段)を教えてもらう。(2)次に、移動計算機欄で自分がやりたい検索スクリプトを合成し、(3)情報ベースに送って実行させる。この手法では、個々のユーザに依存した情報検索・加工手続き自体(「関心がある情報を探す」など)は移動計算機に載ってネットワーク内を移動し、必要な時に情報ベースへ送られる。従って情報ベース側ではそれ固有の検索手段以外はスクリプト言語の処理系だけを用意すれば良い。 我々はこの機構をPerlに永続オブジェクト操作機能をいれたスクリプト言語Persistent Perlを用いて試作している。しかし、移動計算機上にあらかじめ持っていた検索スクリプトと、情報ベース固有の情報との二つを合わせて使うようなスクリプトをユーザが直接Persistent Perlを使って記述するのは難しい。本稿ではオブジェクト指向SQLを使ってこうした検索スクリプトを合成する方式を論じる。

A 52-year-old woman with a high serum alkaline phosphatase (ALP) level underwent a liver biopsy, which showed diffuse heavy deposition of Aκ amyloid, and was diagnosed as having immunoglobulin light chain (AL) amyloidosis. Although she received high-dose melphalan with stem cell transplantation and achieved a hematologic complete response (CR), her ALP level began to increase one year after treatment. Further examinations revealed that she was still in a CR state with dominant bone-type ALP, and re-biopsied liver specimens demonstrated marked regression of amyliod deposition, providing important evidence that the turnover of hepatic amyloid proteins can actually occur more rapidly than previously thought.

The high level of genetic diversity in the microflora of the gastrointestinal tract has the potential to provide numerous beneficial functions to the host. Thus it is now acknowledged that the complexity in animal functioning is linked to the interacting microbiome in the gut. Despite the importance of gut microbiome, there is a lack of information concerning the microbial communities in the pig gut during the weaning transition. This study describes the fecal microbial shifts of healthy piglets during the weaning transition using barcoded pyrosequencing of the prokaryotic 16S rRNA gene. Fecal samples were obtained from 15 piglets during the pre-weaning period (fourth week after birth) and post-weaning (sixth week after birth) and were subjected to community genomic DNA extraction for pyrosequencing analysis. As the piglets underwent the weaning transition a trend toward increased bacterial diversity was observed, based on species abundance as measured by the Shannon-Weaver index. Firmicutes (54.0%) and Bacteroidetes (59.6%) were the most dominant phyla during pre-weaning and post-weaning, respectively. During the weaning transition a distinct shift from Bacteroides to Prevotella as the most abundant genus was observed. Additionally, we detected a number of abundant gut bacterial species that have not been reported previously. Clostridium rectum, C. clostridioforme, C. lactatifermentans and Butyricimonas virosa were uniquely detected prior to weaning while Roseburia cecicola and Blautia wexlerae were detected during the post-weaning period only.

Maternal undernutrition can affect offspring’s physical status and various health parameters that might be transmittable across several generations. Many studies have focused on undernutrition throughout pregnancy, whereas maternal undernutrition prior to pregnancy is not sufficiently studied. The objective of our study was to explore the effects of food restriction prior to and during pregnancy on body weight and longevity of the second generation offspring. Adult female Wistar rats (“F0” generation) were 50% food restricted for one month prior to pregnancy (pre-pregnancy) or during pre-pregnancy and pregnancy. The third group was fed normally (control). The first generation offspring were normally fed until the 6th month of age to produce the second generation offspring; namely, the first-generation female rats were mated with male breeders from outside the experiment. The second generation offspring thus obtained were observed until natural death (up to 36 months). Compared to the controls, the second-generation male offspring whose “grandmothers (F0 females)” undernourished only during pre-pregnancy were significantly heavier from the 8th month of age, whereas no significant weight difference was found in the male offspring whose “grandmothers” were food-restricted during pre-pregnancy and pregnancy. Shorter lifespan was observed in the second-generation male offspring of “grandmothers” that were food-restricted either during pre-pregnancy or during pre-pregnancy and pregnancy. By contrast, no differences in body weight and lifespan were observed in all second-generation female offspring. In conclusion, maternal caloric restriction prior to pregnancy increases the body weight and shortens the longevity of the second-generation male offspring, indicating the sex-dependent transgenerational effect of maternal caloric restriction.

Switching mode CMOS frequency multipliers are studied in sub-Terahertz regime. Analysis on the multiplier architectures and optimal gate bias at CMOS switch are investigated to maximize output power at designated harmonics. Utilizing a differential pair, a 195 GHz tripler having a hair-pin filter is designed to maximize 3rd harmonics with −14.8 dB of conversion gain (CG) from Pin = +13 dBm of the balanced input, while the 260 GHz quadrupler utilizes quadruple-push pairs which achieves CG = −16 dB from two +13 dBm of the balanced I/Q driving signals in a 65 nm digital CMOS process.

For determination of the endogenous and exogenous causes of somaclonal variation in in vitro culture, a bioassay system was developed using the variegated Saintpaulia (African violet) ‘Thamires’ (Saintpaulia sp.), having pink petals with blue splotches caused by transposon VGs1 (Variation Generator of Saintpaulia 1) deletion in the promoter region of flavonoid 3',5'-hydroxylase. Not only true-to-type but also many solid blue and chimeric plants regenerate in vitro-cultured explants of this cultivar. Using multiplex PCR that enables the determination of these variations, we attempted to evaluate the effects of four candidate triggers of mutation: pre-existing mutated cells, shooting conditions in vitro or ex vitro, cutting treatment of explants, and addition of plant growth regulators (PGRs) to the medium. The percentages of somaclonal variations among total shoots regenerated from leaf segments and stamens were 46.6 and 56.5, which were higher than the percentages expected from pre-existing mutated cells (3.6 and 1.4, respectively). These results indicate that pre-existing mutated cells are not a main cause of somaclonal variations. The percentage of somaclonal variation was independent of culture conditions for mother plants; the mutation percentages of adventitious shoots regenerated from ex vitro- and in vitro-grown leaves were 9.2% and 8.5%, respectively. In addition, the percentage of somaclonal variations of adventitious shoots regenerated under in vitro conditions from the in vitro grown mother plants was also low, at 4.9%. This indicates that the in vitro condition itself is not a main cause of somaclonal variation. However, when adventitious shoots were regenerated from 10 × 5-mm cut-leaf laminas on a PGR-free medium, the percentage of somaclonal variation was 26.4%. In addition, the percentage of somaclonal variations dramatically increased when PGRs were added to the medium for both leaves and leaf segments (39.9 and 46.6, respectively). The bioassay system using Saintpaulia ‘Thamires’ will enable the screening of many environmental factors because of its rapidity and ease of use and will facilitate the development of a new tissue culture technology for avoiding mutation.

Background: Tension-type headache (TTH) is a disease with a great incidence on quality of life and with a significant socioeconomic impact. Objectives: The aim of this review is to determine the effectiveness of physical therapy by using manual therapy (MT) for the relief of TTH. Data sources: A review was done identifying randomized controlled trials through searches in MEDLINE, PEDro, Cochrane and CINAHL (January 2002 - April 2012). Study selection: English-language studies, with adult patients and number of subjects not under 11, diagnosed with episodic tension-type headache (ETTH) and chronic tension-type headache (CTTH) were included. Data extraction: Initial search was undertaken with the words Effectiveness, Tension-type headache, and Manual therapy (39 studies). In addition, a search which included terms related to treatments such as physiotherapy, physical therapy, spinal manipulation was performed (25 studies). Results: From the two searches 9 studies met the inclusion criteria and were analysed finding statistically significant results: 1) myofascial release, cervical traction, neck muscles trigger points in cervical thoracic muscles and stretching; 2) Superficial heat and massage, connective tissue manipulation and vertebral Cyriax mobilization; 3) cervical or thoracic spinal manipulation and cervical chin-occipital manual traction; 4) massage, progressive relaxation and gentle stretching, program of active exercises of shoulder, neck and pericranial muscles; 5) massage, passive rhythmic mobilization techniques, cervical, thoracic and lumbopelvic postural correction and cranio-cervical exercises; 6) progressive muscular relaxation combined with joint mobilization, functional, muscle energy, and strain/counterstrain techniques, and cranial osteopathic treatment; 7) massage focused on relieving myofascial trigger point activity; 8) pressure release and muscle energy in suboccipital muscles; 9) combination of mobilizations of the cervical and thoracic spine, exercises and postural correction. All studies used a combination of different techniques and none analyzed treatments separately, also all the studies have assessed aspects related to TTH beyond frequency and intensity of pain. Conclusions: The findings from these studies showed evidence that physiotherapy with articulatory MT, combined with cervical muscle stretching and massage are effective for this disease in different aspects related with TTH. No evidence was found of the effectiveness of the techniques applied separately.

Recent epidemiological studies showed that coffee consumption is associated with a lower risk of type 2 diabetes, presumably due to suppression of excess fat accumulation in adipocytes. However, the mechanism underlying the effect of coffee on adipocyte differentiation has not been well documented. To elucidate the mechanism, we investigated the effect of coffee on the differentiation of mouse preadipocyte 3T3-L1 cells. Coffee reduced the accumulation of lipids during adipocytic differentiation of 3T3-L1 cells. At 5% coffee, the accumulation of lipids decreased to half that of the control. Coffee also inhibited the expression of the peroxisome proliferator-activated receptor γ (PPARγ), a transcription factor controlling the differentiation of adipocytes. Furthermore, coffee reduced the expression of other differentiation marker genes, aP2, adiponectin, CCAAT-enhancer-binding protein α (C/EBPα), GLUT4, and lipoprotein lipase (LPL), during adipocyte differentiation. Major bioactive constituents in coffee extracts, such as caffeine, trigonelline, chlorogenic acid, and caffeic acid, showed no effect on PPARγ gene expression. The inhibitory activity was produced by the roasting of the coffee beans.

Plasma doping and laser annealing are successfully integrated into the conventional p-metal–oxide–silicon field effect transistor (PMOSFET) process to form ultra shallow junction (USJ). Comparing with the conventional combination of ion implantations and rapid thermal annealing (RTA), junction depth ($X_{\text{J}}$) and sheet resistance ($R_{\text{S}}$) are reduced. Also, significant improvement of the short channel effects without the degradation of on-current is observed.

The title imides possessing a planar acenaphthofluoranthene moiety were prepared by the Diels–Alder reaction of diacenaphtho[1,2-b:1′,2′-d]thiophene and maleic anhydride and subsequent treatment with amines. The reaction with maleimide afforded the corresponding 1:2 adduct displaying intense blue fluorescence in solution. Although the imides crystallize yellow needles, their amorphous solids are red. Concentration-dependent 1H NMR spectra of a N-octyl derivative reveal self-aggregation behavior in CDCl3.

Recently, the control of non-CO2 greenhouse gases has attracted interest as a way to prevent global warming. Several research studies on the development and assessment of technologies to control CH4 and N2O emissions by human activities are under way. In the wastewater treatment field, the development of anoxic/oxic processes and the introduction of bacteria that effectively prevent N2O emission have been studied. Moreover, it has been clarified that eco engineering technologies such as artificial wetlands and soil trench systems are very effective, especially in developing countries. As for landfill disposal, CH4 oxidation by cover soil and the use of CH4 gas as a source of energy have very effectively reduced the emission of CH4. In the combustion field, it was clarified that N2O emissions vary according to the structure of each combustor and its operational conditions. And research and development concerning combustors and their optimum operational conditions are under way. Concerning automobile sources, characteristics of N2O emission from gasoline fueled vehicles installed with a catalyst were examined. Three way catalysts and so on are now being developed. Chemical industries, especially adipic acid production are another non-negligible N2O source, and cracking process related countermeasures have been applied. CH4 and N2O emissions from agricultural land are strongly dependent on the oxidation-reduction environment of soil and on fertilization, so they must be suitably managed. The development and management of ruminants feeding and suitable treatment of animal waste are very important ways to control CH4 and N2O from livestock. Research projects to improve the CH4 and N2O emission-absorption inventory, and to estimate the suitable technologies to control CH4 and N2O emission are now in progress. Combining these projects appropriately will develop technologies and systems and will make great contributions to the control of the emission of GHGs and to global warming.
