Objective: Although generic drugs are considered bioequivalent to the original drugs, for formulations such as adhesive agents, the sensation during use (hereinafter, “sensation”) and handling characteristics (hereinafter, “handling”) are usually not investigated during the approval process.  Therefore, we established new drug evaluation criteria for transdermal adhesive agents, which included those relevant to handling and sensation, and investigated their usefulness.Methods: This study assessed four test samples: the original loxoprofen sodium 100 mg tape (hereinafter, “LX-P 100 mg”) and three generic versions considered economically acceptable by our hospital.  The formulations were evaluated based on our criteria for generic drugs.  The handling and sensation were evaluated using a questionnaire with six parameters related to sensation and seven parameters related to handling.  The enrolled subjects comprised 20 pharmacists from our hospital.  Furthermore, a parallel evaluation using loxoprofen sodium 50 mg tape (hereinafter, “LX-P 50 mg”) was performed to explore the variance in the results of each individual criterion.Results: The evaluation revealed differences between the formulations with regard to the thickness of the adhesive agent and stability after opening the package.  Various differences were observed by evaluation of the handling and sensation.  Based on the parameters used, the differences between the various formulations of LX-P 100 mg occurred in the following criteria: tactile sensation commensurate to the degree of adhesion; superficial resemblance to other agents; ease of adhesion; and ease of package opening.  For LX-P 50 mg, the differences were found in superficial resemblance to other agents and ease of liner removal.Conclusion: Our newly established criteria for the evaluation of LX-P 100 mg revealed differences between the samples, which were used to identify products with poor handling and sensation.  Thus, this study successfully demonstrated the usefulness of the criteria for the purpose of medicine selection.

Chemical substitution is an effective way to improve electrocatalytic properties in transition metal oxides. We investigate the synergistic effect between Fe4+ and Co4+ ions on the catalytic activity for oxygen evolution reaction (OER) in the Fe–Co-mixed perovskite oxide CaFe1−xCoxO3. The OER activity of CaFe1−xCoxO3 is substantially increased by small amounts of Co (Fe) doping into CaFeO3 (CaCoO3), leading to the superiority compared to the pure Fe and Co perovskite oxides. The x dependences of the OER overpotential and specific activity for CaFe1−xCoxO3 (0.05 ≦ x ≦ 0.95) are expressed by constant offset from the weighted average between CaFeO3 and CaCoO3, which can be interpreted to be the synergistic effect between Fe4+ and Co4+ ions on OER activity. The absence of the optimum x for the highest activity for CaFe1−xCoxO3 contrasts with the volcano-like plots reported in various mixed-metal oxides. First-principle calculations using the special quasirandom structure models on CaFe1−xCoxO3 (x = 0.03–0.5) demonstrate that about half the amount of Fe4+ is electronically activated to possess smaller charge-transfer energies, corroborating the enhancement of catalytic activity in CaFe1−xCoxO3. These findings provide new insight into the synergistic effects in complex transition metal oxide catalysts.

A new method to produce highly pure 99mTc of maximum 500Ci(1.85e+13Bq) per batch from a low specific activity 99Mo made from natural-Mo(n, γ) method is presented in this study.Authors developed a novel procedure to provide 99mTc of high specific activity and purity from 99Mo with a low specific activity obtained by the(n, γ) method. This process named TcMM(technetium master milker) with a new principle found out by the authors involves the selective adsorption of 99mTc onto an activated charcoal column, followed by an elution of 99mTc by warm alkaline solution, and final purification through an alumina column, which would be applicable to routine production of a large amounts of 99mTc from natMo(with 99Mo) within a short processing time and with a low cost.By the experiment using a high radioactivity of 99Mo of 1e+12Bq levels, a highly pure 99mTc free from natMo(with 99Mo) was obtained in a sterile saline solution within 30 - 50min. as the chemical form of 99mTcO4−, and the average 99mTc milking rate repeated 10 times got 93.5% and the radiochemical purity of the range of 6N - 7N.In the labeling experiments using several radiopharmaceutical kits,99mTc collected from the TcMM process gave the target medicines in a highly radiochemical purity.As the result of investigating the disposition behaviors of the above-mentioned 99mTc-pharmaceuticals using small animals(mouse), 99mTc collected from TcMM gave the same result as it eluted from the conventional 99mTc generator.The findings in this study suggest that the present method using natural isotopic 98Mo(n, γ)99Mo reaction would constitute as an alternative to the Fission Method using HEU to provide 99mTc useful for clinical studies.

Hydrolyzed proteins are often prescribed for dogs with food hypersensitivity in food elimination programs. However, the potential of these diets to stimulate lymphocyte-mediated hypersensitivity is currently unknown. In this study, two commercially available hydrolyzed diets for dogs, D-1 (Aminopeptide Formula Dry, Royal Canin Japon, Tokyo, Japan), and D-2 (Canine z/d Ultra Dry, Hill’s-Colgate (Japan) Ltd., Tokyo, Japan), were analyzed to identify residual proteins or peptides, as well as activated helper T-lymphocyte reactions in dogs with suspected food hypersensitivity. Proteins and peptides with molecular weights >1 kDa (majority 1.5–3.5 kDa) were detected in both diet extracts with sodium dodecyl sulfate polyacrylamide gel electrophoresis, and size exclusion chromatography. When peripheral blood mononuclear cells (PBMC’s) from 316 dogs with suspected food allergies were cultured with hydrolyzed diet extracts, flow cytometry analysis revealed detectable levels of CD25low helper T-lymphocytes stimulated by D-1 and D-2 in 91 of 316, (28.8%), and 75 of 316 (23.7%) samples, respectively. These data indicated that the extracts contained proteins or peptides large enough to activate the lymphocytes. The percentages of CD25low helper T-lymphocytes stimulated by D-1 and D-2 extracts increased to 38.7% and 29.6%, respectively, in 186 of the original 316 samples (186/316, 58.9%), also reactive to poultry-related antigens. Thus, both poultry-related antigens, and D-1 and D-2 diet extracts may activate helper T-lymphocytes. These results demonstrate that hydrolyzed diets may contain proteins that stimulate helper T-lymphocytes, and may not be effective for treating all dogs with food hypersensitivity.

Aim: Previous clinical trials have demonstrated the effectiveness of eicosapentaenoic acid (EPA) in preventing cardiovascular events. The aim of the present study was to investigate the effects of EPA treatment on the accumulation of coronary atherosclerotic plaque using optical coherence tomography (OCT). Methods: A total of 46 acute coronary syndrome (ACS) patients without dyslipidemia were divided into two groups: those who received 1,800 mg/day of EPA (n=15) or the control group (n=31). Serial OCT examinations were performed at baseline and after eight months of follow-up. The target for the OCT analysis was non-culprit plaque with a percent diameter of stenosis of 30% to 70% in non-culprit vessels of ACS. Results: Between the baseline and follow-up visits, the serum EPA levels increased (50±26 mg/dL to 200±41 mg/dL, p<0.001) in the EPA group, although they did not change in the control group. According to the OCT analysis, the lipid arc did not change in the EPA group (131±52 degrees to 126±54 degrees, p=0.106) or the control group (137±50 degrees to 138±50 degrees, p=0.603). In contrast, the fibrous cap thickness significantly increased in both the EPA group (169±70 μm to 201±49 μm, p<0.001) and the control group (164±63 μm to 174±72 μm, p=0.018); however, the relative change in the fibrous cap thickness was significantly greater in the EPA group than in the control group (131±35% vs. 106±15%, p=0.001). Conclusions: In the present study, the administration of EPA for eight months significantly increased the fibrous cap thickness in patients with coronary atherosclerotic plaque.
