Liquid water is considered to be critical for life. Gibbs free energy is another factor that is important to sustain life for long durations. Gibbs free energy is obtained by reactions between reductants and oxidants, or from any other non-equilibrium state of matter. As an example, aerobic organisms use carbohydrates and oxygen to obtain energy. Many types of chemoautotrophic mechanisms are known for this process as well. On the surface of Mars, methane and oxidative compounds such as ferric oxide, sulfate and perchloride, which could provide redox-derived Gibbs free energy, have been detected. Iron-dependent methane oxidizing bacteria have been found in marine environments on Earth. This finding suggests the possible presence of methane-oxidizing bacteria on the Mars surface, if the local thermal environment and other resources permit proliferation and metabolism of bacteria. Our project aims to search for methane-oxidizing microbes on the Mars surface. Martian soil will be sampled from a depth of about 5 - 10 cm below the surface, where organisms are expected to be protected from the harsh hyper-oxidative environment of the Mars surface. Small particles less than 0.1 mm or 1 mm will be sieved from the sample, before being transferred to the analysis section by a micro-actuator. The particles will be stained with a cocktail of fluorescent reagents, and examined by fluorescence microscopy. A combination of fluorescent dyes has been selected to identify life forms in samples. A membrane-specific dye or a combination of dyes will be used to detect membranes surrounding the "cell". An intercalating fluorescent dye such as SYBR Green will be used to detect genetic compounds such as DNA. A substrate dye that emits fluorescence upon cleavage by a catalytic reaction will be used to detect the catalytic activity of the "cell". A combination of staining reagents has been chosen based on the definition of life. A membrane separating a cell from the ambient environment may lead to identification of an "individual". DNA or genetic material is required for "replication" of the life form. Catalytic reactions carried out by enzymes drive "metabolism". This combination of strategies will also be useful for detecting pre-biotic organic material as well as remnants of ancient life. Hydrolysis of the polymers in the "cell" followed by HPLC or soft ionization MS for amino acid analysis will be effective for examining whether Martian life is identical to or different from terrestrial life. The number and type of the amino acids as well as their chirality will be analyzed to distinguish whether the polymers are contaminants from Earth.

Background:Prasugrel has been shown to provide more potency and less variability than clopidogrel, but its potential temporal variability has not been described.Methods and Results:We conducted a prospective open-label study, evaluating platelet reactivity overtime in acute coronary syndrome (ACS) patients on aspirin and clopidogrel (n=60) or prasugrel (n=61), after a percutaneous coronary intervention (PCI). Blood samples were taken at discharge and at 3 and 6 months. Platelet function tests included VerifyNow (VN-P2Y12), and Multiplate Aggregometry (MEA). By means of VN-P2Y12, prasugrel patients displayed significantly (P<0.001) higher platelet inhibition than clopidogrel patients over time, although there were not significant differences using MEA. Prasugrel patients showed higher platelet inhibition at baseline than at 3 months (59.3±8.1 vs. 105.0±49.2; P<0.001), without significant change at 6 months (107.9±72.0; P=0.919 vs. 3 months). Clopidogrel patients showed a similar trend (160.1±65.1, 184.8±62.7 and 185.0±53.3; baseline vs. 3 months P=0.060; 3 months vs. 6 months P=0.974). High platelet reactivity (HPR) was shown in 16.3% prasugrel patients, with no patient consistently remaining in HPR over time. HPR was detected in 36.6% of the clopidogrel patients, being consistently observed in 15.0% of them. Low platelet reactivity (LPR) was detected in 60.5% prasugrel and 9.8% clopidogrel patients.Conclusions:Prasugrel patients showed less temporal variation than patients on clopidogrel in terms of HPR. In contrast, higher variability in LPR was detected in prasugrel patients for up to 6 months’ follow-up.

The purpose of this study was to evaluate and to compare the fracture load and the fracture pattern of monolithic and veneered zirconia posterior fixed dental prostheses (FDPs). Twenty standardized steel dies were prepared to receive posterior 3-unit FDPs. Specimens were randomly divided into 2 groups (n=10): (1) Lava Zirconia, and (2) Lava Plus. All FDPs were cemented using glass ionomer cement and subjected to thermal and mechanical cycling at 5–55ºC with a 30-s dwell time for 120,000 masticatory cycles. All specimens were subjected to a three-point bending test until fracture. Data were statistically analyzed using Student’s t test, paired t-test and Weibull statistics (α=0.05). No differences were observed in fracture load between the groups. Veneering ceramic fractured before than framework in veneered zirconia group. The fracture pattern was different. The tested groups demonstrated clinically acceptable fracture load values. Monolithic zirconia solves the chipping problem.

Fas-mediated apoptosis has been proposed to play an important role in homeostasis. Fas triggers apoptosis after stimulation by its ligand FasL or the Fas ligand agonist anti-Fas antibody through a mitochondria-dependent or -independent pathway, and MSSP has been identified as a transcription factor that regulates the c-myc gene and was later found to positively or negatively regulate a variety of genes, including alpha-smooth actin, MHC class I, MHC class 2 and the thyrotropin receptor. We further found that expression of the Fas gene was repressed, resulting in abrogation of the Fas-mediated induction of apoptosis both in Mssp-knockout mice and primary thymocytes. MSSP was then found to stimulate promoter activity of the Fas gene by binding to a specific region. In this study, to identify the MSSP-dependent Fas-induced apoptosis pathway, primary fibroblasts from MSSP (+/+) and MSSP (-/-) cells were treated with the combination of interleukin 1-beta and interferon-gamma and expression of the Fas gene was examined. The results showed that the Fas gene was expressed at the same levels in the two cell types. Furthermore, when these cells were treated with the anti-Fas antibody, it was found that cytochrome C was not released in the cytosol and that activations of caspase 8 and caspase 3 occurred in primary fibroblasts from MSSP (+/+) cells but not from MSSP (-/-) cells. These results indicate that Fas-mediated apoptosis induced by MSSP occurs independently of mitochondria.

Eddy transport of atmospheric water vapor from the tropics is important for rainfall and related natural disasters in the middle latitudes. Atmospheric rivers (ARs), intense moisture plumes typically associated with extratropical cyclones, often produce heavy precipitation upon encountering topography on the west coasts of mid-latitude North America and Europe. ARs also occur over the northwestern Pacific and sometimes cause floods and landslides over East Asia, but the climatological relationship between ARs and heavy rainfall in this region remains unclear. Here, we evaluate the contribution of ARs to the hydrological cycle over East Asia using high-resolution daily rainfall observations and an atmospheric reanalysis during 1958–2007. Despite their low occurrence, ARs account for 14–44% of total rainfall and 20–90% of extreme heavy-rainfall events during spring, summer and autumn seasons. AR-related extreme rainfall is especially pronounced over western-to-southeastern slopes of terrains over the Korean Peninsula and Japan, owing to strong orographic effects and a stable direction of low-level moisture flows. A strong relationship between warm-season AR heavy rainfall and preceding-winter El Niño is identified since the 1970s, suggesting the potential of predicting heavy-rainfall risk over Korea and Japan at seasonal leads.

Short chain fatty acids (SCFAs) play an important role in the maintenance of colonic homeostasis, and their depletion has been reported in various gastrointestinal disorders. Inflammatory colorectal polyps (ICRPs) are a recently recognized disease specific to miniature dachshunds (MDs), and fecal dysbiosis with a reduction of SCFA-producing bacteria has been reported with this disease. Therefore, this study was performed based on the hypothesis that a reduced SCFA concentration associates with the development of ICRPs. We recruited 11 ICRP-affected MDs and 25 control MDs. Their fecal SCFA concentrations and bacterial proportions were quantified using high performance liquid chromatography and quantitative real-time PCR, respectively. The feces of ICRP-affected MDs contained lower amounts of propionic acid and lower proportions of Bifidobacterium than the feces of control MDs. Furthermore, fecal proportions of Bifidobacterium, Firmicutes and Lactobacillus exhibited significant positive correlations with fecal concentrations of total SCFAs and/or propionic acid; fecal Escherichia coli proportions correlated negatively with fecal concentrations of total SCFAs, as well as acetic, propionic and butyric acid. This result indicates an association between fecal dysbiosis and fecal SCFA concentrations; these phenomena may contribute to ICRP pathogenesis in MDs. Potential therapeutic targeting of the reduced propionic acid concentration using probiotics, prebiotics or SCFA enemas merits further study.

Canine histiocytic sarcoma (HS) is a malignancy originating from the histiocytic cell lineage and characterized by poor response to chemotherapy and short survival time. Mutation of the TP53 gene and its association with poor prognosis has been reported in several canine tumors. However, the mutation of this gene has not been investigated in canine HS. The aim of this study was to examine a TP53 gene mutation in dogs with HS. Aberrations of the TP53 gene were examined by polymerase chain reaction-single strand conformational polymorphism analysis and DNA sequence analysis, revealing mutations of the TP53 gene in 12 (46%) of 26 dogs affected by HS. The incidence of the TP53 gene mutation was relatively high in canine HS compared with other canine tumors. Among these mutations, 10 of 12 dogs (83%) with a TP53 gene mutation harbored the same mutation: a 2-base (AT) insertion in exon 5, resulting in the introduction of a stop codon (c.446_447insAT, p.Tyr150SerfsX8). Further studies are needed to examine the functional change due to the mutation and its association with the pathogenesis of canine HS.

Extracorporeal elimination therapy of metabolic diseases may be taken into consideration if the substrate to be removed is present in the circulation in an elevated concentration and if the endogenous synthe-sis rate does not exceed the amount of re-moval. Systemic amyloidoses are characte-rized by the presence of specific protein pre-cursor molecules in the serum. Polymeriza-tion leads to the formation of fibrils which are deposited as amyloid in the extracellular space. Depending on the variables of the disease to be treated, it may generally be possible to delay its progression, achieve a halt in progression or even initiate regression of amyloid deposition. Two patients, a mother and her son, 54 and 25 years of age, with familial amyloidosis due to the transthyretin met30 variant (Por-tuguese type), after a disease duration of 9 and 6 years, respectively, received regular plasma exchanges in two-week intervals over a period of P/2 years. Patients' plasma was exchanged at volumes of 155% and replaced by 5% human albumin solution. Transthyretin levels were reduced by 50 - 70% during each plasma exchange procedure. Clinical, neurological, opthalmological, laboratory, histological, and electron microscopical examinations were performed prior to, and after one year of plasma exchange therapy. The overall results indi-cate only slight progression in the older pa-tient and stability in the younger patient. Athird patient, 56 years of age, with immunglobulin-type (AL) amyloidosis due to monoclonal gammopathy of unknown significance, with cardiac involvement and rapid deterioration of renal function under chemotherapy was treated initially by plasma exchange over a period of 1 1/2 years without further cytotoxic treatment, and subsequently by Ig adsorption using protein A or anti-IgG columns. Para-protein levels were reduced to 20-25% of the pretreatment values by plasma exchange on two consecutive days. Renal function, clinical and echo-cardiographical signs of cardiac failure remained stable. Our experience having patients with two different forms of systemic amyloidosis sufficiently is encouraging to discuss the possible therapeutic role of extracorporeal elimination in delaying the progression of amyloidosis.

A number of familial and twin studies have assessed the relative contribution of genetic and environmental factors to physical performance or its-related traits, and a significant genetic component has been predicted to affect phenotypes. A twin study indicated that the heritability of athletic status is 66%. Thus, genetic factors appear to be important for determining elite sporting performance. To date, over 200 genes in both nuclear DNA and mitochondrial DNA (mtDNA) have been reported to be associated with physical performance and health-related fitness. However, most studies have been reported in European populations. In the Asian population, only 3 genetic loci have been linked to physical performance, including angiotensin I-converting enzyme and α-actinin-3 genetic polymorphisms and mtDNA polymorphisms (mitochondrial haplogroups). In this review, we discuss the genetics of elite sporting performance, particularly in Asian populations, and their effects on lifestyle-related diseases such as type 2 diabetes and age-related muscle wasting (i.e., sarcopenia).

[Purpose] The purpose of this study was to examine the effects of galvanic vestibular stimulation on event-related potentials. [Subjects and Methods] Forty normal female adult subjects were randomly distributed to a galvanic vestibular stimulation application group (20 subjects) and sham group (20 subjects). For galvanic vestibular stimulation application, a positive electrode was applied to the right mastoid process, and a negative electrode was applied to the left mastoid process; simulation was applied for 10 minutes. A test was conducted on the N100 and P300 components of the event-related potentials before and after galvanic vestibular stimulation. [Results] The N100 latency showed statistically significant differences in interaction effects between time and group in the F3, F4, Fz, and Pz areas. The P300 latency showed the same results in the Fp1 and Fp2 areas, the N100 amplitude showed the same results in the Fp2, Fz, and Pz areas; and the P300 amplitude showed the same results in the Pz area. [Conclusion] These results suggest that galvanic vestibular stimulation may play a positive role in the N100 and P300 components of the event-related potentials of the cerebral cortex related to decision-making in matching words with images.

The Autism-Spectrum Quotient (AQ) children's version has confirmed reliability and validity in the UK. In the current study, the children's AQ was administered in Japan to investigate whether the UK results are found in a very different culture. Two groups of children from primary and secondary schools were assessed: Group 1 (n=81) children with Autism Spectrum Disorders (ASD, including Asperger Syndrome and high-functioning autism); Group 2 (n=372) randomly selected controls, age-matched with Group 1. The children with ASD had a mean AQ score of 31.9 (SD=6.69), which was significantly higher than controls (mean AQ=11.7, SD=5.94). Males scored significantly higher than females in the control group, but not in the ASD group. The pattern of difference between the Japanese clinical group and the control group was remarkably similar to the findings in the UK.

[Purpose] The objective of this study was to investigate the effects of spinal support device (SSD) on pain and hamstring extensibility in patients with non-specific low back pain (NSLBP). [Subjects and Methods] 20 patients with NSLBP were recruited and randomly assigned to either the SSD group or the control group. In the SSD group, SSD was applied; in the control group, bed rest in supine position was performed. Both groups underwent treatment 20 min/day, 3 times a week, for a duration of 4 weeks. To assess the hamstring extensibility, sit and reach test (SRT) was performed. To assess pain pressure threshold (PPT) of the sacroiliac joint, a pressure algometer was used. Visual analog scale (VAS) was used to quantify pain. [Results] The SSD group showed a significant improvement in sacroiliac joint pain with increased VAS, and the control group showed a significantly increased VAS after intervention. In the SSD group, VAS was significantly increased, but SRT was not changed compared with the control group. [Conclusion] These results demonstrated that an application of SSD effectively attenuates low back pain. Therefore, SSD may be a suitable intervention for pain control in patients with NSLBP.

The non-photosynthetic diatoms Nitzschia spp. are known to have evolved from photosynthetic species to heterotrophic species by the loss of photosynthesis. We investigated their ability to tolerate wide ranges of temperatures and salinities. Nitzschia spp. were capable of surviving or thriving even at 5°C and 35°C. In addition, these diatoms were also capable of surviving at salinities of 0.5 and 12.0, while thriving at those from 1.0 to 9.0. Such tolerance to a wide range of temperatures and salinities would allow these non-photosynthetic diatoms to thrive in mangrove estuaries, where environmental conditions often drastically fluctuate. Our experiments revealed that the growth rates of the non-photosynthetic diatoms were larger than those estimated based on cell volumes and temperature, suggesting that these non-photosynthetic diatoms may be an important group of organisms contributing to material circulation by growing heterotrophically in mangrove estuaries.

低温・低圧・無加圧下のAg焼結接合技術の革新を進め、その基本メカニズムを高分解能TEM観察とシミュレーションから明らかにした。Ag粒界へ酸素の吸収、粒界液相形成、粒界液相が噴火し表面堆積する。この一連の反応で粒子間低温焼結や接合界面形成が実現しており、これを”Nano Volcanic Eruption”と名付けた。新技術により、250℃を超える耐熱性が達成されている。また、雰囲気との反応をCu粒子へ展開し、その可能性を見出している。以上のように、極限環境に対応する新接合技術の開発、そのメカニズム解明、さらに、Cuへの展開など、学術的に新たな現象解明から実用レベルの技術開発まで繋げている。

The aim of this study was to describe a ventral laparoscopic technique for bilateral medial iliac lymphadenectomy in dogs. Twelve intact male purpose-bred research dogs weighting less than 15 kg were positioned in dorsal recumbency, and a 3-portal technique was used. Bilateral dissection was performed with vessel-sealing devices while tilting the surgical table by up to 30° towards the contralateral side of the target medial iliac lymph node (MILN) without changing the surgeon’s position. Using a ventral laparoscopic approach, bilateral MILNs were identified and excised in all dogs. The mean time for unilateral and bilateral MILN dissections were 9.7 ± 3.8 min and 21.0 ± 6.0 min, respectively. The mean times for the right and left MILN dissections were 10.8 ± 4.3 min and 9.8 ± 2.5 min, respectively. The mean total surgery time was 43.7 ± 7.7 min. In total, 26 MILNs were dissected. Several complications, including, mild to moderate capillary hemorrhage from perinodal fat and vessels (controlled laparoscopically), mild spleen trauma caused by the first trocar insertion, and capsular damage of MILNs, were observed. However, there were no other major complications. All MILN samples were evaluated and deemed suitable for histopathologic diagnosis. Laparoscopic excision of MILNs is a useful method of excisional biopsy for histopathologic diagnosis. Using this ventral laparoscopic approach with the 3-portal technique, bilateral MILN dissection suitable for obtaining histopathologic samples could be achieved in a short time in dogs weighing less than 15 kg.

Shiba dogs are predisposed to chronic enteropathy (CE) and have poorer prognosis than other dog breeds. The objective of this study was to investigate the significance of polymerase chain reaction for antigen receptor rearrangement (PARR) results on clinical findings and prognosis of Shiba dogs with CE. We retrospectively collected data on 22 Shiba dogs diagnosed as having CE. Fifty-nine percent of the dogs had clonality-positive results on PARR analysis. Furthermore, on histopathology, epitheliotropic behavior of small lymphocytes of the intestinal mucosa was observed significantly more frequently in dogs with clonal rearrangement of antigen receptor genes (P=0.027). The median overall survival time of clonality-positive dogs was 48 days (range, 4–239 days), compared to 271 days (range, 45–1,316+ days) in clonality-negative dogs. The median overall survival time of epitheliotropism-positive dogs was 76 days (range, 30–349 days) compared to 239 days (range, 4–1,316+ days) for epitheliotropism-negative dogs. Statistical analysis revealed that the clonality-positive result was associated with significantly shorter survival time (P=0.036). In contrast, presence or absence of epitheliotropism had no statistically significant effect on survival time (P=0.223). These cases might appropriately be diagnosed as small T-cell intestinal lymphoma; there are some common clinical and pathogenic features with human enteropathy-associated T-cell lymphoma type 2. The pathogenesis and poor prognosis for Shiba dogs with CE seem to be associated with this type of lymphoma, although further investigation is warranted.

Contrast-enhanced ultrasonography (CEUS) with microbubbles as a contrast agent allows the visualization and quantification of tissue perfusion. The assessment of canine intestinal perfusion by quantitative CEUS may provide valuable information for diagnosing and monitoring chronic intestinal disorders. This study aimed to assess the repeatability (intraday variability) and reproducibility (interday variability) of quantitative duodenal CEUS in healthy dogs. Six healthy beagles underwent CEUS three times within one day (4-hour intervals) and on two different days (1-week interval). All dogs were sedated with a combination of butorphanol (0.2 mg/kg) and midazolam (0.1 mg/kg) prior to CEUS. The contrast agent (Sonazoid®) was administered using the intravenous bolus method (0.01 ml/kg) for imaging of the duodenum. Time-intensity curves (TIC) were created by drawing multiple regions of interest (ROIs) in the duodenal mucosa, and perfusion parameters, including the time-to-peak (TTP), peak intensity (PI), area under the curve (AUC), and wash-in and wash-out rates (WiR and WoR, respectively), were generated. Intraday and interday coefficients of variation (CVs) for TTP, PI, AUC, WiR, and WoR were <25% (range, 2.27–23.41%), which indicated that CEUS was feasible for assessing duodenal perfusion in healthy sedated dogs. Further study of CEUS in dogs with chronic intestinal disorders is necessary to evaluate its clinical applicability.
