Our previous study showed that oridonin isolated from <i>Rabdosia rubescens</i> enhanced phagocytosis of apoptotic cells by macrophage-like U937 cells through tumor necrosis factor (TNF) α and interleukin (IL)-1β release. In this study, we further investigated signaling events involved in oridonin-augmented phagocytosis. Phagocytic stimulation was significantly suppressed by inhibitors, including a phosphoinositide 3-kinases (PI3K) inhibitor (wortmannin), a protein kinase C (PKC) inhibitor (stauroporine), and a phospholipase C (PLC) inhibitor (U73122). Exposure of U937 cells to oridonin caused an increase in PKC activity time- dependently, which was prevented by pretreatment with inhibitors of PI3K and PLC. Simultaneously, the activation of protein kinase B (PKB/Akt) and the increased expression of PLCγ2 were also blocked by wortmannin. In addition, an extracellular signal-regulated kinase (ERK) MAPK inhibitor, PD98059, suppressed oridonin-augmented phagocytosis, whereas the p38 MAPK inhibitor (SB203580) and c-Jun N-terminal kinase (JNK) MAPK inhibitor (SP98059) had no inhibitory effect. Furthermore, pretreatment of U937 cells with anti-TNFα and anti-IL-1β antibodies blocked oridonin-induced phagocytic stimulation as well as phosphorylation of ERK, but did not block the activation of PKC, indicating that these signaling events are triggered by oridonin, whereas secreted TNFα or IL-1β only activate the ERK-dependent pathway. Taken together, oridonin is suggested to enhance phagocytosis of apoptotic bodies by activating PI3K, PKC, and ERK-dependent pathways.<br>

Transpiration, the evaporation from aerial parts of plants, is major driving force for the roots to uptake water from the soil. Aquaporins, the water channel proteins, are thought to play crucial roles in regulation of root hydraulic conductivity. Here we demonstrate that the evaporative demand plays a dominant role in the induction of specific aquaporins in rice (Oryza sativa) roots in seedlings grown hydroponically and then transferred to an open field. We found that the abundance of the root-specific aquaporin transcripts, OsPIP2;4, OsPIP2;5 and OsTIP2;1 at 8 a.m. is highly correlated with the evaporative demand in the morning (evaluated from potential evaporation averaged over 4 h before root sampling). Aquaporin isoforms with higher correlation to potential evaporation showed higher diurnal amplitude of their expression. Our study suggests that rice plants sense daily weather and responded to it by adjusting the expression of specific root aquaporin genes.

A sufficient number of functional live hepatocytes delivered to a recipient is necessary for cell therapy. Preventing cell viability loss during the cell injection process is important to improve the clinical outcomes of hepatocyte transplantation. The critical location of cell viability loss is important to identify the causal relationship between the viability loss and cell injection process. In this study, the critical location of cell viability loss was determined experimentally in a rectangular microchannel by microscopic high-speed camera observations. Live hepatocyte distributions were investigated upstream and downstream, and measured on three planes, top, center, and bottom, under horizontally or vertically supplied conditions of the syringe orientation. Sedimented and uniform dispersion conditions of the live hepatocyte distribution at upstream of the microchannel were classified according to observations at horizontal and vertical syringe orientations, respectively. Higher hepatocyte viability loss was found under the sedimented condition. The results suggested that the critical location of hepatocyte viability loss was on the bottom plane of the microchannel. Furthermore, physical causes of the hepatocyte viability loss were found by micro-scale observations of the cell velocity and diameter during the cell injection process. This information may contribute to development of a guideline for the cell injection process to improve hepatocyte transplantation.

A 16-year-old castrated male mongrel cat presented with swelling under the left pinna and a 3 -month history of voice change. Laryngeal endoscopy revealed circumferential oedema around the arytenoid cartilages and hypersecretion of saliva. Histopathological examination of the mass around the left ear canal was considered the primary lesion that originated from cutaneous apocrine adenocarcinoma or parotid gland adenocarcinoma, and it metastasized to the larynx, lung and medial retropharyngeal lymph nodes. This report provides new insights into feline laryngeal diseases which could result in laryngeal metastasis with slight mucosal irregularity alone and without obvious radiographic abnormalities. Therefore, histopathological examination should be performed when a cat presents clinical signs such as stridor, dysphonia, or voice change without any mass-forming laryngeal lesion.

Over 700 bacterial species have been detected in the oral cavity. Several studies have suggested that periodontitis is associated with systemic disorders such as diabetes mellitus, indicating a key role for oral microbiota in human health. However, the relationship between oral microbiota and diabetes has not been well clarified. Therefore, we conducted microbiome analysis of saliva samples obtained from 15 elderly residents (3 with type 2 diabetes mellitus [DM] and 12 without diabetes [non-DM]) at three different nursing homes, as well as 9 young healthy controls (HC). Genomic DNA was extracted from each sample, and then the V4 region of the 16S rRNA gene was amplified and sequenced. Alpha diversity, in terms of operational taxonomic unit richness, was significantly higher in samples from the non-DM group than in those from the HC group. Weighted UniFrac distance analysis showed that salivary microbial communities in the DM group were separately clustered. Furthermore, in the DM group, Actinomyces and Selenomonas showed significantly higher abundance, whereas Alloprevotella showed significantly lower abundance, relative to the non-DM group. Although our findings were limited by the small sample size, oral bacterial diversity in the DM group was clearly different from that in the non-DM group.

The purpose of this research was to compare the accuracy of newly described P wave-related parameters (P wave area, Macruz index and mean electrical axis) with classical P wave-related parameters (voltage and duration of P wave) for the assessment of left atrial (LA) size in dogs with degenerative mitral valve disease. One hundred forty-six dogs (37 healthy control dogs and 109 dogs with degenerative mitral valve disease) were prospectively studied. Two-dimensional echocardiography examinations and a 6-lead ECG were performed prospectively in all dogs. Echocardiography parameters, including determination of the ratios LA diameter/aortic root diameter and LA area/aortic root area, were compared to P wave-related parameters: P wave area, Macruz index, mean electrical axis voltage and duration of P wave. The results showed that P wave-related parameters (classical and newly described) had low sensitivity (range=52.3 to 77%; median=60%) and low to moderate specificity (range=47.2 to 82.5%; median 56.3%) for the prediction of left atrial enlargement. The areas under the curve of P wave-related parameters were moderate to low due to poor sensitivity. In conclusion, newly P wave-related parameters do not increase the diagnostic capacity of ECG as a predictor of left atrial enlargement in dogs with degenerative mitral valve disease.

To investigate the postprocedural cardiovascular events and vascular outcomes, including hand ischemia and neurological compromise, after transulnar (TU) catheterization in ipsilateral radial artery occlusion.Previous randomized trials have shown that the transulnar (TU) approach for coronary angiogram and intervention has safety and outcomes similar to those of the transradial (TR) approach. However, the safety of the TU procedure when ipsilateral radial artery occlusion occurs is unknown.We retrospectively reviewed 87 TU cases with ipsilateral radial artery occlusion confirmed by a forearm angiogram. Eighty percent of these patients had a history of ipsilateral radial artery cannulation or surgery. We avoided the use of over-sized sheaths or applied a sheathless approach during surgery.No ulnar artery occlusion was observed by subsequent Doppler ultrasound or pulse oximetry. No patient developed hand ischemia or serious complications requiring surgery or blood transfusion during the follow-up period of 32.2 ± 24.0 months. Review of the preprocedural forearm angiograms showed that 95.7% of the patients possessed significant collaterals supplying flow from the interosseous artery to the occluded radial artery remnant. Thus, the blood circulation to the palmar arch and digital vessels was maintained even when the ulnar artery was temporarily occluded by an in-dwelling ulnar arterial sheath.TU catheterization was safe in patients with coexisting ipsilateral radial artery occlusions and feasible for use in complex intervention procedures. Cautious manipulation of ulnar artery cannulation and hemostasis helped decrease the risk of hand ischemia.

<b>Background</b> The aim of the present study was to assess the feasibility and usefulness of the arterial stiffness index (ASI) measured non-invasively by computerized oscillometry and by comparing it with the pulse wave velocity (PWV). <b>Methods and Results</b> The study group comprised 60 consutive patients who underwent coronary angiography and whose aorto-femoral PWV were obtained with a Judkins catheter. The ASI was obtained using Cardio Vision <sup>®</sup> MS-2000 (IMDP, Las Vegas, NV, USA): (i) baseline (ASI-B); (ii) hyperemia induced by compression of the arm with cuff pressure for 5 min (ASI-H); and (iii) sublingual nitroglycerin (ASI-N). In total, 34 patients had significant coronary artery disease (CAD). The PWV and all ASI were higher in patients with CAD than in those without CAD (ASI-B, 85.9±57.8 vs 48.2±24.5, p=0.001; ASI-H, 98.1±49.8 vs 48.1±21.3, p<0.01; ASI-N, 66.7±55.7 vs 33.2±27.9, p=0.002). However, only ASI-B and ASI-H were positively correlated to the PWV (ASI-B, r=0.27, p=0.03; ASI-H, r=0.49, p=0.001; ASI-N, r=0.19, p=0.16). The ASI was increased after hyperemia in patients with CAD (ASI-H, 85.9±57.8 to 98.1±49.8, p=0.01), but not in patients without CAD (ASI-H, 48.2±24.5 to 48.1±21.3, p>0.01). After adjusting their age, only ASI-H was correlated to the presence of CAD (r=0.33, p<0.01). <b>Conclusions</b> It is feasible and useful to use the ASI for detection of atherosclerotic coronary disease. The findings of ASI-H suggests that in addition to stiffening of the arterial wall itself, the impairment of flow mediated vasodilation, because of endothelial dysfunction, further increases the arterial stiffness. This paper considers the infinite alternative case to prove the existence of continuous social welfare aggregation that is anonymous and respects the unanimity. It clarifies the controversy between Chichilnisky (1982, QJE) and Huang for their contradictory results for the continuum case. Compared to their topological frameworks, the infinite alternative case is easier to understand and pinpoint their difference.

Although mutations in ACAN, FGFR3, NPR2, and SHOX typically lead to skeletal dysplasia, and mutations in GHRHR, GH1, GHR, STAT5B, IGF1, IGFALS, and IGF1R usually underlie hormonal defects of the growth hormone (GH)-insulin-like growth factor 1 (IGF1) axis, such mutations have also been identified in patients with idiopathic short stature (ISS). Of these, SHOX abnormalities are known to account for a certain percentage of ISS cases, whereas the frequency of mutations in the other 10 genes in ISS cohorts remains unknown. Here, we performed next-generation sequencing-based mutation screening of the 10 genes in 86 unrelated Japanese ISS patients without SHOX abnormalities. We searched for rare protein-altering variants. The functional significance of the identified variants was assessed by in silico analyses. Consequently, we identified 18 heterozygous rare variants in 19 patients, including four probable damaging variants in ACAN, six pathogenicity-unknown variants in FGFR3, GHRHR, GHR, and IGFALS, and eight possible benign variants. Pathogenic variants in NPR2, GH1, and IGF1 were absent from our cohort. Unlike previously reported patients with ACAN mutations, our four patients with ACAN variants manifested non-specific short stature with age-appropriate or mildly delayed bone ages, and had parents of normal stature. These results indicate that ACAN mutations can underlie ISS without characteristic skeletal features, and that such mutations are possibly associated with de novo occurrence or low penetrance. In addition, our data imply that mutations in FGFR3, NPR2, and GH-IGF1 axis genes play only limited roles in the etiology of ISS.

The problem of beamforming and related beamspace high resolution direction-of-arrival (DOA) estimation is studied in this paper. All beamspace processing methods are based on the beam outputs and the beampattern design plays an important role in providing high quality beam output data for further processing. Three typical situations which are frequently encountered in practical sonar system working environment and the most widely studied MUSIC algorithm are considered herein. First, when isotropic noise is the dominant noise at sensors, conventional beamforming techniques provide the optimum performance in the sense that DOA estimate is the ML estimate. Good DOA estimates are obtainable by applying MUSIC to the beam outputs directly. Then, uncorrelated interferes with much higher strength than the wanted signals are assumed to be present in the sidelobe region, and low sidelobe Dolph-Chebyshev and adaptive MVDR beampatterns are designed to guarantee the performance of MUSIC. And finally, the robustness of conventional techniques is combined with the adaptivity of MVDR beamforming to deal with the situation when the interfere in the sidelobe region is strongly correlated with one of the wanted sources. Performance in all three situations is studied with numerical examples.

目的 乳幼児期の栄養摂取は,個々の子どもの成長•発達段階に合わせて適切に対応することが大切である。そこで,今回,乳幼児期の食生活状況について,乳歯萌出状況と調理形態•調理方法などとの関連について検討を行った。方法 東京都 K 区保健所および各保健センターにおいて,1 歳 2 か月児歯科健診を受診した455人に,歯科医師による歯科健診と保護者への自記式調査票を用いて,離乳食の開始時期,離乳食の進行の目安,現在の食事の調理形態などについて調査を行った。記入漏れがあった18人および在胎期間が36週未満の出生児17人を除く420人を解析対象とした。結果 離乳食の開始時期は,生後 5, 6 か月齢頃が81.4%と最も多く,離乳の進め方の目安は,「月齢」と回答した者が最も多かった(71.2%)。乳歯萌出状況により,前歯上下 8 本(乳中切歯 4 本と乳側切歯 4 本)が生え揃っていない段階(ステージI:27.4%),前歯上下 8 本が全て生え揃っているが奥歯(第一乳臼歯)がまだ生え揃っていない段階(ステージII:61.9%),奥歯(第一乳臼歯)が上下 4 本すべて生え揃っている段階(ステージIII:10.7%)の 3 段階に分類した。「おかずの固さの目安」は,ステージI,II,IIIすべてにおいて「歯ぐきでかみつぶせる」がそれぞれ53.5%, 54.4%, 40.0%と最も多かったが,まだ第一乳臼歯 4 本が生え揃っていないステージI,IIにおいて,「奥歯でかみつぶせる」と回答した者が,それぞれ14.0%, 15.1%も認められた。さらに,「大人と同じ固さ」と答えた割合が,ステージI,II,IIIで,それぞれ7.0%, 9.7%, 24.4%であった。また,調理の味付け(塩味,しょうゆ味)については,「大人と同じ」と答えた割合がステージI,II,IIIで,それぞれ13.2%, 17.3%, 22.2%であった。結論 今回の調査結果により,乳歯萌出状況は個人差が大きく,個々の口腔の発達段階•咀嚼機能を把握せずに,調理形態•調理方法が進められていることが推察され,今後の食育支援の必要性が示された。

Quisqualis indica (QI) has been used for treating disorders such as stomach pain, constipation, and digestion problem. This study was aimed to evaluate the therapeutic efficacy of QI extract on treating benign prostatic hyperplasia (BPH) in LNCaP human prostate cancer cell line and a testosterone-induced BPH rat model. LNCaP cells were treated with QI plus testosterone propionate (TP), and androgen receptor (AR) and prostate specific antigen (PSA) expression levels were assessed by Western blotting. To induce BPH, the rats were subjected to a daily subcutaneous injection of TP (3 mg/kg) for 4 wk. The rats in treatment group were orally gavaged with QI (150mg/kg) together with the TP injection. In-vitro studies showed that TP-induced increases in AR and PSA expression in LNCaP cells were reduced by QI treatment. In BPH-model rats, the prostate weight, testosterone in serum, DHT concentration and 5α-reductase mRNA expression in prostate tissue were significantly reduced following the treatment with QI. TP-induced prostatic hyperplasia and the expression of proliferating cell nuclear antigen (PCNA) and cyclin D1 were significantly attenuated in QI-treated rats. In addition, QI induced apoptosis by up-regulating caspase-3 and -9 activity and decreasing the B-cell lymphoma 2 (Bcl-2)/Bcl-2-associated X protein (Bax) ratio in prostate tissues of BPH rats. Further investigation showed that TP-induced activation of AKT and glycogen synthase kinase 3β (GSK3β) was reduced by QI administration. Therefore, our findings suggest that QI attenuates the BPH state in rats through anti-proliferative and pro-apoptotic activities and might be useful in the clinical treatment of BPH.

The objective of this study was to measure differences between arterial and venous blood gas parameters and to evaluate whether arterial blood gas values can be estimated from venous blood in Asiatic black bears (ABBs). Twelve healthy captive ABBs (8 males and 4 females; 8–16 years; 76.8–220 kg) were included in this study. The bears were immobilized with medetomidine and zolazepam-tiletamine using a dart gun. Arterial and venous samples were collected simultaneously at 5 and 35 min after recumbency (5- and 35-min points). Partial pressure of oxygen (PO2), partial pressure of carbon dioxide (PCO2), pH, bicarbonate (HCO3–), total carbon dioxide (TCO2), oxygen saturation of hemoglobin (SO2) and base excess (BEecf) were analyzed using a portable blood gas analyzer. There was no marked difference in measured and calculated variables over time in both venous and arterial blood except for PO2. However, arterial PO2, SO2 and pH were significantly higher and arterial PCO2, TCO2 and HCO3– were lower than those of venous samples at both 5- and 35-min points. In the regression analysis to estimate arterial values from venous values, PCO2, TCO2, HCO3–, BEecf and pH significantly showed over 0.45 in coefficient of determination value (R2), and there were little differences between actual and predicted arterial values. Although there were limits in venous gas values replaced those of arterial blood, if we could not get the arterial samples, the regression formulas for arterial values from venous blood in this study would be useful clinically, except for PO2 and SO2.

A new blue petunia cultivar, TX-794 bred by Takii & Co., Ltd, has a sweet floral fragrance different from conventional petunia cultivars that typically have no odor or have a somewhat unfavorable smell. Since fragrant bedding plants suitable for summer cultivation are rare, we expect that the fragrance will enhance the value of TX-794. The characteristics of emitted scent compounds from TX-794 were evaluated in this study. The major scent compound was phenylacetaldehyde, a C6-C2 aromatic compound that was emitted at higher levels in TX-794 compared to conventional petunia cultivars. iso-Eugenol, a C6-C3 aromatic compound, is the major scent compound in conventional petunia cultivars. These results suggest that TX-794 has a high capacity to biosynthesize C6-C2 aromatic compounds, resulting in a significantly different scent compound composition compared to conventional petunia cultivars. Time-course analysis of scent compound emission revealed that the largest release of scent compounds by TX-794 occurs during the switch from light periods to dark periods. Soon after flowering, that is, in the light period on the day of anthesis, the major scent compound was p-cresol. Thereafter, the primary scent compound was phenylacetaldehyde or methyl benzoate with 2-phenylethanol occupying the next position. Since a bed or a container of petunias has flowers that are primarily over 2-days old, the basic fragrance of TX-794 plants is derived from phenylacetaldehyde, which has a hyacinth-like scent, 2-phenylethanol, which has a rose-like scent, and methyl benzoate, which has a dry fruit-like scent.

Objective The predictors of in-hospital outcomes after primary percutaneous coronary intervention (PCI) for acute myocardial infarction (AMI) complicated with heart failure or cardiogenic shock at presentation remain unclear. Methods Using the AMI-Kyoto Multi-Center Risk Study database, the clinical background characteristics, angiographic findings, primary PCI results, and in-hospital prognoses were retrospectively compared between primary PCI-treated AMI patients with a Killip class status of ≥2 (Killip 2-4 patients, n=390) and those with a Killip class 1 status (Killip 1 patients, n=1,057). Results The Killip 2-4 patients were more likely to have a higher age and proportion of women and exhibited a higher prevalence of previous myocardial infarction, diabetes mellitus and chronic kidney disease or anemia on admission, lower systolic blood pressure (SBP) values on admission, a higher rate of multivessels or left main trunk as the culprit artery, a larger number of diseased vessels, a lower Thrombolysis In Myocardial Infarction (TIMI) grade in the infarct-related artery (IRA) before/after primary PCI and a significantly higher in-hospital mortality rate than the Killip 1 patients. According to a multivariate analysis, age was found to be an independent positive predictor of in-hospital mortality, while admission SBP was an independent positive predictor of in-hospital survival in both groups. In contrast, anemia on admission was found to be an independent predictor of in-hospital death, while the TIMI 3 flow in the IRA after PCI was found to be an independent factor for survival in the Killip 2-4 patients, but not the Killip 1 patients. Conclusion Anemia on admission and the final TIMI 3 flow in the IRA are critical determinants of in-hospital death in AMI patients with a Killip class status of ≥2 undergoing primary PCI.

The risk of invasive fungal infections (IFIs) is extremely high in patients with hematological malignancies due to the prolonged and profound neutropenia and immunosuppression after chemotherapy and hematopoietic stem cell transplantation. There has been increasing interest in mucormycosis despite its relatively uncommon occurrence, because occasional breakthrough infections have been observed under anti-<I>Aspergillus</I> prophylaxis. The aggressive nature of mucormycosis easily leads to high mortality because of delays in diagnosis and incorrect treatment decisions, which are due in part to lack of adjunctive diagnostic tools and having similar clinical and radiological features with invasive aspergillosis. The only currently available antifungals against Mucorales in Japan are amphotericin B formulations. Thus, comprehensive therapeutic strategies, including surgery, should be considered to achieve a successful outcome.

In the wiretap model of secure communication, Alice is connected to Bob and Eve by two noisy channels. Wyner's insight was that the difference in noise between the two channels can be used to provide perfect secrecy for communication between Alice and Bob, against the eavesdropper Eve. In Wyner's model, the adversary is passive. We consider a coding-theoretic model for wiretap channels with active adversaries who can choose their view of the communication channel and also add adversarial noise to the channel. We give an overview of the security definition and the known results for this model, and discuss its relation to two important cryptographic primitives: secure message transmission and robust secret sharing.
