Angiotensin-converting enzyme (ACE) is a key enzyme in the renin-angiotensin system (RAS). ACE2 is a newly identified member of the RAS. The present immunohistochemical study focused on changes in intrarenal ACE and ACE2 immunoreactivity in feline and canine chronic kidney disease (CKD). ACE immunoreactivity was predominantly observed in the brush border of the proximal tubules in dogs and cats. ACE immunoreactivity was lower in CKD kidneys than in normal kidneys, and quantitative analysis demonstrated negative correlations between ACE and renal tissue damage in dogs. ACE2 immunoreactivity was also detected in the proximal tubules; it increased or decreased with CKD in dogs, depending on the renal region assessed. The changes in ACE and ACE2 in CKD were associated with the plasma creatinine concentration in dogs. Findings from dogs with glomerulonephritis were similar to those from dogs with non-glomerulonephritis. The present study suggests that changes in the intrarenal expression of ACE and ACE2 contribute to the pathological mechanisms of canine CKD, but not to the mechanisms of feline CKD.

Copper(I)–dioxygen reactivity studies utilizing new binucleating ligands are described. When catalytic quantities of either CuI or CuII salts were introduced to LH2, which is a dinucleating ligand with two tridentate donors connected via a methylene group at a bis(2-pyridyl)methane junction, dioxygen addition caused complete oxygenation to give bis(2-pyridyl)ketone product L=O. Isolated dicopper(I) complex [CuI2(LH2)]2+ (1) also reacted with excess O2 to give L=O. Additional observations were consistent with a mechanism that does not involve “oxygen-activation” in this process. Ketone hydration and gem-diol(ate) copper(II)-coordination occurred by addition of copper(II)–perchlorate to L=O, giving [CuII2{L(OH)(O−)}Cu2II(−OClO3)]2+ (4), with a bridging alkoxide moiety (X-ray). Oxygenation of [CuI2(L=O)(CH3CN)2]2+ (2) (2/O2 = 0.5, i.e., Cu/O2 = 4:1 manometry), gave [CuII2{L(O−)2}(−OClO3)]+ (5), where both oxygen atoms of the doubly deprotonated gem-diolate bind separate copper(II) ions; mass spectrometric data showed that one of the two oxygen atoms of the gem-diolate L(O−)2 came from O2. An oxo–dicopper(II) species is suggested as an important intermediate, which is supported by the observation that reaction of 2 with NO or iodosylbenzene also produced 5 in high yields. Complexes 4 and 5 are acid–base conjugate pairs and can be readily interconverted using Et3N or HClO4(aq) reagents.

Fcα/μR (CD351) is an Fc receptor for both IgA and IgM and forms an atypical dimer that is resistant to reduction by 2-mercaptoethanol or boiling. We previously demonstrated that the cytoplasmic portion of Fcα/μR is required for dimer formation and for its efficient cell-surface expression. However, the biochemical nature of these phenomena has not been determined. By using a BW5147 mouse cell line expressing deletion mutants of the cytoplasmic region of Fcα/μR, we found that the region spanning amino acids 504–523 was required for efficient cell-surface expression, whereas the region spanning amino acids 481–490 was required for dimmer formation. Immunoblotting analyses of transfectants simultaneously expressing Flag-tagged Fcα/μR and hemagglutinin-tagged Fcα/μR suggested that Fcα/μR does not form homodimers. Instead, our data suggest that Fcα/μR forms heterodimers with an as-yet-unknown molecule with a molecular weight of 60–70 kDa.

Mutations and polymorphisms of factor H gene (FH1) are known to be closely involved in the developmentof atypical hemolytic uremic syndrome (aHUS). Several groups have identified disease risk mutations andpolymorphisms of FH1 for the development of aHUS, and have investigated frequencies of aHUS in a numberof ethnic groups. However, such studies on Japanese populations are limited. In the present study,we analyzed FH1 in Japanese aHUS patients and healthy volunteers, and examined whether those variantsimpacted on a tendency for the development of aHUS in Japanese populations. Similar to previousstudies, we found that a high frequency of FH1 mutations, located in exon 23 of FH1, encodes short consensusrepeat 20 in C-terminal end of factor H molecule in patients with aHUS (40%), but not in healthyvolunteers. Interestingly, no significant differences in frequency of well-known disease risk polymorphismsfor aHUS were observed between healthy volunteers and aHUS patients. Our results suggestedthat although FH1 mutations relates to the development of Japanese aHUS in accordance with other ethnicstudies, other factor may be required for factor H polymorphism to be a risk factor of Japanese aHUS.

Neutrophils play a central role in host defenses against infectious microbial pathogens. However, molecular basis for their triggering signals has been incompletely understood. Here, we show molecular and functional characteristics of myeloid-associated immunoglobulin-like receptor (MAIR)-IV. MAIR-IV was preferentially expressed on neutrophils in the peripheral blood, bone marrow, peritoneal cavity and spleen. MAIR-IV physically associates with immunoreceptor tyrosine based activating motif (ITAM)-bearing adaptor FcRγ chain. Cross-linking MAIR-IV with anti-MAIR-IV monoclonal antibody significantly induced secretion of proinflammatory cytokines TNF-α and IL-6 from neutrophils. Thus, MAIR-IV may regulate activation of neutrophils and play an important role for innate immunity.

Fcα/μR is expressed not only in lymphoid, but also in non-lymphoid organs, including kidney. However, molecular and functional characteristics of Fcα/μR, particularly expressed in non-lymphoid organs, have remained unclear. Here we identified an isoform of Fcα/μR in the murine kidney on the C57BL/6J background. The kidney expressed only the isoform, which was not expressed in other lymphoid and non-lymphoid organs, and this isoform binds both IgA and IgM. Immunohistochemical analyses suggested that the Fcα/μR isoform was expressed in the tubular epithelial cells, but not in the glomeruli. This was confirmed by flow cytometry analysis of isolated tubular epithelial cells and by RT-PCR analyses using the separately excised glomerular and tubular regions by the laser microdissection system. These results suggest that Fcα/μR may not be involved in IgA deposition in glomerular mesangium cells in IgA nephropathy. Rather, it may play an important role in immunity in the renal tubular regions.

The protective effect of cetraxate, an antiulcer and antigastritis agent, on HCl·ethanol-induced gastric lesions was investigated in rats. Oral administration of 1 ml of HCl·ethanol (60% ethanol in 150 mM HCI) induced within 1 hr linear hemorrhagic necrosis in the gastric mucosa. Either oral or intraperitoneal treatment with cetraxate (30-300 mg/kg) significantly inhibited such macroscopic gastric lesions in a dose-related manner, and the inhibition at the oral highest dose (300 mg/kg) was practically complete. Histological analysis also confirmed that cetraxate effectively prevented deep mucosal necrosis, but showed that it was without protective effect on the surface epithelial disruption and submucosal edema in response to HCl·ethanol. The antilesion activity of cetraxate was of statistically significance for at least 3 hr after a single injection, and it was hardly affected by the removal of the gastric contents just prior to application of the necrotizing agent. However, subcutaneous treatment of rats with indomethacin (5 mg/kg) resulted in a partial but significant attenuation in the protection afforded by cetraxate, suggesting that dual mechanisms related and unrelated to endogenous prostaglandins may be involved in its protective activity. The results demonstrate that cetraxate is a potent cytoprotective agent effectively preventing the formation of gastric mucosal necrosis induced by HCl·ethanol.

本研究は、グラフェンを利得媒質とする新規なテラヘルツ(THz)レーザーの創出に挑んだものである。第一に、光学励起したグラフェンの過渡応答におけるTHz帯誘導放出の室温観測に成功し、自ら発見したレーザー理論を実証した。第二に、グラフェン表面プラズモンポラリトンの巨大利得増強作用を理論発見し、独自の光ポンプ・近接場THzプローブ分光法により初めて実証に成功した。第三に、独自開発した高品質エピタキシャルグラフェン製膜技術とレーザー素子加工技術を用いて電流注入型グラフェンレーザー素子を試作評価し、100Kの低温下ながら、5.2THzの単一モード発振に初めて成功した。第四に、グラフェンTHzレーザー設計論を構築するとともに、より高利得化が可能な独自構造を理論実験両面から明らかにし、室温THzレーザー発振実現の見通しを得た。

Intraparticle distribution of adsorbate amount in cyclic adsorption and desorption is simulated by two methods: rigorous numerical solution of the particle-phase diffusion equation and the linear driving force (LDF) approximation. It becomes clear that the conventional value of 15 <i>D</i><sub>s</sub>/<i>R</i><sup>2</sup> as the intraparticle mass transfer coefficient, <i>k</i><sub>s</sub><i>a</i><sub>v</sub>, in the LDF method is not advisable for the simulation of transient adsorption and desorption. A mass transfer coefficient defined by including the cycle time is thus proposed. In this new conception, <i>k</i><sub>s</sub><i>a</i><sub>v</sub> increases with decreasing cycle time and approaches π<sup>2</sup><i>D</i><sub>s</sub>/<i>R</i><sup>2</sup> with increasing cycle time. Simulations of cyclic mode by the LDF method using this new <i>k</i><sub>s</sub><i>a</i><sub>v</sub> agree well in the steady state with that obtained by numerical solution of the diffusion equation. In unsteady-state operation, however, the two simulations do not coincide with each other because of overestimation of driving force for adsorption and underestimation for desorption in the LDF method.

若年性汎発性ニキビダニ症は犬で頻繁にみられる疾患であり,遺伝的素因や免疫学的素因が関与する疾患である。本症は伝染性疾患でも人獣共通感染症でもないが,重篤で時に生命予後に影響することもある。本症はあらゆる犬種に発症しうるが,一部の報告では好発犬種に関する記載が見られる。本報告では1998年から2006年までの間に,アルゼンチン・ラブラタ国立大学獣医学部附属小動物病院で記録された499例について解析した。全ての症例において,ニキビダニ症があらゆる皮膚疾患よりも初発したと考えられ,最終的には本症と診断された。499例中28例が若年性汎発性ニキビダニ症と診断された。他の研究データと比較したところ,本研究ではボクサー,ジャーマン・シェパードおよび雑種犬に,本症が好発することが示された。本研究の成果は,若年性汎発性ニキビダニ症が一部の犬種に好発するという仮説を支持するものと考えられた。

Evidence indicates low 25-hydroxyvitamin D [(25(OH)D] concentrations in European adolescents. Identification of potential determinants is therefore essential to guide public health initiatives aiming at optimizing vitamin D status across Europe. The aim of the study was to identify potential influencing factors of 25(OH)D concentrations in European adolescents aged 12.5 to 17.5 y, participating in the multi-centre cross-sectional Healthy Lifestyle in Europe by Nutrition in Adolescence (HELENA) study. A subset of 1,006 participants (46.8% males) was drawn from the main study. Measures of body composition, biochemical markers, socioeconomic status, dietary intake, physical activity, fitness, sleep time and vitamin D genetic polymorphism (rs1544410) were assessed. Stepwise multivariate linear regression analysis was conducted stratified by gender. In males, linear regression of 25(OH)D, suggested that (1) winter season (β=−0.364; p<0.01), (2) higher latitudes (β=−0.246; p<0.01), (3) BMI z-score (β=−0.198; p<0.05) and (4) retinol concentration (β=0.171; p<0.05) independently influenced 25(OH)D concentrations. In females, (1) winter season (β=−0.370; p<0.01), (2) sleep time (β=−0.231; p<0.01), (3) supplement intake (β=0.221; p<0.05), (4) flexibility (β=0.184; p<0.05), (5) body fat % (β=0.201; p<0.05) (6), BMI z-score (β=−0.272; p<0.05), (7) higher latitudes (β=−0.219; p<0.01) and (8) handgrip strength (β=0.206; p<0.05) independently influenced 25(OH)D concentrations. Season, latitude, fitness, adiposity, sleep time and micronutrient supplementation were highly related to 25(OH)D concentrations found in European adolescents.

Trichothecenes are a large family of sesquiterpenoid secondary metabolites of Fusarium species (e.g., F. graminearum) and other molds. They are major mycotoxins that can cause serious problems when consumed via contaminated cereal grains. In the past 20 years, an outline of the trichothecene biosynthetic pathway has been established based on the results of precursor feeding experiments and blocked mutant analyses. Following the isolation of the pathway gene Tri5 encoding the first committed enzyme trichodiene synthase, 10 biosynthesis genes (Tri genes; two regulatory genes, seven pathway genes, and one transporter gene) were functionally identified in the Tri5 gene cluster. At least three pathway genes, Tri101 (separated alone), and Tri1 and Tri16 (located in the Tri1-Tri16 two-gene cluster), were found outside of the Tri5 gene cluster. In this review, we summarize the current understanding of the pathways of biosynthesis, the functions of cloned Tri genes, and the evolution of Tri genes, focusing on Fusarium species.

We describe and characterize a multiplex CARS microspectroscopic system that uses a nanosecond supercontinuum generated from a photonic crystal fiber and a sub-nanosecond pulse laser. This system has a high spectral resolution (<0.1 cm−1) and an ultrabroadband spectral coverage (>2500 cm−1). The estimated spatial resolutions are 0.45 μm (lateral) and 4.5 μm (axial), respectively. This system enables us to obtain CARS spectra and corresponding images in the fingerprint region as well in the CH stretch region. Using this system, we have successfully obtained label-free and multi-mode vibrational images of a yeast cell.

The dynamics of an atom in a strong infrared laser field (1013 W/cm2) result in substantial changes to the field-free electronic energy levels, which can be probed on time scales shorter than the laser cycle using isolated attosecond pulses. Here, we measure the transient absorption of an isolated attosecond pulse by laser-dressed bound states of neon near the first ionization threshold. The observed subcycle changes in the absorption spectrum result from both laser-induced ac Stark shifts and from quantum interferences between different multiphoton excitation pathways. We further demonstrate the ability to experimentally turn off the quantum interference mechanism by eliminating one of the interfering pathways.

The novel silica nanotubes with flower-like morphology and hexagonal outer surface morphology were fabricated via a simple ZnS nanowires templates-assisted CVD method. The as-synthesized nanotubes exhibited strong visible photoluminescence at 615 nm and indicated that the method was useful to produce silica nanotubes.

1.昨年度に引き続き、イランの現代詩人シャームルーAhmad Shamlu(1925-2000)に関する研究を行った。今年度は、シャームルーの政治的思想と作品との関わりから、さらに進んで、彼が1950-60年代におけるイラン詩の状況の中でうち立てた、独自のジャンルと詩論についての研究を行った。2.シャームルーは、イラン詩において初めて、she'r-e mansur「非韻律詩」のジャンルを確立した詩人である。本研究では、歴史的にshe'r-e mansur(「散文の詩」)と呼ばれてきた作品を分析することによって、ペルシア詩におけるshe'r-e mansur概念の再考を試みた。これによれば、ペルシア語のshe'r-e mansurは、従来、prose poetryと訳されてきたが、形態的には、文学研究一般で言われるところの「散文詩」ではなく、avant garde free verse(「完全自由詩」)と呼ばれるものに近い形であることが明らかになった。しかし、同時に、she'r-e mansurが、「詩」において必要不可欠であるとされてきた「韻律」を排除したために、「散文」であるとみなされ、かつ、she'r-e mansur自体もまた、韻律の存在に関わりなく、詩が「詩たらしめるもの」を追求するという点で、「散文詩」と同様の文学史的課題を背負ってきたことを明らかにした。3.韻律形式は、現代においてもなお、ペルシア文学において、絶対的な価値をもつものであり、これを侵すことの意味もまた重大であった。シャームルーが、詩から韻律を排除するに際して、うち立てた概念のひとつが、「純粋詩」である。本研究担当者は、シャームルーの「純粋詩」の思想を、ペルシア詩において、新しい「詩」概念の形成として捉え、これを分析した。これによれば、第一に、シャームルー詩作論においては、「それ自体の形態をもった詩が、おのずから生まれてくる」という、彼の個人的な感覚が、非韻律詩創出の契機となっていることが指摘できる。また、第二に、シャームルーの主張する無意識性が、たとえば、「自動記述」において見られるような、無意識の利用ではなく、外界における体験が詩人の感覚を支配する力の強さを確信している点で、特徴的であることが明らかになる。4,さらに、シャームルーが事実上、詩作のうえでの師として位置づけたニーマー・ユーシージNima Yushij(1897-1960)の詩論の分析を通し、両者の詩学上の対立点が、詩における韻律リズムの存在に関するものだけでなく、詩と詩のフォルムとの関係をめぐるものであった点を明らかにした。

経年的な子どもの発達、社会適応、健康状態、問題行動の発現への影響を踏まえ、科学的な根拠に基づく「気になる子ども支援プログラム」の開発を目的とした。全国の0~6歳児と保護者約36,000組の12年間パネルコホート調査を用い、子どもの特性別に発達の軌跡と関連要因について分析した。その結果、家庭環境要因、子ども特性要因、家族特性要因、地域サポート要因の子どもの発達への影響の大きさと軌跡を明らかにした。
