Trachelospermi caulis is used widely as an herbal medicine in oriental countries to attenuate fever and pain. We wished to reveal the novel function of this herb and its active component on barrier function in intestinal epithelial cells. Monolayers of intestinal epithelial cells (Caco-2) were used to evaluate the transepithelial electrical resistance (TEER) and quantity of permeated ovalbumin (OVA) as indices of barrier function. T. caulis increased TEER values on cell monolayers and decreased OVA permeation across cell monolayers. To ascertain the active component of T. caulis, the extract was isolated to five fractions, and the effect of each of these fractions on intestinal barrier function examined. Chloroform and ethyl acetate fractions showed increased TEER values and decreased OVA flux. Chloroform and ethyl acetate fractions contained mainly trachelogenin and its glycoside, tracheloside. Trachelogenin increased TEER values and decreased OVA flux by enhancing the tight-junction protein occludin (but not tracheloside) in Caco-2 monolayers. These findings demonstrated that trachelogenin, an active component of T. caulis, might help to attenuate food allergy or inflammatory bowel disease through inhibition of allergen permeation or enhancement of the intestinal barrier.

[Purpose] The purpose of this study was to determine the effects of task-oriented training with whole body vibration (WBV) on the sitting balance of stroke patients. [Subjects] The subjects were 30 stroke patients who were randomly divided into experimental (n1=15) and control (n2=15) groups. [Methods] Subjects in both groups received general training five times per week. Subjects in the experimental group practiced an additional task-oriented training program with WBV, which was performed for 15 minutes, five times per week, for four weeks. The center of pressure (COP) path length and average velocity were used to assess subjects static sitting balance, and the Modified Functional Reach Test (MFRT) was used to assess their dynamic sitting balance. The paired t-test was performed to test the significance of differences between before and after the intervention. The independent t-test was conducted to test the significance of differences between the groups. [Results] Following the intervention, the experimental group showed a significant change in MFRT. [Conclusion] The results of this study suggest that task-oriented training with WBV is feasible and efficacious for stroke patients.

Caffeine (1,3,7-trimethylxanthine) is a popular mild central nervous system stimulant found in the leaves, seeds and fruits of various plants and in foodstuffs such as coffee, tea, and chocolate, among others. Caffeine is widely used and is not associated with severe side effects when consumed at relatively low doses. Although rarely observed, overdoses can occur. However, only a few fatal caffeine intoxication cases have been reported in the literature. Herein, we report the pathological examination results and information on caffeine concentrations in the blood, urine and main organs in a fatal caffeine intoxication case. Even though high caffeine concentrations were found in the systemic organs, no caffeine-related pathological changes were detected.

A number of descriptive reports suggest that Cybister larvae feed on tadpoles, fish, and aquatic insects; however, no quantitative study on their feeding habits has been reported. In order to elucidate the feeding ecology of C. brevis larvae, field observations and laboratory experiments were carried out. In the field, all C. brevis larvae fed on invertebrates, such as insects and isopoda, but did not eat vertebrates, such as fish and anuran larvae. A rearing experiment demonstrated that all C. brevis larvae provided with tadpoles died. Larvae provided with Odonata nymphs had a longer total body length than larvae reared with a mixture of tadpoles and Odonata nymphs. In addition, larvae of C. brevis could search for and eat motionless Odonata nymphs, but all larvae died from starvation when they were supplied with motionless tadpoles. These results suggested that C. brevis larvae mainly preyed upon invertebrate animals and did not eat vertebrate animals, such as tadpoles and fish.

This study aimed to establish culture conditions which are able to give the differentiation of induced pluripotent (iPS) cells to the hepatocytes. To this end, we examined the usefulness of culture medium containing the components involved in the intermediary metabolism in the liver. More specifically, we examined the effect of the 'modified L-15 medium' containing galactose, phenylalanine and ornitine, but deprived of glucose, tyrosine, arginine and pyruvic acid. The medium was altered according to changes in the expression of enzymes that participate in liver-specific pathways. After 25 days of differentiation, the differentiated cells expressed hepatocyte markers and drug-metabolizing enzymes. These expression levels were increased using modified L-15 medium. The survival of human fetal liver cells and the death of human fibroblasts were observed during culture in modified L-15 medium. Most of the cells that differentiated from human iPS cells using modified L-15 medium were stained by anti-human albumin antibody. These results suggest that iPS cells can be converted to high purity-differentiated hepatocytes by cultivating them in modified L-15 medium.

The aim of study was to evaluate the effects of insulin-like growth factor I (IGF-I) on canine sperm function during cooled and freeze-thaw storage. Extenders supplemented with different IGF-I concentrations (0, 100 and 200 ng/ml) were added to canine spermatozoa and the sperm samples were stored at 4°C for 48 hr or freeze-thawed. Sperm motility, morphology, plasma-membrane integrity (PMI) and mitochondrial membrane potential (MMP) were evaluated. IGF-I had no effect on PMI or morphology during cooling and freeze-thawing. However, IGF-I alleviated the reduction in progressive motility and MMP caused by cooled storage and led to an improvement in MMP after freeze-thawing. In conclusion, IGF-I can be helpful to maintain progressive motility of canine spermatozoa during hypothermic storage via increased MMP.

Electron spin resonance (ESR) dosimetry of teeth is used extensively for dose estimation following exposure to radiation. The population inhabiting the northeast region of India is prone to different cancers of the head and neck, and their prevalence is several times the national average. The objective of this study was to determine the role of radiation in the causation of this high cancer incidence by performing ESR spectroscopic measurements of tooth samples collected from the general population living in and around the city of Silchar. Nineteen tooth samples were used, and the age of the patients was 13-60 years. The excess dose, determined by subtraction of the natural background dose from the dose absorbed by the enamel, was found to the extent of 123 ± 43 mGy. However, the individual excess dose was found to be higher in subjects who consumed lime (5/6) than in non-lime-consuming subjects (2/13). It is not entirely clear if radiation is the cause of this excess cancer in this region of India. Therefore there is a need for wider studies including consideration of tobacco consumption as well as a larger number of samples for tooth enamel dosimetry.

Peroxiredoxins (Prxs) are a family of antioxidant enzymes. Here, we cloned a 2-Cys Prx, BgTPx-1, from the canine Babesia parasite B. gibsoni. Sequence identity between BgTPx-1 and 2-Cys Prx of B. bovis was 81% at the amino acid level. Enzyme activity assay by using recombinant BgTPx-1 (rBgTPx-1) indicated that BgTPx-1 has antioxidant activity. Antiserum from a mouse immunized with rBgTPx-1 reacted with parasite lysates and detect a protein with a monomeric size of 22 kDa and also a 44 kDa protein, which might be an inefficiently reduced dimer. BgTPx-1 was expressed in the cytoplasm of B. gibsoni merozoites. These results suggest that the BgTPx-1 may play a role to control redox balance in the cytoplasm of B. gibsoni.

The effects of a silk amino acid (SAA) preparation on the physical stamina and male reproductive function of mice were investigated. Eight-week-old male ICR mice (29—31 g) were orally administered SAA (50, 160 or 500 mg/kg) for 44 d during 30-min daily swimming exercise. The mice were subjected to a weight-loaded (5% of body weight) forced swimming on the 14th, 28th and 42nd day to determine maximum swimming time, and after a 2-d recovery period (treated with SAA without swimming exercise), parameters related to fatigue and reproductive function were analyzed from blood, muscles and reproductive organs. Repeated swimming exercise increased the maximum swimming time to some extent, in spite of a marked reduction in body weight gain, and SAA further enhanced the stamina in a dose-dependent manner. Forced swimming exercises increased blood parameters of tissue injury, but depleted blood glucose and tissue glycogen, which were substantially prevented by SAA treatment. In addition, SAA significantly reduced the muscular thiobarbituric acid-reactive substances and blood corticosterone content increased by forced swimming. Swimming exercise decreased the blood testosterone level, which was recovered by SAA, leading to enhanced sperm counts. These combined results indicate that SAA not only enhances physical stamina by minimizing damage to tissues, including muscles, as well as preventing energy depletion caused by swimming stress, but also improves male reproductive function by increasing testosterone and sperm counts.

Existing researches suggest that the code clone (duplicated code) is one of the factors that degrades the design and structure of software and lowers the software quality such as readability and maintainability. However, the influence of code clones on software quality has not been quantitatively clarified yet. In this paper we tried to quantitatively clarify the relation between code clones and the software reliability and maintainability of twenty years old software. As a result, we found that modules having a code clone (clone-included modules) are more reliable than modules having no code clone (non-clone modules) in average. Nevertheless, the modules having very large code clones (more than 200 lines) are less reliable than non-clone modules. We also found that clone-included modules are less maintainable than non-clone modules; and, modules having larger code clone are less maintainable than modules having smaller code clone.

Background: Antibody testing is essential for accurately estimating the number of people infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). This study aimed to investigate the influence of background factors on seroprevalence by testing for anti-SARS-CoV-2 antibodies in blood samples obtained from the staff of three hospitals. Methods: This cross-sectional observational study was conducted from June 8 to July 4, 2020, as part of a mandatory health examination. Leftover blood samples collected during the health examinations at each hospital were used to test for the presence of anti-SARS-CoV-2 antibodies. The Elecsys Anti-SARS-CoV-2 RUO assay was used for antibody detection. The relationship between staff age, gender, body mass index, blood pressure, work environments with different exposure risks, place of residence, and campus location and seroprevalence was investigated. The data were anonymized prior to analysis. Results: A total of 3,677 individuals were included in the study, comprising 2,554 females (69.5%) and 1,123 males (30.5%). Anti-SARS-CoV-2 antibody (immunoglobulin G) was detected in 13 participants (0.35%). Seroprevalence was slightly higher in males than females (0.62% vs. 0.23%, P=0.08). By occupation, anti-SARS-CoV-2 antibodies were found in 6 (0.75%) physicians, 6 (0.31%) nurses, and one individual (0.11%) in the medical personnel group, with slightly higher levels in physicians. No significant difference was noted in the seroprevalence in terms of all background factors. Conclusions: Our study shows that the background factors do not impact seropositivity rates. Thorough daily infection control and adherence to recommended health guidelines were found to reduce infection risk.

Gaucher disease (GD) is a recessively inherited lysosomal storage disorder characterized by a deficiency of lysosomal glucocerebrosidase (GBA1). This deficiency results in the accumulation of its substrate, glucosylceramide (GlcCer), within lysosomes. Here, we investigated lysosomal abnormalities in fibroblasts derived from patients with GD. It is noteworthy that the cellular distribution of lysosomes and lysosomal proteolytic activity remained largely unaffected in GD fibroblasts. However, we found that lysosomal membranes of GD fibroblasts were susceptible to damage when exposed to a lysosomotropic agent. Moreover, the susceptibility of lysosomal membranes to a lysosomotropic agent could be partly restored by exogenous expression of wild-type GBA1. Here, we report that the lysosomal membrane integrity is altered in GD fibroblasts, but lysosomal distribution and proteolytic activity is not significantly altered.Key words: glucosylceramide, lysosome, Gaucher disease, lysosomotropic agent

土壌に添加した易分解性有機物が分解される際に、土壌に元から含まれる有機物の分解が促進される現象をプライミング効果と呼ぶ。本研究では淡水産巻貝マルタニシが這う際に残す粘液がプライミング効果を引きこすか実験を行った。スクロースを寒天で固めたものと、水田から採集された底泥を有機物源として用いて、両有機物源の表面にマルタニシを這わせ粘液を塗布したものと粘液のないものを準備し、それぞれの有機物分解量をクーロメーターで評価した。その結果、両サンプルにおいて粘液を含む系で酸素消費量が多く、有機物の分解量が多くなることが示された。マルタニシの這う際に残す粘液はプライミング効果を引き起こすことが明らかとなった。

This paper is concerned with the embedding constant of the Sobolev type inequality for fractional derivatives on $\Omega\subset\mathbb{R}^{N}~(N\in\mathbb{N})$. The constant is explicitly described using the analytic semigroup over L2(Ω) generated by the Laplace operator. Some numerical examples of estimating the embedding constant are also provided.

This paper aims to propose a Fallback Control System isolated from cyber-attacks against networked control systems. The fallback control system implements maintaining functionality after an incident. Since cyber-attacks tamper with the communication contents of the networked control systems, the fallback control system is installed in a control target side. The fallback control system detects the incident without the communication contents on field network. This system detects an incident based on a bilinear observer and a switched Lyapunov function. When an incident is detected, normal operation is switched to fallback operation automatically. In this paper, a practical experiment with Ball-Sorter simulating a simple defective discriminator as a part of Factory Automation systems is shown. Assumed cyber-attacks against Ball-Sorter are Man In The Middle attack and Denial of Service attack.
