In transgenic experiments, we often face fundamental requirements such as overexpressing a certain gene, developing organelle markers, testing promoter activities, introducing large genomic fragments, and combinations of them. To fulfill these multiple requirements in rice, we developed simple binary vectors with or without maize ubiquitin (UBQ) promoter, Gateway cassette and fluorescent proteins. First, we compared stabilities of cauliflower mosaic virus 35S and maize UBQ promoters for constitutive gene expression in transgenic rice. We show that the 35S promoter was frequently silenced after shoot regeneration, whereas maize UBQ promoter achieved stable expression in various young tissues. Binary vectors with Gateway cassettes under the control of the UBQ promoter allowed us to develop stable organelle markers for nuclei, microtubules and P-bodies in rice. The maize UBQ promoter can be easily replaced with any promoters of interest as exemplified by reporters of mitotic cells and provascular bundles. Finally, by introducing two genomic fluorescent reporters, we showed utilities of the Gateway cassette and two selection markers in large DNA fragment transfer and sequential transformations, respectively. Thus, these binary vectors provide useful choices of transgenic experiments in rice.

Background: The impact of meeting leisure time physical activity (LTPA) recommendations and household physical activity (HPA) on all-cause mortality in the Taiwanese population is unclear. We aimed to investigate the relationship between sufficient LTPA and all-cause mortality in middle-aged and older Taiwanese adults and the role of HPA in those with insufficient LTPA.Methods: This nationwide prospective cohort study included 4,960 participants aged ≥50 years from the Taiwan Longitudinal Study in Aging (TLSA) survey. Physical activity patterns were assessed in 2003 and then followed up until 2015 for mortality through the National Death Registration Record. Cox proportional hazards regression was conducted to evaluate hazard ratios (HRs) and 95% confidence intervals (CIs) for all-cause mortality.Results: Of the 4,960 participants, 1712 died of all-cause mortality. Compared to those who had insufficient LTPA, participants who engaged in sufficient LTPA showed a significantly lower risk of all-cause mortality (HR=0.84, 95% CI, 0.73-0.97). For those with insufficient LTPA, HPA also had a significantly reduced risk of all-cause mortality (HR=0.85, 95% CI, 0.75-0.96) among general population. Similar associations were observed in subsequent sensitivity analyses. The subgroup analysis showed that the relationship between HPA and reduced mortality risk was only found in the women with insufficient LTPA group.Conclusion: This study confirmed that sufficient LTPA is associated with a lower risk of all-cause mortality. If sufficient LTPA cannot be performed, additional HPA is related to lower mortality.

Purpose: Our aim was to evaluate the development of new significant mitral regurgitation and long-term survival after mitral repair surgery in functional mitral regurgitation.Methods: A retrospective observational analysis of the recurrence of functional mitral regurgitation (ischemic and nonischemic) and global mortality during follow-up of 176 patients who underwent mitral repair surgery between 1999 and 2018 in our center was conducted.Results: The etiology of functional mitral regurgitation was ischemic in 55.7% of cases. After surgery, mitral regurgitation was 0-I in 92.3% of cases. We conducted a long-term clinical follow-up of a mean 42.2 months and an echocardiographic follow-up of a mean 41.8 months. We observed mitral regurgitation of at least grade II in 52 patients (36.9%). Survival at 1, 3, and 5 years was 78.8%, 66.7%, and 52.3%, respectively. Predictive factors for global mortality were age (hazard ratio = 1.038, p = 0.01) and a depressed preoperative ejection fraction. After a competing risk analysis, we found the only predictive factor for the recurrence of mitral regurgitation in our series to be age (sub-hazard ratio = 1.03, 95% confidence interval = 1.01–1.06, p = 0.016).Conclusion: Repair surgery for functional mitral regurgitation shows age as the only independent predictor of recurrence. Age and depressed ejection fraction were predictors of mortality.

Aim: Accumulating evidence reveals that sedentary behavior is associated with mortality and cardiometabolic disease; however, there are potential age and sex differences in sedentary behavior and health outcomes that have not been adequately addressed. This study aimed to determine the association of sedentary behavior with cardiometabolic diseases such as hypertension, dyslipidemia, diabetes mellitus, and its risk factors in a large Japanese population according to age and sex. Methods: Using data from the Japan Multi-Institutional Collaborative Cohort Study obtained from baseline surveys, data of 62,754 participants (27,930 males, 34,824 females) were analyzed. This study uses a cross-sectional design and self-administered questionnaires to evaluate sedentary time and anamnesis. For the logistic regression analysis, sedentary time <5 h/day was used as the reference and then adjusted for age, research areas, leisure-time metabolic equivalents, and alcohol and smoking status. From the analysis of anthropometric and blood examinations, 35,973 participants (17,109 males, 18,864 females) were analyzed. Results: For hypertension and diabetes, sedentary time was associated with a significantly higher proportion of male participants. Both sexes were associated with a significantly higher proportion of participants with dyslipidemia. Participants who had longer sedentary time tended to have increased levels of blood pressure, triglycerides, and non-high-density lipoprotein cholesterol (HDL-C), and decreased levels of HDL-C, especially in the 60–69 years group. Conclusions: Independent of leisure-time physical activity, sedentary time was associated with cardiometabolic diseases in a large Japanese population classified by age and sex. Our findings indicate that regularly interrupting and replacing sedentary time may contribute to better physical health-related quality of life.

In Taiwan, the early harvesting of young ginger is a cultivation technique for domestic ginger, which can be harvested early by forced sprouting. At present, ethylene is used as the sprouting agent. Despite its favorable sprouting effect, the technique can still be improved. Experimental results revealed that 1% citric acid, 300-ppm and 450-ppm ethephon treatments effectively facilitated the formation of 2.4, 2.4, and 2.6 large buds (diameter of ≥ 1 cm), respectively, in the rhizome. The stems and leaves of the ginger seed rhizomes that were subjected to forced sprouting emerged from the soil one month after planting, and emergence rates of 46.7% and 83.3% were achieved using citric acid and ethephon, respectively. Although the fresh weight of semi-matured ginger rhizomes obtained by forced sprouting treatment was lower, the results can still provide a reference for the early harvest of young ginger in Taiwan. Among the examined gingers, semi-matured ginger that was subjected to 50-ppm GA3 forced sprouting treatment exhibited the most favorable growth, and the average weight of its rhizomes reached 1,567 g, which was not significantly different from the weight obtained by conventional cultivation, but significantly greater than that obtained by other treatments. The GA3 forced sprouting treatment was not very effective, but it had an excellent effect on improving the yield of semi-matured ginger rhizomes. In the future, this treatment will be conducive for the production of semi-matured or matured ginger.

A circular surface is a one parameter family of round circles in Euclidean space and which is defined in [2]. The circular surface is determined by a space curve of circular center and planes containing circles. First we study the sufficient condition that the special circular surfaces with constant radius whose normal directions of the planes are rotating around the curve, are embeddings. Next we study the condition that circular surface with constant radius are immersions. Moreover, we discussed the relation between the double circular surfaces (i.e. at least two circles through any point) and quadratic surfaces.

This study illustrates the effectiveness of slightly acidic hypochlorous water (SAHW) in comparison with sodium hypochlorite (NaOCl) in reducing biomass and viable cells in biofilms established by the dual species, Listeria monocytogenes and Escherichia coli, on a microtiter plate and stainless-steel coupon. The SAHW and NaOCl treatments exhibited significant efficacy against biofilms (p<0.05) on both surfaces. Additionally, compared with NaOCl treatment, SAHW treatment significantly reduced biofilm formation (p<0.05). With its high antibiofilm activity, SAHW not only reduced the biomass of biofilms, but also significantly decreased viable biofilm cells to 5 log CFU/mL or ≤1 log CFU/cm2 on microtiter plates and stainless-steel surfaces, respectively. These results indicate that SAHW is a potential candidate for disinfectants against biofilms on various food contact surfaces.

Little Tern Sternula albifrons sinensis was been categorized as Endangered Class II by the 2020 Ministry of the Environment's Red List in Japan. For an effective conservation program for the taxon, the unit of conservation needs to be understood. In this study, we investigated genetic and morphological diversity as a clue to clarify the units to be protected. Blood samples were collected from 47 individuals in four different breeding sites in Japan (Tokyo, Chiba, Fukuoka, Okinawa). The nucleotide sequences of the control region of mitochondrial DNA were used to analyze their genetic diversity and structure. Basic morphological features were also compared. Nucleic acid base substitution, deletion, or insertion were identified at 30 sites of the 1029-bp sequence alignment of the control region, leading to identification of 30 different haplotypes. Genetic diversity was high in the four breeding populations, and haplotype frequencies showed no indication that specific genotypes were present at extreme frequencies. This suggests that the population of Little Tern nesting in Japan has been maintained without recent bottleneck events. The haplotype network formed two major haplotype groups from the 30 haplotypes. Haplotypes of one group were abundant in the Tokyo, Chiba, and Fukuoka populations. In contrast, those of the other group were frequent in the Okinawa population. Pairwise haplotype analysis between populations also revealed that the genetic variations at Okinawa differ from those of the remaining three populations. Furthermore, the Okinawa population showed different morphological characteristics compared to the Tokyo/Chiba populations. These results collectively suggest that the population of Little Terns in Japan is divided into at least two different populations.

In heterogeneous landscapes, birds may mistime their breeding attempts relative to local food abundance. How birds cope with this challenge has been little studied. In this study, we compared the provisioning behavior and nestling condition of the Green-backed Tit Parus monticolus nesting in a landscape mosaic comprised of native mixed-oak forests and exotic Japanese Cedar Cryptomeria japonica plantations. Parent birds in the larvae-poor plantations time their breeding as they do in the larvae-rich forests and have clutches of similar size. We found that parent birds were flexible in adjusting their criteria for prey choice on the basis of larval abundance. In early spring, in cedar plantations where larval abundance was low, male parents maintained constant nearby feeding trips to the early broods, just as they did in the mixed-oak forests, but reduced their dietary specialization on larvae, while female birds increased their provisioning frequency. Combining both parents' efforts, the early brood nestlings raised in the cedar plantations received food deliveries more frequently and with a higher proportion of non-larval prey than their counterparts did in the mixed-oak forests. In contrast, during late brood rearing, when larvae were abundant, both parents in the plantations foraged heavily on larvae at the expense of other prey, just as they did in the mixed-oak forests. We found that parent birds reared heavier nestlings as the contribution of large larvae to the diet increased. Moreover, the nestlings during early broods in the plantations had lower body weights than their counterparts in the mixed-oak forests. We suggest that the provisioning flexibility of Green-backed Tits helps them gather food for raising nestlings when their breeding attempts are less synchronized with the larval food supply in their habitat. As a result, they can breed successfully in the exotic cedar plantations.

When we use a Programmable Microfluidic Device (PMD), we need to wash some contaminated area to use the chip for further experiments. Recently, a novel washing technique called Block-Flushing has been proposed. Block-Flushing washes contaminated area in PMDs by using buffer flows. In Block-Flushing, we need to keep a buffer flow from an input port to an output port of a PMD for a long period to dissolve residual contaminants. Thus, we may need a lot of buffer fluids and washing time even if the contaminated area is small. Another disadvantage of the washing method by Block-Flushing is such that we may not able to clean residual contaminants at valves completely by only buffer flows. To address the above-mentioned issues, this paper proposes a totally new idea to wash PMDs; our method does not use buffer flows, but washes contaminated area by using mixers. By using a mixer, we can dissolve residual contaminants at valves in the area of the mixer very efficiently. In this paper, we propose two methods to wash PMDs by using mixers. The first method can wash the whole chip area by using only four times of a single 2x2-mixer time. We also propose the second method which is a heuristic to reduce the number of moving valves because valves may wear down if they are used many times. We also show some experimental results to confirm that the second method can indeed decrease the number of used valves.

La globalisation de la R&D des firmes multinationales articule au sein de divers réseaux d’une part des flux d’informations et de connaissances codifiées, d’autre part une participation contributive à des dispositifs localisés et collectifs de production de connaissances tacites. Cette seconde démarche, par laquelle les FMN implantent des laboratoires dans des territoires spécifiques, dépend pour son efficacité de leur capacité à mettre en place des dispositifs de partage des savoirs propres à chacun des réseaux de coproduction de connaissances tacites dans lesquels elles sont insérées. De plus, pour favoriser ensuite l’intégration de ces dispositifs, elles doivent aussi développer des méta-dispositifs articulant les dynamiques propres à chaque réseau. Cette architecture complexe, qui allie procédures établies et action ad’hoc de quelques acteurs stratégiques, est illustrée ici par deux études du cas : la firme americaine Motorola et la firme japonaise Canon.

The emu is the second largest ratite; thus, their sera and egg yolks, obtained after immunization, could provide therapeutic and diagnostically important immunoglobulins with improved production efficiency. Reliable purification tools are required to establish a pipeline for supplying practical emu-derived antibodies, the majority of which belongs to the immunoglobulin Y (IgY) class. Therefore, we generated a monoclonal secondary antibody specific to emu IgY. Initially, we immunized an emu with bovine serum albumin multiply haptenized with 2,4-dinitrophenyl (DNP) groups. Polyclonal emu anti-DNP antibodies were partially purified using conventional precipitation method and used as antigen for immunizing a BALB/c mouse. Splenocytes were fused with myeloma cells and a hybridoma clone secreting a desirable secondary antibody (mAb#2-16) was established. The secondary antibody bound specifically to emu-derived IgY, distinguishing IgYs from chicken, duck, ostrich, quail, and turkey, as well as human IgGs. Affinity columns immobilizing the mAb#2-16 antibodies enabled purification of emu IgY fractions from sera and egg yolks via simple protocols, with which we succeeded in producing IgYs specific to the severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2) spike protein with a practical binding ability. We expect that the presented purification method, and the secondary antibody produced in this study, will facilitate the utilization of emus as a novel source of therapeutic and diagnostic antibodies.

The relationship between fruit Brix and the electrical conductivity (EC) of the nutrient solution was investigated under gradually increasing EC conditions to predict and control tomato fruit Brix in commercial greenhouses in Japan. Based on the three experiments, fruit Brix was significantly and highly correlated with the cumulative EC of the drainage during the period from anthesis to harvest (cECd). This relationship followed a linear regression function. We then modelled fruit Brix based on cECd and validated this model to predict and control fruit Brix in four other experiments in different growing seasons using two cultivars, slab substrates, and irrigation systems. Using this model, we calculated the target cECd (cECdt) to achieve a target fruit Brix of 6% or higher and used cECdt as an indicator to manipulate the EC of the nutrient solution. In the validation experiments, cECd was lower than cECdt at the beginning of harvest in all experiments. cECd reached cECdt at 72.3–214.0°C·day after the first harvest. When cECd was higher than cECdt, more than 86.9% of the fruit had a higher than Brix 6%. In addition, the marketable yield was higher than 88.2%. RMSEs between the observed fruit Brix and predicted fruit Brix were 0.60–1.25. These results indicate that our model can predict and control fruit Brix.

A 3-year-old female Miniature Schnauzer dog with a week-long history of generalized intention tremor and progressive weight loss for several months was admitted. Mild anemia, fever, splenomegaly, aseptic cerebral meningitis and systemic lymph nodes enlargement were examined through erythrogram, ultrasonography, computed tomography and magnetic resonance imaging. Mycobacterium bovis was identified via molecular microbiology having the same molecular type as that of isolates from a cattle farm previously identified. However, the dog was raised in a city. The M. bovis had multidrug resistance (MDR)-bearing mutations in both katG and rpoB genes toward first-line antibiotics. To the best of our knowledge, this is the first report describing an MDR M. bovis infection of a dog in Korea.

It is important to consider curvature properties around the control points to produce natural-looking results in the vector illustration. C2 interpolating splines satisfy point interpolation with local support. Unfortunately, they cannot control the sharpness of the segment because it utilizes trigonometric function as blending function that has no degree of freedom. In this paper, we alternate the definition of C2 interpolating splines in both interpolation curve and blending function. For the interpolation curve, we adopt a rational Bézier curve that enables the user to tune the shape of curve around the control point. For the blending function, we generalize the weighting scheme of C2 interpolating splines and replace the trigonometric weight to our novel hyperbolic blending function. By extending this basic definition, we can also handle exact non-C2 features, such as cusps and fillets, without losing generality. In our experiment, we provide both quantitative and qualitative comparisons to existing parametric curve models and discuss the difference among them.
