Species with a chitinous exoskeleton are overrepresented among the aquatic organisms carrying Mycobacterium ulcerans (MU) in nature and laboratory experiments have demonstrated the enhancing effects of chitin on the growth of MU. Field surveys identified pH as one of the key parameters delineating the distribution of MU in tropical regions. The present study investigated the relationship between chitin and pH in MU growth. By focusing on pH variations in the field, our results revealed that chitin enhanced MU growth in acidic environments. The present study provides new information on the ecological conditions favoring the development of this mycobacterium in nature.

[Purpose] This study aimed to conduct experiments to examine the effects of wrist eccentric control exercise or shoulder stabilization exercises after a basic direct treatment of the elbow in the treatment of tennis elbow patients in terms of pain and grip strength. [Subjects and Methods] The subjects were divided into two groups: one group conducted wrist eccentric control exercise and was comprised of 5 male and 4 female subjects, and the other group received shoulder stabilization exercise and was comprised of 5 male and 4 female subjects. [Results] In the intragroup comparison, both groups showed a significant decrease in pain level and a significant increase in the measurement of the tenderness thresholds of the upper trapezius muscle, lateral epicondyle, and grip strength. In the intergroup comparison, the shoulder stabilization exercise group showed a significantly greater increase in the measurement of the tenderness thresholds of the upper trapezius muscle and grip strength, and the differences were not significant in the pain level and tenderness threshold of the lateral epicondyle. [Conclusion] Wrist eccentric control exercise and shoulder stabilization exercises can be useful as intervention methods for relief from pain due to lateral epicondylitis and for the improvement of functions impaired by tennis elbow.

A 16-year-old female Indian peafowl (Pavo cristatus) died two days after recognition of conjunctivitis in the right eye, anorexia and depression. Gross necropsy revealed a thick pseudomembrane under the eyelid and hydropericardium. Histopathological examination revealed hepatocellular necrosis, sinusoidal and vascular congestion and infiltrated inflammatory cells. Infiltration by inflammatory cells was noted in the epicardium. The lungs had mild interstitial pneumonia with the extensive congestion within the capillaries of the air sacs. Tubular interstitial congestion and necrosis was noted in the kidneys. Bacterial culture and nucleotide sequencing of the inflammatory specimens identified the causative agent as Serratia marcescens, an uncommon bacterium in birds. In summary, this study describes the sudden death of an Indian peafowl due to S. marcescens infection, which is rarely seen in animals.

A 2- to 4-year-old uncastrated male Siberian tiger (Panthera tigris altica) bred in a local wild animal park presented with generalized clinical signs including abdominal pain, fever, lethargy, and anorexia, along with subcutaneous nodules along the trunk. The patient subsequently died of chronic, progressive dyspnea despite 45 days of antibiotic treatment. At necropsy, mesenteric fat inflammation and multiple subcutaneous, peritoneal, and intraabdominal nodules were observed. The lungs demonstrated congestion and heavy coagulation, and necrotic foci were observed on the cut surface. Histopathologically, the nodules were identified as granulomatous fatty tissue with numerous lymphocytes, infiltration with lipid-laden macrophages, and fibrosis. These changes were also noted in the lung. The etiology of this condition remains undetermined.

Lung surfactant, besides alveolar stability, also provides defence against pathogens by surfactant proteins (SP), SP-A and SP-D. The hydrophobic proteins SP-B and SP-C enhance surface activity. An unusual and paradoxical effect of bovine LS and synthetic model LS with SP-B/-C was bactericidal to Staphylococcus aureus and Escherichia coli. Bacterial proliferation were investigated with bovine lung surfactant extract (BLES), dipalmitoylphosphatdylcholine, palmitooleylglycerol, in combination with SP-B/-C using standard microbiological colony forming unit (CFU) counts and structural imaging. BLES and other surfactant-SP-B/-C mixtures inhibit bacterial growth in the concentration range of 0 -7.5 mg/mL, at > 10 mg/mL paradoxical growth of both the bacterial species suggest antibiotic resistance. The lipid only LS have no effect on bacterial proliferation. Smaller peptide mimics of SP-B or SP-B1-25, were less efficient than SP-Cff. Ultra structural studies of the bacterial CFU using electron and atomic force microscopy suggest some membrane damage of S. aereus at inhibitory concentration of BLES, and some structural alteration of E. coli at dividing zones, suggesting utilization and incorporation of surfactant lipid species by both bacteria. The results depicted from in vitro studies are also in agreement with protein-protein interactions obtained from PatchDock, FireDock and ClasPro algorithm. The MD-simulation decipher a small range fluctuation of gyration radius of the LS proteins and their peptide mimics. The studies have alarming implications in using of high dosages (100 mg/mL/kg body weight) exogenous surfactant for treatment of respiratory distress syndrome, genetic knock-out abnormalities associated with these proteins, and the novel roles played by SP-B/C as bactericidal agents.

進行性多巣性白質脳症(PML)は,宿主の免疫低下に伴いJCウイルスが再活性化して起こる脱髄脳症である。臨床的に免疫低下の原因が不明瞭で,髄液ポリメラーゼ連鎖反応(PCR)でウイルス陰性でもなお,画像上PMLの可能性を否定できず脳生検を施行する場合がある。こうした症例では,病理診断の指標となる典型的な核内ウイルス封入体を有する細胞に乏しく,高度な炎症細胞浸潤を伴う場合がある。JCウイルスに対する宿主免疫応答が保たれている状態と考えられ,予後は良好である。本稿では,炎症反応を伴ったPMLについて,近年問題となっている免疫再構築症候群も含め,概説する。

To predict the seasonal vegetative growth of potted seedlings of Satsuma mandarin, the effects of initial plant conditions (age, shoot pruning), environment (greenhouse, greenhouse + shading, open-field culture), and pot size on growth were researched for 1.5–2.5 years. The growth pattern was evaluated using a curve fitting with the 4-parameter logistic (4L) model, biomass allocation, and classical (interval) growth analysis, and a generalized linear model analysis based on thermal time (tt). Growth delays from shoot pruning were confirmed by growth trajectories with the 4L model based on tt. Plant growth was positively affected by enlarging the pot size (from 20-L to 45-L), while shading significantly suppressed the growth of 45-L potted Satsuma mandarin seedlings in the greenhouse. In the growth analysis, the relative growth rate was not always determined by the net assimilation rate (NAR), which included both shoot-pruning and pot size effects. To predict the plant mass of Satsuma mandarin seedlings (MP), we proposed several generalized linear models using a log link function assuming that MP followed a Gamma-distribution. The best model to predict MP was selected based on Akaike information criterion (AIC) values, and contained explanatory variables for initial plant biomass, NAR, specific leaf area (SLA), leaf mass ratio (LMR), pot size, and tt. Some simpler models excluding NAR, but including SLA and/or LMR as explanatory variables, were more useful than a model lacking growth analysis parameters (NAR, SLA, and LMR).

Extracellular polymeric substances (EPS) are reported to be the major foulant in membrane bioreactor (MBR) processes. It is important to understand the EPS fractions which cause irreversible fouling to reduce operation cost and energy consumption in MBR. In this study, we developed polarity-molecular weight profiling, in which EPS components were plotted on two-dimensional matrix of its polarity and molecular weight, and applied it to investigate EPS of bulk sludge and cake layers from a lab-scale MBR. The EPS components were also characterized via three-dimensional excitation-emission matrix (3D-EEM) spectroscopy. The result showed that hydrophilic substances as large as 100 − 670 kDa was found only in loosely-bound EPS (LB-EPS) of bulk sludge but not in that of cake layers nor in permeate. Hydrophobic substances smaller than 20 kDa were mainly found in soluble microbial products (SMP) in bulk sludge. Hydrophilic substances larger than 670 kDa was mainly found in tightly-bound EPS (TB-EPS) of bulk sludge and in LB- and TB-EPS of cake layers. These findings suggest that, after conditioning of micropores of virgin membrane by hydrophobic substances smaller than 20 kDa in SMP, hydrophilic biopolymers as large as 100 − 670 kDa in bulk sludge clog the narrowed micropores inside membrane, causing irreversible fouling.

Inflammation of many different aetiologies is a condition affecting almost 100% of all patients presenting at dental clinics, and the complete management of inflammation is often extremely important before any orthodontic surgery can be carried out. The lymphatic system is one of the body’s main lines of defence against inflammation, with the lymph nodes playing an extremely major role. laser therapy, or LLLT, has become much more common in dental offices, and is associated with pain attenuation, wound healing and anti-inflammatory properties. The last of these is mediated amongst others by the effect of LLLT on the lymphatic system, which has been shown to enhance lymphatic drainage concomitantly with increasing local blood flow to the treated area. Infected areas and those with viral contamination have been suggested as contraindications for laser therapy. The possibility of treating these difficult-to-manage entities through laser therapy-accelerated drainage of the affected lymphatics offers another approach. Although lymphatic drainage is frequently used in the clinical setting, its application in dentistry is less common than it should be. An illustrated review of the lymphatic system of the head and neck is presented, with relevance to the use of infrared diode laser therapy in inducing lymphatic drainage in the management of inflammation associated with acute infectious processes such as pericoronitis, endodontic abscesses, alveolitis and herpes. A case report is presented illustrating the success of this therapeutic approach for labial herpes simplex. Infrared diode laser therapy-mediated lymphatic drainage offers an interesting and efficacious approach in the management of infectious and contaminated areas, without actually irradiating the areas themselves.

Sea surface temperature (SST) predictability in the Pacific on decadal timescales is examined in hindcast experiments using the coupled atmosphere-ocean model MIROC with low, medium, and high resolutions. In these hindcast experiments, initial conditions are obtained from an anomaly assimilation procedure using the observed oceanic temperature and salinity while prescribing natural and anthropogenic forcing based on the IPCC concentration scenarios. Our hindcast experiments show the predictability of SST in the western subtropical Pacific, the Indian Ocean, and the tropics to the North Atlantic. Previous studies have examined the SST predictability in the Indian Ocean and the Atlantic, but SST predictability in the western subtropical Pacific has not been evaluated. In the western Pacific, the observed SST anomalies in the subtropics of both hemispheres increased rapidly from the early 1990s to the early 2000s. While this SST warming in the western subtropical Pacific is partly explained by global warming signals, the predictions of our model initialized in 1995 or 1996 tend to simulate the pattern of the SST increase and the associated precipitation changes. This large climate change around the late 1990s may be related to phenomena such as the recent increase in the typhoon frequency in Taiwan and the weakened East Asian monsoon reported by recent studies.

This experimental article explores the question of whether it is possible to examine the experience of playing fighting games (video games) as a form of self-cultivation or practice and, in so doing, whether it becomes possible to shift the debate about the potential impact of violent video games on the people who play them (and on society around them). The article draws on five years of surveys and interviews with gamers from around the world, but seeks to interpret this data through a critical and creative reading of the games themselves as well as a reading of the so-called bushidō tradition (of texts about the intersection between Zen and the martial arts). The article concludes that fighting games might be experienced as forms of martial arts in themselves, complete with potentials for self-transformation, but that this form of engagement requires appropriate intentionality from players, which provokes a space for a manifesto to guide players' intentions.

Objective: Comprehensive profiling of gene expression in peripheral blood leucocytes (PBLs) in patients with acute coronary syndrome (ACS) as a prognosticator is needed. We explored the specific profile of gene expression in PBLs in ACS for long-term risk stratification. Methods: 30 patients with ACS who underwent primary percutaneous coronary intervention (PCI) and 15 age-matched adults who participated in medical check-ups were enrolled from three centres. Peripheral blood samples were collected to extract RNA for microarray analyses. Results: During the 5-year follow-up, 36% of this cohort developed the expected non-fatal coronary events (NFEs) of target lesion revascularisation (TLR) and PCI for a de novo lesion. Class comparison analysis (p<0.005) demonstrated that 83 genes among 7785 prefiltered genes (41 upregulated vs 42 downregulated genes) were extracted to classify the patients according to the occurrence of NFE. Pathway analysis based on gene ontology revealed that the NFEs were associated with altered gene expression regarding the T-cell receptor signalling pathway in ACS. Univariate t test showed that the expression level of death-associated protein kinase1 (DAPK1), known to regulate inflammation, was the most significantly negatively regulated gene in the event group (0.61-fold, p<0.0005). Kaplan-Meier curve analysis and multivariate analysis adjusted for baseline characteristics or clinical biomarkers demonstrated that lower DAPK1 expression in PBL emerged as an independent risk factor for the NFEs (HR: 8.73; CI 1.05 to 72.8, p=0.045). Conclusions: Altered gene expression in T-cell receptor signalling in PBL in ACS could be a prognosticator for secondary coronary events. © Published by the BMJ Publishing Group Limited.

An optical design is developed for a Blu-ray pickup head with a laser expander that can synchronously detect the linear displacement and two-dimensional angular tilts at the test plane. To improve the signal accuracy of the photo detector, a laser expander is used to transform the shape of the laser beam from elliptical to circular. A quadrant detector as used to detect the relative relationship between the $V_{\text{A}}$, $V_{\text{B}}$, $V_{\text{C}}$, $V_{\text{D}}$ signals, can be used to measure the tilt angle and displacement at the test plane. Measurement detection and analysis is based on one normalized focus error signal (NFES) and two angular signals. The optical efficiency of the Blu-ray pickup head with laser expander is 59.29%. The optical efficiency of the currently used commercial pickup head is only 8.08%. Tolerance analysis is also considered in the performance requirements.

The distribution of Y chromosomal haplotypes in Japanese native horse populations was investigated to obtain genetic information on these populations. Here, 159 male/gelded horses from eight local populations were investigated, and three Y haplotypes (JHT-1, JHT-2, and JHT-3) were identified by analyzing five Y-linked loci. Five populations had only JHT-1, whereas two populations had only JHT-2. One population had JHT-1 and JHT-3. Based on the geographical distribution of these haplotypes and previously reported haplotypes for other Asian horses, JHT-1 is considered to be a major haplotype in ancestral native horses. The fixation of each haplotype suggests the influence of independent breeding and genetic drift in each population. These findings complement the results from previous genetic studies of Japanese native horses.

Anthelmintics are used as anti-worming agents. Although known to affect their target organisms, nothing has been published regarding their effect on other digestive tract organisms or on metabolites produced by them. The current work investigated effects of fenbendazole, a benzimidazole anthelmintic, on bacteria and ciliates in the equine digestive tract and on and their major metabolites. Animals receiving anthelmintic treatment had high faecal egg counts relative to controls. Analysis was performed over two weeks, with temporal differences detected in bacterial populations but with no other significant differences detected. This suggests fenbendazole has no detectable effect on organisms other than its targets. Moreover it does not appear to make a contribution to changing the resulting metabolome.
