The aim of this paper is to report two cases of sternal dislocation (SD) in cats and the long-term outcomes with and without surgery. In a cat with poly-traumatized SD (Case 1), mandibular, radial, and ulnar fractures were corrected first, and the SD was allowed to heal without intervention for 14 months. However, normal healing did not occur and sternal instability remained. Therefore, the SD was corrected surgically, and the cat recovered fully within 4 weeks. In a cat with isolated SD (Case 2), surgery was performed, and normal posture and gait were regained after 5 weeks. Furthermore, in both cases, no postoperative complications were observed during follow-up. Therefore, surgical correction of SD in cats is recommended.

A 2-year-old, exotic shorthair cat presented with baldness and mild scaling on trunk that was confirmed as Microsporum canis (M. canis) infection by the following methods. Wood’s lamp and trichogram were used to demonstrate fungal elements suggestive of dermatophytosis consistent with M. canis. Dermatophyte test medium (DTM) and polymerase chain reaction (PCR) were used for identification. E-test and broth microdilution test were then utilized to estimate antifungal minimal inhibitory concentrations (MICs) towards ITZ and TRF respectively. The strain was isolated from the patient and revealing TRF MIC >32 μg/ml and ITZ MIC 0.023 μg/ml. Patient was cured of dermatophytosis with systemic ITZ.

A series of halogenated pyrrolo [2, 1-b] [1, 3] benzoxazines (<b>1</b>-<b>9</b>) was isolated from fermentations of an actinomycete strain X10/78/978 (NCIMB40808), identified as <i>Streptomyces rimosus</i>, during a microbial extract screening programme to identify inhibitors of bacterial histidine kinase. The structures of these compounds were elucidated by spectroscopic methods including the HMQC, HMBC and INADEQUATE NMR experiments. The structure of <b>1</b> was confirmed by X-ray crystallographic studies. Compounds <b>5</b> and <b>6</b> were produced in fermentations in the presence of NaBr and Nal respectively. The most abundant member of the series, streptopyrrole, <b>1</b>, inhibited the nitrogen regulator II (NRII) histidine kinase from <i>Escherichia coli</i> with an IC<sub>50</sub> of 20μM and exhibited antimicrobial activity against a range of bacteria and fungi.

Polymorphisms in MSTN have previously been associated with equine performance. Therefore, the aim of this study was to identify variants in MSTN intron 1 in 16 Brazilian Sport Horses selected for competition in eventing and their possible effects of selection on performance. Among the nine variants identified, eight had already been reported in previous studies or genomic databases, although they showed differences in frequencies when compared with other horse breeds. Moreover, a new mutation was identified in two horses, both in heterozygous form. Considering the absence of molecular studies in this valuable Brazilian breed, these findings represent an important contribution to the characterization of its genetic profile and may possibly aid in further genotype-phenotype association studies.

The petroleum ether extract of the red alga Laurencia obtusa afforded three new C15 acetogenins (cyclic ether enyne): (12Z)-cis-maneonene-D (1), (12E)-cis-maneonene-E (2), and (12Z)-trans-maneonene-C (3), along with one known cis-maneonene-A (4). Blood neutrophils were prepared, cultured, and incubated for 24, 48, and 72 h in medium with and without isolated compounds. Blood neutrophils were prepared, cultured, and incubated for 24, 48 and 72 h in medium with and without the isolated compounds. Both morphology and DNA fragmentation methods assessed the percentage of neutrophils apoptosis in each culture. In the present study, several observations have been made concerning the apoptosis-inducing or inhibiting effect of 1 and 2. Both compounds had no inhibition of apoptosis but apoptosis was enhanced significantly by aging. However, 1 stimulated apoptosis of normal only at the initial 24 h. After that there was no significant difference in apoptosis with or without compound 1, while 2 stimulated apoptosis at all the times. The apoptosis induced by these two compounds was demonstrated by DNA fragmentation assay and microscopic observation. These observations suggest that compounds 1 and 2 may be involved in regulation of programmed death in the initiation and propagation of inflammatory responses.

Background:The effectiveness of statins remains to be examined in patients with heart failure (HF) with preserved ejection fraction (EF).Methods and Results:Among 4,544 consecutive HF patients registered in the Chronic Heart Failure Registry and Analysis in the Tohoku district-2 (CHART-2) between 2006 and 2010, 3,124 had EF ≥50% (HFpEF; mean age 69 years; male 65%) and 1,420 had EF <50% (HF with reduced EF (HFrEF); mean age 67 years; male 75%). The median follow-up was 3.4 years. The 3-year mortality in HFpEF patients was lower in patients receiving statins [8.7% vs. 14.5%, adjusted hazard ratio (HR) 0.74; 95% confidence interval (CI), 0.58–0.94; P<0.001], which was confirmed in the propensity score-matched cohort (HR, 0.72; 95% CI, 0.49–0.99; P=0.044). The inverse probability of treatment weighted further confirmed that statin use was associated with reduced incidence of all-cause death (HR, 0.71; 95% CI, 0.62–0.82, P<0.001) and noncardiovascular death (HR, 0.53; 95% CI, 0.43–0.66, P<0.001), specifically reduction of sudden death (HR, 0.59; 95% CI, 0.36–0.98, P=0.041) and infection death (HR, 0.53; 95% CI, 0.35–0.77, P=0.001) in HFpEF. In the HFrEF cohort, statin use was not associated with mortality (HR, 0.87; 95% CI, 0.73–1.04, P=0.12), suggesting a lack of statin benefit in HFrEF patients.Conclusions:These results suggest that statin use is associated with improved mortality rates in HFpEF patients, mainly attributable to reductions in sudden death and noncardiovascular death. Hyperproduced prostaglandin E2 by cyclooxygenase-2 and microsomal prostaglandin E synthase-1 evokes several pathophysiological responses such as inflammation and carcinogenesis. Our recent study demonstrated that Dioscorea japonica extract suppressed the expression of cyclooxygenase-2 and microsomal prostaglandin E synthase-1 and induced apoptosis in lung carcinoma A549 cells. In the present study, we investigated the effects of Dioscorea japonica on squamous cell carcinoma of mouse skin. Dioscorea japonica feeding and Dioscorea japonica extract topical application suppressed the expression of cyclooxygenase-2, microsomal prostaglandin E synthase-1, interleukin-1β and interleukin-6 and inhibited tumor formation, hyperplasia and inflammatory cell infiltration. Immunohistochemical analyses showed the immunoreactivities of cyclooxygenase-2 and microsomal prostaglandin E synthase-1 in tumor keratinocytes and stronger immunoreactivities of cyclooxygenase-2 and hematopoietic prostaglandin D synthase in epidermal dendritic cells (Langerhans cells). Treatment with Dioscorea japonica decreased the immunoreactivity of cyclooxygenase-2 and microsomal prostaglandin E synthase-1. These results indicate that Dioscorea japonica may have inhibitory effects on inflammation and carcinogenesis via suppression of the prostaglandin E2 synthetic pathway.

This paper describes analysis and recognition of eight Korean isolated vowels based on the distributions of formant frequencies. In the distributions of vowels on the F1-F2 plane, we found overlappings between the vowels /u/, /o/, /∂/ and /w/, /ε/ and /e/ overlapped heavily as if they are the same vowels. In the comparative investigation of the dialect of Seoul and Kyungsangdo, Kyungsangdo dialect showed a heavy overlapping between /∂/ and /w/. As a recognition experiment, the speaker-independent recognition with Bayes decision method was carried out for 1, 760 isolated vowels spoken by 20 speakers. A recognition rate of 81.2% correct was obtained with the formant frequencies F1 and F2while the rate with LPC cepstrum coefficients was only 76.4%. Normalization for eliminating inter-speaker differences showed good results for vowels uttered by speakers of the same dialect. The highest recognition rate of 99.8 % was obtained using a supervised learning method.

Miamiensis avidus is the causative agent of scuticociliatosis in various marine fish species. The virulence factors of the parasite have not been identified, so far. In this study, we examined M. avidus extracellular proteinases (ECPs) as potential virulence factors, using culture supernatants as an ECPs source. We investigated the substrate specificity of ECPs using artificial peptides, and the cytotoxicity of the ECPs was examined using CHSE-214 cells. To elucidate the role of ECPs in ciliate growth, M. avidus was cultured on CHSE-214 cells in the presence of proteinase inhibitors. We detected proteinase activities from the supernatant of M. avidus. Viable CHSE-214 cells decreased significantly in number, when incubated in a medium supplemented with the culture supernatant of M. avidus. The growth of ciliates on CHSE-214 cells was delayed in the presence of PMSF (serine proteinase inhibitor) and E-64 (cysteine proteinase inhibitor). These results suggested that the culture supernatant contained ECPs showing cytotoxicity, and the proteinases facilitated nutrient uptake by the ciliates. Thus, ECPs may be responsible for virulence factors of M. avidus.

The exact pathogenesis of diarrhea-dominant irritable bowel syndrome (IBS) is not known, but the abnormal microbiota of the gastrointestinal tract is considered to be one of the important contributing factors as in other gastrointestinal diseases such as inflammatory bowel disease, antibiotic-associated diarrhea, and colorectal cancer as well as systemic diseases. Though diverse trials of probiotics had been continued in the treatment of diarrhea-IBS, only a few proved by randomized clinical trial. To prove the efficacy of Lactobacillus gasseri BNR17 isolated from breast milk in patients with diarrhea-IBS, prospective, randomized, placebo controlled clinical trial was done including health related-quality of life analysis, colon transit time, and the changes of fecal microbiota. BNR17 significantly improved the symptoms of diarrhea compared to control group. Health related-QOL analysis showed significant improvement of abdominal pain, distension, disturbed daily life, and mean defecation frequency with BNR17. On comparative CTT before and after BNR17, 6 out of 24 subjects showed significant correction of rapid colon transit pattern, while only 2 out of 24 in placebo (p<0.01). Upon fecal microbiota analysis, BNR17 significantly increased B. fecalis, E. rectale, C. aerofaciens, F. prausnitzil and B. steroris. Conclusively, Lactobacillus gasseri BNR17 can be a potential probiotics to ameliorate diarrhea-IBS.

1.チミジンホスホリラーゼが血管新生因子であるPD-ECGFと同一分子であることを明らかにし、さらにチミジンホスホリラーゼ阻害剤である6-アミノ-5-クロロウラシルがチミジンホスホリラーゼによる血管新生を阻害することをゼラチンスポンジアッセイにより明らかにした。この実験結果は非常に大きな意味を持つため、大腸菌のチミジンホスホリラーゼ酵素活性中心部位と相同性を有するPD-ECGFの部位にsite directed mutagenesisにより突然変異を入れ、チミジンホスホリラーゼ活性のない3種類のPD-ECGF分子を作製した(K115E,Lys115→Glu;L148R,Leu148→Arg;R202S,Arg202→Ser)。突然変異を有するPD-ECGFcDNAをCOS細胞にトランスフェクトして、発現レベルを調べると、wild typeのPD-ECGFと同程度発現がみられたが、チミジンホスホリラーゼ活性はほとんど認められなかった。ゼラチンスポンジ法により、これらのPD-ECGFの血管新生を調べたところ、突然変異PD-ECGFはいずれも血管新生活性を有していなかった。チミジンホスホリラーゼ阻害剤がチミジンホスホリラーゼの血管新生活性を阻害することと考えあわせると、チミジンホスホリラーゼの酵素活性が同酵素による血管新生に必須であることが明らかとなった。そこで、同酵素によるチミジンの分解産物が血管新生活性を有しているのではないかと考え調べたところ、デオキシリボースが血管新生活性を持つことが鶏卵漿尿膜法で判明した。また、デオキシリボースは血管内皮細胞の遊走性を亢進した。チミジンホスホリラーゼには、内皮細胞の増殖を促進する作用はなかったが、チミジンを分解してチミジンの組織内濃度を低下させ、血管内皮細胞の増殖のために有利な条件を作り出している可能性がある。このように、チミジンホスホリラーゼの酵素活性が血管新生に必要であることが明らかになると、その酵素活性の強力な阻害剤はチミジンホスホリラーゼによる血管新生を阻害し、その結果として腫瘍の増殖を抑制するのではないかと考えられる。我々は、6-アミノ-5-クロロウラシルよりもさらに強力なチミジンホスホリラーゼ阻害剤を見出し、この阻害剤が腫瘍の増殖や転移にどのような影響を与えるのか検討中である。2.ヒトの固型腫瘍では、チミジンホスホリラーゼ活性がしばしば上昇している。固型腫瘍におけるチミジンホスホリラーゼ活性と血管新生の関連性を調べるため、ヒト大腸癌21症例、アデノーマ13例について、チミジンホスホリラーゼ活性と血管内皮細胞のマーカー蛋白質であるトロンボモジュリンの発現レベルを調べ相関性を検討した。その結果、大腸癌でのチミジンホスホリラーゼ活性とトロンボモジュリンの発現レベルの間には相関性のあることが判明した。このことは、大腸癌においてチミジンホスホリラーゼが血管新生に関与している可能性を示唆している。我々はさらに大腸癌におけるチミジンホスホリラーゼの発現と予後との関係を調べた。チミジンホスホリラーゼを発現している大腸癌の症例は、発現していない症例に比べ予後が悪いことが明らかとなった。またチミジンホスホリラーゼ陽性の大腸癌では、リンパ節へ転移する頻度が高いことがわかった。チミジンホスホリラーゼの単クローン抗体を用いた免疫組織化学染色法にり、大腸癌やその他の固型腫瘍内では癌細胞のみならず浸潤したリンパ球もチミジンホスホリラーゼによる血管新生に関与している可能性が示された。血管新生因子はチミジンホスホリラーゼ以外にもVEGF,bFGFなどがあり、個々の腫瘍でどの血管新生因子が腫瘍血管の新生に関与しているかを調べる必要がある。我々はVEGF、bFGFなどについてもそれらの発現レベルを各々の腫瘍で調べ、血管新生との関連性について解析を進めている。

This paper develops a novel and efficient algorithm for Bayesian inference in inverse Gamma Stochastic Volatility models. It is shown that by conditioning on auxiliary variables, it is possible to sample all the volatilities jointly directly from their posterior conditional density, using simple and easy to draw from distributions. Furthermore, this paper develops a generalized inverse Gamma process with more flexible tails in the distribution of volatilities, which still allows for simple and efficient calculations. Using several macroeconomic and financial datasets, it is shown that the inverse Gamma and Generalized inverse Gamma processes can greatly outperform the commonly used log normal volatility processes with student-t errors or jumps in the mean equation.

Budgerigars use contact calls and warble-songs. Previous studies have found that they acquire these songs through vocallearning, but to date, there have been no long term experimental studies of their vocalizations or social interactions. Thepurpose of this study was to establish a basic understanding of budgerigar vocal ethnology. We recorded all sounds made byfour budgerigars (male=4, female=3) kept in separate cages in a same experimental box for fixed periods (12-h light/darkcycle). There was great diurnal variation in vocalizations, but the variation decreased from lights up to off and during thelights-off period. The variation in vocalizations was also positively related to changes in humidity. The frequency of soundsproduced by the group of males was higher than that of the group of males and females mixed together. The response rate forcontact call varied among individuals which suggests that hierarchy existed in vocal communication. However, the hierarchychanged depending on the membership in a group.

Currently, biofortification breeding programs are being conducted to increase the selenium (Se) content of crops. Se is an element essential to humans and is mostly acquired via consumption of vegetables. However, the effects of Se on the main plant metabolisms such as nitrogen metabolism are unclear. The main objective of this study was to determine the effect of the application of different concentrations of Se (10, 20, and 30 μM) as Na2SeO4 on nitrogen metabolism in Pak-choi grown hydroponically. Supplemental Se increased all enzyme activities analyzed, including nitrate reductase (NR), glutamine synthetase (GS), and glutamate synthase (GOGAT). In 20-μM Se treatment of two cultivars, the fresh weight was significantly increased and the foliar NO3− content was significantly decreased. Compared to the control, the data on Se treatments showed stronger activations of NR, GS, and GOGAT, as well as greater concentrations of total ammonium, amino acid, and protein, and a higher nitrogen use efficiency, resulting in increased biomass production. This suggests the mechanism of Se applied to Pak-choi to decrease NO3− content was due to increasing nitrogen metabolism and protein synthesis to biomass.

We measured and compared the physicochemical properties (pH, yield value, and squeeze force) of a drug for dermatomycosis, terbinafine hydrochloride-containing cream (brand-name product), and 12 generic products to clarify the characteristics of each product. On pH measurement, the pH value of the brand-name product, Lamisil, was 4.8, and those of the generic products ranged from 4.3 to 5.5, showing no marked difference. Furthermore, the yield value of Lamisil, as an index of cream ductility, was 122.2 dyn/cm2, and those of the generic products ranged from 42.1 to 1,621.5 dyn/cm2. In particular, the value of a generic product, Taiyo (42.1 dyn/cm2), was significantly lower, whereas that of another one, Viras (1,621.0 dyn/cm2), was significantly higher. In addition, the squeeze force was measured by attaching a HapLog® to the thumb and second finger. The value of Lamisil was 12.9 N, and those of the generic products ranged from 8.0 to 15.4 N. The values of generic products, Mylan (8.6 N), Tebinaceil (9.0 N), and Kelger (8.0 N), were significantly lower, whereas that of another one, Viras (15.4 N), was significantly higher. These results showed that there were marked differences in the pharmaceutical properties between the generic and brand-name products. The above pharmaceutical characteristics of drugs facilitated the presentation of reasons for differences in the sense of use, which characterizes external preparations, suggesting that products appropriate for individual patients can be recommended.
