Countermeasures against carbon dioxide emissions are a concern in the construction field as well as in society. To solve this problem, a concept for new calcium carbonate concrete is proposed, and this concept is validated experimentally. In the proposed concept, calcium carbonate comprising Ca originating from demolished concrete or other Ca-containing industrial wastes and HCO3- from CO2 gas collected from the air or emitted by industrial plants acts as a binder for aggregates, which can be natural rocks or crushed demolished concrete. This short paper describes the details of the process of making calcium carbonate concrete and discusses future perspectives.

The use of the Internet-of-Things has improved glycemic control in individuals with diabetes in several small-scale studies with a short follow-up period. This large-scale randomized controlled trial investigates whether a smartphone-based self-management support system prevents the worsening of glycemic control in individuals with type 2 diabetes. Individuals with type 2 diabetes (age range 20-74 years; n = 2,000) will be recruited, enrolled, and randomly assigned to two groups: the intensive therapy group and the conventional therapy group. Participants in the intensive therapy group will be supervised to use an automated Internet-of-Things system that demonstrates a summary of lifelogging data (e.g., weight, blood pressure, and daily activities) obtained from each measurement device and will receive feedback messages via smartphone applications to encourage them to increase their physical activity and to monitor weight and blood pressure. Participants in the conventional therapy group are allowed to use the same measurement devices as part of the routine diabetes care but without the Internet-of-Things system. The primary endpoint is the between-group difference in HbA1c levels from baseline to 52 weeks. This randomized controlled study will test the hypothesis that an Internet-of-Things-based self-monitoring system could effectively prevent the worsening of diabetes in individuals with type 2 diabetes. The expected results of the study should facilitate the development of novel strategies for both diabetes treatment and social health.

<p>In the antibiotic screening studies directed by Prof. Sumiki, an antifungal substance was found in a culture filtrate of a fungus, No. K-5201, which was isolated from a soil collected at Karuizawa, Nagano Prefecture. The cultural characteristics of the strain No. K-5201 were investigated by Abe and others<sup>1)</sup>. The strain was designated as <i>Paecilomyces varioti</i> Bainier var. <i>antibioticus</i>, and the antibiotic produced by this strain was named variotin.</p><p>Variotin is a new antifungal antibiotic, having low toxicity and exhibiting inhibition against <i>Trichophyton</i> and other pathogenic fungi. This paper describes processes of production, extraction and purification of variotin and its properties.</p>

Canine gastroesophageal intussusception (GEI) is a rare and life-threatening condition that requires prompt diagnosis and treatment. A 19-day-old Siberian Husky with a 4-day history of regurgitation was diagnosed with GEI based on the findings of computed tomography (CT) performed without anesthesia. Endoscopic reduction of intussusception was impossible; thus, surgical reduction by traction of the duodenum was performed. CT revealed improvement of megaesophagus 82 days postoperatively. Eleven months postoperatively, fluoroscopy showed recovery to nearly normal esophageal motility. Two years postoperatively, no clinical signs were reported. CT is useful to diagnose GEI in neonate puppies with poor abdominal fat and to assess the gastric edema and the anatomical association of stomach with other organs. Fluoroscopy is helpful for evaluating postoperative esophageal motility.

The increase in the number of imported cases of dengue fever in Japan is of particular concern as Aedes albopictus is a vector of dengue fever. Due to the potential for insecticide resistance and the impact of insecticides on non-target species, increased attention is being paid to alternative methods of pest control. Placing salt in used tires has been recommended by the Ministry of Health, Labor and Welfare in Japan as a means of controlling mosquitoes. However, the effectiveness of salt as a larvicide against Ae. albopictus are currently unclear. This study examined the acute toxicity of sodium chloride against first and fourth larval instars of Ae. albopictus. Acute toxicity tests were conducted according to World Health Organization guidelines. The susceptibility of Ae. albopictus larvae was tested against 0.25%, 0.5%, 0.75%, 1.00%, 1.25% and 1.5% NaCl solutions. Larval mortality was correlated with an increase in NaCl concentration and exposure duration. First instar Ae. albopictus larvae were more sensitive to NaCl than fourth instar, and 72-h LC90 values for first and fourth larval instars were 0.49% and 1.01% NaCl, respectively. Our results suggest that the application of 0.5% NaCl to a habitat for 3 days is effective for Ae. albopictus control.

The production of recombinant proteins is important in academic research to identify protein functions. Moreover, recombinant enzymes are used in the food and chemical industries, and high-quality proteins are required for diagnostic, therapeutic, and pharmaceutical applications. Though many recombinant proteins are produced by microbial or mammalian cell-based expression systems, plants have been promoted as alternative, cost-effective, scalable, safe, and sustainable expression systems. The development and improvement of transient expression systems have significantly reduced the period of protein production and increased the yield of recombinant proteins in plants. In this review, we consider the importance of plant-based expression systems for recombinant protein production and as genetic engineering tools.

Twenty-three of 42 European rabbits (Oryctolagus cuniculus), belonging to the same rabbit colony, died in March 2020 (55% mortality) in Chiba prefecture, Japan. The disease course was extremely acute without indicators of death or hemorrhage. Necropsy revealed liver swelling, discoloration, cloudiness and fragility, and pulmonary edema. Histologically, severe hepatocellular necrosis (mainly peripheral) and intra-glomerular capillary hyalin thrombi were observed. On molecular-biological examination, reverse transcription polymerase chain reaction analysis of RNA from tissues detected a rabbit hemorrhagic disease virus, confirmed as a RHDV-2 VP60 fragment, which shared 99.42% nucleotide identity with the homologous fragment of RHDV-2 German isolate by nucleotide sequence analysis. This report shows the outbreak of rabbit hemorrhagic disease caused by RHDV-2, an emerging infectious disease, in Japan.

Background:Optimal intensity is unclear for P2Y12receptor blocker therapy after percutaneous coronary intervention (PCI) in real-world clinical practice.Methods and Results:From the CREDO-Kyoto Registry, the current study population consisted of 25,419 patients (Cohort-2: n=12,161 and Cohort-3: n=13,258) who underwent their first PCI. P2Y12receptor blocker therapies were reduced dose of ticlopidine (200 mg/day), and global dose of clopidogrel (75 mg/day) in 87.7% and 94.8% of patients in Cohort-2 and Cohort-3, respectively. Cumulative 3-year incidence of GUSTO moderate/severe bleeding was significantly higher in Cohort-3 than in Cohort-2 (12.1% and 9.0%, P<0.0001). After adjusting 17 demographic factors and 9 management factors potentially related to the bleeding events other than the type of P2Y12receptor blocker, the higher bleeding risk in Cohort-3 relative to Cohort-2 remained significant (hazard ratio (HR): 1.52 95% confidence interval (CI) 1.37–1.68, P<0.0001). Cohort-3 compared with Cohort-2 was not associated with lower adjusted risk for myocardial infarction/ischemic stroke (HR: 0.96, 95% CI: 0.87–1.06, P=0.44).Conclusions:In this historical comparative study, Cohort-3 compared with Cohort-2 was associated with excess bleeding risk, which might be at least partly explained by the difference in P2Y12receptor blockers.

Near-infrared (NIR) spectroscopy has been used to predict fruit quality. However, thereʼs few previous studies which use small and lightweight devices and they didnʼt construct models for each cultivar although each tomato cultivar has its own characteristics such as color and size. In this study, models to nondestructively determine fruit quality of tomato cultivars A (orange-colored cherry tomato), B (red-colored cherry tomato), C (red-colored cocktail tomato), and D (orange-colored cocktail tomato) were constructed using an inexpensive, portable near-infrared (NIR) spectroscopic device (spectral response range: 590-1,050 nm). We irradiated the tomato fruits and analyzed the spectra of the scattered light. The spectral data were preprocessed using four different methods (min-max normalization, 1st differential, 2nd differential and combination of normalization and 1st differential). The soluble solid content (SSC) and citric acid content (CAC) of the fruits were determined by destructive methods. Partial least-squares regression was used to construct models for predicting the SSC and CAC from the preprocessed spectra. The best preprocessing method differed by cultivar. For the best and worst models of SCC, the R2 values were 0.80 and -0.1, respectively. Model performance may differ according to the color of the pericarp. Also, selection of an appropriate preprocessing method is important when using small spectroscopic devices to measure tomato fruit quality.

Coumarin is a dietary-derived substance that is extensively metabolized by human liver to excretable 7-hydroxycoumarin. Although coumarin under daily dietary consumption is generally regarded as nontoxic, the substance is of toxicological and clinical interest because of its potential association with hepatotoxicity, which is especially evident in rats. In this study, the pharmacokinetics of coumarin were modeled after virtual oral administration in humans. The adjusted monitoring equivalents of coumarin, along with the biotransformation of coumarin to o-hydroxyphenylacetic acid (via 3,4-epoxidation) based on reported plasma concentrations from rat studies, were scaled to human coumarin equivalents using known species allometric scaling factors. Using rat and human liver preparations, data on the rapid in vitro metabolic clearance for humans (~50-fold faster than in rats) were obtained for in vitro–in vivo extrapolation. For human physiologically based pharmacokinetic (PBPK) modeling, the metabolic ratios to o-hydroxyphenylacetic acid and 7-hydroxycoumarin were set at minor (0.1) and major (0.9) levels for the total disappearance of coumarin. The resulting modeled plasma concentration curves in humans generated by simple PBPK models were consistent with reported simulated coumarin maximum concentrations. These results provide basic information to simulate plasma levels of coumarin and its primary metabolite 7-hydroxycoumarin or its secondary activated metabolite o-hydroxyphenylacetic acid (via 3,4-epoxidation) resulting from dietary foodstuff consumption. Under the current assumptions, little toxicological impact of coumarin was evident in humans, thereby indicating the usefulness of forward dosimetry using PBPK modeling for human risk assessment.

Osteoarthritis (OA) is one of the most prevalent disorders in today’s society, resulting in significant socio-economic costs and morbidity. MRI is widely used as a non-invasive imaging tool for OA of the knee. However, conventional knee MRI has limitations to detect subtle early cartilage degeneration before morphological changes are visually apparent. Novel MRI pulse sequences for cartilage assessment have recently received increased attention due to newly developed compositional MRI techniques, including: T2 mapping, T1rho mapping, delayed gadolinium-enhanced MRI of cartilage (dGEMRIC), sodium MRI, diffusion-weighted imaging (DWI)/ diffusion tensor imaging (DTI), ultrashort TE (uTE), and glycosaminoglycan specific chemical exchange saturation transfer (gagCEST) imaging. In this article, we will first review these quantitative assessments. Then, we will discuss the variations of quantitative values of knee articular cartilage with cartilage layer (depth)- and angle (regional)-dependent approaches. Multiple MRI sequence techniques can discern qualitative differences in knee cartilage. Normal articular hyaline cartilage has a zonal variation in T2 relaxation times with increasing T2 values from the subchondral bone to the articular surface. T1rho values were also higher in the superficial layer than in the deep layer in most locations in the medial and lateral femoral condyles, including the weight-bearing portion. Magic angle effect on T2 mapping is clearly observed in the both medial and lateral femoral condyles, especially within the deep layers. One of the limitations for clinical use of these compositional assessments is a long scan time. Recent new approaches with compressed sensing (CS) and MR fingerprinting (MRF) have potential to provide accurate and fast quantitative cartilage assessments.
