Summary Many attempts to utilize ICT (Information and Communication Technology) tools have been made to promote active and interactive learning between teachers and students. A major area has been the introduction of education using clickers for dental students, and some reports indicated their effects. However, the introduction of mobile devices and software and their maintenance and management very costly. On the other hand, almost allundergraduate students of Matsumoto Dental University have a smartphone, and can connect to the Internet in the lecture room. Therefore, we conducted online exercises to pre- pare for the National Board Dental Examination using students' smartphones and the online real–time voting service"Mentimeter" at the lecture of Operative Dentistry for 6th–year dental students of Matsumoto Dental University, in the school year of 2019. We also conducted some anonymous online questionnaires using Mentimeter. Thirty percent of the participating students answered that they were not good at conservation and restoration studies. Although 97% of students answered that the test questions conducted in the exercise using Mentimeter were difficult, 92% of the students answered that the exercise was effective in deepening their knowledge of Operative Dentistry. The results of the questionnaires suggested the effectiveness of the interactive lecture using Mentimeter in order to deepen and retain knowledge of Operative Dentistry.

The purpose of this paper is to consider the relations between the reproduction of modern rationalized social system and its resistant, differentiating or dissimilating components through two famous authors’ two famous works of the same period. Michel de Certeau’s L’invention du quotidian: Arts de faire (published in 1980, English title: “the Practice of Everyday Life”) and Paul Willis’s Learning to Labour: How Working Class Kids get Working Class Jobs (published in 1977). In L’invention du quotidian de Certeau tries to delineate the ‘creativity’ of ‘users’ or consumers in everyday practice, who are ‘commonly assumed to be passive and guided by established rules.’ According to de Certeau, ‘users’ not only consume various products – from commodity to urban space – of modern rationalized and expansionistic technocracy and capitalism, but also bricolent(bricolage) those products in their practices of everyday life where users dissimilate or transgress the control, ‘discipline’ and plans of producers and technocrats, in which de Certeau finds out the ‘creativity’ of ‘users’ and the resistant moments with in the existing systems of dominance relationships of ‘elite’ and ‘popular culture.’ In Learning to Labour , Willis tries to identify the resistant moments in working class culture, especially ‘failed’ working class kids and their counter-school culture which oppose to authorities of school, teachers and academic carrier-based pseudo egalitarian institutions and society. He expounds on the reproductive processes of ‘working class kids get working class jobs, because of various ‘limitations’ which distort the ‘penetrations’ of working class culture which are, he thinks, ‘potential materials…for a thoroughly critical analysis of society and political action for the creation of alternatives.’ Willis situates ‘creativity’ of working class culture in their collective level of the ‘penetrations’ which exist like Freudian ‘unconsciousness’ and cannot be seized in utterance or conscious level of working class people, while de Certeau finds the creativity of resistance in everyday surface level practices like speaking, walking, cooking. This paper has scrutinized the differences between de Certeau,’s and Willis’s ideas of resistant alternative ‘creativity’ in everyday practices and popular culture, and their positions within the ‘reproduction’ of modern capitalist class society.

Dioscorea (Dioscoreaceae) is a diverse genus of more than 600 species. To understand relationships and character evolution within the genus, 273 samples from 183 species (including 28 newly sequenced species) based on four cpDNA regions were analyzed phylogenetically. The phylogenetic tree obtained comprised eleven well-supported major clades, most of which further consisted of more than two subclades. Comparisons with previously proposed infrageneric taxa (23 to 58 sections and associated ‘genera’) showed that some sections/‘genera’ are monophyletic and others polyphyletic. As in previous studies, ‘D. sect. Stenophora’ was sister to the rest of the genus. The present analyses of character state distribution on the tree confirmed that ‘D. sect. Stenophora’ is characterized by having rhizomes, monosulcate pollen and a diploid chromosome number based on x = 10 (plesiomorphies), whereas the rest of the genus has tubers and bisulcate pollen (apomorphies), but is diverse in regard to chromosome number, stem twining direction, fruit types and seed wing morphology. Based on molecular and morphological evidence, two subgenera, Dioscorea (= ‘D. sect. Stenophora’) and Helmia, are proposed. For subgenus Helmia, a revision of the infrageneric classification, especially for the species in the Old World, is needed.

Purpose: To assess the diagnostic performance of the tumor contact length (TCL) and apparent diffusion coefficient (ADC) for predicting extraprostatic extension (EPE) of prostate cancer with capsular abutment (CA).Methods: Ninety-three patients with biopsy-proven prostate cancer underwent 3-Tesla MRI, including diffusion-weighted imaging (b value = 0, 2000 s/mm2) and radical prostatectomy. Two experienced radiologists, blinded to the clinicopathological data, retrospectively assessed the presence of CA on T2-weighted imaging (T2WI). TCL on T2WI and ADC values were measured on detecting CA in prostate cancer. We used the receiver operating characteristic curves to assess the diagnostic performance of TCL and ADC values for predicting EPE.Results: CA was present in 58 prostate cancers among 93 patients. The cut-off value for TCL was 6.9 mm, which yielded an area under the curve (AUC) of 0.75. This corresponded to a sensitivity, specificity, and accuracy of 84.2%, 61.5%, and 69.0%, respectively. The cut-off value for ADC was 0.63 × 10–3 mm2/s, which yielded an AUC of 0.76. This, in turn, corresponded to a sensitivity, specificity, and accuracy of 84.2%, 59.0%, and 67.2%, respectively. The combined cut-off value of TCL and ADC yielded an AUC of 0.82. The specificity (84.6%) and accuracy (81.0%) of the combined value were superior to their individual values (P < 0.05).Conclusion: A combination of TCL and ADC values provided high specificity and accuracy for detecting EPE of prostatic cancer with CA.

A new variety, Goodyera schlechtendaliana Rchb. f. var. yakushimensis Suetsugu & Hiros. Hayak. (Orchidaceae), from Yakushima and Okinawa, is described. It is distinguished from G. schlechtendaliana var. schlechtendaliana by a combination of morphological characters, such as straighter lateral sepals, straighter lip, sparsely hairy inside the concave-saccate hypochile, and slender leaves.

A stone-embedded fish passage (SEF) allows fish to pass a barrier by reducing the velocity in the slope. The efficiency of SEFs has however not been evaluated well and the design parameters are not clear. The efficiency of SEFs for fish therefore needs to be evaluated and analyzed. In this research, we attempted to develop a simple equation to estimate the passability of an SEF for Ayu (Plecoglossus altivelis), which is applicable in the SEF's designing stage. We reproduced the SEF conditions experimentally. The set-up consisted of a pool and a channel, and evaluated its passability based on physical conditions such as air bubbles, velocity, and depth. With the results, we developed a decision tree to explain the passability using design variables such as discharge, channel slope, and pool size and depth. Then, to estimate the whole SEF, we proposed an equation which calculated the average of the decision tree outputs throughout the SEF's whole ascending routes. Lastly, we verified the equation through field experiments. The equation showed good agreement with the observed fish ascent success rate. This technique could therefore be used to evaluate a SEF in its designing stage.

For large and complex systems, full and enough investigation is required not to damage the public or operators, before design, construction and operation. Especially chemical plant, nuclear power plant and ships etc. must not cause any large accidents because of the safety of the public or lots of people working there. So, the new assessment, PSA (Probabilistic Safety Assessment), becomes popular in addition to deterministic safety assessment for these complex systems. In this paper, as an example of an application of PSA for the marine field, the event tree analysis method was applied to the Titanic accident. Factors which related to the Titanic accident were all selected under the those day's surrounding condition. The occurrence probabilities of the factors have been estimated from various data. The probability that the Titanic encounters the sever accidents, and number of fatality have been calculated. This report has shown the detailed procedure of event tree analysis for a marine accident. It has also shown the effectiveness of event tree analysis for the analysis of accident causes and for the improvement of the safety for various large complex systems.

Intracellular sedimentation of highly dense, starch-filled amyloplasts toward the gravity vector is likely a key initial step for gravity sensing in plants. However, recent live-cell imaging technology revealed that most amyloplasts continuously exhibit dynamic, saltatory movements in the endodermal cells of Arabidopsis stems. These complicated movements led to questions about what type of amyloplast movement triggers gravity sensing. Here we show that a confocal microscope equipped with optical tweezers can be a powerful tool to trap and manipulate amyloplasts noninvasively, while simultaneously observing cellular responses such as vacuolar dynamics in living cells. A near-infrared (λ=1064 nm) laser that was focused into the endodermal cells at 1 mW of laser power attracted and captured amyloplasts at the laser focus. The optical force exerted on the amyloplasts was theoretically estimated to be up to 1 pN. Interestingly, endosomes and trans-Golgi network were trapped at 30 mW but not at 1 mW, which is probably due to lower refractive indices of these organelles than that of the amyloplasts. Because amyloplasts are in close proximity to vacuolar membranes in endodermal cells, their physical interaction could be visualized in real time. The vacuolar membranes drastically stretched and deformed in response to the manipulated movements of amyloplasts by optical tweezers. Our new method provides deep insights into the biophysical properties of plant organelles in vivo and opens a new avenue for studying gravity-sensing mechanisms in plants.

To date, little is known about the prognostic significance of ultrasonographic findings in dogs with protein-losing enteropathy (PLE). The aim of this retrospective study was to examine the prognostic value of ultrasonographic findings in dogs with PLE. A total of 26 dogs with PLE were included: 20 dogs with chronic enteropathy and 6 dogs with gastrointestinal lymphoma. The presence of small intestinal dilatation was associated with shorter survival time in dogs with PLE (P=0.003). The presence of hyperechoic intestinal mucosal striations was associated with longer survival time in dogs with PLE (P=0.0085). The results of the current study indicate that the presence of small intestinal dilatation might be associated with poor prognosis in dogs with PLE.

Since thoracic MR imaging was first used in a clinical setting, it has been suggested that MR imaging has limited clinical utility for thoracic diseases, especially lung diseases, in comparison with x-ray CT and positron emission tomography (PET)/CT. However, in many countries and states and for specific indications, MR imaging has recently become practicable. In addition, recently developed pulmonary MR imaging with ultra-short TE (UTE) and zero TE (ZTE) has enhanced the utility of MR imaging for thoracic diseases in routine clinical practice. Furthermore, MR imaging has been introduced as being capable of assessing pulmonary function. It should be borne in mind, however, that these applications have so far been academically and clinically used only for healthy volunteers, but not for patients with various pulmonary diseases in Japan or other countries. In 2020, the Fleischner Society published a new report, which provides consensus expert opinions regarding appropriate clinical indications of pulmonary MR imaging for not only oncologic but also pulmonary diseases. This review article presents a brief history of MR imaging for thoracic diseases regarding its technical aspects and major clinical indications in Japan 1) in terms of what is currently available, 2) promising but requiring further validation or evaluation, and 3) developments warranting research investigations in preclinical or patient studies. State-of-the-art MR imaging can non-invasively visualize lung structural and functional abnormalities without ionizing radiation and thus provide an alternative to CT. MR imaging is considered as a tool for providing unique information. Moreover, prospective, randomized, and multi-center trials should be conducted to directly compare MR imaging with conventional methods to determine whether the former has equal or superior clinical relevance. The results of these trials together with continued improvements are expected to update or modify recommendations for the use of MRI in near future.

Microbial risk assessment in food safety is a valuable tool to reduce the risks of infection by pathogens. The dose-response relation is aimed to establish the relationship between the dose of a pathogen that populations are exposed to and the probability of the adverse health effect by the pathogen. Among many dose–response models ever proposed, the exponential and beta-Poisson models have been internationally applied, but the decision on which model is selected between them solely depends on the goodness of fit to specific data sets. On the other hands, the log-logistic model, one of the alternative models, has been little studied on the dose–response relation. In the present study, thus, the application of the log-logistic model to dose–response relation was studied with hypothetical and experimental data sets of infection (or death), comparing to the above two models. Here the experimental data sets were for pathogenic organisms such as pathogenic Escherichia coli, Listeria monocytogenes, and Cryptosporidium pavrum. Consequently, this model successfully fit to those data sets in comparison to the two models. These results suggested that log-logistic model would have the potential to apply to the dose–response relation, similar to the exponential and beta-Poisson models.

An off-season community influenza outbreak with high prevalence of amantadine-resistant influenza A/H3N2 occurred during September-October 2005 in Nagasaki Prefecture, Japan, prior to standard influenza circulation. A total of 48 patients with influenza-like-illness (ILI) visited a clinic during the outbreak and 27 (69.2%) of 39 ILI patients were positive for influenza A with rapid antigen testing (Quick Vue Rapid SP Influ). Nine patients were not tested because their symptoms were compatible for influenza without examination. Nasopharyngeal swabs were obtained from 4 of 27 rapid test positive patients, and influenza H3N2 strain was isolated from one out of four. The 4 nasopharyngeal samples were positive for influenza A M2 gene in polymerase chain reaction, and sequencing results all showed identical mutation at position 31, serine to asparagine (S31N) in the gene, conferring amantadine resistance. The phylogenetic tree analysis demonstrated that the hemagglutinin (HA) gene sequences of the 4 samples formed a distinct cluster (named clade N) from recent circulating H3N2 strains, characterized by dual mutations at position 193, serine to phenylalanine (S193F), and at position 225, asparatic acid to asparagine (D225N). Our findings suggested that an off-season community influenza outbreak in Nagasaki was caused by a distinct clade in H3N2 (named clade N), which possessed characteristics of amantadine resistance.

Heterosis refers to the improved agronomic performance of F1 hybrids relative to their parents. Although this phenomenon is widely employed to increase biomass, yield, and stress tolerance of plants, the underlying molecular mechanisms remain unclear. To dissect the metabolic fluctuations derived from genomic and/or environmental differences contributing to the improved biomass of F1 hybrids relative to their parents, we optimized the growth condition for Arabidopsis thaliana F1 hybrids and their parents. Modest but statistically significant increase in the biomass of F1 hybrids was observed. Plant samples grown under the optimized condition were also utilized for integrated omics analysis to capture specific changes in the F1 hybrids. Metabolite profiling of F1 hybrids and parent plants was performed using gas chromatography-mass spectrometry. Among the detected 237 metabolites, 2-oxoglutarate (2-OG) and malate levels were lower and the level of aspartate was higher in the F1 hybrids than in each parent. In addition, microarray analysis revealed that there were 44 up-regulated and 12 down-regulated genes with more than 1.5-fold changes in expression levels in the F1 hybrid compared to each parent. Gene ontology (GO) analyses indicated that genes up-regulated in the F1 hybrids were largely related to organic nitrogen (N) process. Quantitative PCR verified that glutamine synthetase 2 (AtGLN2) was upregulated in the F1 hybrids, while other genes encoding enzymes in the GS-GOGAT cycle showed no significant differences between the hybrid and parent lines. These results suggested the existence of metabolic regulation that coordinates biomass and N metabolism involving AtGLN2 in F1 hybrids.

Bokashi fertilizer, an organic fertilizer made of plant residue, has been used in Japan not only to fertilize plants but to regulate their growth. Lactic acid bacteria have been found to play an important role in the fermentation process of Bokashi, but the relationship between these bacteria and plant growth activity has not been clarified. Using the adzuki rooting assay, this study identified 3-phenyllactic acid (PLA) produced by lactic acid bacteria as a root promoting compound in Bokashi. PLA showed synergistic effect with tryptophan, but no stem elongation activity. Lactic acid bacteria produced equal quantities of the L- and D-forms of PLA, which have similar root promoting activity. PLA did not significantly affect the amount of endogenous indole-3-acetic acid (IAA), although the chemical structure of PLA is highly similar to that of L-2-aminooxy-3-phenypropionic acid (L-AOPP), which inhibits IAA biosynthesis. These results indicate that the root promoting activity of PLA is not simply due to its increase in the amount of active auxin.

The constitutive active/androstane receptor (CAR) is a nuclear receptor that functions as a xenobiotic sensor, which regulates the expression of enzymes involved in drug metabolism and of efflux transporters. Evaluation of the binding properties between CAR and a drug was assumed to facilitate the prediction of drug–drug interaction, thereby contributing to drug discovery. The purpose of this study is to construct a system for the rapid evaluation of interactions between CAR and drugs. We prepared recombinant CAR protein using the Escherichia coli expression system. Since isolated CAR protein is known to be unstable, we designed a fusion protein with the CAR binding sequence of the nuclear receptor coactivator 1 (NCOA1), which was expressed as a fusion protein with maltose binding protein (MBP), and purified it by several chromatography steps. The thus-obtained CAR/NCOA1 tethered protein (CAR-NCOA1) was used to evaluate the interactions of CAR with agonists and inverse agonists by a thermal denaturation experiment using differential scanning fluorometry (DSF) in the presence and absence of drugs. An increase in the melting temperature was observed with the addition of the drugs, confirming the direct interaction between them and CAR. DSF is easy to set up and compatible with multiwell plate devices (such as 96-well plates). The use of DSF and the CAR-NCOA1 fusion protein together allows for the rapid evaluation of the interaction between a drug and CAR, and is thereby considered to be useful in drug discovery.

A male 25-month-old Dalmatian dog attended our veterinary hospital because of anorexia and high circulating liver enzyme activities. Abdominal computed tomography showed a slightly small liver with rounded edges, and laparoscopic examination showed that the liver was yellowish. Histopathological examination revealed multifocal necrosis of hepatocytes and severe chronic hepatitis. Rhodanine staining showed severe copper accumulation in hepatocytes and a quantitative analysis of the copper content of the liver showed substantial accumulation (10.3 mg/g dry mass), suggesting a diagnosis of copper-associated hepatitis. Previously reported canine mutation in the COMMD1, the gene responsible for the copper-associated hepatitis in the Bedlington terrier, was not identified. To our knowledge, this is the first report of copper-associated hepatitis in a Dalmatian in Japan.
