Lipopolysaccharide (LPS)-induced inflammation is the leading cause of multiple organ failure in sepsis. Pyruvate kinase 2 (PKM2) is a protein kinase and transcriptional coactivator that plays an important role in glycolysis. Recent studies have confirmed that glycolysis maintains the M1 differentiation and induces immune activation in macrophages. Lycium barbarum polysaccharide (LBP), the main bioactive component of Chinese wolfberry, suppresses glycolysis and inflammation. Here, RAW264.7 macrophages were treated with LBP for evaluating its effects against LPS-induced inflammation. The differentiation of M1/M2 macrophages was assessed by flow cytometry for assessing the cell surface markers, CD86 and CD206. The enrichment of hypoxia inducible factor (HIF)-1α and ubiquitin in the PKM2 protein complex was determined by co-immunoprecipitation. LBP suppressed LPS-induced glycolysis, differentiation of M1 macrophages, and the production of interleukin (IL)-1β, tumor necrosis factor (TNF)-α, and high mobility group (HMG) 1 proteins. The suppressive effects of LBP were similar to those of PKM2 knockdown, but were abolished by the overexpression of PKM2. LPS elevated the mRNA and protein levels of PKM2. LBP reduced the LPS-induced expression of PKM2 protein, but had no effects on the expression of PKM2 mRNA. LPS inhibited the ubiquitination of PKM2, probably by downregulating the expression of ubiquitin ligases, including Nedd4L, Nedd4, and Gnb2. LBP interfered with the inhibition of PKM2 ubiquitination by upregulating the expression of Nedd4L, Nedd4, and Gnb2. In conclusion, LBP suppressed the LPS-induced inflammation by altering glycolysis and the M1 differentiation of macrophages. The effects of LBP were mediated by the downregulation of PKM2 via enhanced ubiquitination.

A high purity powder of Sr2MgMoO6−δ (SMM) was synthesized by a solid-state reaction under atmosphere-controlled conditions. The powder was sintered at a relatively low temperature (1200°C) to prepare compacts with high grain boundary density. SMM is a promising anode material for solid oxide fuel cells and hence, the samples were characterized under conditions relevant to this application. The electrical conductivity values of SMM under various oxygen partial pressures (pO2 = 10−15–104.3 Pa) over the temperature range of 300–850°C were measured by AC impedance spectroscopy. The grain boundary resistivity and bulk resistivity exhibited different oxygen-partial-pressure-dependent behavior. Electron conduction is proposed as the primary mechanism for electrical conductivity in the bulk of the sintered body of SMM under both low and high oxygen partial pressures. Electron conduction and oxygen ion conduction were found to be the primary mechanisms for electrical conductivity at the grain boundary of SMM under low and high oxygen partial pressures, respectively.

ヒトを含む社会的動物は,同種他個体と親密で永続的な関係(以下,社会的絆)を築く。この社会的絆を強く築く個体は繁殖において有利であることが報告されてきている(e.g., Cameron, Setsaas & Linklater, 2009)。また,de Waal & Luttrell(1986)では,個体は自分と類似した集団内の他個体を親和的な相互作用の相手として選び,社会的絆を形成し始める可能性が示唆されている。実際,ウマ(Equus caballus)においてはより年齢の近い個体間で強い社会的絆が築かれる(ワイルズ,2019)。しかし,ウマが他個体と相互作用をする前に,似た年齢の個体を選好しているかどうかは明らかになっていない。本研究では,ウマが,年齢の異なる未知のウマの顔写真(幼若・同齢・老齢)を単呈示された時に,同年齢の同種他個体に対して視覚的選好を示すかどうかを検討した。その結果,写真のウマの年齢は参加個体の刺激に対する注視行動・接近行動・接触行動に影響しなかった。これらの結果は,ウマは相互作用をする前に,その顔写真に対して同齢他個体への視覚的選好を示さないことを示唆している。

Immunochromatography assay is an easy and rapid on-site detection method. However, conventional sandwich immunochromatographies using two antibodies can only detect target molecules above a threshold size. Small molecules below 1000 in molecular weight are usually detected using competitive immunoassay. However, competitive immunoassay is not suitable for visual detection of low concentration samples. Based on the principles of open sandwich immunoassay, which detects small molecules via interchain interaction of separated variable region fragments (VH and VL) from a single antibody, we developed non-competitive open sandwich immunochromatography. Bone Gla protein (BGP)-C7, a peptide containing the seven C-terminal amino acids of human osteocalcin, was selected as the target. By using VL fragments fixed on a nitrocellulose membrane, and colored cellulose bead-labeled VH fragments, we specifically detected 10 ng/mL of BGP-C7. This is the first report of open sandwich immunochromatography, which is an easy and rapid method for on-site, signal-on detection of small molecules.

The repellent efficacy of 17 essential oils against the German cockroach, Blattella germanica was examined using a T-tube olfactometer. Five oils repelled B. germanica with good efficacy, ranging from 70.0 to 96.7%. Four of these oils, grapefruit, lemon, lime, and orange, were from the citrus family Rutaceae. These citrus essential oils showed similar repellent activity against two more cockroach species, such as Periplaneta americana and P. fuliginosa. Gas chromatography (GC) and GC-mass spectrometry analyses revealed that the major components responsible for the repellent activity of the citrus oils were limonene, β-pinene and γ-terpinene. Limonene appears to be the main component responsible for the repellent activity rather than β-pinene and γ-terpinene. The repellent efficacy of these components varied with different doses and the cockroach species tested. It is likely that minor components of the oils also contributed to the overall repellent activity of citrus essential oils, except orange oil. The activity of orange oil is almost solely attributed to the activity of limonene. Also, the repellent activity of citrus oil and that of each of the terpenoids makes little difference to the efficacy of a repellant against the three species of cockroaches.

Background:Data regarding the clinical features, outcomes and prognostic factors in patients presenting with acute total/subtotal occlusion of the unprotected left main coronary artery (LMCA) remain limited.Methods and Results:From a multi-center registry, 134 patients due to acute total/subtotal occlusion of the unprotected LMCA were reviewed. Emergency room (ER) status classification was defined according to the presence of cardiogenic shock and cardiopulmonary arrest (CPA) in the ER (class 1=no cardiogenic shock; class 2= cardiogenic shock but not CPA; and class 3=CPA). In-hospital mortality and cerebral performance category (CPC) as the endpoints were evaluated. One-half (67/134) of the enrolled patients presented with total occlusion of the unprotected LMCA. Regarding ER status classification, class 1, 2, and 3 were observed in 30.6%, 45.5%, and 23.9% of the patients, respectively. In-hospital mortality occurred in 73 (54.5%) patients; of the remaining patients, 52 (85.3%) could be discharged with a CPC 1 or 2. ER status classification (odds ratio 4.4 [95% confidence interval: 2.33–10.67]; P<0.001) and total occlusion of the unprotected LMCA (odds ratio 8.29 [95% confidence interval 2.93–23.46]; P<0.001) were strong predictors of in-hospital mortality.Conclusions:Acute total/subtotal occlusion involving the unprotected LMCA appeared to be associated with high in-hospital mortality. ER status classification and initial flow in the unprotected LMCA were significant predictive factors of in-hospital mortality.

Behavioral synchronization is shown not only between intra-species but also between inter-species. Previous studies reported that behavioral synchronization occurs between dogs and their owners by affiliative bonds and dogs' social skills for communicating with humans acquired by domestication. Horses also have such bonds and skills like dogs, however, there have been few empirical studies of behavioral synchronization between horses and humans. Therefore, we investigated whether the gait synchronization occurs between the horse and his/her familiar human participant while they were walking together by using angular statistics and how horses and umans interacted by analyzing entrainment. Only when human gait rhythm was controlled to be constant, gait synchronization between horse and human participants was found, though entrainment occurred only in humans. When horses and humans were walking in a more natural and uncontrolled situation, entrainment occurred in both horses and humans, though their walk adjustment did not reach synchronization. These results suggest that gait synchronization occurs between horses and humans and walk adjustment from humans is involved in such synchronization.

Background:The aim of this study was to investigate the impact of anatomical site status and major vascular complication (MVC) severity on the outcome of transfemoral transcatheter aortic valve replacement (TF-TAVR).Methods and Results:The FinnValve registry enrolled consecutive TAVR patients from 2008 to 2017. MVC was divided into 2 groups: non-access site-related MVC (i.e., MVC in aorta, aortic valve annulus or left ventricle); and access site-related MVC (i.e., MVC in iliac or femoral arteries). Severity of access site-related MVC was measured as units of red blood cell (RBC) transfusion. Of 1,842 patients who underwent TF-TAVR, 174 had MVC (9.4%; non-access site related, n=29; access site related, n=145). Patients with MVC had a significantly higher 3-year mortality than those without MVC (40.8% vs. 24.3%; HR, 2.01; 95% CI: 1.16–3.62). Adjusted 3-year mortality risk was significantly increased in the non-access site-related MVC group (mortality, 77.8%; HR, 4.30; 95% CI: 2.63–7.02), but not in the access site-related MVC group (mortality, 32.6%; HR, 1.38; 95% CI: 0.86–2.15). In the access site-related MVC group, only those with RBC transfusion ≥4 units had a significantly increased 3-year mortality risk (mortality, 51.8%; HR, 2.18; 95% CI: 1.19–3.89).Conclusions:In patients undergoing TF-TAVR, MVC was associated with an increased 3-year mortality risk, incrementally correlating with anatomical site and bleeding severity.

秋田県において菌類の子実体から得られた<i>Ceratophysella mediolobata</i> Nakamori, sp. nov. を記載した.また,沖縄県から本邦初記録となる<i>Ceratophysella liguladorsi</i> (Lee, 1974) が菌類の子実体から得られた. これらの2 種および<i>Ceratophysella tergilobata</i> (Cassagnau, 1954) は腹部第5 節に突起をもつ点で類似するが,<i>C</i>. <i>tergilobata</i> は他の2 種と上顎外片に1 本の副片毛をもつ点で区別でき(他の2 種は2 本),<i>C</i>. <i>mediolobata</i> sp. nov. は<i>C</i>. <i>liguladorsi</i> と腹部第4 節背面の後列の第3 毛を欠く点で区別できる(<i>C</i>. <i>liguladorsi</i> は第3 毛をもつ).DNAバーコードによる同定を可能にするために,<i>C</i>. <i>mediolobata</i> sp. nov. および<i>C</i>. <i>liguladorsi</i> のミトコンドリアのチトクロームC オキシダーゼサブユニットI 遺伝子および16S リボソームRNA 遺伝子の一部分の塩基配列を決定した.
