Relatively young (4-week-old) selenium deficient (SeD) mice, which lack the activity of selenium-dependent glutathione peroxidase (GSH-Px) isomers, were prepared using torula yeast-based SeD diet. Mice were fed the torula yeast-based SeD diet and ultra-pure water. Several different timings for starting the SeD diet were assessed. The weekly time course of liver comprehensive GSH-Px activity after weaning was monitored. Protein expression levels of GPx1 and 4 in the liver were measured by Western blot analysis. Gene expression levels of GPx1, 2, 3, 4, and 7 in the liver were measured by quantitative real-time PCR. Apoptotic activity of thymocytes after hydrogen peroxide (H2O2) exposure was compared. Thirty-day survival rates after whole-body X-ray irradiation were estimated. Pre-birth or right-after-birth starting of the SeD diet in dams was unable to lead to creation of SeD mice due to neonatal death. This suggests that Se is necessary for normal birth and healthy growing of mouse pups. Starting the mother on the SeD diet from 2 weeks after giving birth (SeD-trial-2w group) resulted in a usable SeD mouse model. The liver GSH-Px activity of the SeD-trial-2w group was almost none from 4 week olds, but the mice survived for more than 63 weeks. Protein and gene expression of GPx1 was suppressed in the SeD-trial-2w group, but that of GPx4 was not. The thymocytes of the SeD-trial-2w group were sensitive to H2O2-induced apoptosis. The SeD-trial-2w group was sensitive to whole-body X-ray irradiation compared with control mice. The SeD-trial-2w model may be a useful animal model for H2O2/hydroperoxide-induced oxidative stress.

While lead-free (Li,Na,K)NbO3 piezoceramics have a high Curie temperature (TC > 400 °C) and an excellent piezoelectric constant (d33 > 200 pC/N), the process window regarding the sintering temperature and dwelling time is narrow. Inappropriate sintering conditions can induce the volatilization of A-site alkali ions, leading to a deterioration in dielectric and piezoelectric properties. However, the effect of alkali volatilization on the ferroelastic properties is still uncertain. In this study, (Li,Na,K)NbO3 piezoceramics were deliberately exposed to the sintering temperature for varying times to compare the susceptibility of the effect of alkali volatilization on the ferroelastic properties and compared to changes in the ferroelectric and ferroelastic and dielectric properties.

Vitamin A is an essential nutrient necessary for human growth and development, with critical roles in vision, immune function reproduction and maintenance of epithelial cellular integrity. Inadequate intake of vitamin A places populations at risk of developing diseases associated with vitamin A deficiency (VAD). VAD is highly prevalent across the Eastern Mediterranean Region (EMR) in children under 5 y and women of childbearing age. Therefore, infants and young children, pregnant women and postpartum women are commonly targeted by supplementation programs. Although, vitamin A supplementation has been shown to decrease preventable childhood diseases and deaths related to VAD, supplementation of vitamin A has been greatly misused in several countries within the EMR raising concern around the process of supplementing the target population. Countries across the EMR have reported different supplementation practices depending on the income level of the country, the availability of vitamin A and the prevalence rates of VAD. Although some countries had higher supplementation rates than others, the concern lies in the middle-income countries and their supplementation practices. Some of the countries across the region do not follow the World Health Organization’s (WHO) guidelines for vitamin A supplementation for the recommended age groups. The objective of this study is to assess the vitamin A supplementation practices across the countries in the EMR, determine the gaps in the supplementation practices and the issue with supplementing to healthy populations where VAD is not a public health concern, and provide recommendations for proper vitamin A supplementation within the region.

The “Frog in Space” (FRIS) experiment marked a major step for Japanese space life science, on the occasion of the first space flight of a Japanese cosmonaut. At the core of FRIS were six Japanese tree frogs, Hyla japonica, flown on Space Station Mir for 8 days in 1990. The behavior of these frogs was observed and recorded under microgravity. The frogs took up a “parachuting” posture when drifting in a free volume on Mir. When perched on surfaces, they typically sat with their heads bent backward. Such a peculiar posture, after long exposure to microgravity, is discussed in light of motion sickness in amphibians. Histological examinations and other studies were made on the specimens upon recovery. Some organs, such as the liver and the vertebra, showed changes as a result of space flight; others were unaffected. Studies that followed FRIS have been conducted to prepare for a second FRIS on the International Space Station. Interspecific diversity in the behavioral reactions of anurans to changes in acceleration is the major focus of these investigations. The ultimate goal of this research is to better understand how organisms have adapted to gravity through their evolution on earth.

Purpose: This study aimed to examine novel techniques using prototype endodontic obturators to obturate a resin-based sealer.Methods: Powder-liquid ratios of MetaSEAL Soft were changed to obtain suitable root canal sealing, and the physical properties for various powder-liquid ratios were analyzed according to ISO-6876. Tensile bond strength was also examined. Prototype endodontic obturators with a combination of thread numbers and pitch angles were analyzed for sealing ability after MetaSEAL Soft was obturated in simulated root canals.Results: Powder-liquid ratios of 1.0:1, 1.1:1, 1.2:1, and 1.3:1 showed suitable physical properties; however, flow for 1.4:1 was below a standard value. Tensile bond strength increased gradually when the powder-liquid ratio changed from 1.0:1 to 1.3:1, and 1.3:1 and 1.4:1 showed the highest and lowest bond strengths, respectively. Sealing ability increased when pitch angles of the obturators were 5°, 8°, and 11°; 11° showed the best results. Similarly, sealing ability increased when the thread number was 12, 17, and 22 pitches; 22 showed the best results.Conclusion: These findings suggest that the prototype endodontic obturator can be useful for obturating MetaSEAL Soft, and a powder-liquid ratio of 1.3:1 MetaSEAL Soft may be the most suitable for achieving excellent sealing.

犬糸状虫抗原を用いた免疫ブロット検査により, 2000年11月から2001年8月にかけて, 山口県内で飼育されている猫315頭 (雄136頭, 雌168頭, 不明11頭) を対象に犬糸状虫の感染調査をしたところ, 19頭 (6.0%) が検査陽性と判定された. 雄 (12頭/136頭, 8.8%) は雌 (7頭/168頭, 4.2%) より陽性率が高かったが有意差は認められなかった (P=0.095). 屋内外を自由行動する猫 (11頭/199頭, 5.5%) と屋内飼育の猫 (5頭/90頭, 5.6%) との間の陽性率に有意差は認められなかった (P=0.594). 猫の年齢別では, 2歳以下 (5頭/117頭, 4.3%), 3~6歳 (6頭/86頭, 7.0%) および7歳以上 (7頭/92頭, 7.6%) の順に年齢が高くなるとともに陽性率は上昇したが有意差は認められなかった (P=0.559). 東部 (7頭/106頭, 6.6%), 中部 (6頭/123頭, 4.9%), 西部 (5頭/65頭, 7.7%) および北部 (1頭/11頭, 9.1%) の地域間の陽性率に差は認められなかった.

本研究の目的は,1人のトライアスリートによる長期間のトレーニングと連続する4大会のトライアスロン競技がDNA損傷に及ぼす影響を検討することであった。被検者は,成人男性トライアスリート(年齢:38 year,身長:185.4 cm,体重:78.8 kg)で,ハーフアイアンマン(1.9 km水泳, 90 km自転車,21.1 kmランニング)とフルアイアンマン(3.9 km水泳,180 km自転車,42.2 kmランニング)を含む連続する4競技会を完走した。競技会AとCは,ハーフアイアンマン,競技会BとDは,フルアイアンマンから構成された。競技会B,C,及びDで,8-ハイドロキシディオキシグアノジン(8-OHdG)をマーカーとしたDNA損傷と修復のバランスを定量化するために,競技会出場前日と出場後1,2,4日目に,24時間尿が採取された。競技会B,C,及びDでは,競技出場後1日目ではDNA損傷と修復が多く見られたが,4日目ではほぼ通常値に戻る傾向を示した。また,連続する競技会で,運動時間および距離が異なるにも関わらず,同様な傾向が見られた。本研究結果により,長期間のトレーニングと連続する競技会出場によって,抗酸化能力が高まることが明らかとなった。

Background: Inflammation is thought to be a risk factor for kidney disease. However, discussion is controversial whether inflammatory status is either a cause or an outcome of chronic kidney disease. We aimed to investigate the causal relationship between high-sensitivity C-reactive protein (hs-CRP) and estimated glomerular filtration rate (eGFR) using mendelian randomization (MR) approaches.Methods: A total of 10,521 participants of the Japan Multi-institutional Collaborative Cohort Study was analyzed in this study. We used two-sample MR approaches (the inverse-variance weighted (IVW), the weighted median (WM), and the MR-Egger method) to estimate the effect of genetically determined hs-CRP on kidney function. We selected four and three hs-CRP associated single nucleotide polymorphisms (SNPs) as two instrumental variables (IV): IVCRP and IVAsian, based on SNPs previously identified in European and Asian populations. IVCRP and IVAsian explained 3.4% and 3.9% of the variation in hs-CRP, respectively.Results: Using the IVCRP, genetically determined hs-CRP was not significantly associated with eGFR in the IVW and the WM methods (estimate per 1 unit increase in ln(hs-CRP), 95%CI: 0.000, –0.019 to 0.020 and –0.003, –0.019 to 0.014). For IVAsian, we found similar results using the IVW and the WM methods (estimate, 95% CI: –0.005, –0.020 to 0.010 and –0.004, –0.020 to 0.012). The MR-Egger method also showed no causal relationships between hs-CRP and eGFR (IVCRP: –0.008, –0.058 to 0.042; IVAsian: 0.001, –0.036 to 0.036).Conclusions: Our two-sample MR analyses with different IVs did not support a causal effect of hs-CRP on eGFR.

本総説において以下の知見を得たのでこれらについて述べる。1)マウスを用いた金線連の薬理活性、特に肝障害改善作用について検討した。2)ボランテイアによる脂質関連障害の改善傾向を認めた。3)金線連の粗エキスから脂質代謝改善作用を指標として kinsenoside を単離し、本化合物が活性本体であることを明らかにした。ラン科に属する金線連(Anoectochilus formosanus Hayata)は台湾から沖縄にかけて自生しており、高血圧、肺障害、肝障害、小児の発育障害等に用いられてきた。原料の枯渇から金線連の近縁種が代用として用いられるようになってきた。かかる現状から研究材料を確保する目的で、金線連の完熟種子を培地へ無菌的に播種し、発芽後幼植物を得た。このものを液体培地で培養し、植物体を得て実験材料とした。金線連のエキスは四塩化炭素で誘導する肝細胞障害を抑制することが判明した。また、マウスの体重増加や肝重量増加を抑制した。さらにマウスの肝臓や血清中の脂質を低下させる作用が認められた。ボランテイアによる金線連エキスの臨床試験の結果、健常者には作用しないが、脂質やコレステロール、またその両者が高い患者に対しては VLDL や LDL、ALT、AST 値を下げることが明らかとなった。培養金線連を抽出しカラムクロマトにて成分の精製単離を行い、8種の化合物を単離し構造を明らかにした。その中で量的にも多い kinsenoside についてはX-線解析を行い、絶対構造を明らかにした。金線連エキスにつき脂質代謝を指標として分画を行い、kinsenoside がその活性本体であることを明らかにした。Kinsenoside を金線連エキス同様の評価系にて評価した結果、肝臓や子宮の脂質量低下作用があることが明らかとなった。脂肪肝を発症させたマウスに kinsenoside を与えて顕微鏡により調査した結果、0.2%の kinsenoside を与えることにより脂肪肝は改善されることが明らかとなった。

[Purpose] The purpose of this study was to determine the concurrent validity of Wii Fit center-of-pressure parameters with Clinical Test of Sensory Integration for Balance and to evaluate the discriminant validity of the Wii Fit center-of-pressure parameters for adults and the elderly for the assessment of postural sway. [Participants and Methods] This study used an observational cross-sectional correlational design. All 70 participants were required to complete a questionnaire for eligibility screening, followed by a center-of-pressure assessment using three Wii Fit balance activities (snowboard slalom, ski slalom, and balance bubble) and Clinical Test of Sensory Integration for Balance using a force platform. [Results] For center-of-pressure assessment, our results showed there was concurrent validity of Wii Fit ski slalom and balance bubble with Clinical Test of Sensory Integration for Balance conditions 6 and 4, respectively. Our results also demonstrated that the three Wii Fit balance activities selected in this study could be used to discriminate between adults and the elderly. [Conclusion] Our results suggest that Wii Fit is a viable and affordable alternative method for center-of-pressure assessment.

Lo digo porque, comparando los casos de Jap6on y Puerto Rico, al darse en cada sociedad la transformaci6n de pais agricola a industrial, la situacion de las mujeres cambio en cada pais de manera similar. Se puede esperar que el parecido entre las situaciones favorezca una unificaci6n de intereses y tienda lazos de comunicacion fuertes, traspasando fronteras.

Cell analysis is of great significance for the exploration of human diseases and health. However, there are not many techniques for high-throughput cell analysis in the simulated cell microenvironment. The high designability of the microfluidic chip enables multiple kinds of cells to be co-cultured on the chip, with other functions such as sample preprocessing and cell manipulation. Mass spectrometry (MS) can detect a large number of biomolecules without labelling. Therefore, the application of the microfluidic chip coupled with MS has represented a major branch of cell analysis over the past decades. Here, we concisely introduce various microfluidic devices coupled with MS used for cell analysis. The main functions of microfluidic devices are described first, followed by introductions of different interfaces with different types of MS. Then, their various applications in cell analysis are highlighted, with an emphasis on cell metabolism, drug screening, and signal transduction. Current limitations and prospective trends of microfluidics coupled with MS are discussed at the end.

Iwateite, Na2BaMn(PO4)2 is a new mineral, which was found in the manganese ore of the No. 3 (Matsumaezawa) deposit in the Tanohata mine, Tanohata Village, Iwate Prefecture, Japan. Iwateite has a glaserite structure, which occurs as granular to anhedral inclusions of 10-100 μm in the main constituent minerals in manganese ore and sérandite crystal. It is colorless with white streaks, non-fluorescent, and it displays very weak pleochroism. Iwateite crystallizes in the trigonal system and belongs to the P3 space group where, a = 5.3642(10), c = 7.039(2) Å, V = 175.41(8) Å3, and Z = 1, with a calculated density of 4.06 g/cm3.

[Purpose] The present study examined the effects of treatment using PNF extension techniques on the pain, pressure pain, and neck and shoulder functions of the upper trapezius muscles of myofascial pain syndrome (MPS) patients. [Subjects] Thirty-two patients with MPS in the upper trapezius muscle were divided into two groups: a PNF group (n=16), and a control group (n=16) [Methods] The PNF group received upper trapezius muscle relaxation therapy and shoulder joint stabilizing exercises. Subjects in the control group received only the general physical therapies for the upper trapezius muscles. Subjects were measured for pain on a visual analog scale (VAS), pressure pain threshold (PPT), the neck disability index (NDI), and the Constant-Murley scale (CMS). [Results] None of the VAS, PPT, and NDI results showed significant differences between the groups, while performing postures, internal rotation, and external rotation among the CMS items showed significant differences between the groups. [Conclusion] Exercise programs that apply PNF techniques can be said to be effective at improving the function of MPS patients.

Mesenchymal stem cells (MSCs) possess regenerative and immunomodulatory properties and can control the immune dysregulation that leads to β-cell destruction. Stem-cell transplantation could thus manage insulin-dependent diabetes mellitus (IDDM) in dogs. In this pilot study, we aimed to assess canine adipose tissue-derived MSCs (cAT-MSCs) transplantation as a treatment for canine diabetes mellitus. This study included four dogs with over a year of insulin treatment for IDDM, following diagnosis at the Veterinary Medicine Teaching Hospital of Seoul National University. Allogenic cAT-MSCs were infused intravenously three or five times monthly to dogs with IDDM. Blood and urine samples were obtained monthly. General clinical symptoms, including changes in body weight, vitality, appetite, and water intake were assessed. Three of the four owners observed improvement of vitality after stem cell treatment. Two of the four dogs showed improvement in appetite and body weight, polyuria, and polydipsia. C-peptide has increased by about 5–15% in three of the cases, and fructosamine and HbA1c levels have improved in two of the cases. Hyperlipidemia was resolved in two of the dogs, and there was no concurrent bacterial cystitis in any of the dogs. C-peptide secretion and lipid metabolism are associated with diabetic complications. Improvement in these parameters following the treatment suggests that cAT-MSC transplantation in dogs with IDDM might help to improve their insulin secretory capacity and prevent diabetic complications.

Plasma N-terminal pro-atrial natriuretic peptide (NT-proANP) concentration increases with progression of myxomatous mitral valve disease (MMVD) in dogs. This multicentre, prospective study compared plasma NT-proANP, N-terminal pro-brain natriuretic peptide (NT-proBNP), ANP, and cardiac troponin I (cTnI) concentrations in dogs with MMVD for their characteristics and discriminatory ability to detect cardiac dilatation and congestive heart failure (CHF). Thirty-six healthy dogs and 69 dogs with MMVD were included. Clinical variables were obtained via physical examination, thoracic radiography, and echocardiography. The discriminatory ability of each cardiac biomarker (CB) to determine the presence or absence of cardiac dilatation (event 1) and CHF (event 2) was evaluated using the receiver operating characteristic curves. Plasma NT-proANP, NT-proBNP, and ANP concentrations showed a significant association with the left atrium/aorta ratio (P<0.01). The area under the curve of plasma NT-proANP and NT-proBNP concentrations were 0.72 and 0.75, respectively in event1 and 0.72 and 0.76, respectively in event2. Plasma NT-proANP and NT-proBNP concentrations showed sensitivity 80.0 and 80.0%; specificity 67.6 and 64.7% in event1 (cutoff value; 8,497.81 pg/ml and 1,453.00 pmol/l, respectively) and sensitivity 85.7 and 81.0%; specificity 60.4 and 64.6% in event2 (cutoff value; 8,684.33 pg/ml and 1,772.00 pmol/l, respectively). In dogs with MMVD, plasma NT-proANP, NT-proBNP, and ANP concentrations increase with left atrial enlargement.
