親密なパートナーからの暴力は,社会的,経済的,宗教的あるいは文化的背景に関わらず,公衆衛生上の健康課題となっている.日本においても,他の諸国と比較して低い状況とはいえ,親密なパートナーからの暴力が発生している.本研究では,大学1年生に対して,交際相手からのデートDV被害を受けた経験とその後の援助希求行動について,25の被害経験シナリオを含む無記名自記式質問票を用いて横断研究を行った.371人から回答を得た.男子148人(60.2%)および女子81人(64.8%)が,交際経験があると回答した(χ2=0.754, df=1, p=0.385).交際経験がある学生中,男子69人(46.6%)および女子49人(60.5%)が,何らかのデートDV被害を受けており,女子の方が統計的有意にデートDV被害を経験していた(χ2=4.033, df=1, p=0.045).最も回答が多かった被害経験は,「交際相手から『ブス/ブサイク』や『バカ』などと言われたことがある」であり,男子24人(16.2%)および女子26人(31.2%)であった.デートDV被害後,援助希求行動について回答した115人中,19人(16.5%)が相談していたが,相談先としては,19人全員が「友人」,4人が「母親」と回答した.専門職・機関に相談した者はいなかった.自分の意志に反したキスあるいはセックスといった性行為の経験があると回答した11人のうち,誰かに相談した者は4人,またその後交際を解消した者は4人であった.デートDVの予防や早期に適切な相手・機関への援助希求行動を促進するための介入プログラムを講じる際には,性別や文化的要素について考慮する必要性が示唆された.

Electrochemical bio-sensing using paper-based detection systems is the main focus of this review. The different existing designs of 2-dimensional and 3-dimensional sensors, and fabrication techniques are discussed. This review highlights the effect of adopting different sensor designs, distinct fabrication techniques, as well as different modification methods, in order to produce reliable and reproducible reading. The use of various nanomaterials have been demonstrated in order to modify the surface of electrodes during fabrication to further enhance the signal for subsequent analysis. The reviewed sensors were classified into categories based on their applications, such as diagnostics, environmental and food testing. One of the major advantages of using paper-based electrochemical sensors is the potential for miniaturization, which only requires relatively small amount of samples, and the low cost for the purpose of mass production. Additionally, most of the devices reviewed were made to be portable, making them well-suited for on-site detection. Finally, paper-based detection is an ideal platform to fabricate cost-effective, user-friendly and sensitive electrochemical biosensors, with large capacity for customization depending on functional needs.

A mutant of a strain of the SR organisms (SRO, transovarially transmitted male-killing spiroplasmas) of Drosophila is described. The mutant, NSRO-A, has lost the male-specific killing activity on host flies, but maintains other properties such as the clump forming spectrum and the extractable titer of the associated virus as original NSRO strain.

The aim of this study was to determine whether serum symmetric dimethylarginine (SDMA) and cystatin C (CysC) levels can be utilized as more accurate markers of early kidney dysfunction in dogs. Forty-one client-owned dogs with chronic kidney disease (CKD), which were clinically stable, and ten beagles as healthy controls were included. All dogs underwent physical examination, systemic blood pressure measurement, complete blood cell count (CBC), and plasma biochemistry analyses. Frozen serum was used for SDMA and CysC analyses. Data analysis was performed using Kruskal Wallis, Pearson’s correlation, Bland-Altman plots, and receiver operating characteristic (ROC) curve. SDMA and CysC levels were significantly higher in patients with CKD at various International Renal Interest Society (IRIS) stages than in the healthy controls. In particular, CysC level was the only biomarker that could indicate the earliest stage of CKD (IRIS stage I). Similar to these results, CysC level showed better sensitivity and specificity compared to the other biomarkers in early CKD dogs.

We examined the effects of 8 macrolide antimicrobial agents, including 14-, 15- and 16-membered ring lactone and ketoride derivatives, on hemolysis induced by lysophosphatidylcholine (LPC) in human erythrocytes. LPC induced hemolysis at concentrations above the critical micelle concentration (4 µM). Vitamin E (α-tocopherol), used as a reference drug, attenuated the 50% hemolysis induced by 6 µM LPC when present at concentrations between 1 µM and 100 mM. Clarithromycin significantly attenuated LPC-induced hemolysis at a wider range of concentrations (100 nM to 1 mM), but other macrolides attenuated hemolysis only at high concentrations (100 µM and/or 1 mM). Since vitamin E tends to stabilize membranes due to its high lipophilicity, it appears that the high lipophilicity of clarithromycin is responsible for its protective action against damage induced by LPC. However, rokitamycin was not effective, although it is more lipophilic than clarithromycin, indicating that factors other than high lipophilicity are responsible for the protective effects of macrolide antimicrobial agents against LPC-induced hemolysis. Neither vitamin E nor clarithromycin attenuated hypotonic hemolysis (60 mM NaCl) at concentrations that inhibit LPC-induced hemolysis. On the other hand, both vitamin E and clarithromycin affected LPC micelle formation, suggesting that these drugs directly bind to LPC. We therefore believe that the protective effects of clarithromycin on LPC-induced hemolysis may be related physicochemically to its high lipophilicity and 14-membered ring lactone structure, which help maintain erythrocyte membrane integrity by preventing LPC micelle formation. These drugs likely do not act by a mechanism that protects against osmotic imbalance.

A novel cold-adapted methane-oxidizing bacterium, termed TFB, was isolated from the thermoglacial Arctic karst spring, Trollosen, located in the South Spitsbergen National Park (Norway). The source water is cold and extremely low in phosphate and nitrate. The isolate belongs to the Methylovulum genus of gammaproteobacterial methanotrophs, with the closest phylogenetic affiliation with Methylovulum miyakonense and Methylovulum psychrotolerans (96.2 and 96.1% 16S rRNA gene sequence similarities, respectively). TFB is a strict aerobe that only grows in the presence of methane or methanol. It fixes atmospheric nitrogen and contains Type I intracellular membranes. The growth temperature range was 2–22°C, with an optimum at 13–18°C. The functional genes pmoA, mxaF, and nifH were identified by PCR, whereas mmoX and cbbL were not. C16:1ω5c was identified as the major fatty acid constituent, at an amount (>49%) not previously found in any methanotrophs, and is likely to play a major role in cold adaptation. Strain TFB may be regarded as a new psychrotolerant or psychrophilic species within the genus Methylovulum. The recovery of this cold-adapted bacterium from a neutral Arctic thermal spring increases our knowledge of the diversity and adaptation of extremophilic gammaproteobacterial methanotrophs in the candidate family “Methylomonadaceae”.

Cryptosporidium species infect domestic animals, livestock, and humans. These protozoan parasites are frequently reported as major environmental contaminants in many countries despite their differing climatic, socioeconomic, and demographic factors. This review focuses on the research findings that relate to Cryptosporidium epidemiology, genetic diversity, and associated risk factors relating to animals, contaminated water sources, and humans in Japan. Adequate knowledge of these factors is essential for understanding the economic and public health importance of cryptosporidiosis in Japan so that effective control strategies against it are implemented. Cryptosporidium infections are highly prevalent in animals in Japan. Among the different animal species, cattle infections stand out because of their economic importance and zoonotic potential. Living circumstances in Japan restrain Cryptosporidium transmission between humans, but there is evidence to suggest that animals, especially those in close contact with humans, can be potential sources of human infections. Water sampling studies have provided clues about how environmental contamination with Cryptosporidium oocysts can cause infections in livestock and wild animals. There is some evidence of person-to-person transmission of cryptosporidiosis, but only occasionally and under certain circumstances. By identifying the major role played by animals in Cryptosporidium transmission to people in Japan, we highlight the urgent need for disease control against this pathogen.

Epidemiologic studies have shown that low vitamin D levels are associated with reduced insulin sensitivity and increased risk of developing type 2 diabetes mellitus (T2DM). However, there is little evidence that vitamin D supplementation improves glucose intolerance. We evaluated the glucose-lowering effect of vitamin D in Korean T2DM subjects. We enrolled 158 T2DM patients who had stable glycemic control [hemoglobin A1c (HbA1c) <8.5%] and vitamin D levels less than 20 ng/mL. The participants were randomized into two groups: Placebo (100 mg daily of elemental calcium administered twice a day) or Vitamin D (1000 IU daily of cholecalciferol combined with 100 mg of elemental calcium administered twice a day). We compared outdoor physical activity, glycemic control, homeostasis model of assessment - insulin resistance (HOMA-IR), and parathyroid hormone (PTH), during the 24-week intervention. We analyzed the data of 129 participants (placebo =65, vitamin D =64) who completely followed the protocol. Outdoor physical activity and oral anti-diabetic drugs did not differ between the groups. While there were significant differences in the vitamin D levels (15.6 ± 7.1 ng/mL vs 30.2 ± 10.8 ng/mL, P<0.001) and change in PTH levels (1.4 ± 15.3 pg/mL vs -5.5 ± 9.8 pg/mL, P=0.003) between the placebo and vitamin D groups, there were no differences in HbA1c (7.27 ± 0.87% vs 7.40 ± 0.90%) (P=0.415) and HOMA-IR. Serum calcium and kidney function results showed that the vitamin D supplementation was safe. While vitamin D supplementation is safe and effective in the attainment of vitamin D sufficiency, it had no effect on long-term glycemic control for T2DM in our study.

Aim: This study aimed to investigate the association between the serum high-sensitivity C-reactive protein (hs-CRP) levels and incident atrial fibrillation risk in the general Japanese population, who have lower hs-CRP levels than the Western population, and assess whether the association is modified by sex, overweight, hypertension, and smoking status. Methods: We conducted a prospective study in 6517 Japanese men and women aged 40–79 years without atrial fibrillation at baseline and enrolled in the Circulatory Risk in Communities Study (2002–2008). The hs-CRP levels were measured using the latex particle-enhanced immunonephelometric assay. Atrial fibrillation was identified using standard 12-lead electrocardiograms and information on physician-diagnosed atrial fibrillation history from the follow-up surveys. We used a Cox proportional hazard regression stratified by community. Results: During a median follow-up of 11 years, 127 new cases of atrial fibrillation (74 and 53 cases among men and women, respectively) were found. Compared to the lowest quintile of hs-CRP levels, the multivariable hazard ratios (95% confidence intervals) were 2.54 (1.17–5.50), 2.28 (1.06–4.93), 2.92 (1.37–6.23), and 2.77 (1.30–5.91) for the second, third, fourth, and fifth (highest) quintiles, respectively. There was no significant effect modification by sex, overweight, hypertension, and smoking status (P for interaction >0.05). Conclusions: Elevated hs-CRP levels were significantly associated with increased risk of atrial fibrillation in the Japanese population. The association of hs-CRP levels with incident atrial fibrillation did not vary according to sex, overweight, hypertension status, or smoking status.

Testosterone and 5α-dihydrotestosterone (DHT) are the principal male hormones (androgens) in mammals. The enzyme, steroid 5α reductase catalyzes the conversion of testosterone (T) to its biologically potent steroid (DHT) in androgen dependent tissues. Two 5α reductase isoenzymes have been identified in rat tissues. The type I isoenzyme has been shown to be predominately expressed in the rat liver, whereas androgen target tissues of the genital tract express mainly isoenzyme II. The effects of androgens and glucocorticoids on the abundance of steroid 5α reductase type I (5αR1) messenger RNA in the rat liver were examined. Steady state levels of 5αR1 mRNA decreased dramatically to 1.5% of control levels 14 days following adrenalectomy (ADX), whereas dexamethasone (Dex) administered (0.5 mg/100 g) to ADX animals enhanced the expression of 5αR1 to twice its' normal values within 40 hours. Bilateral orchiectomy induced, within eight days, the expression of 5αR1 mRNA in the rat liver to 1.75 fold the normal value while testosterone injection failed to reduce this enhanced expression in castrated animals. Addition of Dex (1 μM) to primary cultures of rat hepatocyte resulted in a five- and three-fold reduction in the mRNA expression of 5αR1 after 24 and 48 hours, respectively. DHT (0.5 μM) however, induced the expression of 5αR1 mRNA by two- and seven-fold 24 and 48 hours post-treatment, respectively. In vitro nuclear run-on analysis of the 5αR1 gene showed no correlation between the rate of synthesis and steady state levels of this mRNA either in the intact liver or in cultured hepatocytes. These results appear to suggest that glucocorticoids and androgens differentially regulate 5αR1 mRNA in the rat liver. Moreover, our findings appear to indicate that regulation of 5αR1 gene is primarily at the post-transcriptional level.

Background: Daily step count is the most simple measure of physical activity. However, little is known about how daily step count related to time spent in different intensities of physical activity (PA) and sedentary behavior (SB).Methods: These cross-sectional data were derived from 450 older Japanese adults (56.7% men, mean age 74.3 years) who were randomly selected from three communities and responded a survey. Daily step count and time spent in moderate-to-vigorous PA (MVPA), light-intensity PA (LPA), and SB were measured using a validated wearable technology (HJA-350IT). Associations of daily step count with time spent in measured behaviors were examined by linear regression models using isometric log-ratio transformations of time-use composition, adjusting for gender, age and residential area.Results: Participants averaged 5,412 (SD: 2,878) steps/d and accumulated MVPA, LPA, and SB corresponding to 4.0, 34.8, and 61.2% of daily waking time, respectively. Daily step count significantly increased with increase in time spent in MVPA relative to other behaviors (i.e. LPA and SB) and in the ratio of LPA to SB after allowing for MVPA. After stratification, daily step count was significantly related to the ratio of LPA to SB in those taking <5,000 steps/d, but not in those taking 5,000-7,499 and ≥7,500 steps/d.Conclusions: Higher daily step count can be an indicator of not only larger relative contribution of time spent in MVPA, but also higher ratio between LPA and SB, particularly among those who are the least physically active.

Selective laser melting (SLM) is widely used for the additive manufacturing (AM) of metal components, which can produce net shape complex geometries. Novel titanium alloy/bioactive glass composites were successfully fabricated via SLM method for biomedical applications. The fabricated composites contained a Ti5Si3 phase, which resulted from the reaction between the titanium alloy and the bioactive glass. The phase can improve the bonding strength between composite and bone. Additionally, the remained amorphous bioactive glass phase could improve bioactivity. The present study opens a new avenue for developing new titanium alloy/bioactive glass composites with optimal bioactivity and bonding strength with the bone.

本稿では、日本語・英語のテキストを論理式に変換し、自動推論を行う意味解析システムccg2lambdaの基本的な機能を解説し、特にこのシステムを金融ドキュメントの処理へと応用する試みについて紹介する。ccg2lambdaでは、統語・意味解析から推論までの各モジュールが明確に区別されており、統語情報・意味合成・意味表現をCCG導出木としてグラフィカルに表現する機能をサポートしている。このため、処理プロセスのどの部分で解析エラーが起こったのかを容易に同定することが可能である。ccg2lambdaの意味解析と推論システムについて紹介した上で、特に金融ドキュメントを対象とした含意関係認識と矛盾検知について具体例に基づいて説明する。

Background:This study aimed to assess the relationship between hospital-acquired functional decline and the risk of mid-term all-cause death in older patients undergoing transcatheter aortic valve implantation (TAVI).Methods and Results:In total, 463 patients (mean age 85 years, interquartile range [IQR]: 82, 88) undergoing elective TAVI at Sakakibara Heart Institute between 2010 and 2018, who were followed up for 3 years, were enrolled in the study. Hospital-acquired functional decline after TAVI, which was defined by at least a 1-point decrease on the Short Physical Performance Battery before discharge compared to the preoperative score, was assessed. A total of 113 patients (24.4%) showed hospital-acquired functional decline after TAVI, and 50 (11.3%) patients died over a mean follow-up period of 1.9±0.8 years. Kaplan-Meier survival curves indicated that hospital-acquired functional decline was significantly associated with all-cause mortality (log-rank test, P=0.001). On multivariate Cox regression analysis, hospital-acquired functional decline was associated with a higher risk of all-cause mortality (OR 2.108, 95% CI 1.119–3.968, P=0.021) independent of sex, body mass index, advanced chronic kidney disease, and preoperative frailty, as assessed by the modified essential frail toolkit.Conclusions:Hospital-acquired functional decline is associated with mid-term all-cause mortality in older patients following TAVI. Trajectory of functional status is a vital sign, and it is useful for risk stratification in older patients following TAVI.

The recombinant production of glycoenzymes plays a significant role in industrial biocatalysis. The selection of an expression system which allows the high-level production of recombinant glycoenzymes using extracellular and intracellular expression, and the need for correct protein folding make Gram-positive bacteria interesting alternatives to the commonly used Gram-negative expression host Escherichia coli. In this overview we summarize recent approaches using Gram-positive bacteria as cell factories for the recombinant production of glycoenzymes, with a special focus on the optimization of glycoprotein production in lactic acid bacteria.

Dimethyl trisulfide (DMTS) develops in wine during storage, creating an unpleasant aroma. However, the mechanisms of DMTS formation during the wine-making process are poorly understood. We attempted to identify the factors that affect DMTS development during storage. We found that the death of yeast cells, followed by the leakage of their cell contents into the must, enhanced DMTS formation, but this effect varied between grape cultivars. Oxidation of the must and insufficient clarification of the grape juice can also lead to DMTS development during storage. However, DMTS-P1 and methanethiol, precursors of DMTS in other beverages, contributed little to DMTS formation in wine. When synthetic grape must was fermented instead of grape juice, less DMTS developed, despite most yeast cells having died. These findings indicate that unknown compounds contained in grape juice are involved in DMTS formation and that the key compounds involved in DMTS formation in wine originate from grapes.

ミドリシャミセンガイ殻よりヒドロオキシアパタイトに強く吸着するタンパク画分を単離した。これは分子量14万で, アスパラギン酸, グルタミン酸, グリシンに富む酸性タンパクである。またニンヒドリン陽性の末同定の酸性成分を含む。一方, γ-カルボキシグルタミン酸は含まない。このタンパクのアミノ酸組成はオステオネクチンやボーンモルフォジェネテックプロティン (bone morphogenetic protein) のアミノ酸組成に類似している。このタンパクはカルシウム親和性を有し, α-ヘリックス, β-構造をかなり含み, そのコンフォメーションはカルシウムイオン濃度を高めても, ほとんど変化しなかった。このタンパク画分は疎水結合, S-S結合によって会合した多量体タンパクである。

[Purpose] This study was performed to investigate the effects of yogic eye exercises on eye fatigue in undergraduate nursing students. [Subjects and Methods] The study used a pretest-posttest design with a non-equivalent control group. Forty undergraduate nursing students were selected by convenience sampling, with 20 assigned to an exercise group and 20 assigned to a control group. The yogic eye exercise intervention was performed for 60 minutes, two days a week for 8 weeks. It consisted of 8 steps: palming, blinking, sideways viewing, front and sideways viewing, rotational viewing, up and down viewing, preliminary nose tip gazing, and near and distant viewing. Eye fatigue was measured using a questionnaire for evaluating ocular fatigue. [Results] The exercise-group measurements revealed a significantly decreased eye-fatigue score compared with that of the control group. [Conclusion] These findings indicate that yogic eye exercises could reduce the eye fatigue score in undergraduate nursing students.
