Avocado fruit ripen with ethylene production after harvest and the flesh becomes soft and edible due to degradation of cell wall polysaccharides during ripening. α-l-Arabinofuranosidase is a hydrolytic enzyme known to digest arabinose-containing cell wall polysaccharides. It has been shown that its activity increased with fruit ripening. However, our previous study showed that an α-l-arabinofuranosidase gene (PaArf/Xyl3A) is expressed in the avocado fruit before ethylene production. In addition, the transcripts were detected in some organs in which the level of ethylene was extremely low. These results indicate that the gene expression is independent of ethylene. In the present study, we carried out immunoblot analyses of α-l-arabinofuranosidase at the protein level. Using a polyclonal antibody raised against Japanese pear α-l-arabinofuranosidase, two α-l-arabinofuranosidase proteins with molecular masses of 72 kDa and 68 kDa, presumably belonging to glycoside hydrolase family 3, were detected in ripening avocado fruit. The protein levels in ethylene or 1-methylcyclopropene (1-MCP)-treated fruits were examined and the results indicated that the two proteins responded to ethylene in opposite ways; the 68 kDa protein showed a temporary accumulation, whereas the 72 kDa protein exhibited dissipation possibly caused by a loss of stability. The total enzyme activity of α-l-arabinofuranosidase was elevated faster in the ethylene-treated fruit throughout ripening and was slower in the 1-MCP-treated fruit, suggesting the existence of another α-l-arabinofuranosidase, which did not cross-react with the antibody and was positively regulated by ethylene, in ripening avocado fruit.

We developed a habitat suitability model for wild boar Sus scrofa in the Mt. Baekwoonsan region of Korea. We recorded wild boar field signs (dust baths, bedding sites, digging areas, feces and tracks) and habitat variables such as the nearest distance to watercourses (DWATER) and trails (DTRAIL), slope, aspect (ASPECT), forest type and forest age. Field signs and habitat characteristics were assessed within a 25 × 25 m quadrat along seven survey routes from August 1999 to July 2000. We conducted a CATMOD regression analysis based on 416 field signs at 50 points and classified them into 3 levels of habitat importance (high, medium and low) based on the relative importance of field signs. Habitat suitability indices (P) were calculated based on significant relationships between parameters such as DWATER, DTRAIL and ASPECT resulting in the following function: P = 1/(1 + exp – (3.82 × (DWATER) – 1.21 × (DTRAIL) + 1.35 × (ASPECT))). Important habitats of wild boar were distributed near watercourses and far from trails along east and south-facing slopes.

[Purpose] We investigated the effects of upper extremity resistance exercise with elastic bands on respiratory function in children with cerebral palsy. [Subjects and Methods] Fifteen children were divided into two groups: the experimental (n=8) and control (n=7) groups. Both groups performed general exercises for 30 minutes per session, two times a week during the intervention period. The experimental group performed an additional upper extremity resistance exercise with elastic bands for 20–30 minutes per session, twice weekly for 8 weeks. Pulmonary function, and respiratory muscle and grip strength were measured before and after the exercise. [Results] There was no significant difference in forced vital capacity, forced expiratory volume in one second, the ratio of forced expiratory volume in one second to forced vital capacity, and peak expiratory flow before and after the intervention in either group. The increment of maximal expiratory pressure was significantly greater in the experimental group, compared with the control group. In addition, grip strength was significantly increased in the experimental group after the intervention than before. [Conclusion] We found that upper extremity resistance exercise with elastic bands has a positive effect on expiration and improves grip strength in children with cerebral palsy.

Improving the process of cell injection during hepatocyte transplantation requires an understanding of the causal relationships that shear, direct contact cells with a solid surface, and cell deformation have on cell viability loss. A linear shear model was used to model this loss of cell viability during their movement on a solid surface as part of the injection step of hepatocyte transplantation. Rat hepatocytes were studied under linear shear using two parallel plates, with a ”tight” condition that had a 25 μm gap, and a ”loose” gap condition with a > 25 μm gap, to determine the effects of cell deformation, and simulate cell viability loss during injection. Cell morphology and deformation were also observed using time-lapse images. Direct contact with a solid surface is deleterious for cells, and live cells became deformed under shear stress until they lost viability. The cell size could decrease or increase during deformation, and a loss of viability could occur due to a loss of membrane integrity or cell rupture. The space limitations in the tight gap could prevent cell expansion, which delayed the process of cell viability loss. In summary, preventing the direct contact of hepatocytes with a solid surface is recommended to improve the cell injection process during transplantation.

An 11-year-old female golden retriever dog had a mass at the right corner of the upper lip, which gradually increased in size and protruded into the oral cavity. The mass was removed surgically. The cut surface of the mass was smooth, whitish and solid, and covered by the oral mucosal membrane. Histopathologically, the mass consisted mainly of reticular pattern of short spindle cells that stained positively for cytokeratin AE1/AE3, α-smooth muscle actin and p63, suggestive of a myoepithelial cell phenotype. Between the neoplastic cords, there was myxoid or edematous connective tissue. Additionally, neoplastic cells with luminal epithelial and basal cell phenotypes were arranged in ducts and small islands, respectively. Based on the diverse histological and immunohistochemical features, the tumor was diagnosed as pleomorphic adenoma of the labial gland. To our knowledge, the reticular proliferation pattern of myoepithelial cells has not been described in salivary gland tumors of domestic animals.

We propose a lambda calculus λenv→ where it is possible to handle first-class environments. This calculus is based on the idea of explicit substitution, that is; λσ-calculus. Syntax of λenv→ is obtained by merging the class of terms and the one of substitutions. Reduction is made from the weak reduction of λσ-calculus. Its type system also originates in the one of λσ-calculus. Confluence of λenv→ is proved by Hardin's interpretation method which is originally used for proving confluence of λσ-calculus. We proved strong normalizability of λenv→ by reducing it to strong normalizability of a simply typed record calculus. Finally, we propose a type inference algorithm which produced a principal typing for each typable term.

We herein report a woman in her 50s with systemic lupus erythematosus (SLE) who developed swelling and pain in her fingers; the symptoms were more prominent in winter. Magnetic resonance imaging (MRI) revealed bone edema in the phalanges of both hands, which was compatible with phalangeal microgeodic syndrome (PMS). This is the first reported case of PMS in a patient with SLE and suggests that performing MRI should be considered for patients with SLE in order to assess the nature of finger symptoms and signs more precisely.

Our previous study showed that oridonin isolated from <i>Rabdosia rubescens</i> enhanced phagocytosis of apoptotic cells by macrophage-like U937 cells through tumor necrosis factor (TNF) α and interleukin (IL)-1β release. In this study, we further investigated signaling events involved in oridonin-augmented phagocytosis. Phagocytic stimulation was significantly suppressed by inhibitors, including a phosphoinositide 3-kinases (PI3K) inhibitor (wortmannin), a protein kinase C (PKC) inhibitor (stauroporine), and a phospholipase C (PLC) inhibitor (U73122). Exposure of U937 cells to oridonin caused an increase in PKC activity time- dependently, which was prevented by pretreatment with inhibitors of PI3K and PLC. Simultaneously, the activation of protein kinase B (PKB/Akt) and the increased expression of PLCγ2 were also blocked by wortmannin. In addition, an extracellular signal-regulated kinase (ERK) MAPK inhibitor, PD98059, suppressed oridonin-augmented phagocytosis, whereas the p38 MAPK inhibitor (SB203580) and c-Jun N-terminal kinase (JNK) MAPK inhibitor (SP98059) had no inhibitory effect. Furthermore, pretreatment of U937 cells with anti-TNFα and anti-IL-1β antibodies blocked oridonin-induced phagocytic stimulation as well as phosphorylation of ERK, but did not block the activation of PKC, indicating that these signaling events are triggered by oridonin, whereas secreted TNFα or IL-1β only activate the ERK-dependent pathway. Taken together, oridonin is suggested to enhance phagocytosis of apoptotic bodies by activating PI3K, PKC, and ERK-dependent pathways.<br>

A sufficient number of functional live hepatocytes delivered to a recipient is necessary for cell therapy. Preventing cell viability loss during the cell injection process is important to improve the clinical outcomes of hepatocyte transplantation. The critical location of cell viability loss is important to identify the causal relationship between the viability loss and cell injection process. In this study, the critical location of cell viability loss was determined experimentally in a rectangular microchannel by microscopic high-speed camera observations. Live hepatocyte distributions were investigated upstream and downstream, and measured on three planes, top, center, and bottom, under horizontally or vertically supplied conditions of the syringe orientation. Sedimented and uniform dispersion conditions of the live hepatocyte distribution at upstream of the microchannel were classified according to observations at horizontal and vertical syringe orientations, respectively. Higher hepatocyte viability loss was found under the sedimented condition. The results suggested that the critical location of hepatocyte viability loss was on the bottom plane of the microchannel. Furthermore, physical causes of the hepatocyte viability loss were found by micro-scale observations of the cell velocity and diameter during the cell injection process. This information may contribute to development of a guideline for the cell injection process to improve hepatocyte transplantation.

<b>Background</b> The aim of the present study was to assess the feasibility and usefulness of the arterial stiffness index (ASI) measured non-invasively by computerized oscillometry and by comparing it with the pulse wave velocity (PWV). <b>Methods and Results</b> The study group comprised 60 consutive patients who underwent coronary angiography and whose aorto-femoral PWV were obtained with a Judkins catheter. The ASI was obtained using Cardio Vision <sup>®</sup> MS-2000 (IMDP, Las Vegas, NV, USA): (i) baseline (ASI-B); (ii) hyperemia induced by compression of the arm with cuff pressure for 5 min (ASI-H); and (iii) sublingual nitroglycerin (ASI-N). In total, 34 patients had significant coronary artery disease (CAD). The PWV and all ASI were higher in patients with CAD than in those without CAD (ASI-B, 85.9±57.8 vs 48.2±24.5, p=0.001; ASI-H, 98.1±49.8 vs 48.1±21.3, p<0.01; ASI-N, 66.7±55.7 vs 33.2±27.9, p=0.002). However, only ASI-B and ASI-H were positively correlated to the PWV (ASI-B, r=0.27, p=0.03; ASI-H, r=0.49, p=0.001; ASI-N, r=0.19, p=0.16). The ASI was increased after hyperemia in patients with CAD (ASI-H, 85.9±57.8 to 98.1±49.8, p=0.01), but not in patients without CAD (ASI-H, 48.2±24.5 to 48.1±21.3, p>0.01). After adjusting their age, only ASI-H was correlated to the presence of CAD (r=0.33, p<0.01). <b>Conclusions</b> It is feasible and useful to use the ASI for detection of atherosclerotic coronary disease. The findings of ASI-H suggests that in addition to stiffening of the arterial wall itself, the impairment of flow mediated vasodilation, because of endothelial dysfunction, further increases the arterial stiffness. Liquid water is considered to be critical for life. Gibbs free energy is another factor that is important to sustain life for long durations. Gibbs free energy is obtained by reactions between reductants and oxidants, or from any other non-equilibrium state of matter. As an example, aerobic organisms use carbohydrates and oxygen to obtain energy. Many types of chemoautotrophic mechanisms are known for this process as well. On the surface of Mars, methane and oxidative compounds such as ferric oxide, sulfate and perchloride, which could provide redox-derived Gibbs free energy, have been detected. Iron-dependent methane oxidizing bacteria have been found in marine environments on Earth. This finding suggests the possible presence of methane-oxidizing bacteria on the Mars surface, if the local thermal environment and other resources permit proliferation and metabolism of bacteria. Our project aims to search for methane-oxidizing microbes on the Mars surface. Martian soil will be sampled from a depth of about 5 - 10 cm below the surface, where organisms are expected to be protected from the harsh hyper-oxidative environment of the Mars surface. Small particles less than 0.1 mm or 1 mm will be sieved from the sample, before being transferred to the analysis section by a micro-actuator. The particles will be stained with a cocktail of fluorescent reagents, and examined by fluorescence microscopy. A combination of fluorescent dyes has been selected to identify life forms in samples. A membrane-specific dye or a combination of dyes will be used to detect membranes surrounding the "cell". An intercalating fluorescent dye such as SYBR Green will be used to detect genetic compounds such as DNA. A substrate dye that emits fluorescence upon cleavage by a catalytic reaction will be used to detect the catalytic activity of the "cell". A combination of staining reagents has been chosen based on the definition of life. A membrane separating a cell from the ambient environment may lead to identification of an "individual". DNA or genetic material is required for "replication" of the life form. Catalytic reactions carried out by enzymes drive "metabolism". This combination of strategies will also be useful for detecting pre-biotic organic material as well as remnants of ancient life. Hydrolysis of the polymers in the "cell" followed by HPLC or soft ionization MS for amino acid analysis will be effective for examining whether Martian life is identical to or different from terrestrial life. The number and type of the amino acids as well as their chirality will be analyzed to distinguish whether the polymers are contaminants from Earth.

[Purpose] The purpose of this study was to investigate the effects of combined exercise training with aerobic and functional strengthening exercises on balance ability of hemiplegic stroke patients. [Subjects] The subjects were 21 patients with chronic stroke who were attending a rehabilitation center. [Methods] Participants were randomly allocated to two groups: a combined exercise training group (n=11) and a conventional exercise group (n=10). Both groups did 60 minutes exercise per day, 5 times a week for four weeks. [Results] Both groups showed significant improvements in static and dynamic balances, and the combined exercise showed greater improvement in dynamic balance. [Conclusion] The result of this study suggests that combined exercise training with functional strengthening exercise and aerobic exercise was effective at improve static and dynamic balance ability, and was more effective than conventional exercise at improving dynamic balance. This suggests that combined exercise training can be prescribed for stroke patients to reduce their risk of falls and lead to independent ADL.
