Based on the Law for the Humane Treatment and Management of Animals, the Ministry of the Environment specifies “Standards Relating to the Care and Management of Laboratory Animals and Relief of Pain”. Regarding the use of laboratory animals, Japan has avoided stringent laws and regulations and adopted a system of rationalization based on voluntary management in each institution. These measures are intended to reinforce the independence of institutions to promote creative and flexible research. By legalization of the 3R-principle, ministries promoting science and technology have established basic policies for rationalization of animal experimentation in research institutions under their jurisdiction. The Science Council of Japan formulated the “Guidelines for Proper Conduct of Animal Experiments”. These guidelines were elaborated by collaboration of representatives from all scientific fields related to animal experiments including biological, pharmaceutical, agricultural, veterinary and medical sciences. To verify the voluntary management of laboratory animal care and use, 1) a collaborating committee of laboratory animal facilities of national, public and private universities, 2) Japan Health Sciences Foundation and 3) Japanese Society for Laboratory Animal Resources are investigating or testing third party evaluation systems, respectively. Accreditation by AAALAC International has been completed for seven facilities in Japan.

The epidermis is a self-renewing tissue, the homeostasis of which is dependent upon the tight balance between proliferation and differentiation based on appropriate regulation of the cell cycle. The cell cycle regulation is dependent on the interactions among a number of cell cycle regulatory molecules, including the pituitary tumor-transforming gene 1 (PTTG1), also known as securin, a regulator of sister chromatid separation and transition from metaphase to anaphase. This study was conducted to clarify the less-known functions of PTTG1 in the epidermis by the use of keratinocytes cultured under two-dimensional (2D) or three-dimensional (3D) conditions. Forced overexpression of PTTG1 caused upregulation of cyclin B1, cyclin-dependent kinase 1 (CDK1), and c-Myc, resulting in enhanced proliferation and suppression of early differentiation without apparent alterations in terminal differentiation, and the exogenous PTTG1 was downregulated in association with cell cycle exit. In contrast, depletion of PTTG1 caused their downregulation and constrained proliferation with retention of differentiation capacity. These findings suggested that PTTG1 could alter the proliferation status by modulating the expression levels of the other cell cycle regulatory proteins, and excess PTTG1 primarily affects early differentiation of keratinocytes under the stability regulation associated with cell cycle exit.

A series of chiral three-ring type compounds with rigid shape was employed as chiral inducers for induction of chiral cholesteric liquid crystal (cholesteric LC) from achiral nematic LC. Helical twisting power of the chiral compounds was estimated with the Cano wedge method. Cholesteric LC electrolyte solution was prepared by adding the chiral compounds. Subsequently, polymerization in the cholesteric LC was carried out to produce chiroptically active polymer films. This method is different from conventional methods for synthesizing chiral polymers because neither chiral monomers nor asymmetric catalysts are employed. Surface structure and optical properties of the polymer thus prepared were examined.

The click reaction is one of the latest techniques for the chemical modification of bioactive compounds. Chemical modifications of α-amino acid side chains are gaining significance as useful and important tools for biochemical research. Biotinylation at side chain of propargylglycine using click reaction was examined. The detail quantitative analysis of Cu(I) concentration are performed to proceed the click reaction effectively.

The coastal line of our country is long, approximately 35,000km, and various forms such as a muddy beach, a sandybeach, a rocky beach and a cliff. The coastal area was very important area for our life, industry, transportation, etc. from ancienttime. It is important to understand "the mindscape" which a modern Japanese holds for coastal landscape because of the decrease of the natural coast and the terrible recreational use of the coastal area. In this study, it was intended to identify the structure ofcoastal "mindscape" and factors that would influence their mindscape. The questionnaire survey was conducted to graduate andundergraduate students from August to October 2010. The questionnaire was consisted with two main part, survey about figureof the coastal mindscape and individual background of respondents. As a result, the "sandy beach" and sea were the majorcomponent of mindscape. Because most respondents enjoyed sea bathing as recreational use, it was guessed that the viewpointfrom the beach were dominated and inland area, like coastal dune, was not described. About the mindscape of half respondents were influenced by real scenery they had seen, but the remaining respondents were influenced by some kind of media such asphotographs or TV programs.
