This study comprises a survey regarding the thoughts of Japanese teachers working in various educational institutions on improving the teaching approach of Japanese pronunciation in class and phonetics in training Japanese teachers. In this study, the relationship between teachers' beliefs (regarding language learning and teaching Japanese pronunciation) and backgrounds will be discussed on the basis of the results of the pilot survey. In conclusion, three patterns of beliefs and four groups of informants were found: Two groups teaching advanced level Japanese and taking pronunciation into consideration and other two groups teaching primary level Japanese, and not taking pronunciation into consideration.

Homoserine dehydrogenase catalyzes the conversion of L-aspartate-4-semialdehyde to L-homoserine. Homoserine dehydrogenase is required for the biosynthesis of the three essential amino acids, i.e. lysine, methionine, and isoleucine, from aspartic acid. This enzyme attracts attention also as a promising antifungal drug target. We have determined the crystal structures of homoserine dehydrogenase from Thermus thermophilus HB8 in both substrate-free form and homoserine-binding form by X-ray diffraction. Crystallization conditions were surveyed at 293 K by using a hanging-drop vapor-diffusion method. The substrate-free form was solved to 1.4 Å resolution and the homoserine-binding form was solved to 2.0 Å resolution. The homoserine dehydrogenase was a dimer with each subunit composed of three distinct domains, nucleotide-binding, dimerization, and substrate-binding domain. In the homoserine-binding forms, the amino group of homoserine made a hydrogen bond to the side chain carboxylate of Glu180. This hydrogen bond induced the conformation change of Thr165-Pro183 loop, which made the loop close to the active site. The substrate homoserine was rigidly recognized by several amino acid residues in the enzyme active site, indicating that Lys99 or Lys195 would be an essential catalytic residue to facilitate hydride transfer.

本研究は、乳児の母親を対象に「乳児の心に目を向ける傾向」を多角的に測定し、乳児の内的状態に対する目の向けやすさという認知的特徴と、乳児の内的状態に調和した関わり方という行動特徴の関連を検討した。乳児の心の状態を読み取りやすいという母親の認知的特徴は、母子自由遊び場面において、子どもと一緒にやりとりを作り上げていく養育行動と関連することが見出された。

We have applied the teaching of Japanese pronunciation using chunking, posing and 'へ' shaped intonation as an approach that is teachable and learnable. We have already compiled a pronunciation textbook for Japanese beginners and advanced learners and are currently developing teaching material for upper-intermediate learners of Japanese. The upper-intermediate learners have already learnt basic Japanese but need to develop the variation in expression and pronunciation and the ability to use the variation appropriately. Our textbook for the learners, therefore, facilitates the development of learners' pronunciation through various tasks, such as speech and discussion, to acquire the variation in expression and pronunciation. For each task, the learner outcomes are elucidated to both teachers and learners. This paper discusses the process of creating the textbook.

Latrunculin A (LatA) is the widely used reagent to depolymerize actin filaments. Its mechanism has been thought that LatA sequesters actin monomers from polymerization. Recent observation of single actin filaments by TIRF microscopy found that the binding affinity of LatA to actin monomers depends on the nucleotide status on actin. The observation of actin filaments also showed that LatA binds to actin filaments. LatA increased the depolymerization rate at ends of filaments assembled from ATP-actin to the rates of ADP-actin, but did not change these rates of ADP- or ADP-Pi-bound actin filaments. LatA also severed actin filaments. Thermodynamic analysis proposes that LatA accelerates phosphate release only at ends of actin filaments. Rapid depolymerization, severing and sequestering monomers are mechanisms of LatA to inhibit cellular actin cytoskeleton.

システム構築プロジェクトでは,QCDの観点での問題が発生する場合がある.原因として,提案活動時における見積もり項目の漏れや曖昧な前提条件,提案活動を行う担当者からの見積もり根拠の曖昧な引継ぎ等が考えられる.リスクマネジメントの一環として,この原因に働きかけリスク顕在化を未然に防止するため見積もりシートを作成し,提案活動での活用を推進することにより確実な統合変更管理の実施を試みた.そして,提案活動時の検討漏れの防止や前提条件の明確化の重要性と,仕様確定までがプロジェクトサイズを決定する重要なフェーズであり,システム構築プロジェクトを成功裏に導くQCD遵守の鍵であることを再認識した.今回の一連の試みから,実際のリスク対応実施時の問題点と対処,リスク顕在化時の対処,効果について報告する.
