Tissue engineering is a promising approach to supplement existing treatment strategies for craniofacial bone regeneration. In this study, a type I collagen scaffold made from a recombinant peptide (RCP) with an Arg-Gly-Asp motif was developed, and its effect on regeneration in critical-size mandibular bone defects was evaluated. Additionally, the combined effect of the scaffold and lipid-free dedifferentiated fat (DFAT) cells was assessed. Briefly, DFAT cells were separated from mature adipocytes by using a ceiling culture technique based on buoyancy. A 3 cm × 4 cm critical-size bone defect was created in the rat mandible, and regeneration was evaluated by using RCP with DFAT cells. Then, cultured DFAT cells and adipose-derived stem cells (ASCs) were seeded onto RCP scaffolds (DFAT/RCP and ASC/RCP) and implanted into the bone defects. Micro-computed tomography imaging at 8 weeks after implantation showed significantly greater bone regeneration in the DFAT/RCP group than in the ASC/RCP and RCP-alone groups. Similarly, histological analysis showed significantly greater bone width in the DFAT/RCP group than in the ASC/RCP and RCP-alone groups. These findings suggest that DFAT/RCP is effective for bone formation in critical-size bone defects and that DFAT cells are a promising source for bone regeneration.

米酢は米よりつくる醸造酢の一種であり, 単なる食用酢としてではなく, 東洋では民間薬として健康維持に役立ってきた歴史を持っている。著者らは, マウスを用い急性毒性と脂質代謝に及ぼす影響を検討した。その結果, 1) マウスでの急性毒性は21.5ml/kg (p. o.) でその死因は含有する酢酸による上部消化管に対する障害作用に基づくことが認められた。2) 通常食および高コレステロール食で飼育したマウスの血清コレステロール値を米酢は 2.5ml/kg (p. o.) 以上で低下させた。また, 4%酢酸水溶液でも, ほぼ同様の効果が認められた。3) 抗生物質アドリアマイシソによる心臓中の過酸化脂質 (LPO) 上昇に対し, 米酢は2.5ml/kg (p. o.) で抑制した。また, 正常マウス心臓中のLPOも5ml/kg (p. o.) 以上で低下させた。一方, 4%酢酸水溶液のこれらLPOに対する作用は弱いものだった。

東京語における「動詞未然形+ウ・ヨウ」と「動詞連用形+マショウ」は同じ文法「意志」と「勧誘」との二つの意味を表すことができる。この二つの表現ではアクセント核は元来後ろから二拍目のところにあるが、最近、特に若い女性の場合にアクセント核が消失した上昇調のイントネーションがよく聞かれる。この現象については、先行研究が「浮き上がり調」と名付けた。本研究は東京語の意志表現と勧誘表現のイントネーションがアクセント核を保ったままであるか、アクセント核が消失した「浮き上がり調」であるかどうかについて、気分による相違や男女差、年齢差との関連などを考察する。浮き上がり調ピッチパターン心理的事情

TRP channels are expressed in various cells in skin. As an organ system to border the host and environment, many nonneuronal cells, including epidermal keratinocytes and melanocytes, express several TRP channels functionally distinct from sensory processing. TRPV1 and TRPV3 in keratinocytes of the epidermis and hair apparatus inhibit proliferation, induce terminal differentiation, induce apoptosis, and promote inflammation. Activation of TRPV4, 6, and TRPA1 promotes regeneration of the severed skin barriers. TRPA1 also enhances responses in contact hypersensitivity. TRPCs in keratinocytes regulate epidermal differentiation. In human diseases with pertubered epidermal differentiation, the expression of TRPCs are altered. TRPMs, which contribute to melanin production in melanocytes, serve as significant prognosis markers in patients with metastatic melanoma. In summary, not only act in sensory processing, TRP channels also contribute to epidermal differentiation, proliferation, barrier integration, skin regeneration, and immune responses. In diseases with aberrant TRP channels, TRP channels might be good therapeutic targets.

Microbiology at the high altitude atmosphere is important for assessing the chances and limits of microbial transfer from the earth to extraterrestrial bodies. Among the microorganisms isolated from the high-atmospheric samples, spore formers and vegetative Deinococci were highly resistant against harsh environment at high altitude. From limited knowledge available to date, it is suggested that terrestrial microorganisms may have had chances to be ejected and transferred to outer space. Survival of these organisms during their space travel and proliferation on other planets might be also feasible. Directed Panspermia from Earth to extraterrestrial bodies is discussed on the basis of findings reported in literatures.

Background: Much effort has been expended on interpreting the mechanism of influenza epidemics, so as to better predict them. In addition to the obvious annual cycle of influenza epidemics, longer-term incidence patterns are present. These so-called interepidemic periods have long been a focus of epidemiology. However, there has been less investigation of the interepidemic period of influenza epidemics. In the present study, we used spectral analysis of influenza morbidity records to indentify the interepidemic period of influenza epidemics in Japan.Methods: We used time series data of the monthly incidence of influenza in Japan from January 1948 through December 1998. To evaluate the incidence data, we conducted maximum entropy method (MEM) spectral analysis, which is useful in investigating the periodicities of shorter time series, such as that of the incidence data used in the present study. We also conducted a segment time series analysis and obtained a 3-dimensional spectral array.Results: Based on the results of power spectral density (PSD) obtained from MEM spectral analysis, we identified 3 periodic modes as the interepidemic periods of the incidence data. Segment time series analysis revealed that the amount of amplitude of the interepidemic periods increased during the occurrence of influenza pandemics and decreased when vaccine programs were introduced.Conclusions: The findings suggest that the temporal behavior of the interepidemic periods of influenza epidemics is correlated with the magnitude of cross-reactive immune responses.

Fatty acids in triacylglycerol (TAG) are catabolized after digestion. However, the catabolic rates of several fatty acids bound to the α (sn-1, 3) or β (sn-2) position of TAG have not been thoroughly compared. In this study, the catabolic rates of 13C-labeled palmitic acid, oleic acid, linoleic acid, α-linolenic acid, eicosapentaenoic acid (EPA), or docosahexaenoic acid (DHA) bound to the α and β position of TAG were compared using isotope ratio mass spectrometry. The catabolic rates of the studied fatty acids were evaluated using the ratio of 13C and 12C in carbon dioxide expired from mice. The results indicated that palmitic acid, oleic acid, or α-linolenic acid bound to the β position was slowly catabolized for a long duration compared to that when bound to the α position. In contrast, EPA bound to the β position was quickly catabolized, and EPA bound to the α position was slowly catabolized for a long time. For linoleic acid or DHA, no difference in the catabolic rates was detected between the binding positions in TAG. Furthermore, EPA and DHA were less catabolized than the other fatty acids. These results indicate that the catabolic rates of fatty acids are influenced by their binding positions in TAG and that this influence on the catabolic rate differed depending on the fatty acid species.

本研究の目的は、日本語を母語とする聴覚障害児の韻律知覚能力を調べることである。そこで、2モーラ単語アクセントと2語文イントネーション(肯定/疑問)を用いて、弁別テストと識別テストを含む検査用バッテリを新たに開発した。これらの検査を東京都と千葉市にある小学校きこえの教室に通級する児童16名を対象に実施した。主な結果は、以下の通りである。1)アクセント、およびイントネーションの知覚能力と平均聴力レベルとの相関に関しては、中程度から高い負の相関(r=-0.494~-0.778)が認められた。平均聴力レベルが91dB以上(WHO の分類による最重度)であれば、アクセントとイントネーションの知覚は困難になることが示唆された。2)アクセント、およびイントネーションの知覚能力と語音明瞭度との相関に関しては、高い正の相関(r=0.694~0.966)が認められた。語音明瞭度が80%以上であれば、アクセントとイントネーションの知覚はより高いレベルになることが示唆された。

災害時情報共有のためのプロトコル、およびそのプロトコルを扱うサーバシステムを提案する。災害時において情報共有をいかに行うかは迅速な災害対応を行う上で重要であるが、実際の災害現場では紙ベースあるいは閉鎖的なシステムによる情報管理が行われており、十分な情報共有が行なわれていない。提案するシステムは、プロトコルとしてオープンな標準規格を用い、既存システムを柔軟に接続する事ができる。

The postharvest physiology of cut astilbe inflorescences (Astilbe × arendsii), which consist of many small florets with a short vase life, was studied in response to treatments to extend their vase life. Exogenous ethylene treatment at 0.3 μL·L−1 for 7 h did not affect the senescence of inflorescences or leaves of five cultivars and 0.2 mM silver thiosulfate for 2 h did not improve the quality of inflorescences of three cultivars, which indicated that ethylene is not a critical factor for senescence in astilbe florets. Continuous treatment with 2% sucrose or 2% trehalose solutions prolonged the cut inflorescence vase life of one or two of five astilbe cultivars, respectively. Pulse treatment with 2% trehalose in combination with 6% sucrose increased total soluble sugar contents from 11.4 to 57.6 mg·g−1 FW and raised the respiration rate of inflorescences from 15.3 to 28.4 μmol CO2·h−1·g−1 FW at 2 days after harvest (DAH) in the cultivar ‘Gloria Purpurea’. However, the effects of pulse treatment diminished at 4 DAH and extended the vase life from 4.0 days to a maximum of 5.6 days, which suggested that pulse treatments were inadequate to maintain sugar contents and respiration activity. Continuous treatment with 6% sucrose extended the vase life from 4.3 to 10.0 days and raised the chroma (C*) value of florets from 28.7 to 54.9 at 8 DAH. Continuous treatment with 2% trehalase + 4% sucrose markedly prolonged the vase life to 11.5 days and increased the C* value to 53.9 at 8 DAH. Under stereomicroscopic observation, continuous treatment with 2% trehalase + 4% sucrose maintained more vivid pink color of petals, styles, filaments and receptacles than those in control florets at 9 DAH. Combined treatment with 2% trehalose and 30 μM validamycin A, a potent inhibitor of trehalose metabolizing activity, induced severe wilting of florets and necrotic spots on leaves. Exogenous trehalose may be hydrolyzed by trehalose metabolizing activity in cut astilbe inflorescences. The results suggest that continuous treatment with trehalose and sucrose solutions is effective to maintain development and delay senescence of florets to extend the vase life of cut astilbe inflorescences.

In healthy dogs, amino acid infusion significantly attenuates the decrease in body temperature during anesthesia by facilitating insulin secretion, suggesting that such an increase in insulin secretion is related to increased heat production. In dogs, selective cyclooxygenase-2 (COX-2) inhibitors, which are used for pain relief in veterinary medicine, possess anti-pyretic action. And, in mice and humans, selective COX-2 inhibitors increase insulin secretion and sensitivity. Therefore, treatment with COX-2 inhibitors may negate or accelerate the attenuating effect on decreased body temperature during anesthesia by amino acid infusion. In the present study, influences on insulin secretion and body temperature by treatment with meloxicam or robenacoxib at therapeutic dose were evaluated in healthy dogs. Treatment with meloxicam or robenacoxib did not affect insulin secretion in the unanesthetized and anesthetized dogs, and did not affect body temperature and heart rate under the anesthetized condition with amino acid infusion. In conclusion, COX-2 inhibitors at therapeutic doses did not affect body temperature during anesthesia in dogs administered amino acids.

We have reported a simple method for separating lactic acid bacteria (LAB) from yogurt. This method is based on the process of destructions and denaturation of casein micelle aggregates by vortexing, and can supply samples containing only LAB. Recovered LAB were clearly observable by microscopy, meaning that morphological changes could be directly detected at the single-cell level. This method will be a helpful tool for the analyzing various LAB, including their enzyme activity and protein expression.

The activation of epidermal growth factor receptor (EGFR) involves the geometrical conversion of the extracellular domain (ECD) from the tethered to the extended forms with the dynamic rearrangement of the relative positions of four subdomains (SDs); however, this conversion process has not yet been thoroughly understood. We compare the two different forms of the X-ray crystal structures of ECD and simulate the ECD conversion process using adiabatic mapping that combines normal mode analysis of the elastic network model (ENM-NMA) and energy optimization. A comparison of the crystal structures reveals the rigidity of the intradomain geometry of the SD-I and -III backbone regardless of the form. The forward mapping from the tethered to the extended forms retains the intradomain geometry of the SD-I and -III backbone and reveals the trends to rearrange the relative positions of SD-I and -III and to dissociate the C-terminal tail of SD-IV from the hairpin loop in SD-II. The reverse mapping from the extended to the tethered forms complements the promotion of ECD conversion in the presence of epidermal growth factor (EGF).

The paracrine function of mesenchymal stem cells (MSCs) during transplantation has been recently studied due to its poor differentiation ratio. Dimethyloxalylglycine (DMOG) has been used to promote angiogenesis in experimental animal models, however, comparable approaches for canine MSCs are not sufficient. In the present study, we assessed whether DMOG improves angiogenesis in canine adipose tissue-derived mesenchymal stem cells (cAT-MSCs). cAT-MSCs were treated with DMOG and their effect on angiogenesis was investigated by cell proliferation assay, western blotting, and tube formation assay. Dimethyloxalylglycine preconditioning enhanced the expression of vascular endothelial growth factor (VEGF) among pro-angiogenic factors in cAT-MSCs via hypoxia-inducible factor-1α stabilization. Dimethyloxalylglycine primed-cAT-MSC-conditioned media increased angiogenesis in human umbilical vein endothelial cells. These results suggest that DMOG conditioning of cAT-MSCs augmented the secretion of VEGF, which acted as a prominent pro-angiogenic factor during angiogenesis. DMOG-primed cAT-MSCs may have the potential to induce beneficial effects in ischemic diseases in clinical trials.

Integrins, transmembrane molecules that facilitate cell-to-cell and cell-to-extracellular matrix interactions, are heterodimers that consist of an α- and β-subunit. The integrin α4 gene (itgα4) is expressed in various type of cells and tissues. Its biochemical functions and physiological roles have been revealed using cultured cell assays. In contrast, the primary effect caused by itgα4 deletion on vertebrate development is poorly understood, because knockout mice exhibit multiple defects that can lead to embryonic lethality in the uterus. Zebrafish are a convenient vertebrate model to investigate morphogenesis during embryogenesis, because of their external fertilization and subsequent development outside the female's body. Here, we generated a zebrafish mutant line named itgα4 ko108 using the CRISPR/Cas9 genome editing system; the mutant genome harbored an approximately 2.0-kb deletion in the itgα4 locus. A truncated transcript was detected in itgα4 (+/-) or (-/-) fish but not in (+/+) fish. The mutant transcript was hypothesized to encode a truncated Itgα4 protein due to a premature stop codon. itgα4 (-/-) embryos obtained from the mating of heterozygous parents exhibited no apparent phenotype during development at 24 hours post-fertilization (hpf). However, approximately half of them exhibited cephalic hemorrhage at 48 hpf. The incidence ratio was significantly higher than that in (+/+) or (+/-) embryos. Embryonic hemorrhage has also been reported previously in Itgα4 knockout mice. In contrast, embryonic lethality with the other defects reported in the knockout mice was not observed in our zebrafish model. Therefore, the mutant line itgα4 ko108 should be a useful model to investigate a physiological function for Itgα4 in the blood circulation system.

Objective: Alpha-actinin (ACTN) 3 R577X polymorphisms have three genotypes: RR, RX and XX. Only RR and RX genotypes express ACTN3 protein in type II fibers. The purpose of this study was to clarify whether there are differences in ACTN3 protein expression levels and myosin heavy chain (MyHC) composition between RR and RX genotypes in Japanese college-level male sprinters.Materials and Methods: Forty-three Japanese college-level male sprinters participated in this study. Subjects were genotyped for ACTN3 R577X using a real-time polymerase chain reaction assay. Furthermore, muscle biopsies from the vastus lateralis muscle were obtained from a subset of subjects who had R allele and gave their consent (4 RR and 9 RX). ACTN3 protein expression levels were assessed by western blotting. MyHC composition was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis.Results: There was no difference in 100-m sprint performance among RR, RX and XX genotypes in all 43 subjects. In 13 biopsy-sampled subjects, there was also no difference in 100-m sprint performance between RR and RX and ACTN3 protein expression levels tended to be higher in RR genotype than in RX genotype. Although there were no differences in the proportion of type I and II MyHC isoforms between both genotypes, RR genotype had a significantly higher proportion of type IIx MyHC isoform and a significantly lower proportion of type IIa MyHC isoform than RX genotype.Conclusions: ACTN3 protein expression levels and the proportion of type IIx MyHC isoform are higher in RR genotype compared with RX genotype in Japanese college-level male sprinters.

<p>Multiple factors have been associated with the honey bee colony losses in the past 25 years. Accumulating evidence indicates that at sublethal doses, neonicotinoids cause honey bee brain dysfunction and reduce immunocompetence, leading to impaired navigation and olfactory learning and memory, and susceptibility to pathogens. Moreover, such pesticides disturb the reproductive system of queen bees, i.e., the number of eggs and viability of sperm stored in the spermatheca of queen bees, thereby reducing the numbers of adult bees and broods. Pesticide exposure during the larval development stages prolongs larval development and shortens adult longevity. In addition, the density of synaptic units in the calyces of mushroom bodies in the heads decreases; this effect has been further associated with the abnormal olfactory learning ability of adult honey bees exposed to sublethal doses of imidacloprid during the larval stage. Therefore, numerous physiological aspects of honey bees might be altered after exposure to pesticides at sublethal doses, regardless of the developmental stage. We evaluated the impact of the most widely used neonicotinoids—imidacloprid—on different developmental stages of honey bees by profiling the transcriptomes of worker bees with imidacloprid during only the larval stage. Our results confirm that even intaking 1 ppb imidacloprid during only the larval stage would be enough to severely impact a bee's gene expression. The existence of many differentially expressed genes may reflect or result in honey bee disorder.</p>
