Maternal immune activation (MIA) by an infection is considered to be an important environmental factor of fetal brain development. Recent animal model on MIA induced by polyinosinic:polycytidylic acid, a mimic of viral infection, demonstrates that maternal IL-17A signaling is required for the development of autism spectrum disorder (ASD)-like behaviors of offspring. However, there is little information on bacterial infection. In this study, we aim to elucidate the influence of MIA induced by lipopolysaccharide (LPS) to mimic a bacterial infection on fetal brain development. We demonstrated that LPS-induced MIA promoted ASD-like behaviors in mouse offspring. We further found that LPS exposure induced acute phase immune response: elevation of serum IL-17A levels in MIA mothers, upregulation of Il17a mRNA expression and increase of IL-17A-producing γδ T cells in the uterus, and upregulation of Il17ra mRNA expression in the fetal brain. Blocking of IL-17A in LPS-induced MIA ameliorated ASD-like behaviors in offspring. Our data suggest that bacterial-induced maternal IL-17A pathway promotes ASD-like behaviors in offspring.

Background:The second-generation cryoballoon (CB2) has demonstrated high procedural efficacy and convincing clinical success rates for pulmonary vein isolation (PVI). Nevertheless, data on the impact of different ablations protocols on durability are limited. The aim was to comparing the durability of PVI following 3 different ablation strategies in patients with recurrence of atrial fibrillation or atrial tachycardia undergoing repeat procedures.Methods and Results:In 192 patients, a total of 751 PVs were identified. All PVs were successfully isolated during index PVI. Thirty-one out of 192 (16%) patients were treated with a bonus-freeze protocol (group 1), 67/192 (35%) patients with a no bonus-freeze protocol (group 2), and 94/192 (49%) patients with a time-to-effect-guided protocol (group 3). Persistent PVI was documented in 419/751 (55.8%) PVs, and in 41/192 (21%) patients, all PVs were persistently isolated. The total rate of PV reconnection was not significantly different between the groups (P=0.134) and the comparison of individual PVs revealed no differences (P-values for RSPV: 0.424, RIPV: 0.541, LSPV: 0.788, LIPV: 0.346, LCPV: 0.865). The procedure times were significantly reduced by omitting the bonus-freeze and applying individualized application times (group 1: 123.4±31.5 min, group 2: 112.9±39.8 min, group 3: 86.67±28.4 min, P<0.001).Conclusions:Comparing 3 common ablation protocols, no differences for durable PVI were detected. Procedure times were significantly reduced by omitting the bonus-freeze cycle and by applying individualized application times.

Thus far, there are few computed tomography (CT) characteristics that can distinguish benign and malignant etiologies. The criteria are complex, subjective, and difficult to use in clinical applications due to the high level of experience needed. This study aimed to identify practical CT variables and their clinical relevance for broadly classifying histopathological diagnoses as benign or malignant. In this prospective study, all dogs with liver nodules or masses that underwent CT examination and subsequent histopathological diagnosis were included. Signalments, CT findings and histopathological diagnoses were recorded. Seventy liver nodules or masses in 57 dogs were diagnosed, comprising 18 benign and 52 malignant lesions. Twenty-three qualitative and quantitative CT variables were evaluated using univariate and stepwise multivariate analyses, respectively. Two variables, namely, the postcontrast enhancement pattern of the lesion in the delayed phase (heterogeneous; odds ratio (OR): 14.7, 95% confidence interval (CI): 0.82–262.03, P=0.0429) and the maximal transverse diameter of the lesion (>4.5 cm; OR: 33.3, 95% CI: 2.29–484.18, P=0.0006), were significantly related to the differentiation of benign from malignant liver lesions, with an area under the curve of 0.8910, representing an accuracy of 88.6%. These findings indicate that features from triple-phase CT can provide information for distinguishing pathological varieties of focal liver lesions and for clinical decision making. Evaluations of the maximal transverse diameter and postcontrast enhancement pattern of the lesion included simple CT features for predicting liver malignancy with high accuracy in clinical settings.

Polymorphism information generated by next-generation sequencing (NGS) technologies has enabled applications of genome-wide markers assisted breeding. However, handling such large-scale data remains a challenge for experimental researchers and breeders, calling for the urgent development of a flexible and straightforward analysis tool for NGS data. We developed “IonBreeders” as bioinformatics plugins that implement general analysis steps from genotyping to genomic prediction. IonBreeders comprises three plugins, “ABH”, “IMPUTATION”, and “GENOMIC PREDICTION”, for format conversion of genotyping data, preprocessing and imputation of genotyping data, and genomic prediction, respectively. “ABH” converts genotyping data derived from NGS into the ABH format, which is acceptable for our further plugins and with other breeding software tools, R/qtl, MapMaker, and AntMap. “IMPUTATION” filters out non-informative markers and imputes missing marker genotypes. In “GENOMIC PREDICTION”, users can use four statistical methods based on their target trait, quantitative trait locus effect, and number of markers, and construct a prediction model for genomic selection. IonBreeders is operated in Torrent Suite, but can also handle genotype data in standard formats, e.g., Variant Call Format (VCF), by format conversion using free software or our provided scripts.

Background:Patients with severe aortic stenosis (AS) might be at high risk for adverse cardiovascular events at the time of non-cardiac surgery.Methods and Results:The current study population included 348 patients who underwent elective non-cardiac surgery under general or spinal anesthesia during the follow up of 3,815 patients in the CURRENT AS (Contemporary outcomes after sURgery and medical tREatmeNT in patients with severe Aortic Stenosis) registry. There were 187 patients with untreated severe AS at time of surgery (untreated severe AS group) and 161 patients who had undergone aortic valve replacement (AVR) before surgery (prior AVR group), including 23 patients with prophylactic AVR. The primary outcome measure was 30-day mortality after non-cardiac surgery. At 30 days after non-cardiac surgery, 8 patients (4.3%) died in the untreated severe AS group, while no patients died in the prior AVR group (P=0.008). The causes of death were cardiovascular in 6 out of 8 patients. Mortality at 30 days was higher in untreated severe AS patients with AS-related symptoms before surgery than in those without AS-related symptoms (7.2% vs. 3.1%). Higher surgical risk estimates of the non-cardiac surgery incrementally increased the risk of 30-day mortality in patients with untreated severe AS, though the difference was not statistically significant (low-risk: 0%, intermediate-risk: 4.3%, and high-risk: 6.6 %, P=0.46).Conclusions:Symptomatic and asymptomatic severe AS might be associated with higher risk of 30-day mortality if untreated before elective intermediate- and high-risk non-cardiac surgery, while no patient with prior AVR died after elective non-cardiac surgery.

A 1-year-old male mixed breed dog presented for the evaluation of progressive hindlimb paresis. Neurological examination indicated a spinal cord lesion between the 3rd thoracic and 3rd lumbar vertebrae. Magnetic resonance imaging (MRI) revealed an intramedullary spinal cord lesion located at the level of the 1st and 2nd lumbar vertebrae. Following cytoreductive surgery of the mass, palliative radiation therapy was administered. A diagnosis of nephroblastoma was made based on histological examination. After radiation therapy, the disappearance of the spinal lesion was confirmed by MRI. The dog was improved from gait abnormality and alive at 16 months postoperatively, with slight signs of neurological dysfunction.

Background: The disparity of overall diet quality by personal educational attainment has been a public issue. However, it remains unknown which food groups contribute to the disparity. This cross-sectional study assesses which food groups explain associations between education and overall diet quality in Japanese women.Methods: A total of 3788 middle-aged (mean: 47.7 years) and 2188 older women (mean: 74.4 years), who lived in 47 prefectures in Japan, provided data on their education (low, middle, and high) and dietary intakes from a diet history questionnaire. A diet quality score (possible score 0-70) was calculated based on seven food components. Mean diet quality scores with adjustment for lifestyle and neighborhood variables were estimated by education, and Dunnett’s multiple comparison was conducted. Additionally, mean scores of each food component were estimated by education and compared using the same manner.Results: After adjustment for lifestyle and neighborhood variables, mean diet quality score of high or middle education was higher than low education for both generations. Middle-aged women with high and middle education had higher scores of ‘milk’, ‘snacks, confection, and beverages’, ‘fruits’, and ‘vegetable dishes’ than those with low education. Older women with high and middle education had higher scores of ‘sodium from seasonings’ and ‘fruits’ than those with low education.Conclusions: This study suggests that positive associations between education and diet quality are explained by different food groups in middle-aged and older Japanese women, which are independent of lifestyle and neighborhood variables.

研究論文精神疾患に対する否定的な態度は,援助を必要とする者の専門家への受診を妨げる重大な問題であると指摘されている。しかしながら,そのような態度を測定できる青年版の尺度は存在しないというのが本邦の現況である。そこで本研究は,パブリックスティグマの一側面に位置づけられる社会的距離の概念に基づいた,青年版社会的距離の近さ尺度(Social Distance of Adolescents Scale : SDAS)の作成および信頼性と妥当性の検討を目的として行われた。中根他(2010)の成人を対象とする社会的距離尺度を参考に,教育現場での活用を考慮した上で,青年期の生徒が理解できる表現に改めた6項目が作成された。研究1では中学生を対象に調査を行い,信頼性と妥当性を検討したところ,十分な信頼性と妥当性が確認された。続いて研究2において,高校生を対象にSDASを測定し尺度の因子構造を再度探索的に検討したところ,中学生と同様の結果が得られた。SDASは不安症の子どもに対する態度を示すパブリックスティグマを測る尺度として有用であることが示唆され,SDASの汎用可能性について議論された。

We made an effort to inspect the raindrop size distribution (RSD) characteristics of Indian Ocean and Pacific Ocean tropical cyclones (TCs) using ground-based disdrometer measurements from observational sites in India and Taiwan. Five TCs (2010-2013) from the Indian Ocean and six TCs (2014-2016) from the Pacific Ocean were measured using particle size and velocity disdrometers installed in south India and south Taiwan, respectively. Significant differences between the RSDs of Indian Ocean and Pacific Ocean TCs are noticed. For example, a higher number of small drops is observed in Indian Ocean TCs, whereas Pacific Ocean TCs have more mid-size and large drops. RSDs of Pacific Ocean TCs have higher mass-weighted mean diameter and lower normalized intercept parameter than Indian Ocean TCs. RSD values quantified based on rainfall rate and precipitation types also showed similar characteristics between Indian Ocean and Pacific Ocean TCs. The radar reflectivity and rainfall rate (Z-R) relations and shape and slope (μ-Λ) relations of both oceanic (Indian and Pacific) TCs are found to be distinctly different. Possible causes for the dissimilarities in RSD features between Indian Ocean and Pacific Ocean TCs are due to relative differences in water vapor availability and convective activity between TCs in these two oceanic basins.

The aim of this paper is to show an upper bound for finding defective samples in a group testing framework. To this end, we exploit minimization of Hamming weights in coding theory and define probability of error for our decoding scheme. We derive a new upper bound on the probability of error. We show that both upper and lower bounds coincide with each other at an optimal density ratio of a group matrix. We conclude that as defective rate increases, a group matrix should be sparser to find defective samples with only a small number of tests.

Metagenomic approaches are now commonly used in microbial ecology to study microbial communities in more detail, including many strains that cannot be cultivated in the laboratory. Bioinformatic analyses make it possible to mine huge metagenomic datasets and discover general patterns that govern microbial ecosystems. However, the findings of typical metagenomic and bioinformatic analyses still do not completely describe the ecology and evolution of microbes in their environments. Most analyses still depend on straightforward sequence similarity searches against reference databases. We herein review the current state of metagenomics and bioinformatics in microbial ecology and discuss future directions for the field. New techniques will allow us to go beyond routine analyses and broaden our knowledge of microbial ecosystems. We need to enrich reference databases, promote platforms that enable meta- or comprehensive analyses of diverse metagenomic datasets, devise methods that utilize long-read sequence information, and develop more powerful bioinformatic methods to analyze data from diverse perspectives.

In this article, we propose a method called “continuous noise masking (cNM)” that allows eliminating residual buzziness in a continuous vocoder, i.e. of which all parameters are continuous and offers a simple and flexible speech analysis and synthesis system. Traditional parametric vocoders generally show a perceptible deterioration in the quality of the synthesized speech due to different processing algorithms. Furthermore, an inaccurate noise resynthesis (e.g. in breathiness or hoarseness) is also considered to be one of the main underlying causes of performance degradation, leading to noisy transients and temporal discontinuity in the synthesized speech. To overcome these issues, a new cNM is developed based on the phase distortion deviation in order to reduce the perceptual effect of the residual noise, allowing a proper reconstruction of noise characteristics, and model better the creaky voice segments that may happen in natural speech. To this end, the cNM is designed to keep only voice components under a condition of the cNM threshold while discarding others. We evaluate the proposed approach and compare with state-of-the-art vocoders using objective and subjective listening tests. Experimental results show that the proposed method can reduce the effect of residual noise and can reach the quality of other sophisticated approaches like STRAIGHT and log domain pulse model (PML).

Objective: Among various adverse reactions of atypical antipsychotics, weight gain and impaired glucose tolerance are clinically significant. The aim of this study is to analyze quantitatively the contributions of various receptors to these antipsychotics-induced adverse reactions based on the receptor occupancy theory.    Methods: Two indices of antipsychotics-induced weight gain (the values estimated by a meta-analysis and the observed values in clinical trials) and the morbidity rate of type 2 diabetes mellitus during treatment with antipsychotics were taken from the literature. We calculated the estimated mean receptor occupancies of α1 adrenergic, α2 adrenergic, dopamine D2, histamine H1, muscarinic acetylcholine (mACh), serotonin 5-HT1A, 5-HT2A and 5-HT2C receptors by antipsychotics by using the pharmacokinetic parameters and receptor dissociation constants, and analyzed the correlation between the occupancies and the extent of adverse reactions as assessed using the aforementioned indices.    Results: There were statistically significant correlations between the estimated occupancies of H1 and mACh receptors and antipsychotics-induced weight gain estimated by meta-analysis (rs=0.81 and rs=0.83, respectively, p<0.01). There were also statistically significant correlations between these receptor occupancies and observed weight gain in clinical trials (rs=0.66 in each case, p<0.01). The morbidity rate of type 2 diabetes mellitus was highly correlated with H1, mACh, and 5-HT2C receptor occupancies (rs=0.90 in each case, p<0.05). However, H1 receptor occupancy was also highly correlated with mACh receptor occupancy among antipsychotics, so that only one of them may be critically associated with the adverse reactions. Considering that these adverse reactions have not been reported for drugs with mACh receptor antagonistic action, other than antipsychotics, the H1 receptor may contribute predominantly to the antipsychotics-induced weight gain and diabetes mellitus.    Discussion/Conclusion: Model analysis based on receptor occupancy indicates that H1 receptor blockade is the primary cause of antipsychotics-induced weight gain and diabetes mellitus.

In 2018, heatwaves (HWs), which are defined as period of abnormally hot weather with the daily maximum temperature (T_Max) exceeding its 95th percentile threshold for at least three consecutive days, were prevalent during June to August, and temperature records were broken in many countries over East Asia (EA) including China (CH), Japan (JP), and the Korean Peninsula (KP). In particular, extreme HWs through July to August lasted for the longest duration with 21.3 days with T_Max reaching 36.9°C. The highest T_Max, recorded since 1907, exceeded to 41°C in Hongcheon, located east of Seoul in the KP. Here, we examine the factors that influenced the 2018 HW, and how these relate to the 1994 HW, which was second longest HW recorded in the KP. The results show that abnormally strong and northwestward extended anticyclone features observed in July 2018 lasted as a persistent North Pacific anticyclone anomaly until August 2018 centered at the northern KP. These anticyclone features subsequently formed as a modon-like blocking with a cyclonic anomaly in the East China Sea. Meanwhile, in August 1994, the North Pacific High (NPH) extended to eastern EA, which broked the meridional dipole structure and HWs do not persist. The NPH, which persisted till August 2018, was accompanied by a sinking motion, suppression of precipitation, anomalous maximum temperature, weakening of the westerly jet stream, and increased insolation due to clear sky. We find that the prolonged and northwestward-shifted NPH including the KP drove the extraordinarily hot 2018 summer in Korea. In addition, low precipitation and massive evapotranspiration with the persistent insolation in July 2018 influenced the dry condition at the surface. We suggest that the predictions for the location and duration of the NPH associated with the HWs are required to reduce heat-related mortality and the impact on agriculture due to excessive evapotranspiration.

A 72-year-old man had type 2 diabetes (T2D) that had been diagnosed at 54 years old. Macroalbuminuria was first detected at age 64. While his HbA1c had been kept below 7%, his estimated glomerular filtration rate (eGFR) was declining rapidly. At 70 years old, his eGFR dropped below 50 ml/min/1.73 m2. A renal biopsy revealed diabetic nephropathy. sodium glucose transporter 2 inhibitors (SGLT2i)/glucagon-like peptide-1 receptor agonists (GLP-1RA) combination therapy substantially improved his eGFR and urinary albumin level, and the renoprotective effect persisted for the two-year study period. These findings suggest that SGLT2i and GLP-1RA can additively improve the renal function in patients with T2D.

Using special data from the field program of “Impact of Typhoons on the Ocean in the Pacific” (2010) and an ensemble Kalman filter-based vortex initialization method, this study explores the impact of the Taiwan terrain on the uncertainty in forecasting track, intensity and rainfall of Typhoon Fanapi (2010) based on ensemble simulations. The results show that the presence of Taiwan topography leads to rapid growths of the simulation uncertainty in track and intensity during the landfall period, in particular at the earlier landfall period. The fast moving ensemble members show an earlier southward track deflection as well as the weakening of intensity, resulting in a sudden increase of standard deviation in track and intensity. During the period of offshore departure from Taiwan, our analysis suggests that the latitudinal location of the long-lasting and elongated rainband to the south of tropical cyclone (TC) center has a strong dependence on the latitude of the TC center. In addition, the rainfall uncertainty in southern Taiwan is dominated by the uncertainty of simulated TC rainband, and the latitude of TC track can be regarded as a good predictor of the rainband's location at departure time. It is also found that the rainband develop farther to the south as the topography is elevated. Considering the fact that the rainband impinging the high mountains in the southern Central Mountain Range generates the greatest accumulated rainfall, positions where the rainband associated circulation and its interaction with topography appear to offer an explanation on the uncertainty of the simulated rainfall.

縄文時代人骨3体から DNA を抽出し, PCR 法を用いてミトコンドリア DNAを増幅して解析を行い,その方法論的な問題点を考察した。今回用いた方法では,試料の保存状態•部位にかかわらず,ほぼ安定して DNA の分離と増幅が可能であり,その有効性が確かめられた。分離された DNA は,そのほとんどがヒト由来のものではなかったが, PCR 法によりヒトミトコンドリア由来の DNA の増幅を確認することができた。ミトコンドリア DNAのV 領域および D ループ領域に対する制限酵素を用いた解析では多型は検出できなかった。しかし増幅した V 領域の塩基配列を決定したところ,1個体では1箇所の変異が見出された。
