Gene doping is prohibited in horseracing and equestrian sports. In previous studies, we developed non-targeted transgene and genome editing detection methods based on whole genome resequencing (WGR) using genomic DNA extracted from whole blood. In this study, we aimed to develop a WGR method using DNA extracts from hair roots. Hair roots are a preferred substrate because their collection is less invasive than blood collection. Hair is also easier to store for long periods of time. Although almost all genomic DNA extracted from hair root samples stored for years at room temperature was degraded, the quality of genomic DNA from samples stored for years at refrigerated temperatures (4–8°C) was maintained. High-molecular-weight genomic DNA was isolated from hair roots using a magnetic silica beads method of extraction, enabling WGR from horsehair root extracts. Nucleotide sequencing results and numbers of single-nucleotide polymorphisms and insertions/deletions concurred with those previously reported for WGR of DNA extracted from whole blood. Therefore, we consider that storing hair samples at refrigerated temperatures prevents degradation of DNA, allowing the detection of gene doping in these samples based on WGR. It is likely this finding will also have a deterrent effect, as it is now possible to test horses with archived samples even if they or their parents are deceased. To our knowledge, this is the first report employing WGR on horsehair roots stored for a long term.

Background: Second-hand smoke exposure has been associated with poor mental health. However, among Japanese adults, little is known about the association between second-hand smoking and depressive symptoms. We examined this association in a cross-sectional study among a Japanese general adult population sample.Methods: Japanese adults were recruited from the Japan Multi-Institutional Collaborative Cohort Study in the Okazaki area between 2012 and 2017. Second-hand smoke exposure and smoking status were assessed using a self-administered questionnaire. Based on their frequency of exposure to second-hand smoke, non-smokers and smokers were categorized as “almost never,” “sometimes,” and “almost every day”. Depressive symptoms were defined by a Kessler 6 score ≥5 points. We performed a multivariable Poisson regression analysis to obtain adjusted prevalence ratios (PRs) and 95% confidence intervals (CIs) for depressive symptoms.Results: Overall, 5,121 participants (4,547 non-smokers and 574 smokers) were included whose mean age was 63.6 (standard deviation [SD], 10.3) years for non-smokers and 59.33 (SD, 10.2) years for smokers. The association between second-hand smoking and depressive symptoms was significant among non-smokers, but not among smokers. Among non-smokers, PRs compared with “almost never” were 1.25 (95% CI, 1.09–1.42) for “sometimes” and 1.41 (95% CI, 1.09–1.84) for “almost every day” (P for trend <0.001); among smokers, PRs compared with “almost never” were 1.30 (95% CI, 0.82–2.06) for “sometimes” and 1.44 (95% CI, 0.90–2.33) for “almost every day” (P for trend = 0.144).Conclusions: Second-hand smoking and depressive symptoms were associated among non-smokers. Our findings indicate the importance of tobacco smoke control for mental health.

A 13-year-old spayed female Labrador Retriever was presented with severe progressive tetraparesis. The neuroanatomic localization was the C1–C5 spinal cord segments with brainstem or cranial nerve involvement. Magnetic resonance imaging revealed diffuse T1-weighted and T2-weighted hyperintense lesions with strong contrast enhancement spreading through meninges of the cervical spinal cord and the brain. Few small round areas showing T1-weighted hyperintensity and T2-weighted hypointensity were scattered within the lesions. Cerebrospinal fluid analysis revealed neoplastic round cells and possible melanocytes. Malignant melanoma was suspected. At necropsy, the brain and the entire spinal cord were covered with thick, dark membranous tissue. Based on histopathologic findings, a positive response against Melan-A, and no melanoma identified outside the central nervous system, primary meningeal melanomatosis was diagnosed.

A 3-month-old male Scottish Fold kitten with pleural fluid and low ratio of albumin to globulin (A/G ratio) was brought to our small animal hospital. Since RNA from the type I feline coronavirus (FCoV) were detected in drained pleural fluid, the cat was tentatively diagnosed with effusive feline infectious peritonitis (FIP). Following the administration of itraconazole and prednisolone, the A/G ratio increased, and the pleural fluid mostly disappeared. The fecal FCoV levels temporarily decreased. However, the cat showed neurological manifestations and was eventually euthanized due to status epilepticus after 38 days of treatment. In conclusion, itraconazole partly exerted a beneficial effect in a cat with FIP. However, further investigation of a possible role of itraconazole in FIP treatment is warranted.

Background:Drug-coated balloon strategies in endovascular therapy often result in severe dissection, so lesion preparation must be improved. We evaluated the efficacy of a gradual inflation method, termed “super slow inflation” (SSI), in preparing lesions to avoid severe dissections.Methods and Results:The association between balloon pressure and the dilatation of a model constricted vessel, as well as the load applied to the balloon surface, were determined using a burst leak detector for a quick inflation (QI; 1 atm/s) protocol and SSI (1 atm/20 s). A retrospective, single-center, non-randomized study evaluated differences in vessel dissection patterns after balloon angioplasty depending on inflation method in 81 consecutive patients (mean [±SD] age 74.6±9.2 years; 54 males) who underwent balloon angioplasty for de novo femoropopliteal lesions between January 2017 and March 2019. In the constricted vessel model, vessel dilatation increased gradually using SSI, with the maximum dilatation load being approximately 100 g lower for the SSI than QI protocol. In patients, the rate of severe vessel dissection was significantly lower in the SSI than non-SSI group (17.6% vs. 55.2%, respectively; P<0.001). Multivariate regression analysis revealed that SSI was an independent factor preventing severe dissection (odds ratio 0.18; 95% confidence interval 0.06–0.53; P=0.002).Conclusions:SSI is a gentle and effective method for the preparation of femoropopliteal lesions to reduce the incidence of severe angiographic dissection when using drug-coated balloons.

We examined fragmentary human skeletal remains from Escalon Cave near Surigao City, northeastern Mindanao, the Philippines, with respect to the morphology of bones and teeth, radiocarbon dating, and mitochondrial DNA (mtDNA) haplogroup. These remains contained parts of the left temporal bone, the right humerus, the right femur, the upper right first and second premolars, and the first molar. They are presumed to belong to an early-middle adult male, whom we named the Escalon Man. Using the femur sample, we estimated the 14C date of the individual at 2692 ± 39 years BP (uncalibrated). His stature, estimated from the maximum femur length, was about 170 cm—this makes him much taller than the Aeta and the Mamanwa, who are indigenous negrito hunter-gatherers of the Philippines. The femur is sturdy and shows well-developed muscle markings. Numerous narrow grooves on the humerus were found in close proximity to each other, but we could not prove whether these are artificial cut marks made by humans with sharp-edged tools or scratches from some non-human entity. The mesiodistal crown diameters of the molar and premolars are large and resemble those of native Oceanians. Analysis of mtDNA haplogroup was carried out using a DNA sample extracted from a molar. A next-generation sequencer was used to determine the nucleotide sequences of the mtDNA genome. The results indicated that the Escalon Man belongs to the haplogroup E1a1a, which is known to be the marker of Austronesian-speaking agriculturist populations that originated in Taiwan and spread southwards through the Philippines to the Western Pacific, since about 4000 BP. Thus, the Escalon Man was likely a member of the late Neolithic or early Metal Age agriculturist peoples who settled in northeastern Mindanao and who may be the ancestors of the territory’s present-day occupants, such as the Manobo.

During a taxonomic study of the snapping shrimp genus Alpheus from Japan and the Gulf of Thailand, it has been found that two species have been confounded under A. digitalis De Haan, 1844, which was originally described based on a single specimen possessing abnormal chelipeds. Differences between the two species are found in both morphology and coloration. Careful examination of published information of the holotype and supplemental material enable us to establish the true identity of A. digitalis. The new species, A. longiforceps, can be distinguished from A. digitalis by the more slender major chela with a sinuous, rather than convex, flexor margin, the longer male minor chela with a more elongate dactylus, and the possession of a single, rather than two, obliquely transverse white bands on the carapace and of a conspicuous dark brown spot on the lateral surface of the fourth abdominal tergum.

Background:Patients with cancer-associated venous thromboembolism (VTE) are at high risk for recurrent VTE and are recommended to receive prolonged anticoagulation therapy if they are at a low risk for bleeding. However, there are no established risk factors for bleeding during anticoagulation therapy.Methods and Results:The COMMAND VTE Registry is a multicenter retrospective registry enrolling 3,027 consecutive patients with acute symptomatic VTE among 29 Japanese centers. The present study population consisted of 592 cancer-associated VTE patients with anticoagulation therapy. We constructed a multivariable Cox proportional hazard model to estimate the hazard ratio (HR) and 95% confidence interval (CI) of the potential risk factors for major bleeding. During a median follow-up period of 199 days, major bleeding occurred in 72 patients. The cumulative incidence of major bleeding was 5.8% at 3 months, 13.8% at 1 year, 17.5% at 2 years, and 28.1% at 5 years. The most frequent major bleeding site was gastrointestinal tract (47%). Terminal cancer (adjusted HR, 4.17; 95% CI, 2.22–7.85, P<0.001), chronic kidney disease (adjusted HR, 1.89; 95% CI 1.06–3.37, P=0.031), and gastrointestinal cancer (adjusted HR, 1.78; 95% CI, 1.04–3.04, P=0.037) were independently associated with an increased risk of major bleeding.Conclusions:Major bleeding events were common during anticoagulation therapy in real-world cancer-associated VTE patients. Terminal cancer, chronic kidney disease, and gastrointestinal cancer were the independent risk factors for major bleeding.

The genre of boys’ love (BL), which generally refers to a body of works that depict fictional relationships between beautiful “boys,” is produced for and consumed mainly by women in Japan. Ludic expressions of sexuality and gender unique to BL have gained popularity on a global scale but have also drawn negative attention. In this article, we employ the concept of asobigokoro (playful spirit/heart) to highlight the importance of regional ideas of play and playfulness in game analysis. We argue that asobigokoro functions as a kind of counter-discourse as it privileges non-Eurocentric ways of knowing, understanding, and “playing” with representations of sexuality. Game analysis through an asobigokoro lens enriches the field of regional gaming by drawing on Japanese sociopolitical contexts to situate a reading of Japanese ludic representations. Asobigokoro stresses the importance of understanding cultural variations of “play” and “playfulness” in order to make sense of “taboo” subjects in culturally nuanced ways. In our textual analysis of Enzai: Falsely Accused, we discover that the simultaneous appropriation and subversion of violent and sexually explicit content, which characterizes the game’s asobigokoro, can be traced to Japanese feminist forms of asobi (play), which are rooted within the Yaoi tradition.

This Letter presents evidence for single top-quark production in the s-channel using proton-proton collisions at a centre-of-mass energy of 8 TeV with the ATLAS detector at the CERN Large Hadron Collider. The analysis is performed on events containing one isolated electron or muon, large missing transverse momentum and exactly two b-tagged jets in the final state. The analysed data set corresponds to an integrated luminosity of 20.3 fb-1. The signal is extracted using a maximum-likelihood fit of a discriminant which is based on the matrix element method and optimized in order to separate single-top-quark s-channel events from the main background contributions, which are top-quark pair production and W boson production in association with heavy-flavour jets. The measurement leads to an observed signal significance of 3.2 standard deviations and a measured cross-section of σs=4.8±0.8(stat.)-1.3+1.6(syst.) pb, which is consistent with the Standard Model expectation. The expected significance for the analysis is 3.9 standard deviations.

Strong winds are one of the several factors contributing to natural disasters. Although in recent years, the intensity and frequency of strong winds has decreased, different areas are differently affected by such winds; these winds still have the potential to cause adverse impacts on life and property. In Korea, strong winds are also responsible for the incidence of increased number of accidents and forest fires. Therefore, in this study, relationship between wind speed and damage was analyzed, and the threshold value of damage-causing wind speed was estimated. We first analyzed the relationship between wind speed and damage occurrences based on the daily maximum wind speed and daily maximum instantaneous wind speed data, and data on damage related to strong winds. Second, we examined the validity regarding the regional segmentation of the Korean criteria of special weather report for strong winds using the critical success index and cumulative percentile distributions to estimate the damage-causing threshold value for each region. We found that damage resulting from very strong wind speeds in Korea had not occurred in recent times. In addition, considerable damage had occurred because of low-speed wind compared to the current criteria for high wind advisory. However, the incidence of damage was higher when wind speed was stronger than the current criteria for high wind advisory. Based on threshold estimation, the study areas were categorized into areas with high threshold values (coastal, mountainous, and island), and those with low threshold values (inland areas). A notable difference was observed between the threshold values of the two categories of areas. This necessitated the regional segmentation of the criteria of special weather report on strong winds.

[Purpose] This study aims to describe a protocol based on neurocognitive therapeutic exercises and determine its feasibility and usefulness for upper extremity functionality when compared with a conventional protocol. [Subjects and Methods] Eight subacute stroke patients were randomly assigned to a conventional (control group) or neurocognitive (experimental group) treatment protocol. Both lasted 30 minutes, 3 times a week for 10 weeks and assessments were blinded. Outcome measures included: Motor Evaluation Scale for Upper Extremity in Stroke Patients, Motricity Index, Revised Nottingham Sensory Assessment and Kinesthetic and Visual Imagery Questionnaire. Descriptive measures and nonparametric statistical tests were used for analysis. [Results] The results indicate a more favorable clinical progression in the neurocognitive group regarding upper extremity functional capacity with achievement of the minimal detectable change. The functionality results are related with improvements on muscle strength and sensory discrimination (tactile and kinesthetic). [Conclusion] Despite not showing significant group differences between pre and post-treatment, the neurocognitive approach could be a safe and useful strategy for recovering upper extremity movement following stroke, especially regarding affected hands, with better and longer lasting results. Although this work shows this protocol’s feasibility with the panel of scales proposed, larger studies are required to demonstrate its effectiveness.

Biotin is a member of the vitamin B-complex family that acts as a cofactor of carboxylases and is essential in the metabolism of all organisms. In addition to its essential metabolic functions, biotin has been suggested to play a critical role in regulating gene expression in plants and animals. However, all studies in plants have been conducted under biotin deficiency. Therefore, we hypothesized that exogenously supplied biotin also exerts non-enzymological functions similar to those reported in animals, regulating root growth and development in Arabidopsis thaliana. To test this hypothesis, we evaluated the effects of the biotin supply on seedlings and analyzed the root architecture. Biotin supplementation inhibited root elongation and promoted lateral root formation in a concentration-dependent manner. Inhibited primary root elongation was correlated with decreased expression of the cell cycle genes CycB1;1:uidA and cell expansion gene pAtExp7::uidA, and depended on the concentration of biotin. Viability tests with AtHisH2B:YFP showed that the meristem was depleted. However, biotin supplementation did not affect the concentration of chlorophyll but had a slight inhibitory effect on foliage growth and increased the production of reactive oxygen species (ROS) at the apex of primary roots. Our study provides the first evidence of functions of biotin supplementation in plants in addition to its catalytic role as an enzyme cofactor, likely advancing our understanding of the biological functions of biotin.

This study investigated rainfall trends and their associations with tropical cyclones (TCs) during the period of 1979-2019, using TC best-track data from the Regional Specialized Meteorological Center Tokyo - Typhoon Center and daily rainfall data from 138 meteorological stations in Vietnam. The radius of influence of TCs on local rainfall was limited to 500 km from TC centers. The average annual number of TCs affecting Vietnam has decreased slightly in the last two decades compared to previous decades. The ratio of TC-induced rainfall to total rainfall attained the highest value of 37.3% in the central region, in July. The temporal distribution of TC-induced rainfall coincided with the frequency of TCs, with an active period from June to November. During 1979-2019, the non-TC rainfall was the main contributor to the change in total rainfall, especially in relation to the decline in the north and the increase in the coastal South Central region. The rainfall trend during the entire period was principally explained by the interdecadal shift in the late 1990s. Rainfall intensity and the number of heavy rainfall days were intensified for total rainfall and non-TC rainfall, indicating that TCs contributed minimally to the changes of extreme rainfall events during 1979-2019.
