Considering the scarce information on occurrences of Toxoplasma gondii and Neospora caninum in domestic animals from Turkey, the aim of this study was to investigate the seroprevalence of these parasite infections in cattle, horses, sheep, goats and dogs in Turkey. The specific antibodies against T. gondii and N. caninum were detected by iELISAs based on the recombinant TgSAG2 or NcSAG1 in a total of 2,039 serum samples from eleven provinces. The seroprevalence of T. gondii infections was 46.3%, 4.0%, 20.0%, 12.9% and 19.8%, that of N. caninum infections was 0.3%, 7.4%, 2.1%, 3.2% and 16.6% in the horses, cattle, sheep, goats and dogs, respectively. These results indicated that T. gondii and N. caninum infections are prevalent in Turkish domestic animals.

Contrast-enhanced ultrasonography (CEUS) of the hepatic vein can assess intrahepatic hemodynamic changes, and has been studied as a noninvasive method to assess the severity of portal hypertension and hepatic fibrosis in humans. However, few reports have described its usefulness in veterinary medicine. The purpose of this study was to characterize CEUS findings of the hepatic vein in normal dogs and assess the repeatability of this method both in a conscious group (n=6) and a sedated group (n=6). Sonazoid® (0.01 ml/kg) was used as a contrast agent and scanning of the hepatic vein was performed for 2 min. Time-intensity curves were generated from regions of interest in the hepatic vein. Four perfusion parameters were measured for quantitative analysis: hepatic vein arrival time (HVAT), time to peak (TTP), time to peak phase (TTPP) and wash-out ratio (WR). CEUS examinations were performed three times in each dog. The median (range) values of HVAT, TTP, TTPP and WR in the conscious group were 13.5 sec (9–22 sec), 12.5 sec (6–24 sec), 8 sec (6–13 sec) and 78.0% (60.7–91.7%), respectively. Median (range) values of HVAT, TTP, TTPP and WR in the sedated group were 12 sec (8–17 sec), 12.5 sec (9–17 sec), 9 sec (7–13 sec) and 84.1% (63.0–94.4%), respectively. The coefficients of variation of these parameters in the conscious and sedated groups were 7.6–29.7% and 11.8%–14.8%, respectively.

Cannabis consumption may induce psychotic states in normal individuals, worsen psychotic symptoms of schizophrenic patients, and may facilitate precipitation of schizophrenia in vulnerable individuals. Recent studies provide additional biological and genetic evidence for the cannabinoid hypothesis of schizophrenia. Examinations using [3H]CP-55940 or [3H]SR141716A revealed that the density of CB1 receptors, a central type of cannabinoid receptor, is increased in subregions of the prefrontal cortex in schizophrenia. Anandamide, an endogenous cannabinoid, is also increased in the CSF in schizophrenia. A genetic study revealed that the CNR1 gene, which encodes CB1 receptors, is associated with schizophrenia, especially the hebephrenic type. Individuals with a 9-repeat allele of an AAT-repeat polymorphism of the gene may have a 2.3-fold higher susceptibility to schizophrenia. Recent findings consistently indicate that hyperactivity of the central cannabinoid system is involved in the pathogenesis of schizophrenia or the neural mechanisms of negative symptoms.

The Limulus amoebocyte lysate (LAL) test is the most sensitive and reliable assay for the detection of trace amounts of bacterial endotoxins (lipopolysaccharides, or LPS), and is an accepted in vitro alternative to the rabbit pyrogen test for evaluations of parenteral drugs, biological products, and medical devices. There are three principal LAL tests, which can be categorized as both semi-quantitative and quantitative methods, including gel-clot, turbidimetric, and chromogenic assays. Since the 1970s, these tests have been successfully formulated and commercialized by US and Japanese manufacturers. More recently, in addition to the recombinant factor C-based assay, a novel product containing all of the recombinant coagulation factors from horseshoe crab has been developed, which may lead to the creation of a next generation LAL alternative. Furthermore, there are antimicrobial peptides called “host defense peptides (HDPs)” that play a key role in innate immune responses. The LAL test for HDP-related studies is challenging, because the active site of endotoxin could be masked by the binding with HDPs. Thus, it is very important to properly evaluate the actions of HDPs (human defensins and cathelicidin peptide LL-37) such as the neutralization of LPS, immunostimulatory functions, and anti-endotoxin activity. Moreover, sensitive detection of LPS in cell culture media should be conducted to address the problem of endotoxin contamination in the media. Here, we discuss the progress of LAL-based endotoxin assay technologies, as well as their applications and limitations, with a focus on innovative functional studies of HDPs.

Solvent extracts from the carnivorous plant Dionaea muscipula (Venus flytrap) were prepared using eight different organic solvents, and examined for antibacterial activity against food-related pathogenic and putrefactive bacteria. All solvent extracts showed higher antibacterial activity against gram positive bacteria than against gram negative bacteria. The TLC-bioautography analysis of the extracts revealed that a yellow spot was detected at Rf value of 0.85, which showed strong antibacterial activity. The UV, MS, and NMR analyses revealed that the antibacterial compound was plumbagin.

During apple (Malus × domestica Borkh.) storage, a loss in fruit firmness can occur. This is frequently associated with mealiness, an undesirable trait. There have been studies, such as phenotypic analyses and transcriptomics, as well as others employing a transgenic approach, focusing on this trait. Certain genetic approaches, such as quantitative trait loci (QTL) approach, however, have not been attempted. In this study, to identify and characterize QTLs controlling flesh mealiness and to investigate their application in apple breeding, we performed classical QTL mapping based on a bi-parental population and a genome-wide association study (GWAS) of mealiness. Phenotypic data for mealiness allowed us to identify two QTLs in the bi-parental family located on linkage group 10. The GWAS discovered significant marker-trait associations on chromosomes 2, 9, and 10. The MdPG1 locus, located on chromosome 10, was identified as the locus with the strongest significance by both QTL mapping and GWAS, suggesting its primary contribution to flesh mealiness. Using a tri-allelic simple sequence repeat marker, Md-PG1SSR10kd, 10 kb downstream of the MdPG1 coding sequence, we divided apple accessions into six groups based on their genotypes. Among the six groups, the Md-PG1SSR10kd genotype “2/2” had the least mealy phenotype.

企業は将来にわたって事業を継続することを前提に利潤を追求し自らを発展させてきた。近年、これに加えて企業も社会を構成する一員であるとの考えから、自らの発展と同時に社会貢献が求められるようになった。特に「企業の社会的責任」元年と呼ばれる2003年以降、多くの企業が「企業の社会的責任(Corporate Social Responsibility 以下CSR)」を推進している。企業が行う植樹活動もその多くがCSRの一環として始められ、環境活動を行う環境CSRの具体的な方策として植樹による「企業の森づくり」に参加する企業が急増している。しかし、それらは、都道府県との連携による森づくりであり、短期的な企業主導の森づくりであるため、地域の森林計画や地域住民の意識との乖離といった課題が明らかになっている。本論では、企業のCSR報告書を中心に、資料・文献を整理したうえで、聞き取り調査を行い、これらの課題を解決するために企業が行う新たな活動形態として、ピジョン株式会社が茨城県常陸大宮市の社有地で行う「ピジョン美和の森」植樹活動の特異性を明らかにした。環境CSRの一環として行う植樹活動を自社の保有地で行い、自社の資産として森づくりを進めるためには、他の有形・無形の資産同様、維持管理のための持続的森林管理方法の構築が必要である。A company, developing its business in chase of profits, is required contribution to society as a member of the community. Since 2003, many companies have developed and promoted corporate social responsibility (CSR). In recent years, more and more companies introduce an activity of foresting as a part of CSR. The number of companies with CSR Forest is rapidly increasing. However, other studies have concluded that the creation of a forest is dissociated from the local forest plan because of domination by administrative division and short-term contract.In this article, first of all, the current reality of foresting by companies was clarified by the survey of CSR reports. Secondly, the investigation was made of the company continuing planting trees in its own land as CSR for years to reveal the uniqueness of the activity. The following are the main findings(: a)many companies have lack of knowledge about forestry preservation and(b) it is requested to construct the management system for forest assets like established one for other tangible or intangible assets.

Morphological investagations were carried out on basophilic intranuclear and cytoplasmic inclusion bodies in epithelial cells of the hair bulbs of anagen hair follicles in wild foxes affected with an abnormal hair coat condition. The inclusions, which were DNA-positive, contained numerous minute virus-like particles ("FMVP"). The particles, having a diameter of approximately 13 nm, had an arrangement of capsomer-like subunits of approximately 2-3 nm in diameter and were nonenveloped. The striking resemblance to icosahedral virus was crystallographically and morphologically demonstrated.

Pepleomycin (PEP), 3-[(S)-1'-phenylethylamino]propylaminobleomycin has potent activity and is less pulmonary toxic than bleomycin (BLM). Biological activity and toxicity of the following degradation products of PEP have been studied in detail: the product of carbamoyl migration (ISO), the product of decarbamylation (DC), the product of ring closure of the side chain on the pyrimidine moiety (RC), the depyruvamide product (DP) and the product of an enzymatic inactivation (DA). These degradation products showed much lower activity than PEP in vitro: antimicrobial and anti-HeLa activities, inhibition of DNA synthesis in AH66 cells and the DNA strand cleavage. Acute toxicity and pulmonary toxicity were tested in mice. Results indicated much lower acute toxicity corresponding to the decreased in vitro activity when compared to PEP. DP and RC did not cause lung fibrosis in mice, while ISO and DC showed 1/2.6 and 1/5.7 degree of pulmonary toxicity, respectively, in comparison with PEP.

The kinetics of the reaction of BLM-Fe(II)-O2 with DNA in the absence or presence of 2-mercaptoethanol (2-ME) were studied. The total number of bases released by BLM-Fe(II)-O2 in the presence of 2-ME increased about 6.5 times more than that in the absence of 2-ME in the reaction of 6 hours at 37°C. The molar ratios of the released bases ware little affected by the reaction time, temperature or 2-ME. Among the four bases, thymine was preferentially released. On the basis of a reaction scheme of BLM-Fe(II)-O2 with DNA, the equations were derived by the steady-state method. In the absence of 2-ME, the release of bases from DNA was dependent on the concentration of BLM-Fe(II)-O2, but independent of the concentration of DNA. In the presence of 2-ME, a biphasic reaction was observed; the first one is due to BLM-Fe (II) which originally existed and the second one is due to BLM-Fe(II) produced by the reduction of BLM-Fe(III) with 2-ME. In the second reaction, the rate of the release of bases from DNA was proportional to the concentration of BLM-Fe(II) and 2-ME, but inversely proportional to the concentration of DNA. The rate-determining step in the reaction of BLM-Fe(II)-O2 with DNA in the presence of 2-ME was found to be the reduction of BLM-Fe(III) to BLM-Fe(II). By these kinetic studies, the reaction of BLM-Fe(II)-O2 with DNA in the presence of 2-ME was elucidated to proceed in a catalytic fashion. Furthermore, the maximum number of bases released by BLM from DNA was one base per twelve to thirteen bases.

Bisphosphonates have high affinity for hydroxyapatite (HA), which is abundantly present in bone. Also, platinum complexes are known that have a wide spectrum of antitumor activities. The conjugate of bisphosphonate and a platinum complex might have HA affinity and antitumor activity, and become a drug for metastatic bone tumor. In this study, the authors synthesized platinum complexes that had dialkyl bisphosphonic acid as a ligand, and evaluated the possibility of the synthesized complexes as a drug for metastatic bone tumor. The synthesized dialkyl bisphosphonate platinum(II) complex was characterized, and its stability in an aqueous solution was also confirmed. The synthesized platinum complex showed higher HA affinity than other platinum complexes such as cisplatin and carboplatin in an experiment of adsorption to HA. In vitro, the platinum complex showed tumor growth inhibitory effect stronger than or equal to cisplatin, which is the most commonly used antitumor agent. Moreover, the platinum complex showed a bone absorption inhibitory effect on the osteoclast. These results suggest potential of dialkyl bisphosphonate platinum(II) complexes as a drug for metastatic bone tumor.
