古き薬学生にとって, 薬草園はさまざまな想い出の泉であった.四季折々の花や緑は試薬の臭気から逃れ出るひとときの憩いでもあった.今, 薬草園は学生にとって意外に遠い存在となりつつあるらしい.それにはそれなりの厳しい理由もある.しかし薬学教育の場から, あるいは研究の場から薬草園を無くしてしまった時に, 我々の受ける損失はまた大きいのではないかとも思われる.今日はそのことを考えていただきたい.

The cultivation or trafficking of "Magic mushrooms," containing hallucinogenic psilocin and psilocybin, has been prohibited by the Narcotics and Psychotropics Control Law in Japan since 2002. To identify these mushrooms, we attempted to prepare the monoclonal antibody (mAb) reactive to these hallucinogens. As an antigen inducing an anti-psilocin mAb, N-{4-[3-(2-dimethylaminoethyl)indol-4-yl-oxy]butyl}succinamic acid was synthesized by modifying the 4-hydroxyl moiety of psilocin and coupled to a carrier protein of keyhole limpet hemocyanin. BALB/c mice were immunized five times with the antigen emulsified with an adjuvant, and their spleen cells were fused with mouse myeloma cells. We obtained several hybridoma cells producing mAbs reactive to psilocin, from which fourclones, BA631, CA231, KA422, and MA332 with a higher production of anti-psilocin mAb were selected by limiting dilution. Isotype of CA231 and KA422 mAbs were IgG2a and that of BA631 and MA332 mAbs were IgGl.Enzyme immunoassay (EIA) using BA631 mAb, revealed that BA631 cross-reacted with psilocin and dimethyltryptamine, but not with the other indole derivatives such as psilocybin, 4-hydroxyindole, tryptamine, and tryptophan. Therefore, these antibodies could be used for the identification of magic mushrooms.

The scavenging effects of twenty-five tannins including low-molecular polyphenols on the superoxide anion radical (O_2^-) generated in the hypoxanthine-xanthine oxidase system were estimated by electron spin resonance (ESR) measurements of the adducts formed by 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) and the radical. The scavenging effects of tannins and related polyphenols having ortho-trihydroxyl (pyrogallol) structure [galloyl, hexahydroxydiphenoyl (HHDP) groups in hydrolyzable tannins, galloyl group in acylated proanthocyanidins, and the B-ring of some flavan-3-ols] were stronger than the effects of unacylated proanthocyanidins. The effects of tannins and related polyphenols on the superoxide anion radical were also compared with those on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Each tannin in an ethanol solution of DPPH radical reduced the intensity of the signal of the DPPH radical, and gave a weak signal assignable to a radical derived from that tannin, in a similar way to the appearance of the signal of dl-α-tocopherol radical, accompanied with reduction of the signal of DPPH radical, in a mixture of dl-α-tocopherol and the DPPH radical. In contrast to the case of the superoxide anion radical, the effects of unacylated proanthocyanidins on DPPH radical were comparable with those of the other types of tannins. The scavenging effects of all of the tannins and related polyphenols tested in the experiments on DPPH radical were stronger than that of dl-α-tocopherol.

In the present study, a water/isopropyl alcohol system was selected as the solvent in a hydroxypropylcellulose (HPC) binder solution for the preparation of lactose granules.With a fluidized bed granulation method, the mean granule diameter increased as the adhesion tension (γcosθ) and lactose solubility increased. With an agitating granulation method, the mean granule diameter was independent of the adhesion tension and lactose solubility, but it increased as the viscosity of the binder solution increased.With both granulating methods, greanule hardness increased as the adhesion tension and lactose solubility increased, while the pore volume of the granules decreased as the adhesion tension and lactose solubility increased.The quantitative relationship between the physical properties of the granules and several properties of the solvent solution was investigated by the application of multiple regression analysis.

A method for determination of vanadium in air by flameless atomic absorption spectrophotometry was studied. Atomospheric particulates were collected on a Gelman filter by a high-volume air sampler, and extracted with 5% hydrochloric acid. Cupferron, a chelating agent, reacts with vanadium to form a complex, which can be extracted with methyl isobutyl ketone. The solvent was dried, ashed, and atomized by passing through a high electric current of the atomizer. The concentration of vanadium was calculated directly from the calibration curve by measuring the absorbance of vanadium at 318.5 nm. Cations, except tin, did not interfere the determination of vanadium seriously. Analytical results were in good agreement with those obtained by a nitrous oxide-acetylene flame atomic absorption spectrophotometry. The coefficient of variation was from 4.4% to 9.4%, and the limit of detection was 0.03 ppm of vanadium, when the signal/noise ratio was 2.

Separative determination of α-chymotrypsin and Bromelain, frequently used as an anti -inflammatory enzyme preparation, was examined. p-Chloromercuribenzoic acid (PCMB) was used as the inhibitor of Bromelain. Since Bromelain activity in preparations was completely inhibited by 10^<-4>_M of PCMB in the reaction system, remaining α-chymotrypsin activity was determined with good accuracy. The difference between their total activity and α-chymotrypsin activity agreed with Bromelain activity.

2-Ethoxy-3-methoxy-2-cyclohexenone (II) and 2,3,5-trimethoxy-2-cyclohexenone (III) were prepared from pyrogallol in a satisfactory yield. II was reduced with lithium aluminium hydride to 2-ethoxy-3-methoxy-2-cyclohexenol (XXV), which was not so stable. III was reduced under the same condition to 2,5-dimethoxy-2-cyclohexenol (XXIX) and 2,3,5-trimethoxy-2-cyclohexenol (XXX). XXX was relatively stable but was not so stable as the reduced product of acutumine (I).

The mechanism for the international regulatory control of drugs of abuse under the Single Convention on Narcotic Drugs and the Convention on Psychotropic Substances is outlined. A historical overview of the substances which have been added to the list of controlled drugs is presented, focusing on the review of dependence-producing substances by the World Health Organization (WHO) for international control decisions.

Sympathomimetic amines, amphetamine and methamphetamine, were determined as trifluoroacetyl derivatives by gas chromatography (GC) equipped with a flame-thermionic detector (FTD). Resolution of these derivatives was carried out in a glass column (3 mm×1 m), packed with 1.5% SE-30 on Chromosorb W (AW-DMCS) at 110℃, with helium at a flow rate 50 ml/min. Detection limit of these amines was 0.1 ng. In a case of the autopsy samples from 4.5 month-postmortem suspected the intoxication of methamphetamine, we could detect the amine by FTD-GC, and the content was calculated to be 12.5 μg per g wet weight liver.

A program adapted for use on microcomputers (DCN) has been developed which permits one to perform operations of numerical convolution and deconvolution using polyexponential functions, that are often implemented in pharmacokinetic analysis. The program is written in Microsoft GWBASIC and can be used in personal computers with no modification. The user supplies information relating to the coefficients and exponentials defining the polyexponential equation of the response and weighting functions and the program performs the deconvolution operation by numerical integration using trapezoidal rule and provides numerical and graphic information concerning the input function. The program can be applied to the deconvolution of many linear pharmacokinetic systems and allows one to solve problems related to drug release, absorption, distribution, as well as others. Additionally, the program is able to perform the convolution operation if information about the input and weighting functions and is also able to simulate pharmacokinetic processes. The efficacy of the program was evaluated by comparison with several deconvolution algorithms, in particular that proposed by Veng-Pedersen and Iga.
