Cathepsin L-like enzyme was purified from the body wall of the sea cucumber Stichopus japonicus by an integral method involving ammonium sulfate precipitation and a series of column chromatographies on DEAE Sepharose CL-6B, Sephadex G-75, and TSK-GEL. The molecular mass of the purified enzyme was estimated to be 63 kDa by SDS–PAGE. The enzyme cleaved N-carbobenzoxy-phenylalanine-arginine7-amido-4-methylcoumarin with Km (69.92 μM) and kcat (12.80/S) hardly hydrolyzed N-carbobenzoxy-arginine-arginine 7-amido-4-methylcoumarin and L-arginine 7-amido-4-methylcoumarin. The optimum pH and temperature for the purified enzyme were found to be 5.0 and 50 °C. It showed thermal stability below 40 °C. The activity was inhibited by sulfhydryl reagents and activated by reducing agents. These results suggest that the purified enzyme was a cathepsin L-like enzyme and that it existed in the form of its enzyme-inhibitor complex or precursor.

一般に食用とされていないクロナマコの有効利用を図ることを目的として, クロナマコ摂取がラットの脂質代謝に及ぼす影響を検討した。生のクロナマコの可食部をフードカッターで粉砕し, 凍結乾燥後粉末状にしたものを試料としてラットの餌を調製した。タンパク質レベルを20%とし, クロナマコを用いた食餌ではタンパク質源としてカゼインとクロナマコを窒素含量で3:1の割合にした。対照として, タンパク質源にカゼインのみを用いたコントロール群を設けた。コレステロール (Chol) を0.2%添加したこれら飼料をSD系雄ラットに4週間自由摂食させた。クロナマコは血清および肝臓Chol濃度をコントロール群より有意に低下させ, HDL-Chol/総Chol比を上昇させた。クロナマコ摂取ラットで糞中への中性および酸性ステロイド排泄は促進した。これらの結果より, クロナマコは糞中へのステロイド排泄促進によりChol低下作用を発現することが明らかとなった。

マナマコ人工種苗の成長に伴う管足の数や吸盤部の表面積の変化を計測し,引っ張り試験を実施して管足の基質への固着力を測定した.更に,基質に着底したマナマコ人工種苗の流動耐性を調べた.稚ナマコの管足数と吸盤部の表面積は体長に比例して増加した.管足より発生する稚ナマコの固着力は付着基質の粗度に依存し,管足吸盤部の直径とほぼ等しい相当粗度で最小値をとりながら成長とともに増加した.吸盤部の単位面積当たりの固着力は,成長とともに付着基質の粗度に依存しながら一定値に収束した.定常流の増加とともにマナマコの形態は防水形状に変化し,他の棘皮動物と同等以上の流動耐性をもつことがわかった.マナマコ人工種苗の放流海域における波浪条件をもとに,適正放流サイズや放流水深を決定することが可能である.

Two structurally distinct lectins were purified from the coelomic plasma of holothurian, Stichopus japonicus, by affinity chromatography on a porcine stomach mucin-conjugated agarose column, gel filtration on a Superose 6 column, and ion-exchange chromatography on a HiTrap Q-FPLC. The two lectins showed apparent molecular masses of about 400 kDa (SPL-1) and 60 kDa (SPL-2) on gel filtration, but about 17 kDa on SDS-PAGE under reducing conditions. Both lectins showed hemagglutination activity toward rabbit erythrocytes in the presence of Ca2+ ions. The N-terminal amino acid sequences were highly homologous to but distinct from those of a Ca2+-dependent (C-type) lectin named SJL-I purified from the same species. In addition to porcine stomach mucin, the hemagglutinatioi activity of SPL-1 was strongly inhibited by uronic acids such as galacturonic acid, and glucuronic acid, while the activity of SPL-2 was inhibited by GalNAc and galactosides. Both lectins were adsorbed on clotted coelomocytes in the presence of Ca2+ but not in the presence of inhibitory sugars or EGTA, suggesting the presence of an endogenous carbohydrate ligand(s) for plasma C-type lectins in the clot. However, coelomocyte clotting occurred normally even in the presence of inhibitory sugars, but was strongly inhibited by synthetic GRGDSP peptide or EGTA, suggesting the participation of integrin but not the lectin-carbohydrate interaction in the clotting events.

Two Ca^<2+>-dependent lectins were purified from the sea cucumber Stichopus japonicus by affinity chromatography on lactosyl-Sepharose 4B and ion-exchange chromatography on Q-Sepharose. Their molecular masses were estimated to be 13 kDa (SJL-I) and 15 kDa (SJL-II) on SDS-PAGE. SJL-I agglutinated rabbit erythrocytes as well as human A, B, and O-type erythrocytes, but SJL-II agglutinated only rabbit erythrocytes. Hemagglutination by SJL-I was competitively inhibited by N-acetyl-D-galactosamine and galactose-containing carbohydrates. On the other hand, only lactose, melibiose, and raffinose gave weak inhibition of hemagglutination by SJL-II, suggesting that SJL-II may have high specificity for particular complex carbohydrate(s) on the surface of rabbit erythrocytes. SJL-II was activated at ten times lower Ca^<2+> concentration than SJL-I. Both lectins lost activity in acidic pH, while SJL-I appeared more stable down to pH 4.5.

Environment protection is and will be the most important aspect of the tire development. A study of tire's life cycle assessment (LCA) was conducted under Japan Rubber Manufacturers Association, and revealed that the usage stage accounts for more than 85% of total CO2 emission in an entire life cycle of a common tire. From this standpoint, reduction of rolling resistance (RR) is the most important requirement, followed by extension of service life, which contributes to all other stages. Following the development of silica-filled tread compound, further reduction of hysteresis loss had been achieved by development of new materials for silica system, such as dispersion additive and novel modified polymers. In addition, recent implementation of non-linear viscoelastic properties in the Finite Element Analysis has greatly improved the precision of tire RR prediction. This will allow efficient application of low hysteresis materials in tire design.

The purpose of this study is to clarify the relation between spatial configuration and ceremonies in Heian-kyu Palace.This study presents the following points.1. The Dobisashi is an unique space that only used in Heian-kyu Palace, playing a role in the square with the gate and gallery in the Ugi (the ceremony in rainy day).2. According to the ceremonial documents about Heian-kyu Palace, the usage of the Dobisashi in the Ugi has been developed during the mid - to -late Heian Period.3. During the relation between the Dobisashi and Ugi, it's clarified that the Dobisashi in Heian-kyu palace a special space for Ugi.
