アメリカの科学教育改革が,国家規模で展開されている。「全米科学教育スタンダード」の作成(1996年),科学教育の新しい「フレームワーク」ドラフト版の提示(2011年),「科学に関する次世代スタンダード」の公表(2013年)と,スタンダードに基づくカリキュラム設計を通して改革の遂行が目指されてきた。また,現代科学教育改革のトレンドであるSTEM教育の振興に本格的に取り組んでおり,成果が着実に具体化しつつある。

Objective: Comprehensive profiling of gene expression in peripheral blood leucocytes (PBLs) in patients with acute coronary syndrome (ACS) as a prognosticator is needed. We explored the specific profile of gene expression in PBLs in ACS for long-term risk stratification. Methods: 30 patients with ACS who underwent primary percutaneous coronary intervention (PCI) and 15 age-matched adults who participated in medical check-ups were enrolled from three centres. Peripheral blood samples were collected to extract RNA for microarray analyses. Results: During the 5-year follow-up, 36% of this cohort developed the expected non-fatal coronary events (NFEs) of target lesion revascularisation (TLR) and PCI for a de novo lesion. Class comparison analysis (p<0.005) demonstrated that 83 genes among 7785 prefiltered genes (41 upregulated vs 42 downregulated genes) were extracted to classify the patients according to the occurrence of NFE. Pathway analysis based on gene ontology revealed that the NFEs were associated with altered gene expression regarding the T-cell receptor signalling pathway in ACS. Univariate t test showed that the expression level of death-associated protein kinase1 (DAPK1), known to regulate inflammation, was the most significantly negatively regulated gene in the event group (0.61-fold, p<0.0005). Kaplan-Meier curve analysis and multivariate analysis adjusted for baseline characteristics or clinical biomarkers demonstrated that lower DAPK1 expression in PBL emerged as an independent risk factor for the NFEs (HR: 8.73; CI 1.05 to 72.8, p=0.045). Conclusions: Altered gene expression in T-cell receptor signalling in PBL in ACS could be a prognosticator for secondary coronary events. © Published by the BMJ Publishing Group Limited.

(1)ニラとネギの種間雑種である「なかみどり」の香気特性を,成分組成から解明するため,親植物と合わせ三種の香気成分分析を行った. (2)ニラより香気濃縮物の調製方法を検討しチルド試料から減圧水蒸気蒸留による方法を最適と判断した. (3)香気濃縮物をGC, GC-MSデータより比較検討した結果,「なかみどり」の香気組成は,親ニラのそれと類似し,香気前駆物質としてニンニクと同様アリシンの存在が示唆された.「なかみどり」は親ニラに比して精油量も多く,アリル基を有する化合物は親植物の2倍以上となり,これがニンニク臭の強いニラ新種の原因と考えられる.一方ネギには催涙性の原因物質であるthiopropanal S-oxideが検出されたが「なかみどり」にこの形質は受け継がれていない.

An optical design is developed for a Blu-ray pickup head with a laser expander that can synchronously detect the linear displacement and two-dimensional angular tilts at the test plane. To improve the signal accuracy of the photo detector, a laser expander is used to transform the shape of the laser beam from elliptical to circular. A quadrant detector as used to detect the relative relationship between the $V_{\text{A}}$, $V_{\text{B}}$, $V_{\text{C}}$, $V_{\text{D}}$ signals, can be used to measure the tilt angle and displacement at the test plane. Measurement detection and analysis is based on one normalized focus error signal (NFES) and two angular signals. The optical efficiency of the Blu-ray pickup head with laser expander is 59.29%. The optical efficiency of the currently used commercial pickup head is only 8.08%. Tolerance analysis is also considered in the performance requirements.

A mode of the alkaline and acidic degradation of sodium 6-[D-2-(2-(4-formyl-1-piperazinyl)-5, 8-dihydro-5-oxopyrido [2, 3-d] pyrimidine-6-carboxamido)-p-hydroxyphenylacetamido] penicillanate (1, PL-385) and structures of the degradation products were studied. Treatment of 1 with three equimolar amounts of sodium hydroxide produced a kinetically stable intermediate, (5R)-penicilloic acid (2a), which on kept at 37°C for 65h was converted into a thermodynamically stable product, (5S)-penicilloic acid (2b). Treatment of 1 with an excess of sodium hydroxide gave a deformyl derivative (3) arising from the elimination of formly group via 2. On the acidic treatment of 1, the degradation product, (5R, S)-penilloic acid (4), was yielded.

A series of N-alkylampicillin (2), N-heteroarylampicillin (5) and N-heteroarylcephalexin (6) were synthesized in order to obtain β-lactam detivatives with a broad and orallypotent antibacterial activity similar to that of the injectable N-acylampicillins. 6-[2-[(Pyrido [2, 3-d] pyrimidin-6-yl) methylamino]-2-phenylacetamido] penicillanic acid derivatives (2) were prepared by the reduction of the Schiff base which was derived from the reaction of pyrido [2, 3-d] pyrimidine-6-carboxaldehyde (1) with ampicillin. 6-N-(4-Pyrimidinyl) ampicillin and -cephalexin derivatives (5 and 6) were obtained by the reaction of 4-chloropyrimidine (4) with ampicillin or cephalexin. Compounds 2, 5 and 6 were tested in vitro antibacterial activity and, however, none of them have a broad and potent activity.

A series of N-(pyrido [2, 3-d] pyrimidine-6-carbonyl) ampicillin and -amoxicillin derivatives (1-3) were synthesized and tested for antibacterial activity and acute toxicity in mice. 5, 8-Dihydro-2-(1-piperazinyl)-5-oxopyrido [2, 3-d] pyrimidine-6-carboxylic acid (7) was converted to N-acyl- and -alkylpiperazinyl derivatives (8 and 9) by acylation and alkylation, respectively ; a part of 9 was alternatively prepared by the reactions involving the displacement of N-alkylpiperazines with sulfoxide 11 which was derived from ethyl 5, 8-dihydro-2-methylthio-5-oxopyrido [2, 3-d] pyrimidine-6-carboxylate (10). Treatment of 4, 5, 8 and 9 with ethyl chloroformate followed by the reaction with ampicillin and amoxicillin gave the desired N-acylampicillin (2) and -amoxicillins (1 and 3), respectively. Among compounds 1-3, sodium 6-[D-(-)-2-(2-(4-formyl-1-piperazinyl)-5, 8-dihydro-5-oxopyrido [2, 3-d] pyrimidine-6-carboxamido)-p-hydroxyphenylacetamido] penicillanate (3l, PL-385) was found to be the most excellent in antibacterial activity and to be the less potent in acute toxicity in mice. An alternative route for the synthesis of PL-385 was accomplished, consisting of the reaction of an active ester 13 with amoxicillin. Structure-activity relationships of 1-3 were discussed.

本稿ではユーザが与えた1語のクエリに対して,そのクエリに関する意外な情報を発見する手法の提案を行う.提案手法では,クエリに対して意外度の高い関連語を発見し,クエリと意外度の高い関連語を基に意外な情報を発見する.その際,クエリの関連語の中でもクエリとの関係が非典型的であり,かつ認知度が高い関連語ほど意外度が高いという仮説に基づいて関連語の意外度を求める.たとえば提案手法により,“落合博満”というクエリに対して“ガンダム”という関連語の意外度が高いことが分かり,これを基に“落合博満はガンダムマニアである.”という意外な情報を発見できる.提案手法では,Wikipediaから得られる情報のみを用いてクエリに対する意外な情報の発見を行う.実験では75語のクエリを用いて評価を行い,クエリと関連語間の関係の非典型度,および関連語の認知度を考慮することの有効性を示した.

東日本大震災では,石油精製・輸送施設の損壊を背景に,東北地域は長期にわたる深刻なガソリン不足に直面し,地域全体の社会・経済活動が大きく低下した.本研究では,ガソリン販売統計と港湾間の移出入統計を用いて東北地域における発災後一ヵ月間のガソリン需給ギャップを分析し,ガソリン不足の主要因が供給(輸送)戦略の失敗であったことを示す.その上で,日本海側港湾を活用してガソリンを早期に大量供給する輸送戦略を提案し,それによりガソリン不足がいかに軽減されるかを示す.結果として,提案輸送戦略に必要な追加的陸上輸送費用は高々2~3億円程度である一方,経済損失軽減効果は1500~2500億円に上ることを明らかにする.また,ガソリン価格の操作により経済損失の軽減を図る戦略は大規模災害時には不適当であることも論ずる.
